Abstract

Die vorliegende Erfindung betrifft ein D-Pantothensäure enthaltendes Tierfuttersupplement, ein verbessertes Verfahren zu dessen Herstellung und dessen Verwendung.
Als ein Ausgangsprodukt der Biosynthese von Coenzym A ist D- Pantothenat in der Pflanzen- und Tierwelt weit verbreitet. Im Gegensatz zum Menschen, der die Pantothensäure in ausreichenden Mengen über die Nahrung zu sich nimmt, sind jedoch sowohl für Pflanzen als auch für Tiere häufig Mangelerscheinungen für D-Pantothenat beschrieben. Die Verfügbarkeit von D-Pantothenat ist daher von großem wirtschaftlichen Interesse, insbesondere in der Tierfutterindustrie.
Herkömmlicher Weise erfolgt die Herstellung von D-Pantothenat durch chemische Synthese aus D-Pantolacton und Calcium-ß-Alaninat (Ullmann's Encyclopedia of Industrial Chemistry, 6th edition, 1999, electronic release, Kapitel „Vitamins"). Zur Bereitstellung von D- Pantolacton ist eine aufwendige, klassische Racematspaltung über diastereomere Salze erforderlich. Das aus der chemischen Synthese resultierende Verkaufsprodukt ist meist das Calciumsalz der D- Pantothensäure Calcium-D-Pantothenat.
Gegenüber der chemischen Synthese liegt der Vorteil biotechnologischer Herstellungsverfahren mit Mikroorganismen in der selektiven (enantiomerenreinen) Bereitstellung der für den höheren Organismus verwertbaren D-Form der Pantothensäure. Eine aufwendige Racematspaltung, wie sie bei der chemischen Synthese erforderlich ist, entfällt somit.
Fermentative Verfahren zur Herstellung von D-Pantothensäure mit Mikroorganismen sind zahlreich bekannt, so u.a. aus EP 0 590 857, WO 96/33283, US 6,013,492, WO 97/10340, DE 198 46 499, EP 1 001 027, EP 1 006 189, EP 1 006 192 und EP 1 006 193.
So beschreibt die GB 598,177 die Darstellung von Pantothensäure im Promillebereich als Nebenprodukt der 2,3-Butylenglykolherstellung durch Aerobacter aerogenes. Eine Aufkonzentrierung der Pantothensäure ist jedoch nur durch Adsorption an Holzkohle und anschließende Elution möglich.
Die EP 1 006 189 ein Verfahren zur Herstellung von Pantothenat bei dem in der Fermentationslösung ein Gehalt von max. 1 g/l D-Pantothensäure erreicht wird. Solche geringen Pantothensäure-Gehalte in der Fermentationslösung, also von weniger als 10 Gew.-% bezogen auf den Feststoffgehalt sind jedoch für eine wirtschaftliche Herstellung von D- Pantothensäure enthaltenden Tierfuttersupplementen ungeeignet. Ein weiterer Nachteil bei den bislang beschriebenen Verfahren ist, daß die Isolierung des Produkts aus dem Fermentationsmedium zahlreiche aufwendige Aufarbeitungsschritte erfordert. Ein wirtschaftliches Herstellungsverfahren für den großtechnischen Maßstab ist nicht offenbart.
So beschreibt die US 6,013,492 die Aufarbeitung der D-Pantothensäure aus der Fermentationslösung durch Abfiltern unlöslicher Bestandteile, wie z. B. Zellmaterial vom Kulturmedium, eine Adsorption des Filtrats an Aktivkohle, eine sich anschließende Elution der D-Pantothensäure mit einem organischen Lösungsmittel, bevorzugt Methanol, eine Neutralisation mit Calciumhydroxid und eine abschließende Kristallisation von Calcium-D-Pantothensäure. Ein wesentlicher Nachteil dieser
aufwendigen Aufarbeitungsschritte ist ein zusätzlicher Verlust an Wertprodukt. Darüber hinaus wäre bei einer Produktion im technischen Maßstab eine zusätzliche Anlage zur Rückgewinnung des eingesetzten Lösungsmittels erforderlich. Ein weiterer Nachteil ist der zusätzliche Anfall großer Mengen an Abwasser, welches seinerseits eine kosten intensive Behandlung oder sogar Entsorgung erfordert.
Aus der DE 100 16 321 A1 ist ein Verfahren zur Herstellung von Tierfuttermittel-Additiven bekannt, die durch Fermentation von D- Pantothensäure produzierenden Mikroorganismen hergestellt werden. Allerdings erfordert dieses Verfahren nach der Fermentation die Zugabe von Hydroxiden oder Oxiden der Alkali- oder Erdalkalimatallen.
Aufgabe der vorliegenden Erfindung ist somit die Bereitstellung eines freie D-Pantothensäure und/oder deren Salze enthaltendes
Tierfuttersupplement sowie dessen Herstellung durch ein verbessertes Verfahren, das die zuvor genannten Nachteile nicht mehr aufweist.
Diese Aufgabe wird durch die vorliegende Erfindung in vorteilhafter Weise gelöst.
Gegenstand der Erfindung ist ein Verfahren zur Herstellung eines freie D- Pantothensäure und/oder deren Salze enthaltendes Tierfuttersupplement, wobei
a) ein D-Pantothensäure produzierender Organismus in einem Kulturmedium enthaltend wenigstens eine Kohlenstoff- und eine Stickstoffquelle, ohne die Zufütterung weiterer Vorstufen fermentiert wird und
b) die D-Pantothensäure und/oder deren Salze enthaltende Fermentationslösung ohne die Durchführung weiterer Aufarbeitungsschritte einer Trocknung und/oder Formulierung unterzogen wird.
Das erfindungsgemäße Verfahren zeichnet sich ferner dadurch aus, daß die Fermentation solange durchgeführt wird, bis ein Feststoffgehalt von wenigstens 6 Gew.-%, bevorzugt von 7 - 25 Gew.-% und/oder ein Gehalt an D-Pantothensäure von wenigstens 2 - 15 Gew.-%, bevorzugt 4 - 15 Gew.-% erreicht wird
Dabei kann die Fermentation nach an sich bekannten Vorgehensweisen im batch-, fed-batch- oder repeated-fed-batch-Betrieb oder bei kontinuierlicher Prozeßführung durchgeführt werden. Unter einem Feststoffgehalt im Sinne der vorliegenden Erfindung ist die getrocknete Fermentationslösung enthaltend u.a. getrocknete Biomasse, Mineralien und D-Pantothenat und/oder deren Salze zu verstehen. Zur Bestimmung des Feststoffgehalts wird eine Probe der Fermentationslösung steril entnommen und beispielsweise in einem Vakuumtrockenschrank 12 Stunden bei 120 °C getrocknet.
Ein besonderer Vorteil des erfindungsgemäßen Verfahrens gegenüber dem Stand der Technik ist dabei, daß die Fermentationslösung nicht einer weiteren, aufwendigen Aufarbeitung unterzogen werden muß, wie z. B. adsorptive Verfahren über Aktivkohle, um ein Produkt zu liefern, das die Anforderungen an ein Tierfuttersupplement der gewünschten Art erfüllt. Diese Anforderungen sind beispielsweise ein relativ hoher Gehalt an D-. Pantothensäure und eine gute Verträglichkeit für den Zielorganismus sowie eine biologische Wertigkeit im Sinne der „Vitamin-Wirkung" des erfindungsgemäßen Produktes, die der Wertigkeit der chemisch synthetisierten D-Pantothensäure entspricht.
Der Reinheitsgrad der D-Pantothensäure als solche ist hier als nebensächlich anzusehen, da sie für die Tierernährung in den meisten Fällen in Futtermischungen eingearbeitet wird. Vielmehr ist ein weiterer Vorteil des erfindungsgemäßen Produktes gerade darin zu sehen, daß es aufgrund des zuvor genannten Herstellungsverfahrens neben D- Pantothensäure weitere dem Wohlbefinden der Tiere zuträgliche Bestandteile der Fermentationslösung, wie z. B. einen relativ hohen Proteingehalt, während der Fermentation ggf. in das Medium sekretierte (ggf. sogar essentielle) Aminosäuren, Mineralien, Vitamine u.a. Bestandteile enthält.
