Abstract
Die Erfindung betrifft ein Verfahren zur Transformation von $i(Amycolatopsis sp.) DSM 9991 und DSM 9992 und die Verwendung der so transformierten Stämme zur Herstellung von Vanillin, vorzugsweise zur Herstellung von Vanillin aus Ferulasäure.
Description
Verfahren zur Transformation von Amycolatopsis sp. DSM 9991 und DSM 9992
Die Erfindung betrifft ein Verfahren zur Transformation von Amycolatopsis sp. DSM 9991 oder DSM 9992 und die Verwendung der so transformierten Stämme zur
Herstellung von Vanillin, vorzugsweise zur Herstellung von Vanillin aus Ferula- säure.
Vanillin ist ein wichtiger, in der Nahrungs- und Genussmittelindustrie viel ver- wendeter Aromastoff. Die Herstellung erfolgt auf chemischem Wege hauptsächlich aus in Sulfitablaugen enthaltenem Lignin, sowie durch Oxidation von Eugenol oder Isoeugenol. Auf chemischem Wege hergestelltes Vanillin hat jedoch den Nachteil, dass es im lebensmittelrechtlichen Sinne kein Naturstoff ist und deshalb nicht als natürlicher Aromastoff bezeichnet werden darf.
Der natürliche Aromastoff Vanillin ist bisher nur durch Extraktion von Vanilleschoten erhältlich, das auf diese Weise gewonnene Vanillin ist jedoch sehr teuer. Verschiedene andere Verfahren zur Herstellung von natürlichem Vanillin mit unterschiedlichen Mikroorganismen und Enzymen sind bereits bekannt (s. beispiels- weise EP 405 197 A, EP 453 368 A und EP 542 348 A), sind aber bislang aufgrund der geringen Ausbeuten bzw. Konzentrationen an Vanillin für eine großtechnische Herstellung ungeeignet.
In EP 761 817 A ist ein Verfahren zur fermentativen Herstellung von natürlichem Vanillin aus Ferulasäure beschrieben, wobei zwei Stämme der Gattung
Amycolatopsis (Familie Pseudonorcardiaceae) welche unter den Nummern DSM 9991 und DSM 9992 (Datum der Ersthinterlegung: 2. Mai 1995) bei der Deutschen Sammlung für Mikroorganismen und Zellkulturen GmbH in Braunschweig hinterlegt sind, eingesetzt wurden. Mit Hilfe dieser Stämme und des in EP 761 817 A beschriebenen Verfahrens war es möglich, natürliches Vanillin auf wirtschaftliche
Weise in guten Ausbeuten und hohen Konzentrationen zu erhalten.
Es ist bekannt, dass der Einsatz von genetisch modifizierten Mikroorgarisimen in fermentativen Prozessen oft zu verbesserten Ausbeuten oder Konzentrationen an gewünschten Stoffen führt. Um solche genetisch modifizierten Mikroorganismen zu erhalten, ist es notwendig, ein geeignetes Verfahren zur Transformation dieser
Mikroorganismen zu kennen.
Verfahren zur Transformation verschiedener Amycolatopsis Spezies sind bekannt. In P. Matsushima et al. J Bacteriol. 169, 1987, 2298 - 2300 ist ein Verfahren zur Protoplasten-Transformation von Amycolatopsis orientalis beschrieben. Nachteilig an diesem Verfahren ist, dass es sich nicht auf die Transformation von Amycolatopsis sp. DSM 9991 und DSM 9992 anwenden lässt (s. Beispiele). In R. Lal et al. j. Antibiotics 51, 1998, 161-169 ist ein Elektroporations- Verfahren zur Transformation von Amycolatopsis mediterranei beschrieben. Nachteilig ist, dass bei der Anwendung dieses Verfahrens bei Amycolatopsis sp. DSM 9991 und DSM 9992 nur geringe
Transformationsraten erzielt werden konnten (s. Beispiele).
In J. Madon et al. J Bacteriol. 173, 1991, 6325 - 6331 ist die direkte Mycel- Transformation von Amycolatopsis mediterranei beschrieben. Eine Abwandlung dieses Verfahrens für die direkte Mycel-Transformation von Amycolatopsis methanolica ist in j. W. Vrijbloed et at. Plasmid 34, 1995, 96 - 104 beschrieben Nachteilig an diesen Verfahren sind die niedrigen Transformationsraten, welche sich aufgrund der langen Inkubationsdauer des Mycels von ca. 40 Stunden ergeben.
