Abstract
Die Erfindung betrifft eine Vorrichtung zur Probenaufgabe für miniaturisierte Analysensysteme. Durch gezieltes Design des Kanalsystems und Verdrängen des Flüssigkeitsvolumens eines bestimmen Kanalabschnitts können definierte Volumina von 0,01 νl bis zu 100 νl aufgegeben werden. Die erfindungsgemässe Vorrichtung eignet sich insbesondere für eine anschliessende Analyse der Proben mittels Isotachophorese.

Vorrichtung zur Probenaufgabe
Die Erfindung betrifft eine Vorrichtung zur Probenaufgabe für planare, miniaturisierte Analysensysteme.
In Bereichen wie der Lebensmittelanalytik, Umweltanalytik oder auch der industriellen Qualitätskontrolle besteht zunehmend Bedarf an Analysensystemen, die schnell und ohne großen apparativen Aufwand die genaue und quantitative Analyse komplexer Gemische ermöglichen. Neben Sensoren oder Schnelltests, die auf spezifischen chemischen Reaktionen basieren und daher keine universellen Verfahren darstellen, werden hauptsächlich chromatographische und elektrophoretische Trennverfahren eingesetzt. Im Gegensatz zu den meisten chromatographischen und elektrophoretischen Verfahren bietet die Isotachophorese (ITP) die Möglichkeit, große Probenmengen bei hoher Trennselektivität ohne vorherige Aufarbeitung zu analysieren. Elektrophoretische Trennverfahren wie die ITP eignen sich zudem auch zum Einsatz in miniaturisierten Analysensystemen (MAS), so daß der apparative Aufwand für die Analysen stark reduziert werden kann. Ein wesentlicher Vorteil des Einsatzes von MAS besteht darin, daß diese nach einer Kontaminierung verworfen werden können. Um diesen Vorteil zu realisieren, muß die Reproduzierbarkeit von Analysen in der Serie und zwischen verschiedenen MAS gleichen Types sichergestellt sein.
Neben der Analysenvorrichtung selbst ist einer der wichtigsten Bestandteile eines miniaturisierten Systems die Vorrichtung zur Probenaufgabe. Da Verfahren wie beispielsweise die ITP bezüglich der Beschaffenheit und der Menge der Probe sehr variabel sind, wird durch die Art der Probenaufgabe bestimmt, welches Probenvolumen und welche Art von Probe analysiert werden kann.
In makroskopischen Analysensystemen können ähnlich wie bei Geräten für die Hochdruckflüssigkeitschromatographie oder Geräten für die Isotacho- phorese mechanische Aufgabevorrichtungen zur Aufgabe eines definierten Probenvolumens verwendet werden. In Abbildung 4 wird beispielhaft eine derartige Aufgabevorrichtung aus dem Stand der Technik näher beschrieben. Die Vorrichtungen bestehen zumeist aus komplex aufgebauten Hahnsystemen, teilweise mit integrierten Aufgabeschleifen. Auf miniaturisierte Analysensysteme lassen sich diese Vorrichtungen nicht übertragen, da drehbare Hähne oder sonstige mechanische Vorrichtungen, wie beispiels- weise verschließbare Ventile, nicht entsprechend miniaturisiert werden können.
Deswegen finden sich bei miniaturiserten Analysenvorrichtungen auf Basis von Kapillarelektrophorese (CE) oder ITP Vorrichtungen, bei denen die Probenaufgabe elektrokinetisch unter Ausnutzung des elektroosmotischen Flusses erfolgt. Dies wird im folgenden elektroosmotische Probenaufgabe genannt. Ein schematischer Aufbau einer solchen Vorrichtung aus dem Stand der Technik ist in Abbildung 3 gezeigt. Durch gekreuzte oder versetzt gekreuzte Kapillarstrukturen wird ein Probenvolumen dadurch definiert, daß zunächst ein Kanal mit Probe befüllt wird. Dies kann beispielsweise elektro- osmotisch durch Anlegen einer Spannung erfolgen. Anschließend werden die Elektroden an dem befüllten Kanal auf gleiches Potential geschaltet und eine Spannung an dem senkrecht dazu liegenden Trennkanalsystem angelegt. Auf diese Weise wird das Probenvolumen, das sich an der Schnittstelle der beiden Kanalsysteme befindet, in das Trennkanalsystem transportiert. Das so erzeugte Probenvolumen liegt im Bereich einiger Nanoliter oder weniger.