Ein weiterer Vorteil des erfindungsgemäßen Verfahrens ist die Einsparung aufwendiger Aufarbeitungsschritte der Fermentationslösung zur Herstellung eines Produktes mit guter biologischer Wertigkeit. Insbesondere zeichnet sich das erfindungsgemäße Verfahren dadurch aus, daß die Bereitstellung des gewünschten Wertstoffes völlig ohne die Verwendung organischer Lösungsmittel erfolgt. Ferner wird erfindungsgemäß die Menge an anfallendem Abwasser wesentlich reduziert. Dies resultiert somit in weiteren Einsparungen an aufwendigen Aufbereitungs- und Entsorgungsanlagen. Somit zeichnet sich das erfindungsgemäße Verfahren in vorteilhafter Weise dadurch aus, daß es einfacher, weniger störanfällig, weniger zeitaufwendig, deutlich kostengünstiger und damit wirtschaftlicher ist, als herkömmliche Verfahren.
Hierbei ist unter der Formulierung „produzieren" erfindungsgemäß zu verstehen, daß der Organismus größere Mengen D-Pantothensäure und/oder deren Salze synthetisieren kann, als für den eigenen Stoffwechseibedarf erforderlich sind. In einer erfindungsgemäß vorteilhaften Variante liegt die synthetisierte Menge an D-Pantothensäure und/oder deren Salze nicht zellintern vor, sondern wird idealerweise
vollständig aus dem Organismus in das Kulturmedium abgegeben. Dies Ausschleusung kann aktiv oder passiv nach an sich bekannten Mechanismen erfolgen.
Erfindungsgemäß werden als D-Pantothensäure produzierende Organismen Mikroorganismen eingesetzt. Hierzu zählen erfindungsgemäß Pilze, Hefen und/oder Bakterien. Erfindungsgemäß werden bevorzugt Pilze wie beispielsweise Mucor oder Hefen, wie z. B. Saccharomyces oder Debaromyces und hierbei bevorzugt Saccharaomyces cerevisiae eingesetzt. Vorteilhaft werden erfindungsgemäß coryneforme Bakterien oder Bacillaceae verwendet. Erfindungsgemäß umfaßt sind bevorzugt z. B. Bakterien der Gattungen Corynebacterium, Escherichia, Bacillus, Arthrobacter, Bevibacterium, Pseudomonas, Salmonella, Klebsieila, Proteus, Acinetobacter oder Rhizobium. Besonders bevorzugt sind hierbei beispielsweise Corynebacterium glutamicum, Brevibacterium breve oder Bacillus subtilis, B. licheniformis, B. amyloliquefaciens, B. cereus, B. lentimorbus, B. lentus, B. firmus, B.pantothenticus, B. circulans, B. coagulans, B. megaterium, B. pumilus, B. thuringiensis, B. brevis, B. stearothermophilus und andere Bacillusarten der Gruppe 1 , die durch ihre 16sRNA charakerisiert sind oder Actinum mycetalis. Diese Aufzählung dient der Erläuterung und ist keinesfalls limitierend für die vorliegende Erfindung.
Darüber hinaus umfaßt die vorliegende Erfindung auch den Einsatz genetisch veränderter Organismen zur erfindungsgemäßen Herstellung eines Tierfuttersupplements enthaltend freie D-Pantothensäure und/oder deren Salze. Solche genetisch veränderten Organismen können beispielsweise durch chemische Mutagenese und anschließende Selektion durch eine geeignetes „Screeningverfahren" isoliert werden. Erfindungsgemäß sind auch sogenannte Produktionsstämme umfaßt, die zur Herstellung des Produktes im Sinne der vorliegenden Erfindung
geeignet sind und genetische Veränderungen hinsichtlich des Stoffwechselflusses in Richtung D-Pantothensäure aufweisen, wobei auch Veränderungen hinsichtlich der Ausschleusung von D-Pantothensäure und/oder deren Salzen über die Zellmembran inbegriffen sind.
Denkbar ist auch der Einsatz transgener Organismen, die aus der Übertragung homologer und/oder heterologer Nukleotidsequenzen resultieren, welche zur Synthese des gewünschten Produktes erforderlich sind oder förderlich sein können. Hierbei ist die Überexpression und/oder Deregulation ein oder mehrerer Gene einzeln und/oder in Kombination, lokalisiert im Genom und/oder auf einem Vektor, denkbar. Derartige transgene Organismen können in vorteilhafter Weise zusätzliche Kopien und/oder genetisch veränderte Gene ausgewählt aus der Gruppe von panB, panC, panD, panE und/oder deren Kombinationen und/oder sogar Organisationseinheiten, wie das panBCD-Operon enthalten. Ferner können weitere Stoffwechselwege, wie z. B. der Isoleucin-Valin- Biosyntheseweg, in den Organismen vorteilhaft manipuliert sein, wie beispielsweise in der EP 1 006 189, EP 1 006 192, EP 1 006 193 oder EP 1 001 027 beschrieben ist. Dadurch werden verzweigtkettige Vorläufersubstanzen der Pantothensäure-Biosynthese vermehrt zur Verfügung gestellt. Vorteilhaft werden gegebenenfalls die Gene für diesen Biosyntheseweg d.h. ilvB, ilvN, ilvC und/oder ilvD überexprimiert. Darüber hinaus sind genetische Veränderungen der Aspartat-α- Decarboxylase (panD), z.B. durch Überexpression und/oder Deregulation, in dem eingesetzten D-Pantothensäure produzierenden Organismus erfindungsgemäß umfaßt. In vorteilhafter Weise liegt so ß-Alanin bereits in den Zellen in erhöhten Konzentrationen gegenüber entsprechend nichtgenetisch veränderten Organismen vor und muß somit nicht als Precursor dem Kulturmedium zugesetzt werden, wie es beispielhaft in EP-A-0 590 857 beschrieben ist. Vorteilhaft sind Mikroorganismen, deren
Pantothensäure-(pan)- und/oder lsoleucin-Valin-(ilv)-Biosynthese und/oder Asparat-α-Decarboxylase (panD) dereguliert ist.
Weiterhin ist eine zusätzliche Überexpression der Ketopanthoat- Reduktase (panE) in den Mikroorganismen von Vorteil.
Weiterhin von Vorteil ist, wenn gegebenenfalls das coaA-Gen, das für die Synthese von CoenzymA erforderlich ist, in seiner Aktivität erniedrigt ist oder (beispielsweise in Bacillus-Arten) ganz ausgeschaltet ist. Bacillus enthält nämlich neben coaA ein weiteres Gen für diese enzymatische Funktion (= coaX). Auch die Aktivität dieses Gens coaX oder des korrespondierenden Enzyms kann verändert, bevorzugt erniedrigt, oder sogar deletiert werden, sofern coaA selbst noch eine ausreichende, wenn auch erniedrigte Enzymaktivität aufweist, d.h. die Enzymaktivität von coaA nicht ganz verloren gegangen ist. Neben der Überexpression der verschiedenen Gene ist auch eine genetische Manipulation der Promotorregionen dieser Gene in der Art vorteilhaft, daß diese Manipulation zu einer Überexpression der Genprodukte führt.
In einer Ausführungsvariante der vorliegenden Erfindung finden die gemäß der Anlage (PCT/US-Anmeldung 0025993) beschriebenen Bakterienstämme, wie z.B. PA 668 und/oder Derivate davon Einsatz. In einer bevorzugten Ausführungsvariante findet erfindungsgemäß der Mikroorganismus Bacillus subtilis PA 377, wie gemäß der Anlage (PCT/US-Anmeldung 0025993) beschrieben, Einsatz in dem erfindungsgemäßen Verfahren. Dieser Stamm Bacillus subtilis PA 377 wurde wie folgt hergestellt:
Ausgehend von dem Stamm Bacillus subtilis 168 (Marburg Stamm ATCC 6051 ), der den Genotyp trpC2 (Trp") ausweist, wurde über Transduktion des Trp+ Markers (aus dem Bacillus subtilis Wildtyp W23) der Stamm PY79 erzeugt. In den Stamm PY79 wurden durch klassische gentechnische Methoden (wie z. B. in Harwood, C.R. and Cutting, S.M.