Es bestand daher Bedarf, ein für Amycolatopsis sp. DSM 9991 oder DSM 9992 geeignetes Transformationsverfahren zu finden, mit welchem hohe Transformationsraten erzielt werden können.
Überraschenderweise wurde nun ein Verfahren zur Transformation von Amycolatopsis sp. DSM 9991 oder DSM 9992 durch
(1) Kultivierung von Amycolatopsis sp. DSM 9991- oder DSM 9992-Mycelien in einem Kulturmedium und
(2) Inkontaktbringen dieser Kultur mit einer Mischung enthaltend
a) 0,25 bis 10 μg/ml zu transformierende DNA b) 0,4 bis 0,7 M CsCl c) 0 bis 9 mM MgCl2 d) 30 bis 50 % [w/v] Polyethylenglykol 1000 und e) 10 bis 50 μg/ml DNA, welche von a) verschieden ist,
wobei das Mycel 4,5 bis 9 Stunden nach Eintritt in die stationäre Phase mit der genannten Mischung in Kontakt gebracht wird.
Schritt (1) des erfindungsgemäßen Verfahrens betrifft die Kultivierung von Amycolatopsis sp. DSM 9991 oder DSM 9992-Mycelien in einem Kulturmedium.
Geeignete Kulturmedien sind solche, wie sie in der Literatur für die Kultivierung von Mycelien der Spezies Amycolatopsis bekannt sind. Besonders geeignete Kulturmedien sind Komplexmedien wie beispielsweise YMG Medium (0,4 % [w/v] Hefeextrakt, 1 % [w/v] Malzextrakt, 0,4 % [w/v] Glucose, pH 7,2), TYN Medium (0,25 % [w/v] Hefeextrakt, 1 % [w/v] Trypton, 0,5 % [w/v] NaCl, pH 7,2) oder TSB Medium
(1,7 % [w/v] Trypton, 0,3 % [w/v] Soytone, 0,25% [w/v] Glucose, 0,5 % [w/v] NaCl, 0,25 % [w/v] K2HPO4 pH 7,3). Die Kultivierung wird vorzugsweise bei Temperaturen von 30 bis 48°C, besonders bevorzugt von 39 bis 42°C durchgeführt.
Vorzugsweise wird in Schritt (1) des erfindungsgemäßen Verfahrens so vorgegangen, dass in einem geeigneten Kulturmedium eine Vorkultur von Amycolatopsis sp. DSM 9991- oder DSM 9992-Mycelien herangezogen wird. Vorzugsweise wird die Vorkultur bei einer gleichen Temperatur und in gleichem Kulturmedium herangezogen, wie die eigentliche Kultur. Zur Herstellung einer Kultur von Amycolatopsis sp. DSM 9991- oder DSM 9992-Mycelien wird ein Teil der Vorkultur bevorzugt nach 16 bis
24 Stunden, besonders bevorzugt nach 18 bis 22 Stunden, ganz besonders bevorzugt nach 19 bis 20 Stunden, zur Inokulation verwendet.
Das Wachstum von Amycolatopsis sp. DSM 9991- oder DSM 9992-Mycelien im Kulturmedium kann beispielsweise mit Hilfe spektrometrischer Methoden bestimmt werden. Vorzugsweise wird das Wachstum über die optische Dichte der Kultur bestimmt. Gemäß dem erfmdungsgemäßen Verfahren erfolgt die Transformation von Amycolatopsis sp. DSM 9991- oder DSM 9992-Mycelien 4,5 bis 9 Stunden nach Eintritt in die stationäre Phase. Es wurde überraschenderweise gefunden, dass sich in diesem Zeitfenster besonders hohe Transformationsraten erzielen lassen, welche deutlich höher sind, als bei Anwendung der im Stand der Technik beschriebenen Transformationsverfahren. Besonders bevorzugt erfolgt die Transformation 5 bis 8,5 Stunden nach Eintritt in die stationäre Phase, ganz besonders bevorzugt nach 6,5 bis 7,5 Stunden.
Zur Transformation wird die Mycel-Kultur von Amycolatopsis sp. DSM 9991 oder DSM 9992 mit einer Mischung, enthaltend
a) 0,25 bis 10 μg/ml zu transformierende DNA b) 0,4 bis 0,7 M CsCl c) 0 bis 9 mM MgCl2 d) 30 bis 50 % [w/v] Polyethylenglykol 1000 und e) 10 bis 50 μg/ml DNA, welche von a) verschieden ist,
in Kontakt gebracht. Die genannte Mischung wird im folgenden als Transformationsmischung bezeichnet.