Auf diese Weise ist es zwar möglich, ein durch die Schnittstelle der Kanäle definiertes Probenvolumen aufzugeben, aber die Volumenelemente, in denen durch Diffusion Stoffaustausch mit den Seitenkanälen stattfindet, sind bezogen auf das durch das Schnittvolumen vorgegebene Proben-
volumen sehr groß. Somit ist das tatsächlich eingebrachte Probenvolumen starken Schwankungen unterworfen. Da nur sehr kleine Probenmengen untersucht werden können, sinkt zudem die Konzentration bestimmter Analyte im Detektionsbereich schnell unter die Nachweisgrenze oder das entnommene Probenvolumen kann nicht als repräsentativ für die Probengesamtheit angesehen werden.
Weiterhin kann die Probenaufgabe bei ausreichend großem Kanalquerschnitt durch hydrodynamische Injektion aus einem Probengefäß erfolgen. Dabei wird ein Teil der Probe durch zeitlich gesteuertes Anlegen eines
Druckunterschiedes aus dem externen Probengefäß an den Anfang der Trennkapillare transportiert. Nachteil dieser Technik ist eine starke Abhängigkeit des Probenvolumens von der Beschaffenheit der Probe (z.B. Viskosität), aber auch von der erreichbaren Genauigkeit der Druck- Steuerung. Schon dadurch ist die Aufgabe eines exakt definiertes
Probenvolumen nicht möglich. Zusätzlich bestehen auch hier Probleme durch diffusiven bzw. konvektiven Stoffaustausch an den Grenzflächen zwischen Probevolumen und angrenzenden Volumeneinheiten. Bei kommerziellen, nicht miniaturisierten Systemen ist die hydrodynamische Injektion Stand der Technik, für miniaturisierte Systeme bietet sie keine
Vorteile gegenüber der oben beschriebenen elektrokinetischen Injektion unter Ausnutzung des elektroosmotischen Flusses.
Eine direkte, elektrophoretische Injektion aus einem externen Probengefäß (ohne Ausnutzung des elektroosmotischen Flusses), wie sie ebenfalls in kommerziellen Geräten eingesetzt wird, ist schon vom Prinzip her nicht geeignet, definierte Volumina aufzugeben, da hierbei in der Probelösung kein Volumenfluß erzeugt wird, sondern nur Ionen elektrophoretisch in das Trennsystem überführt werden.
Ein weiterer grundsätzlicher Nachteil aller elektroosmotischen Verfahren ergibt sich aus der eingeschränkten Materialauswahl. Da der Proben-
transport an das Auftreten eines elektroosmotischen Flusses gebunden ist, muß eine hohe Ladungsdichte an der Materialoberfläche vorhanden sein. Zudem erfolgt schon während der Aufgabe eine elektrophoretische Auftrennung der Probe, so daß ein inhomogenes Injektionsprofil entsteht.
Da mittels ITP problemlos größere Probenvolumina analysiert werden können, wird die Analyseleistung der derzeitigen miniaturisierten Analysensysteme größtenteils durch die ungenügende Möglichkeit zur Aufgabe großer, definierter Probenvolumina eingeschränkt.
Aufgabe der vorliegenden Erfindung ist es daher, eine Vorrichtung zur Probenaufgabe zu entwickeln, die es ermöglicht, definierte variable Probenvolumina zwischen 0,01 bis 100 μl in ein miniaturisiertes Analysensystem einzubringen.
Es wurde gefunden, daß eine Aufgabevorrichtung bestehend aus einem Kanalsystem sowie Fluidikanschlüssen zum Flüssigkeitstransport die Aufgabe großer Probenvolumina in planaren Systemen ermöglicht. Durch Öffnen des Systems am Ende eines Kanalabschnitts und gleichzeitiges Befüllen des Kanalabschnitts mit der Probenlösung am anderen Ende wird ein bestimmter Kanalabschnitt mit der Probenlösung gefüllt. Das Volumen des Kanalabschnitts und somit das aufgegebene Probenvolumen ist durch die Geometrie des Kanalabschnitts bestimmt, ansonsten aber frei wählbar.