(editors), Molecular Biological Methods for Bacillus (1990) John Wiley & Sons, Ltd., Chichester, England beschrieben) ΔpanB und ΔpanEI Mutationen eingebracht. Der resultierende Stamm wurde mit genomischer DNA des Bacillus subtilis Stammes PA221 (Genotyp P2 anBCD, trpC2 (Trp")) und genomischer DNA des Bacillus subtilis Stammes PA303 (Genotyp P2ψanE1) transformiert. Der resultierende Stamm PA327 hat den Genotyp P φanBCD, P26panE1 und ist Tryptophan auxotroph (Trp"). Mit dem Bacillus subtilis Stamm PA327 wurde in 10 mL Kulturen mit SVY- Medium (25 g/L Difco Veal Infusion Broth, 5 g/L Difco Yeast Extract, 5 g/L Na-Glutamat, 2,7 g/L Ammoniumsulfat auf 740 mL Wasser auffüllen, autoklavieren, anschließend Zugabe von 200 mL 1 M Kaliumphosphat, pH 7,0 und 60 mL 50% steriler Glucoselösung), das mit 5 g/L ß-Alanin und 5 g/L α-Ketoisovalerat supplementiert war, Pantothensäure-Titer von bis zu 3,0 g/L (24 h) erreicht.
Die Herstellung des Bacillus subtilis Stammes PA221 (Genotyp
P ψanBCD, trpC2 (Trp")) ist in folgendem Abschnitt beschrieben :
Durch klassische gentechnische Methoden wurde mit Hilfe der Sequenzinformation des panBCD Operons von E. coli (siehe Merkel et al., FEMS Microbiol. Lett., 143, 1996:247-252) ausgehend von einer Bacillus subtilis GP275 Plasmid-Bibliothek das panBCD Operon von Bacillus kloniert. Zur Klonierung wurde der E. coli Stamm BM4062 (birts) und die Information, daß das Bacillus Operon in der Nähe des birA Gens liegt, verwendet. Das panBCD Operon wurde in ein in E. coli replizierbares Plasmid eingebracht. Zur Verbesserung der Expression des panBCD Operons wurden starke, konstitutive Promotoren des Bacillus subtilis Phagen SP01 (P2Θ) verwendet und die Ribosomenbindungsstelle (=RBS) vor dem panB-Gen durch eine artifizielle RBS ersetzt. Vor die P2φanBCD Kassette auf dem Plasmid wurde ein DNA Fragment, das unmittelbar upstream des nativen panB Gens in Bacillus liegt, ligiert. Dieses Plasmid
wurde in den Bacillus subtilis Stamm RL-1 (durch klassische Mutagenese erhaltenes Derivat des Bacillus subtilis 168 (Marburg Stamm ATCC 6051), Genotyp trpC2 (Trp")) transformiert und durch homologe Rekombination wurde das native panBCD Operon durch das p26panBCD Operon ersetzt. Der resultierende Stamm heißt PA221 und hat den Genotyp P2φanBCD, trpC2 (Trp").
Mit dem Bacillus subtilis Stamm PA221 wurde in 10 mL Kulturen mit SVY- Medium, das mit 5 g/L ß-Alanin und 5 g/L -Ketoisovalerat supplementiert war, Pantothensäure-Titer von bis zu 0,92 g/L (24 h) erreicht.
Die Herstellung des Bacillus subtilis Stamms PA303 (Genotyp P2φanE1) ist in folgendem Abschnitt beschrieben :
Mit Hilfe der E. coli panE Gensequenz wurde die Bacillus panE Sequenz analog Moniert. Es zeigte sich, daß in B. subtilis zwei Homologe des panE Gens von E. coli existieren, die mit panEl und panE2 bezeichnet wurden. Durch Deletionsanalysen zeigte sich, daß das panEl Gen für 90 % der Pantothensäure Produktion verantwortlich ist, während die Deletion des panE2 Gens keinen signifikanten Effekt auf die Pantothensäure Produktion hatte. Auch hier wurde analog zur Klonierung des panBCD Operons der Promotor durch den starken konstitutiven Promotor P26 ersetzt und die Ribosomenbindungsstelle vor dem panEl Gen durch die artifizielle Bindungsstelle ersetzt. Das P φanE1 Fragment wurde in einen Vektor kloniert, des so gestaltet war, daß das P2φanE1 Fragment in den original panEl locus im Genom von Bacillus subtilis integrieren konnte. Der nach Transformation und homologer Rekombination resultierende Stamm heißt PA303 und hat den Genotyp P &panE1. Mit dem Bacillus subtilis Stamm PA303 wurde in 10 mL Kulturen mit SVY- Medium, das mit 5 g/L ß-Alanin und 5 g/L α-Ketoisovalerat supplementiert war, Pantothensäure-Titer von bis zu 1 ,66 g/L (24 h) erreicht.
Die weitere Stammkonstruktion erfolgte durch Transformation von PA327 mit einem Plasmid, welches das P QUVBNC Operon und das Marker-Gen für Spectinomycin enthielt. Das P2QUVBNC Operon integrierte in den amyE locus, was durch PCR nachgewiesen wurde. Ein Transformante wurde als PA340 (Genotyp P2φanBCD, P2φanE1, P26ilvBNC, specR, trpC2 (Trp")) bezeichnet.
Mit dem Bacillus subtilis Stamm PA340 wurde in 10 mL Kulturen mit SVY- Medium, das nur mit 5 g/L ß-Alanin supplementiert war, Pantothensäure- Titer von bis zu 3,6 g/L (24 h) erreicht, in 10 ml Kulturen mit SVY-Medium, das mit 5 g/L ß-Alanin und 5 g/L α-Ketoisovalerat supplementiert war, wurden Pantothensäure-Titer von bis zu 4,1 g/L (24 h) erreicht.
Des weiteren wurde in den Stamm PA340 eine deregulierte //vD-Kassette eingebracht. Dazu wurde ein Plasmid, welches das ilvD Gen unter der Kontrolle des P26 Promotors mit der artifiziellen RBS2 enthält, in PA340 transformiert. Dabei wurde das P QHVD Gen durch homologe Rekombination in den original ilvD locus integriert. Der resultierende Stamm PA374 hat den Genotyp P2φanBCD, P2φanE1, P26ilvBNC, P QΪIVD, specR und trpC2 (Trp"). Mit dem Bacillus subtilis Stamm PA374 wurde in 10 mL Kulturen mit SVY- Medium, das nur mit 5 g/L ß-Alanin supplementiert war, Pantothensäure- Titer von bis zu 2,99 g/L (24 h) erreicht.
Um mit dem Stamm PA374 ß-Alanin zufütterungsfrei Pantothensäure zu produzieren, wurden zusätzliche Kopien des für die Aspartat-α- decarboxylase kodierenden Gens panD in den Stamm PA374 eingebracht. Dazu wurde chromosomale DNA des Stammes PA401 , der nachfolgend beschrieben wird, in PA374 transformiert. Durch Selektion auf Tetrazyklin wurde der Stamm PA377 erhalten. Der resultierende Stamm PA377 hat den Genotyp P2φanBCD, P2φanE1, P26ilvBNC, P2eilvD, specR, tetR und trpC2 (Trp").
Mit dem Bacillus subtilis Stamm PA377 wurde in 10 mL Kulturen mit SVY- Medium Vorstufen-zufütterungsfrei Pantothensäure-Titer von bis zu 1 ,31 g/L (24 h) erreicht.
Die Herstellung des Bacillus subtilis Stamms PA401 (Genotyp P2φanD) ist in folgendem Abschnitt beschrieben :
Das Bacillus subtilis panD Gen wurde aus dem panBCD Operon in einen Vektor, der das Tetrazyklin Marker Gen trägt, Moniert. Vor das panD Gen wurde der Promotor P26 und eine oben beschriebene artifizielle RBS kloniert. Durch Restrikfionsverdau wurde ein Fragment, das das Tetrazyklin Marker Gen und das P2βpanD Gen enthielt, hergestellt. Dieses Fragment wurde religiert und in den oben beschriebenen Stamm PA221 transformiert. Dabei integrierte das Fragment in das Genom des Stamms PA211. Der resultierende Stamm PA401 hat den Genotyp P26panBCD, P2φanD, tetR und trpC2 (Trp").
Mit dem Bacillus subtilis Stamm PA401 wurde in 10 mL Kulturen in SVY- Medium, das mit 5 g/L α-Ketoisovalerat supplementiert war, Pantothensäure-Titer von bis zu 0,3 g/L (24 h) erreicht. In 10 mL Kulturen mit SVY-Medium das mit 5 g/L D-Pantoinsäure und 10 g/L L-Aspartat supplementiert war, wurden Pantothensäure-Titer von bis zu 2,2 g/L (24 h) erreicht.