Zur Transformation wird vorzugsweise ein Aliquot der Mycel-Kultur abzentrifugiert, gewaschen und anschließend in der Waschlösung oder in einem geeigneten Puffer, vorzugsweise in TE-Puffer resuspendiert. Als Waschlösung können geeignete Puffer verwendet werden, vorzugsweise wird TRIS-EDTA-Puffer (10 mM TRIS-HC1, pH
8,0, 1 mM EDTA) verwendet. Bei der Resuspendierung wird die Mycel-Kultur vorzugsweise auf eine optische Dichte von 25 bis 160 (bei 400 nm) verdünnt, besonders bevorzugt auf eine optische Dichte von 30 bis 100 (bei 400 nm), ganz besonders bevorzugt auf eine optische Dichte von 40 bis 60 (bei 400 nm).
Das Inkontaktbringen der Mycel-Kultur erfolgt vorzugsweise durch Mischen mit der oben genannten Transformationsmischung. Die so erhaltene Mischung wird vorzugsweise für 20 bis 60 Minuten, besonders bevorzugt für 30 bis 40 Minuten bei einer Temperatur von vorzugsweise 30 bis 46°C, besonders bevorzugt bei 37 bis 40°C inkubiert. Als vorteilhaft hat es sich erwiesen, die Mycele nach der Inkubation zu waschen. Als Waschflüssigkeiten werden vorzugsweise isotonische Medien eingesetzt, besonders bevorzugt S27M Medium (7,32 % [w/v] D-Mannitol, 0,5 % [w/v] Pepton, 0,3 % [w/v] Hefeextrakt, 0,2 % [w/v] CaC03) Dabei kann ein oder mehrmals gewaschen werden.
Die im erfindungsgemäßen Verfahren eingesetzte Transformationsmischung enthält die oben genannten Verbindungen a) - e).
Die im erfindungsgemäßen Verfahren eingesetzte Transformationsmischung enthält 0,25 bis 10 μg/ml zu transformierende DNA, vorzugsweise 1 bis 7,5 μg/ml, besonders bevorzugt 2 bis 6 μg/ml. Die zu transformierende DNA kann, in Form von einzelsträngiger oder doppelsträngiger DNA vorliegen, vorzugsweise wird DNA in Form von doppelsträngiger ringförmiger DNA (Plasmide) eingesetzt. Die vorzugsweise bei der Transformation eingesetzten Plasmide enthalten insbesondere folgende Bestandteile: mindestens einen Replikationsursprung, der die effiziente
Vermehrung des Plasmids in Amycolatopsis sp. DSM 9991 oder 9992 ermöglicht. Vorzugsweise enthält das Plasmid zusätzlich einen Replikationsursprung, der die effiziente Vermehrung des Plasmids in einer Zelle, die zur einfachen Produktion und Isolierung des Plasmids geeignet ist, (beispielsweise Escherichia coli) ermöglicht. Weiterhin enthält das Plasmid vorzugsweise ein Resistenzgen, welches die Selektion von Amycolatopsis sp. DSM 9991 oder 9992-Zellklonen, die das Plasmid enthalten,
ermöglicht, vorzugsweise ein Kanamycin-Resistenzgen, nicht aber ein Erythromycin- oder Thiostrepton-Resistenzgen, da Amycolatopsis sp. DSM 9991 oder 9992 eine inherente Erythromycin-bzw. Thiostrepton-Resistenz aufweisen. Vorzugsweise enthält das Plasmid Restriktionsschnittstellen zum Einbau fremder DNA-Fragmente. Vorzugsweise handelt es sich bei der zu transformierenden DNA um eine DNA, welche einen niedrigen Methyherungsgrad aufweist. Dies kann dadurch erreicht werden, dass die zu transformierende DNA aus einen Organismus isoliert wird, der nicht oder nur in geringem Ausmaß zur Modifizierung von DNA in der Lage ist. Diese Organismen sind dem Fachmann bekannt, es können beispielsweise verschiedene Escherichia coli-Stämme wie beispielsweise E. coli ET12567 (dam, dem, hsd) oder E. coli JMllO (dam, dem) oder Amycolatopsis sp. DSM 9991 oder 9992 selbst eingesetzt werden.
Die im erfindungsgemäßen Verfahren eingesetzte Transformationsmischung enthält 0,4 bis 0,7 M CsCl, vorzugsweise 0,5 bis 0,675 M CsCl, besonders bevorzugt 0,575 bis 0,625 M CsCl.