Die genaue Konstruktion der Stämme ist gemäß der Anlage der PCT/US- Anmeldung 0025993 zu entnehmen.
Mit dem oben beschriebenen Stamm PA377 wird bei Glucose-Iimitierter Fermentation in SVY-Medium (25 g/L Difco Veal Infusion Broth, 5 g/L Difco Yeast Extract, 5 g/L Tryptophan, 5 g/L Na-Glutamat, 2 g/L (NH4)2S04) 10 g/L KH2PO4, 20 g/L K2HPO4> 0,1 g/L CaCI2, 1 g/L MgSO4, 1 g/L Natriumcitrat, 0,01 g/L FeSO4*7 H2O und 1 ml/L einer Spurensalzlösung folgender Zusammensetzung : 0,15 g Na2Mo04 x 2 H20, 2,5 g H3BO3, 0,7 g C0CI2 x 6 H2O, 0,25 g CuSO4 x 5 H20, 1 ,6 g
MnCI2 x 4 H2O, 0,3 g ZnSO4 x 7 H20, mit Wasser auf 1 I aufgefüllt)) im 10 L Maßstab bei kontinuierlicher Zufütterung einer Glucose-Lösung in 36 h (48 h) Pantothensäure-Konzentrationen in der Fermentationsbrühe von 18-19 g/L (22-25 g/L) erreicht. Durch Medien-, Stamm- und Fermentationsverfahrensentwicklung können die Pantothensäure-Titer in den Fermentationsbrühen auf über 40, 45, 50, 55, 60, 65, 70, 75, 80, 85, und > 90 g/L gesteigert werden.
Ein wesentlicher Vorteil des erfindungsgemäßen Verfahrens ist, daß die Fermentation in einem Kulturmedium durchgeführt wird, das außer wenigstens einer Kohlenstoff- und Stickstoffquelle als Ausgangsverbindungen keine weiteren Vorstufen (Precursor) enthält. D.h. die Biosynthese von D-Pantothensäure ist von der Zufütterung weiterer Vorstufen unabhängig. Unter solchen Vorstufen sind erfindungsgemäß Substanzen wie z.B. ß-Alanin und/oder L-Aspartat und/oder L-Valin und/oder α-Ketoisovalerat und/oder deren Kombinationen zu verstehen. In einer bevorzugten Variante des erfindungsgemäßen Verfahren wird die Fermentation des D-Pantothensäure produzierenden Organismus in einem Kulturmedium durchgeführt, welches wenigstens eine Kohlenstoff- und eine Stickstoffquelle als Vorstufe, aber kein dem Medium zugesetztes ß-Alanin enthält. Die Unabhängigkeit von der Zufütterung von Vorstufen stellt insbesondere einen wesentlichen wirtschaftlichen Vorteil des erfindungsgemäßen Verfahrens gegenüber bekannten Verfahren dar, da eine Vielzahl von Vorstufen sehr teuer sind.
Beispiele erfindungsgemäß geeigneter Kohlenstoffquellen zum Einsatz in einem Kulturmedium zur Fermentation der zuvor genannten Organismen sind Zucker, wie Stärkehydrolysate (Mono-, Di-, Oligosaccharide), bevorzugt Glucose oder Saccharose sowie Rüben- oder Rohrzuckermelasse, Proteine, Proteinhydrolysate, Sojamehl, Maisquellwasser, Fette, freie Fettsäuren, rückgeführte Zellen aus bereits
durchgeführten Fermentationen oder deren Hydrolysate sowie Hefeextrakt. Auch diese Aufzählungen sind nicht limitierend für die vorliegende Erfindung, ebenso wie die nachfolgenden Beispiele geeigneter Stickstoffquellen, wie Ammoniak, Ammoniumsulfat, Harnstoff, Proteine, Proteinhydrolysate oder Hefeextrakt. Ferner enthält das Fermentationsmedium Mineralsalze und/oder Spurenelemente, wie Aminosäuren und Vitamine. Die genauen Zusammensetzungen geeigneter Fermentationsmedien sind zahlreich bekannt und dem Fachmann zugänglich. Nach Inokkulation des Fermentationsmediums mit einem geeigneten D- Pantothensäure produzierenden Organismus mit dem Fachmann bekannten Zelldichten erfolgt ggf. unter Zusatz eines Antischaummittels die Kultivierung des Organismus. Die Fermentation wird erfindungsgemäß so geführt, daß sie bei Beendigung wenigstens einen Feststoffgehalt der getrockneten Fermentationslösung von wenigstens 6 Gew.-% und einen Gehalt an freier D-Pantothensäure von wenigstens 2 Gew.-%, bevorzugt von wenigstens 4 Gew.-% aufweist. Dazu kann die Fermentation im batch- , fed-batch- oder repeated-fed-batch-Betrieb unter Zudosierung der Kohlenstoffquelle durchgeführt oder kontinuierlich betrieben werden. Die Fermentationstemperatur liegt bei 10 - 70 °C, bevorzugt bei 20 - 50 °C. Die Begasung des Fermenters erfolgt mit Sauerstoff, Luft oder Mischungen mit Stickstoff oder anderen Inertgasen. Der pH-Wert wird auf einen Wert im Bereich von 4 - 8, bevorzugt 5 - 7,5 eingestellt und ggf. durch Zudosierung geeigneter Basen und/oder Säuren reguliert.
Ferner zeichnet sich das vorliegende Verfahren in vorteilhafter Weise dadurch aus, daß der Gesamt-Zuckergehalt bis zum Ende der Fermentation auf ein Minimum reduziert wird, da dieser anderenfalls die spätere Trocknung und/oder Formulierung der Fermentationslösung durch Verkleben erschwert. Dies kann erfindungsgemäß dadurch erreicht werden, daß die Fermentation noch einige Zeit weitergeführt wird,
nachdem die Kohlenstoffquelle verbraucht ist (bei Kultivierung im batch- Betrieb) oder nachdem die Kohlenstoffzufuhr (bei einer Prozeßführung im fed-batch oder repeated-fed-batch-Betrieb) unterbrochen ist und/oder derart reguliert ist, daß die Konzentration der Kohlenstoffquelle nahezu Null ist (bei fed-batch, repeated-fed-batch oder kontinuierlicher Prozeßführung).
Dies erfolgt erfindungsgemäß dadurch, daß nach Unterbrechung der Zudosierung der Kohlenstoffquelle (z. B. Zuckerlösung) die Fermentation bis zum Erreichen der Gelöstsauerstoffkonzentration (PO2) auf wenigstens 80 %, bevorzugt 90 % und besonders bevorzugt 95 % des Sättigungswertes in der Fermentationslösung weitergeführt wird.
Erfindungswesentlich für das erfindungsgemäße Verfahren ist ferner, daß die Fermentationslösung ohne die Durchführung weiterer Aufarbeitungsschritte einer Trocknung und/oder Formulierung unterzogen werden kann. D. h. aufwendige Aufarbeitungsschritte zur Isolierung des gewünschten D-Pantothensäure enthaltenden Wertproduktes aus der Fermentationslösung, wie z. B. adsorptive Aufreinigungen über Aktivkohle, sind nicht erforderlich. Eine Abtrennung der Biomasse aus der Fermentationslösung ist ebenfalls nicht zwingend erforderlich, so daß der Proteingehalt des erfindungsgemäßen Produktes, d.h. des D- Pantothensäure enthaltenden Tierfuttersupplements einen Proteingehalt von bis zu 50 Gew.-% aufweisen kann.
Die Trocknung und/oder Formulierung der Fermentationslösung erfolgt nach an sich bekannten Verfahren, wie beispielsweise Sprühtrocknung, Sprühgranulation, Wirbelschichttrocknung, Wirbelschichtgranulation, Trommeltrocknung oder Spin-flash Trocknung (Ullmann's Encyclopedia of Industrial Chemistry, 6th edition, 1999, electronic release, Kapitel „Drying of Solid Materials"). Die Gaseintrittstemperatur bei Konvektionstrocknung liegt im Bereich von 100 - 280 °C, bevorzugt bei 120 - 210 °C. Die
Gasaustrittstemperatur liegt bei 50 - 180 °C, bevorzugt bei 60 - 150 °C. Zur Einstellung einer gewünschten Partikelgrößenverteilung und der damit verbundenen Produkteigenschaften können Feinpartikel abgetrennt und zurückgeführt werden. Ferner kann Grobgut in einer Mühle gemahlen und ebenfalls anschließend zurückgeführt werden. Das erfindungsgemäße Produkt weist beispielsweise eine beige bis braune Färbung auf. Ferner enthält es einen Restwassergehalt von weniger als 5 Gew.-%, bevorzugt 1 - 3 Gew.-% und besonders bevorzugt von 0,5 - 2 Gew.-%. Um ein Verklumpen des Produktes zu vermeiden, sollte ein Wassergehalt von 5 Gew.-% nicht überschritten werden. Ein schematisches Blockfließdiagramm des zuvor genannten Verfahrens ist in Fig. 1 zusammengefaßt.
In einer Variante des zuvor genannten erfindungsgemäßen Verfahrens erfolgt vor der Trocknung und/oder Formulierung der Fermentionslösung ggf. eine Abtrennung der Biomasse von der Fermentationslösung. Diese Abtrennung kann nahezu vollständig oder nur teilweise erfolgen. Bevorzugt wird eine teilweise Abtrennung der Biomasse, wodurch der Proteingehalt auf unter 10 Gew.-% gesenkt werden kann. Zur Erzielung einer nahezu vollständigen Abtrennung der Biomasse können die Feststoffanteile von der wässrigen Flüssigkeit beispielsweise durch Zentrifugation abgetrennt werden. Bezogen auf das getrocknete Endprodukt kann in einer weiteren Variante der Erfindung sogar ein Proteingehalt von weniger als 5 Gew.-% eingestellt werden. Die abgetrennte Biomasse kann in vorteilhafter Weise genutzt werden, um die natürlichen und in bestimmten Toleranzbereichen auftretenden Schwankungen des D-Pantothensäuregehaltes in den
Fermentationslösungen unterschiedlicher Produktionsansätze auszugleichen. Beispielsweise kann nach Abtrennung der Biomasse mehrerer unterschiedlicher Ansätze durch die erneute Zugabe von zuvor abgetrennter Biomasse ein Produkt mit gleichbleibend konstantem Gehalt
an D-Pantothenat und/oder deren Salze geliefert werden. Dies garantiert ein Produkt mit reproduzierbar konstanter Qualität.
In einer weiteren Ausführungsform des erfindungsgemäßen Verfahrens kann vor der Trocknung und/oder Formulierung der Fermentionslösung und ggf. nach einer Abtrennung der Biomasse eine Aufkonzentrierung der Fermentationslösung zur Erhöhung des Feststoffgehaltes enthaltend D- Pantothensäure und/oder deren Salzen erfolgen. Dies kann beispielsweise durch Wasserentzug durch eine Eindampfung erreicht werden, die aus Kostengründen ggf. mehrstufig ausgeführt sein kann und zur Schonung des Produktes neben Normaldruck auch im Vakuum durchgeführt werden kann. Eine weitere Möglichkeit ist die Nutzung eines Membranverfahrens. Hier können beispielsweise Verfahren wie Nanofiltration und/oder Umkehrosmose verwendet werden. Die Aufkonzentrierung kann bis zu einem D-Pantothensäure-Gehalt von 20 bis 50 Gew.-% erfolgen. Gegebenenfalls kann gleichzeitig eine Rückführung des Wasser in den Fermentationsprozeß vorgenommen werden. Dadurch wird in vorteilhafter Weise die anfallende Menge an Abwasser reduziert, wodurch der Aufwand für die Abwasserbehandlung wesentlich gesenkt wird. Dies ist schematisch in Fig. 2 dargestellt.
In einer bevorzugten Ausführungsvariante der vorliegenden Erfindung liegt eine Kombination von Abtrennung der Biomasse und Aufkonzentrierung der verbleibenden Fermentationslösung, ggf. bei gleichzeitiger Rückführung des Wassers, vor. Eine Darstellung im Blockfließdiagramm ist in Fig. 3 gezeigt.
Hierzu kann zur Einstellung eines konstanten Wertstoffgehaltes im Produkt bei dem erfindungsgemäßen Verfahren nach Ende der Fermentation die Biomasse oder ein Teil davon z. B. mittels Separation, Zentrifugation, Ultra-, Mikro- oder Tiefenfiltration oder Kombinationen abgetrennt werden. Die so erhaltene Biomasse kann ihrerseits nochmals
mittels eines Dekanters weiter entfeuchtet werden. Der Klarlauf des Dekanters wird dann zum Zulauf des Separators zurückgeführt. Durch die Zellabtrennung kann der Gehalt der D-Pantothensäure im Produkt erhöht werden bzw. der Gehalt auf einen konstanten Wert eingestellt werden, in dem verschiedene Fraktionen miteinander vermischt werden, so daß auch schwankende Gehalte aus der Fermentation problemlos verarbeitet werden können. Anschließend kann eine weitere Aufkonzentrierung der Fermentationslösung erfolgen. Die Gehalte bezogen auf die freie D- Pantothensäure und/oder deren Salze liegen bei 20 - 95 Gew.-%, bevorzugt 30 - 90 Gew.-%. Besonders bevorzugt weist das resultierende Produkt einen hohen Gehalt an freier D-Pantothensäure und/oder deren Salze von 60 - 80 Gew.-% und insbesondere von mehr als 80 Gew.-% auf.
In weiteren Varianten des erfindungsgemäßen Verfahrens kann vor der Trocknung und/oder Formulierung der Fermentionslösung wenigstens einer der nachfolgenden Schritte umfassend
1) Lyse und/oder Abtötung der Biomasse und/oder
2) Abtrennung der Biomasse von der Fermentationslösung und/oder 3) Zugabe weiterer Zuschlagstoffe und/oder
4) Konzentrierung der Fermentationslösung, bevorzugt durch Wasserentzug und ggf. gleichzeitiger Rückführung des Wassers in den Fermentationsprozeß und/oder
5) Kombinationen der Schritte 1 ) bis 4) durchgeführt werden.
Gegenstand der vorliegenden Erfindung ist somit auch ein Verfahren, wobei die Lyse und/oder Abtötung der Biomasse noch in der Fermentationslösung oder erst nach der Abtrennung der Biomasse von der Fermentationslösung durchgeführt wird. Dies kann beispielsweise durch eine Temperaturbehandlung, bevorzugt bei 80 - 200 °C und/oder eine Säurebehandlung, bevorzugt mit Schwefelsäure oder Salzsäure
und/oder enzymatisch, bevorzugt mit Lysozym erfolgen. Ein Blockfließschema zur Verdeutlichung ist in Fig. 4 gezeigt.
Eine weitere Ausführungsform des erfindungsgemäßen Verfahrens beschreibt ein Vorgehen, bei dem der Fermentationslösung vor der Aufkonzentrierung und/oder vor der Trocknung und/oder Formulierung weitere Zuschlagstoffe und/oder Mischungen davon zur Einstellung eines einheitlichen Gehaltes an D-Pantothensäure und/oder zur Verbesserung der Produkteigenschaften, wie Staubverhalten, Fließeigenschaften, Wasseraufnahmefähigkeit und Lagerstabilität zugesetzt werden. Beispiele für solche Zuschlagstoffe und/oder Mischungen davon können auf der Basis von Zuckern z. B. Lactose oder Maltodextrin, auf der Basis von Getreide- oder Hülsenfruchtprodukten z. B. Maisspindelmehl, Weizenkleie und Sojaschrot, auf der Basis von Mineralsalzen u.a. Calcium-, Magnesium-, Natrium-, Kaliumsalze, sowie auch D-Pantothensäure oder deren Salze selbst (chemisch oder fermentativ hergestelltes D- Pantothensäuresalz) sein. Die Zugabe kann vor der Trocknung und/oder während des Granulierungs- bzw. Formulierungsschritts selbst erfolgen. Dies ist zusammenfassend in Fig. 5 verdeutlicht.