Die im erfindungsgemäßen Verfahren eingesetzte Transformationsmischung enthält 0 bis 9 mM MgCi2, vorzugsweise 2,5 bis 7,5 mM MgCi2, besonders bevorzugt 3,5 bis 5,5 mM MgCl2.
Die im erfindungsgemäßen Verfahren eingesetzte Transformationsmischung enthält 30 bis 50 % [w/v] Polyethylenglykol 1000 (im folgenden als PEG 1000 bezeichnet), vorzugsweise 31 bis 40 % [w/v] PEG 1000, besonders bevorzugt 32 bis 35 % [w/v] PEG 1000. Der Einsatz von PEG 1000 ist vorteilhaft, da bei Einsatz von PEG, welches ein höheres oder ein niedrigeres Molekulargewicht aufwies, nur geringe Transformationsraten erzielt werden konnten.
Die im erfindungsgemäßen Verfahren eingesetzte Transformationsmischung enthält 10 bis 50 μg/ml DNA, welche von a) verschieden ist, vorzugsweise 12 bis 30 μg/ml, besonders bevorzugt 15 bis 25 μg/ml. Die Anwesenheit von e) in der Transforma-
tionsmischung erlaubt es, die Konzentration der Komponente a) gering zu halten. Bevorzugt wird hierfür Kälberthymus-DNA eingesetzt, besonders bevorzugt ultra- schall-behandelte Kälberthymus-DNA.
In einer bevorzugten Ausführungsform enthält die im erfindungsgemäßen Verfahren eingesetzte Transformationsmischung
a) 0,25 bis 10 μg/ml zu transformierende DNA b) 0,575 bis 0,625 M CsCl c) 2,5 bis 7,5 mM MgCl2 d) 32 bis 35 % [w/v] Polyethylenglykol 1000 und e) 12 bis 30 μg/ml DNA, welche von a) verschieden ist.
In einer besonders bevorzugten Ausführungsform enthält die im erfindungsgemäßen Verfahren eingesetzte Transformationsmischung
a) 2 bis 6 μg/ml zu transformierende DNA b) 0,575 bis 0,625 M CsCl c) 3,5 bis 5,5 mM MgCl2 d) 32 bis 35 % [w/v] Polyethylenglykol 1000 und e) 15 bis 25 μg/ml DNA, welche von a) verschieden ist.
Nach der Transformation wird die Mycel-Kultur vorzugsweise mit einer R2L- Agarose-Lösung, wie in J. Madon et al. J Bacteriol. 173, 1991, 6325 - 6331 beschrieben, vermischt, wobei die R2L-Lösung auf vorzugsweise auf 37 bis 46°C, besonders bevorzugt auf 40 bis 42°C temperiert ist. Die Mischung wird anschließend auf Agar-Platten, vorzugsweise S27M-Agar-Platten, ausgebracht. Die Inkubationszeit beträgt vorzugsweise 14 bis 22 Stunden, besonders bevorzugt 16 bis 20 Stunden, die Inkubationstemperatur beträgt vorzugsweise 30°C. Nach der Inkubation erfolgt die Selektion vorzugsweise durch Überschichten der Platten mit Antibiotika-haltigem
Soft-Agar (0,5 % [w/v] Agar), vorzugsweise S27M-Soft-Agar, und anschließender Inkubation bei vorzugsweise 30 bis 37°C für vorzugsweise 5 bis 10 Tage.
Durch das erfindungsgemäße Verfahren ist es möglich, Amycolatopsis sp. DSM 9991 und DSM 9992 in einfacher Weise zu transformieren, wobei hohe Transformationsraten erhalten werden.

Beispiel 1
Konstruktion und Isolierung des für die Transformation eingesetzten Plasmids
Zur Konstruktion eines für die Tranformation geeigneten Plasmids wurde der Vektor pRL60 (Lal et al, J. Antibiotics 51, 1998, 161-169) eingesetzt. Dieser 10,2 kbp große Vektor enthielt einen Replikationsursprung (pA-rep) für verschiedene Amycolatopsis mediterranei Stämme, einen Replikationsursprung (pBR-ori) für Escherichia coli, ein Kanamycin-Resistenzgen, ein Erythromycin-Resistenzgen und ein α-Amylase-
Markergen. Das Plasmid wurde einem Restriktionsverdau mit EcoRI unterworfen, in einem Agarosegel aufgetrennt und ein 6 kbp großes Fragment, welches das Kanamycin-Resistenzgen, pA-rep und pBR-ori enthielt, wurde daraus isoliert. Das Fragment wurde religiert und das so erhaltene Plasmid pRLΕ6 (5843 bp) wurde in kompetente E. coli ET12567-Zellen transformiert und daraus isoliert. Restriktionsverdau, Auftrennung im Agarosegel, Isolierung des gewünschten DNA-Fragments, Religation, Transformation in E. coli und Isolierung des Plasmids wurden gemäß in der Molekularbiologie üblichen Standardprotokollen durchgeführt (s. beispielsweise J. Sambrook et al, Molecular cloning: a laboratory manual, 2nd ed. Cold Spring Harbor Laboratory, Cold Spring Harbor, NY).