In einer weiteren Variante der vorliegenden Erfindung wird ein Calcium-D- Pantothenat hergestellt, indem in einem möglichst späten Schritt des erfindungsgemäßen Verfahrens, also bevorzugt vor und/oder während der Aufarbeitung der Fermentationslösung, d.h. vor und/oder während der Konzentrierung und/oder Trocknung und/oder Formulierung der Fermentationslösung (siehe Fig. 5) die Zugabe von Calciumsalzen erfolgt. Hierbei wird der Gehalt an Calciumionen durch Zugabe von Calciumsalzen so eingestellt, daß etwa 1 mol Calciumsalz pro 2 mol D- Pantothensäure im formulierten Endprodukt enthalten sind. Dabei kann der Gehalt an Calciumionen, der bereits in der Fermentationslösung enthalten ist, vorteilhaft berücksichtigt werden. Als Calciumsalze können
z.B. Calciumoxid, Calciumhydroxid, Calciumhydrogenphosphat, Calciumcarbonat, Calciumsulfat, Calciumchlorid und/oder ein anderes Calciumsalz verwendet werden.
Die vorliegende Erfindung betrifft somit auch ein Verfahren, bei dem bezogen auf den Gehalt an D-Pantothensäure im formulierten Produkt vor und/oder während der Aufkonzentrierung, Trocknung und/oder Formulierung 1 mol Calciumionen pro 2 mol D-Pantothensäure in Form eines Calciumsalzes als Zuschlagstoff zugesetzt werden.
Darüber hinaus kann in einer anderen Variante des erfindungsgemäß durch die Komposition des Fermentationsmediums und hier insbesondere die Auswahl der Mineralsalze mit einem speziellen Kation ein Produkt hergestellt werden, das vermehrt ein ausgewähltes Salz der D- Pantothensäure enthält. Beispielsweise kann durch die Verwendung von Di-Kaliumhydrogenphosphat/Kaliumdihydrogenphosphat-Puffer bereits im Verlauf der Fermentation ein Produkt hergestellt werden, das weitgehend Kalium-D-Pantothensäure enthält. Denkbare Salze sind z.B. Calcium-, Kalium-, Magnesium-, Natrium- oder Ammoniumsalze der D- Pantothensäure oder beliebige Mischungen davon. Ein Blockfließdiagramm ist in Fig. 6 dargestellt.
Erfindungsgemäß sind alle zuvor genannten Varianten sowie die in den Fig. 1 bis 6 gezeigten Vorgehensweisen frei kombinierbar.
Gegenstand der vorliegenden Erfindung ist ferner ein Tierfuttersupplement hergestellt nach einem der zuvor beschriebenen Verfahren auf der Basis einer durch Fermentation wenigstens eines D-Pantothensäure produzierenden Organismus erhaltenen Fermentationslösung enthaltend wenigstens freie D-Pantothensäure und/oder deren Salze in einer Konzentration von wenigstens 30 - 95 Gew.-%, einen Gesamt-
Zuckeranteil von 0,1 - 15 Gew.-% und einen Proteingehalt von weniger als 5 bis 50 Gew.-% bezogen auf die Trockenmasse.
Das erfindungsgemäße Tierfuttersupplement zeichnet sich dadurch aus, daß es 50 - 95 Gew.-%, bevorzugt 70 - 95 Gew.-%, besonders bevorzugt 60 - 80 Gew.-% und insbesondere mehr als 80 Gew.-% an freier D- Pantothensäure und/oder deren Salze enthält.
Erfindungsgemäß enthält die unbehandelte Fermentationslösung als Basis für das erfindungsgemäße Tierfuttersupplement wenigstens 10 g/l, bevorzugt wenigstens 20 g/l und besonders bevorzugt wenigstens 40 g/l D-Pantothenat und/oder deren Salze.
Ferner kann das Tierfuttersupplement der vorliegenden Erfindung Caicium-, Kalium-, Magnesium-, Natrium- und/oder Ammoniumsalze der D-Pantothensäure und/oder Mischungen davon enthalten.
Eine besondere Variante des erfindungsgemäßen Tierfuttersupplements zeichnet sich durch eine Zusammensetzung der Trockenmasse mit wenigstens folgenden Komponenten aus: a) freie D-Pantothensäure und/oder deren Salze wenigstens 30-95 Gew.-% b) Proteine max. 50 Gew.-% c) Gesamtzucker max. 15 Gew.-% d) Mineralstoffe max. 20 Gew.-%
Erfindungsgemäß kann das Tierfuttersupplement einen Proteingehalt von maximal 50 Gew.-% als Obergrenze und als Untergrenze weniger als 10 Gew.-%, bevorzugt weniger als 7 Gew.-% und besonders bevorzugt von weniger als 5 Gew.-% enthalten. Der Gesamt-Zuckeranteil des Tierfuttersupplements beträgt maximal etwa 15 Gew.-% und kann als
untere Grenze weniger als etwa 0,1 Gew.-% enthalten, wobei alle Zwischenstufen denkbar sind. Hinsichtlich seines Wassergehaltes zeichnet sich das erfindungsgemäße D-Pantothensäure enthaltende Produkt durch einen Restwassergehalt von weniger als 5 Gew.-%, bevorzugt 1 - 3 Gew.-% und besonders bevorzugt von 0,5 - 2 Gew.-% aus.
Gegenstand der vorliegenden Erfindung ist ferner ein Tierfuttersupplement enthaltend unbelebte, lebende und/oder vermehrungsfähige Anteile an D- Pantothensäure produzierenden Organismen. Bevorzugt handelt es sich dabei um Mikroorganismen, bevorzugt Pilzen, Hefen und/oder Bakterien. Besonders bevorzugt enthält das erfindungsgemäße Tierfuttersupplement unbelebte, lebende und/oder vermehrungsfähige Anteile an Pilzen der Gattung Mucor, Hefen der Gattung Saccharomyces und/oder Bakterien der Enterobakteriaceae, wie E. coli, Salmonellen, wie Salmonella typhimurium, Proteus vulgaris, Pseudomonaden, wie Pseudomonas matophila, Bacillaceae, wie Bacillus subtilis oder Bacillus cereus, coryneformer Bakterien, wie Corynebacterium glutamicum oder Brevibacterium breve und/oder Actinum mycetalis und/oder Mischungen davon. Ganz besonders bevorzugt sind Bakterien der Gattung Bacillus und hierbei der Spezies Bacillus subtilis. Ebenso sind genetisch veränderte und/oder transgene Organismen und/oder zur Herstellung von Tierfuttersupplementen geeignete Produktionsstämme erfindungsgemäß umfaßt. Die vorangehende Aufzählung ist dabei nicht limitierend für die vorliegende Erfindung.
Ferner ist erfindungsgemäß ein Tierfuttersupplement eingeschlossen, das weitere Zuschlagstoffe, bevorzugt auf Basis von Zuckern und/oder
Getreide- und/oder Hülsenfrüchten und/oder Mineralsalzen und/oder (separat hergestellte chemisch und/oder fermentativ hergestellte) D-
Pantothensäure und/oder deren Salze und/oder deren Mischungen enthält.
Das erfindungsgemäße Tierfuttersupplement ist ferner charakterisiert durch eine Formulierung mit einer Schüttdichte von 0,35 bis 0,7 kg/1, bevorzugt 0,4 bis 0,6 kg/1. Dabei weist es erfindungsgemäß einen mittleren Kσrndurchmesser im Bereich von 10 - 2000 μm, bevorzugt von
20 - 1500 μm, besonders bevorzugt von 25 - 1000 μm und höchst bevorzugt von 30 - 800 μm auf. Es hat eine beige bis braune Farbe. Das erfindungsgemäße Tierfuttersupplement kann als Pulver, Granulat, Pellet, mit einem Überzug versehen („coated") und/oder als Kombinationen davon vorliegen. Die Formulierung des erfindungsgemäßen
Tierfuttersupplements, beispielsweise durch umhüllende Verbindungen, dient z. B. zur Verbesserung der Produkteigenschaften, wie Staubverhalten, Fließeigenschaften, Wasseraufnahmefähigkeit und
Lagerstabilität.
Gegenstand der vorliegenden Erfindung ist ferner die Verwendung des Tierfuttersupplements mit den zuvor dargestellten Eigenschaften als Zusatz zu Tierfutter und/oder Tierfutterergänzungsmitteln.
Die nachfolgenden Beispiele dienen zur Erläuterung der vorliegenden Erfindung, wirken sich jedoch nicht limitierend aus:
Beispiel 1 : Herstellung von D-Pantothensäure-haltiger
Fermentationslösung mit B. subtilis
In einem Laborfermenter mit Rührer und Begasungseinrichtung mit 14 I Inhalt wurde wassriges Fermentationsmedium mit der folgenden Zusammensetzung vorgelegt:
Hefeextrakt 20 g/ι
Tryptophan 5 g/ι
Ammoniumsulfat 2 g/ι Natriumglutamat 5 g/ι
Nach der Sterilisation wurden folgende Medienkomponenten zusätzlich zugegeben:

0,15 g Na2Mo04 x 2 H20, 2,5 g H3BO3, 0,7 g CoCI2x 6 H20, 0,25 g CuS04 x 5 H20, 1 ,6 g MnCI2 x 4 H20, 0,3 g ZnS04 x 7 H20 werden mit Wasser auf 1 I aufgefüllt.
Die Zugabe der Spurensalzlösung erfolgt über eine Sterilfiltration. Das Anfangsflüssigkeitsvolumen beträgt 6 I. Die oben angeführten Gehalte sind auf diesen Wert bezogen.
Dieser Lösung wird 60 ml Impfkultur (OD6oo = 9,5) von Bacillus subtilis PA 377 zugesetzt und bei 37 °C bei 200 U/min bei einer Begasungsrate von 12 l/min fermentiert. Dieser Stamm ist gemäß der Anlage in der PCT/US- Anmeldung 0025993 beschrieben.
Innerhalb von 72 h wurden 4,5 I einer sterilen, wässrigen Lösung zudosiert. Die Zusammensetzung war:
Glucose 400 g/l
Hefeextrakt 25 g/ι
Während der Fermentation wurde der pH-Wert bei 7,2 durch die Zudosierung von Ammoniak in die Fermenterzuluft bzw. von
Phosphorsäure gehalten. Ammoniak dient gleichzeitig als Stickstoffquelle für die Fermentation. Die Drehzahl des Rührorgans wurde geregelt, indem der Gelöstsauerstoffgehalt auf 30 % des Sättigungswertes gehalten wurde. Nach Abbruch der Zudosierung der Kohlenstoffquelle wurde die Fermentation so lange weitergeführt, bis der Gelöstsauerstoffgehalt (PO2) einen Wert von 95 % des Sättigungswertes erreicht hat. Danach wurde die
Fermentation beendet und der Organismus thermisch abgetötet. Dazu wurde die Fermentationslösung bei 1 h bei 100 °C gehalten. Die Abtötung wurde durch Ausplattieren nachgewiesen. Die Konzentration an D- Pantothensäure bei Abbruch nach 72 h betrug 28 g/l.
In analoger Weise lassen sich auch Fermentationsbrühen erzeugen, die ß-Alanin- zufütterungsfrei Pantothensäure-Titer von über 20, 25, 30, 35, 40, 45, 50, 55, 60, 65, 70, 75, 80, 85, und >90 g/L aufweisen.
Bei dieser Fermentation wurde das Gegenion der D-Pantothensäure durch die Verwendung eines Di-
Kaliumhydrogenphosphat Kaliumdihydrogenphosphat-Puffers in der Fermentation so eingestellt, daß man weitgehend das Kaliumsalz der D- Pantothensäure, also Kalium-D-Pantothenat erhält.
Beispiel 2: Zellabtrennung und Trocknung von D-Pantothensäure- haltiger Fermentationslösung aus E. coli
Die Herstellung von D-Pantothensäure-haltiger Fermentationslösung erfolgt nach Beispiel 1 der US 6,013,492 mit Escherichia coli IFO 814/pFV 31. Im Anschluß daran wird die Fermentation bis zum vollständigen Verbrauch der Kohlenstoffquelle weiter begast, bis der Gelöstsauerstoffgehalt (PO2) auf über 80% angestiegen ist. Anschließend werden die Zellen mit einem Separator abgetrennt. Der Gehalt an D-Pantothensäure liegt hier bei 38,5 g/l. Nach einer Eindampfung mit einem Rotationsverdampfer unter Vakuum (< 100 mbar) auf einen Feststoffgehalt von etwa 45 Gew.-% wird das Konzentrat in einem Laborsprühtrockner unter folgenden Bedingungen getrocknet:
Gaseintrittstemperatur: 100-250 °C
Gasaustrittstemperatur: 60-150 °C
Man erhält ein rieselfähiges Produkt mit einem mittleren Partikeldurchmesser von 20-300 μm.
Beispiel 3: Trocknung von D-Pantothensäure-haltiger
Fermentationslösung aus B. subtilis mit zusätzlicher Abtrennung der Zellmasse
Fermentationslösung (1 I) aus Beispiel 1 wird in einem Laborsprühtrockner unter folgenden Bedingungen getrocknet:
Gaseintrittstemperatur: 100 - 250 °C Gasaustrittstemperatur: 60 - 150 °C
Man erhält ein rieselfähiges Produkt mit einem mittleren Partikeldurchmesser von 20 - 300 μm.
Beispiel 4: Zellabtrennung und Trocknung von D-Pantothensäure- haltiger Fermentationslösung mit Lactose als Zuschlagsstoff
Die Biomasse aus Fermentationslösung (1 I) aus Beispiel 1 wird in einer Zentrifuge abzentrifugiert. Der Überstand wird mit 30 g Lactose versetzt und in einem Laborsprühtrockner unter folgenden Bedingungen getrocknet:
Gaseintrittstemperatur: 100 - 250 °C Gasaustrittstemperatur: 60 - 150 °C
Man erhält ein rieselfähiges Produkt mit einem Partikeldurchmesser von 40 - 500 μm und einem Gehalt an freier D-Pantothensäure > 30 Gew.-%.
Beispiel 5: Trocknung von D-Pantothensäure-haltiger
Fermentationslösung mit chemisch hergestelltem Calcium-D- Pantothenat als Zuschlagsstoff, z.B. zur Einstellung einer festen Konzentration von D-Pantothensäure im Endprodukt
Die Biomasse aus Fermentationslösung (1 I) aus Beispiel 1 wird in einer Zentrifuge abzentrifugiert. Der Überstand wird mit 100 g chemisch hergestelltem Calcium-D-Pantothenat versetzt und in einem Laborsprühtrockner unter folgenden Bedingungen getrocknet:
Gaseintrittstemperatur: 100 - 250 °C Gasaustrittstemperatur: 60 - 150 °C
Man erhält ein rieselfähiges Produkt mit einem Partikeldurchmesser von 40 - 500 μm und einem Gehalt an freier D-Pantothensäure > 60 Gew.-%.
Beispiel 6: Einstellung des Calciumgehaltes in Formulierungen von D- Pantothensäure aus Fermentationslösungen
Eine D-Pantothensäure-haltige Fermentationslösung enthält nach Abtrennung der Biomasse einen Feststoffgehalt von 95 g/l, davon 70 g/l D-Pantothensäure und 25 g/l sonstige Feststoffe (Salze, Reste der Biomasse, weitere feste Bestandteile je nach Fermentationsmedium, keine Calciumionen). '
Nachfolgend sind die sich ergebenden Gehalte an D-Pantothensäure im formulierten Produkt bei Zugabe verschiedener Calciumsalze angegeben. Dabei wurde der Calciumgehalt so eingestellt, daß 1 mol Calciumionen pro 2 mol D-Pantothensäure enthalten sind.

Claims
1. Verfahren zur Herstellung eines freie D-Pantothensäure und/oder deren Salze enthaltendes Tierfuttersupplement, wobei
a) ein D-Pantothensäure produzierender Organismus in einem Kulturmedium enthaltend wenigstens eine Kohlenstoff- und eine Stickstoffquelle, ohne die Zufütterung weiterer Vorstufen fermentiert wird und b) die D-Pantothensäure und/oder deren Salze enthaltende
.Fermentationslösung ohne die Durchführung weiterer Aufarbeitungsschritte einer Trocknung und/oder Formulierung unterzogen wird.
2. Verfahren nach Anspruch 1 , dadurch gekennzeichnet, daß die Fermentation solange durchgeführt wird, bis ein Feststoffgehalt von wenigstens 6 Gew.-%, bevorzugt von 7 - 25 Gew.-% und/oder ein Gehalt an D-Pantothensäure von wenigstens 2 - 15 Gew.-%, bevorzugt 4 - 15 Gew.-% erreicht wird.