Beispiel 2 (nicht erfindungs emäß) Protoplastentransformation von Amycolatopsis sp. DSM 9992
(modifiziert nach Hopwood et al 1985, Genetic manipulation of Streptomyces- a labor manual, The John Innes Foundation, Norwich, England.)
Folgende Methoden, Medien und Puffer wurden verwendet:
Protoplastierungs-Puffer (P-Puffer) für Streptomyceten 103 g Saccharose, 0,25 g MgCl2 x 6 H2O, 2,02 g K2SO4, 2 ml Spurenelementlösung
2 (beschrieben in Hopwood et al 1985, Genetic manipulation of Streptomyces- a
labor manual, The John Innes Foundation, Norwich, England), mit Wasser auf 790 ml auffüllen und autoklavieren. Nach dem Autoklavieren werden folgende sterilisierte Lösungen separat dazugegeben: 100 ml TES (5,73 % [w/v] auf pH 7,2 mit NaOH eingestellt), 100 ml CaCl2 x 2 H2O (3,68 % [w/v]) und 10 ml K2HP04 (0,5 % [w/v])
R3 -Regenerationsmedium für Protoplasten
103 g Saccharose, 10 g Glucose (Monohydrat), 0,5 g KC1, 4 g Pepton, 4 g Hefeextrakt, 8,1 g MgCl2 x 6 H2O, 2,2 g CaCl2 x 2 H2O, mit Wasser auf 880 ml auffüllen, 18 g Bactoagar zugeben und autoklavieren. Nach dem Autoklavieren werden folgende sterilisierte Lösungen separat dazugegeben: 100 ml TES (5,73 % [w/v] auf pH 7,2 mit NaOH eingestellt), 20 ml K2HP04 (1 % [w/v])
Transformations-Puffer für Protoplasten Alle Lösungen werden separat angesetzt und autoklaviert und anschließend wie angegeben gemischt:
25 ml Saccharose (10,3 % [w/v]), 75 ml Wasserfest, 0,2 ml Spurenelementlösung 2 (beschrieben in Hopwood et al 1985, Genetic manipulation of Streptomyces- a labor manual, The John Innes Foundation, Norwich, England) und 1 ml K SO4 (2,5 % [w/v]).
Es werden 9,3 ml von der Mischung entnommen, dazu werden folgende Lösungen zugefügt: 0,2 ml 5 M CaCl2 und 0,5 ml 1 M TRIS-Maleinat, pH 8. Vor der Verwendung werden 3 Teile des Transformations-Puffers mit 1 Teil [w/v] sterilem PEG 1550 gemischt.
Bestimmung des Protoplastentiters und der Regenerationsrate
Um die Regenerationsrate zu ermitteln wurden Verdünnungsreihen (10'1 - 10" ) in P-Puffer bzw. in H2Obidest (+ 0,01 % [w/v] Natriumdodecylsulfat) angelegt und je 100 μl der Verdünnnungsstufen mit 3 ml „Topagar" (38 °C, P-Puffer versetzt mit 0,4 % [w/v] „low melting point'Αgarose, Sigma) auf R3 -Regenerationsmedium für
Protoplasten aufgetragen. Die Platten wurden für 15-20 min unter der sterilen
Werkbank getrocknet, bevor sie bei 37 °C inkubiert wurden. Nach 4-6 Tagen wurde die Anzahl der Kolonien pro Platte ausgezählt.