3. Verfahren gemäß einem der Ansprüche 1 oder 2, dadurch gekennzeichnet, daß die Zudosierung der Kohlenstoffquelle unterbrochen und/oder derart reguliert wird, daß eine Konzentration von nahezu Null eingestellt wird und/oder die Fermentation bis zum Erreichen einer Gelöstsauerstoffkonzentration von wenigstens 80 %, bevorzugt 90 % und besonders bevorzugt 95 % des Sättigungswertes in der Fermentationslösung weitergeführt wird.
4. Verfahren gemäß einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, daß ein D-Pantothensäure produzierender
Organismus eingesetzt wird, dessen Pantothensäure-(pan)- Und/oder lsoleucin-Valin-(ilv)-Biosynthese und/oder Asparat-α-Decarboxylase dereguliert ist.
5. Verfahren gemäß einem der Ansprüche 1 bis 4, dadurch gekennzeichnet, daß vor der Trocknung und/oder Formulierung ggf. eine Abtrennung der Biomasse von der Fermentationslösung erfolgt.
6. Verfahren gemäß einem der Ansprüche 1 bis 5, dadurch gekennzeichnet, daß vor der Trocknung und/oder Formulierung und ggf. nach der Abtrennung der Biomasse eine Aufkonzentrierung der
Fermentationslösung durch Wasserentzug, bei ggf. gleichzeitiger Rückführung des Wasser in den Fermentationsprozeß, erfolgt.
7. Verfahren gemäß einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, daß vor der Trocknung und/oder Formulierung wenigstens einer der nachfolgenden Schritte umfassend
1 ) Lyse und/oder Abtötung der Biomasse und/oder
2) Abtrennung der Biomasse von der Fermentationslösung und/oder 3) Zugabe weiterer Zuschlagstoffe und/oder
4) Konzentrierung der Fermentationslösung, bevorzugt durch Wasserentzug und ggf. gleichzeitiger Rückführung des Wassers in den Fermentationsprozeß und/oder
5) Kombinationen der Schritte 1 ) bis 4) durchgeführt wird.
8. Verfahren gemäß einem der Ansprüche 1 bis 7, dadurch gekennzeichnet, daß die Lyse und/oder Abtötung der Biomasse noch in der Fermentationslösung oder nach der Abtrennung der Biomasse von der Fermentationslösung durchgeführt wird.
9. Verfahren gemäß einem der Ansprüche 1 bis 8, dadurch gekennzeichnet, daß der Fermentationslösung vor und/oder während der Aufkonzentrierung, Trocknung und/oder Formulierung weitere
Zuschlagstoffe und/oder Mischungen von Zuschlagstoffen zugesetzt werden.
10.Verfahren gemäß einem der Ansprüche 1 bis 9, dadurch gekennzeichnet, daß bezogen auf den Gehalt an D-Pantothensäure im formulierten Produkt vor und/oder während der Aufkonzentrierung, Trocknung und/oder Formulierung 1 mol Calciumionen pro 2 mol D-
Pantothensäure in Form eines Calciumsalzes als Zuschlagstoff zugesetzt werden.
11. Tierfuttersupplement auf der Basis einer durch Fermentation wenigstens eines D-Pantothensäure produzierenden Organismus erhaltenen Fermentationslösung enthaltend wenigstens freie D- Pantothensäure und/oder deren Salze in einer Konzentration von wenigstens 30 - 95 Gew.-%, einen Gesamt-Zuckeranteil von 0,1 - 15 Gew.-% und einen Proteingehalt von weniger als 5 bis 50 Gew.-% bezogen auf die Trockenmasse.
12. Tierfuttersupplement gemäß Anspruch 11 , dadurch gekennzeichnet, daß es 50 r 95 Gew.-%, bevorzugt 70 - 95 Gew.-%, besonders bevorzugt 60 - 80 Gew.-% und insbesondere mehr als 80 Gew.-% an freier D-Pantothensäure und/oder deren Salzen enthält.
13. Tierfuttersupplement gemäß einem der Ansprüche 11 oder 12, dadurch gekennzeichnet, daß die unbehandelte Fermentationslösung wenigstens 10 g/l, bevorzugt wenigstens 20 g/l und besonders bevorzugt wenigstens 40 g/l D-Pantothenat und/oder deren Salze enthält.
14. Tierfuttersupplement gemäß einem der Ansprüche 11 bis 13, dadurch gekennzeichnet, daß es Caicium-, Kalium-, Magnesium-, Natrium- und/oder Ammoniumsalze der D-Pantothensäure und/oder Mischungen davon enthält.
15. Tierfuttersupplement gemäß einem der Ansprüche 11 bis 14, gekennzeichnet durch eine Zusammensetzung der Trockenmasse mit wenigstens folgenden Komponenten: a) freie D-Pantothensäure und/oder deren Salze wenigstens 30-95 Gew.-% b) Proteine max. 50 Gew.-% c) Gesamtzucker max. 15 Gew.-% d) Mineralstoffe max. 20 Gew.-%
16. Tierfuttersupplement gemäß einem der Ansprüche 11 bis 15, enthaltend einen Proteingehalt von weniger als 10 Gew.-%, bevorzugt weniger als 7 Gew.-% und besonders bevorzugt von weniger als 5 Gew.-%.
17. Tierfuttersupplement gemäß einem der Ansprüche 11 bis 16, enthaltend einen Gesamt-Zuckeranteil von weniger als 10 Gew.-%, bevorzugt von etwa 0,5 Gew.-% und besonders bevorzugt von etwa 0,1 Gew.-%.
18. Tierfuttersupplement gemäß einem der Ansprüche 11 bis 17, enthaltend einen Restwassergehalt von weniger als 5 Gew.-%, bevorzugt 1 - 3 Gew.-% und besonders bevorzugt von 0,5 - 2 Gew.- %.
19. Tierfuttersupplement gemäß einem der Ansprüche 11 bis 18, dadurch gekennzeichnet, daß es unbelebte, lebende und/oder vermehrungsfähige Anteile an D-Pantothensäure produzierenden Organismen enthält.
20. Tierfuttersupplement gemäß einem der Ansprüche 11 bis 19, dadurch gekennzeichnet, daß es unbelebte, lebende und/oder vermehrungsfähige Anteile an Mikroorganismen, bevorzugt Pilzen, Hefen und/oder Bakterien enthält.
21. Tierfuttersupplement gemäß einem der Ansprüche 11 bis 20, dadurch gekennzeichnet, daß unbelebte, lebende und/oder vermehrungsfähige Anteile an Pilzen der Gattung Mucor, Hefen der Gattung Saccharomyces und/oder Bakterien der Familien der Enterobakteriaceae, Salmonellen, Pseudomonaden, Bacillaceae, coryneformer Bakterien und/oder der Gattung Proteus und/oder Actinum und/oder Mischungen davon enthält.
22. Tierfuttersupplement gemäß einem der Ansprüche 11 bis 21 , dadurch gekennzeichnet, daß es weitere Zuschlagstoffe, bevorzugt auf Basis von Zuckern und/oder Getreide- und/oder Hülsenfrüchten und/oder Mineralsalzen und/oder D-Pantothensäure, Salze davon und/oder deren Mischungen enthält.
23. Tierfuttersupplement gemäß einem der Ansprüche 11 bis 22, dadurch gekennzeichnet, daß es in einer Formulierung mit einer Schüttdichte von 0,35 - 0,7 kg/l, bevorzugt 0,4 - 0,6 kg/l vorliegt.
24. Tierfuttersupplement gemäß einem der Ansprüche 11 bis 23, dadurch gekennzeichnet, daß ^s einen mittleren Korndurchmesser im Bereich von 10 - 2000 μm, bevorzugt von 20 - 1500 μm, besonders bevorzugt von 25 - 1000 μm und höchst bevorzugt von 30 - 800 μm aufweist.
25. Tierfuttersupplement gemäß einem der Ansprüche 11 bis 24, dadurch gekennzeichnet, daß es als Pulver, Granulat, Pellet, mit einem
Überzug versehen und/oder als Kombinationen davon vorliegt.
26. Verwendung des Tierfuttersupplements gemäß einem der Ansprüche 11 bis 25 als Zusatz zu Tierfutter und/oder Tierfutterergänzungsmitteln.