a) Protoplastengewinnung
Die Anzucht des Mycels erfolgte für 15 h in 50 ml YMG-Medium mit 5 % [v/v] PEG 6000 in 300 ml Schikanekolben bei 37°C und 150 Upm. Unter sterilen Bedingungen wurde das Zellmaterial bei 3 000 Upm für 15 min abzentrifugiert und anschließend zwei mal mit je 15 ml einer sterilen 10,3 % [w/v] Saccharoselösung ge- waschen. Das abzentrifugierte Mycelpellet wurde einer Lysozym-Behandlung unterzogen, um einen (partiellen) Verdau des Murein-Sacculus zu erreichen. Hierzu wurde das Pellet in 4 ml Lysozym-Lösung (2 mg/ml gelöst in sterilem P-Puffer, s.u.) resuspendiert und bei 30°C unter leichtem Schütteln (120 Upm) inkubiert. Der Verlauf der Protoplastierung wurde über den gesamten Zeitraum mikroskopisch verfolgt. Bereits nach 15 - 30 min waren Protoplasten erkennbar. Nach 2 bis 2 lA h wurde die Suspension mit einer sterilen 5 ml-Pipette 3 mal auf- und abgesogen um eine bessere Protoplastenfreisetzung zu erzielen. Anschließend wurde die Suspension für weitere 15 - 30 min inkubiert.
Nach Zugabe von 5 ml P-Puffer erfolgte die Abtrennung der Protoplasten von den
Mycelresten durch differenzielle Zentrifugation. Nach 10 min Zentrifugation bei 1 100 Upm (Megafuge 1.0R) - entsprechend ca. 200 g - wurde der Protoplasten- haltige Überstand von den Mycelresten abdekantiert und erneut zentrifugiert. Dieser Schritt erfolgte bei 3 400 Upm (ca. 2 000 g) für 10 min. Die so erhaltenen Proto- plasten wurden in der Restflüssigkeit resupendiert. Zur Kontrolle der Protoplastierung wurden je 50 μl Suspension mit 50 μl P-Puffer, bzw. mit 50 μl ^O^est (+0,01 % [w/v] Natriumdodecylsulfat) versetzt und mikroskopisch betrachtet.) Es folgten 3 Waschschritte. Hierzu wurden 10 ml steriler P-Puffer zu der „cremigen" Protoplastensuspension gegeben und nach leichtem Schütteln folgte eine 10 min Zentrifugation bei 3 400 Upm und 4 °C. Die Protoplasten wurden abschließend P-
Puffer resuspendiert. Das Puffervolumen (ca. 1 - 3 ml) wurde so gewählt, dass ein
Titer von ca. 109 - 1010 Protoplasten pro ml erzielt wurde. Die so behandelten Protoplasten konnten bis zur weiteren Verwendung bei - 70°C für längere Zeit gelagert werden. Die Zellaliquote wurden langsam auf Eis bei - 70 °C eingefroren, Aufgetaut wurden die Protoplasten unter lauwarmen Wasser.
b) Protoplastentransformation
Für die Transformation wurden 100 μl der Protoplastensuspension (Herstellung s.o.) in der Tischzentrifuge zur Pelletierung kurz anzentrifugiert. Der Überstand wurde abdekantiert, das Pellet in der „Restflüssigkeit" resuspendiert (durch Fingerantippen).
Plasmid-DNA (in TE-Puffer; max. 20 μl) wurde zu dieser Protoplastensuspension gegeben. Parallel wurde zur Kontrolle ein Ansatz ohne Plasmid-DNA durchgeführt. Unmittelbar danach wurden 0,5 ml Transformations-Puffer versetzt mit 25 % [v/v] PEG 1 550 zugegeben und mit der Pipette zweimal auf- und abgesogen und je 50 μl der Suspension mit 3 ml „Topagar" (38 °C, P-Puffer versetzt mit 0,4 % [w/v] „low melting point'Αgarose, Sigma) auf Regenerationsmedium R3 ausplattiert.
Zur Selektion transformierter Protoplasten wurde dem Medium R3 Antibiotika (50 μg/ml Kanamycin) zugefügt. Nach Trocknung der Platten unter der sterilen Werkbank (ca. 30 min) wurden die Platten für 4-6 Tage bei 37°C inkubiert.
Anschließend wurde die Koloniezahl ermittelt.
Es wurden keine Kanamycin-resistenten Transformanten erhalten, obwohl die Regeneration der Protoplasten erfolgreich war, was durch einen Kontrollversuch ge- zeigt werden konnte.
Beispiel 3 (nicht erfindungsgemäß) Elektroporation von Amycolatopsis sp. DSM 9992
Für die Elektroporation wurden eine Kultur von Amycolatopsis sp. DSM 9992 in
TYN-Mediurn (Trypton 10 g; Hefeextrakt 2,5 g; NaCl 5 g; H2Obidest ad 1000 ml; pH
7,2) bei 150 Upm für ca. 20 h bei 37°C angezogen. Das Mycel wurde durch Zentrifugation (4 500 Upm, 4°C, 15 min) geerntet und anschließend zweimal mit eiskaltem salzfreiem Wasser (Milli-Q Plus- Aufbereitungssystem für hochreines Wasser; MILLIPORE, Eschborn, Deutschland) gewaschen. Das Mycelpellet wurde in 200 μl Lysozym-Lösung (4 mg/ml; in 10 % [v/v] Glycerin) resuspendiert und für
20 min bei Raumtemperatur inkubiert. Im Anschluss wurde die Mycelsuspension mit 10 % [v/v] Glycerin gewaschen, die Zentrifugation wurde bei 3 000 Upm und 4°C für 10 min durchgeführt. Die so vorbehandelten Zellen wurden in einem entsprechenden Volumen Glycerin (10 % [v/v]) so resuspendiert, dass sich ein Zelltiter von ca. 1 x 1010 CFU/ml ergab.
Je 400 μl der vorbehandelten Mycelsuspension wurden mit der zu übertragenden Plasmid-DNA (0,1-5,0 μg/μl) gemischt und in gekühlte Elekfroporationsküvetten (Eppendorf-Netheler-Hinz, Hamburg) mit 2 mm Elektrodenabstand überführt. Die Elektroporation erfolgte bei einer elektrischen Feldstärke von 7,5 kV/cm (Kapazität
25 μF; Widerstand 600 Ω.). Zeitkonstanten von 3,6 - 5,6 ms wurden erreicht. Direkt nach der Elektroporation wurde 400 μl LB-Medium zu der Mycelsuspension zugefügt. Direkt im Anschluss wurden je 100 μl des Ansatzes auf GYM-Platten ausplattiert. Nach 15 h Inkubation bei 30°C wurden die Platten mit 3 ml Soft- Agar (TYN-Medium versetzt mit 5 g/1 Agar) überschichtet, welcher zur Selektion positiver
Elektroporanden Antibiotika (z.B. 1000 μg/ml Kanamycin) enthielt. Nach 20 min Trockenen der Platten wurden diese für 3 - 5 Tage bei 30°C bzw. 37°C inkubiert. Anschließend konnten die erhaltenen möglichen positiven Elektroporanden weiter untersucht werden.
Eine Transformationsrate von 2 x 102 Transformanten pro μg Plasmid-DNA wurde bei einer Feldstärke von 7,5 kV cm-1 und einem Puls von 4,6 - 5,2 ms erhalten.
Beispiel 4
Erfmdungsgemäße Transformation von Amycolatopsis sp. DSM 9992
Die Zellen wurden TSB-Medium angezogen. Das Zellmaterial wurde nach 20 h Wachstum (7 h stationär) bei 4 500 Upm für 15 min abzentrifugiert. Nach dreimaligem Waschen des geernteten Mycels mit TRIS-EDTA-Puffer wurde durch Resuspension in geeignetem Volumen TE-Puffer eine dichte Mycelsuspension (OD400nrn = 50) erhalten. 100 μl dieser Suspension wurden mit MgCl2 (Merck Darmstadt, Endkonzentration 5 mM), CsCl (ICN Biomedicals, Endkonzentration 0,625 M), Kälberthymus-DNA (Sigma-Aldrich, Endkonzentration 37,5 ng/μl;
Stammlösung 7,5 μg/ml), Plasmid-DNA (Endkonzentration 1,25 μg/ml) und PEG 1 000 (NBS Biologicals, Endkonzentration 32,5 % [w/v]) versetzt. Das Gesamtvolumen des Transformationsgemisches betrug 400 μl. Die verwendete Plasmid- DNA des Modellvektors pRLE6 wurde zuvor wie in Beispiel 1 beschrieben, aus E. coli ET 12567 isoliert.
Das Transformationsgemisch wurde bei 37°C für 40 min inkubiert. Die Zellen wurden zweimal mit je 1 ml S27M-Medium gewaschen Das Mycel wurden in 400 μl S27M-Medium resuspendiert und für 10 min auf Eis inkubiert. Anschließend wurden Aliquots der Zellen mit R2L-Agarose-Lösung (temperiert auf 42°C) vermischt und auf gut getrocknete S27M-Agar-Platten ausgebracht. Gegebenenfalls wurden die Ansätze zuvor mit S27M-Medium verdünnt. Nach 16 - 20 h Inkubation bei 30°C erfolgte die Selektion durch Überschichten der Platten mit Kanamycin-haltigem Soft- Agar (S27M-Medium mit 5 g/1 Agar) und anschließender Inkubation bei 37°C für 5-10 Tage.
Die folgenden Beispiele wurden analog zu Beispiel 4 durchgeführt, wobei jeweils 1 Parameter (s. Tabellen) variiert wurde.

Beispiel 5
Erfindungsgemäße Transformation von. Amycolatopsis sp. DSM 9992
Beispiel 5 wurde analog zu Beispiel 4 durchgeführt, wobei das Mycel 7 h nach Erreichen der stationären Phase geerntet wurde (OD4oonm = 50). PEG 1000 wurde in einer Endkonzentration von 32,5 5 [w/v], MgCl2 in einer Endkonzentration von 5 mM, CsCl in einer Endkonzentration von 0,6 M und Kälberthymus-DNA in einer Endkonzentration von 19 ng/μl eingesetzt, weiterhin wurden 0,5 μg des Plasmids pRLE6 (isoliert nach Beispiel 1) eingesetzt.
Das Beispiel wurde wiederholt, wobei statt des aus E. coli ET12567 isolierten Plasmids pRLE6 die in Tabelle 8 beschriebenen Plasmide eingesetzt wurden. Dabei weist das aus E. coli XLl-Blue isolierte Plasmid einen höheren Methyherungsgrad auf als das aus E. coli ET12567 und Amycolatopsis sp. DSM 9992 isolierte Plasmid.
Claims
1. Verfahren zur Transformation von Amycolatopsis sp. DSM 9991 oder 9992 durch
(1) Kultivierung von Amycolatopsis sp. DSM 9991- oder DSM 9992- Mycelien in einem Kulturmedium und
(2) Inkontaktbringen dieser Kultur mit einer Mischung enthaltend
a) 0,25 bis 10 μg/ml zu transformierende DNA b) 0,4 bis 0,7 M CsCl c) 0 bis 9 mM MgCl2 d) 30 bis 50 % [w/v] Polyethylenglykol 1000 und e) 10 bis 50 μg/ml DNA, welche von a) verschieden ist,
wobei die Kultur 4,5 bis 9 Stunden nach Eintritt in die stationäre Phase mit der genannten Mischung in Kontakt gebracht wird.
2. Verfahren gemäß Anspruch 1, wobei die Kultur 5 bis 8,5 Stunden nach Eintritt in die stationäre Phase mit der genannten Mischung in Kontakt gebracht wird.
3. Verfahren gemäß einem oder mehreren der vorangegangenen Ansprüche 1 und
2, wobei die genannte Mischung 0,5 bis 0,675 M CsCl enthält.
4. Verfahren gemäß einem oder mehreren der vorangegangenen Ansprüche 1 bis
3, wobei die genannte Mischung 2,5 bis 7,5 mM MgCl2 enthält.
5. Verfahren gemäß einem oder mehreren der vorangegangenen Ansprüche 1 bis
4, wobei die genannte Mischung 12 bis 30 μg/ml DNA, welche von a) verschieden ist, enthält.
6. Verfahren gemäß einem oder mehreren der vorangegangenen Ansprüche 1 bis 5, wobei e) Kälberthymus-DNA ist.
7. Verfahren gemäß einem oder mehreren der vorangegangenen Ansprüche 1 bis 6, wobei die genannte Mischung 32 bis 35 % [w/v] Polyethylenglykol 1000 enthält.
8. Verfahren gemäß einem oder mehreren der vorangegangenen Ansprüche 1 bis 7, wobei a) eine DNA mit einem niedrigen Methylierungsgrad ist.
9. Transformierte Amycolatopsis sp. DSM 9991 oder 9992, wobei die Transformation nach einem Verfahren gemäß den Ansprüchen 1 bis 8 durchgeführt wurde.
10. Verwendung von Amycolatopsis sp. DSM 9991 oder 9992 gemäß Anspruch 9 zur Herstellung von Vanillin.
11. Verwendung von Amycolatopsis sp. DSM 9991 oder 9992 gemäß Anspruch 9 zur Herstellung von Vanillin aus Ferulasäure.
12. Verfahren zur Herstellung von Vanillin, dadurch gekennzeichnet, dass transformierte Amycolatopsis sp. DSM 9991 oder 9992 gemäß Anspruch 9 eingesetzt werden.