Gegenstand der Erfindung ist daher eine Vorrichtung zur Aufgabe definierter Probenvolumina über 0,01 μl für miniaturisierte Analysensysteme, umfassend in der Hauptsache mindestens einen Kanalabschnitt, an dessen Enden jeweils Fluidikanschlüsse vorhanden sind.

Probenvolumina zwischen 0,05 und 30 μl aufgegeben werden.
Bevorzugte Ausführungsform ist weiterhin eine Aufgabevorrichtung, die mindestens zwei hintereinanderliegende Kanalabschnitte enthält, die jeweils von Fluidikanschlüssen begrenzt werden. Wenn beide Kanalab- schnitte direkt aneinander grenzen, so sind insgesamt drei Fluidik- anschlüsse vorgesehen.
Bevorzugte Ausführungsform ist auch eine Aufgabevorrichtung, die ein Kanalsystem mit mindestens zwei parallelen Kanalabschnitten enthält, die unabhängig voneinander von Fluidikanschlüssen begrenzt werden.
Eine bevorzugte Ausführungsform ist eine Vorrichtung, die als Fluidik- anschlüsse MikromischerNentile und Mikropumpen oder dichtschließende Mikropumpen besitzt.
Abbildung 1 zeigt eine erfindungsgemäße Aufgabevorrichtung.
In Abbildung 2 ist eine mögliche Vorgehensweise zum Befüllen eines miniaturisierten Analysensystems mit einer erfindungsgemäßen Aufgabevorrichtung gezeigt.
Abbildung 3 zeigt eine Aufgabevorrichtung für miniaturisisierte Analysensysteme aus dem Stand der Technik.
Abbildung 4 zeigt eine Aufgabevorrichtung für makroskopische Analysensysteme aus dem Stand der Technik.
Im Gegensatz zu anderen Aufgabemethoden wird bei der erfindungsgemäßen Vorrichtung das Kanalsystem während der Probenaufgabe an zwei Stellen geöffnet. Eine Öffnung dient zur Einleitung der Flüssigkeit, d.h. beispielsweise der Probenlösung, die andere Öffnung ermöglicht den Austritt der vorher im System befindlichen Flüssigkeit oder Luft. Prinzip der
erfindungsgemäßen Aufgabevorrichtung ist demnach das Verdrängen eines in einem bestimmten Kanalabschnitt befindlichen Flüssigkeits- oder Gas-Volumens durch die Probenlösung.
Durch geeignete Wahl der Eintritts- und Austrittsöffnung wird lediglich die
Flüssigkeit in dem dazwischenliegenden Kanalabschnitt verdrängt bzw. der dazwischenliegende Kanalabschnitt gefüllt. Die Flüssigkeit in eventuell vorhandenen angrenzenden Seitenkanälen wird nicht ausgetauscht, da sich in den Seitenkanälen keine geöffneten Eintritts- oder Austritts- Öffnungen befinden und so die Flüssigkeit in diesen Bereichen weder durch Druck noch durch Sog bewegt wird. Verluste bzw. Verdünnungen durch Flüssigkeitsströme an den Kontaktflächen zu Seitenkanälen sind im Verhältnis zum gesamten Probenvolumen, das typischerweise im μl- Bereich liegt, gering. Bei geeigneter konstanter Dosiergeschwindigkeit kann die Probenaufgabe sehr gut reproduzierbar erfolgen. Dies ist ein großer Vorteil gegenüber Methoden, bei denen sehr kleine Probenvolumina von wenigen Nanolitern aufgegeben werden. Eine erfindungsgemäße Aufgabevorrichtung ist prinzipiell auch für Aufgabenvolumina von weniger als 50 nl geeignet. Jedoch sind dann bezüglich Präzision und Genauigkeit Kompromisse notwendig.
Der Transport der Probenflüssigkeit kann über dicht angeschlossene Pumpen, Spritzen, Mikromischer, Elektroosmose oder hydrostatischen Druck erfolgen, bevorzugt über Mikropumpen- und Ventile.
Diese Vorrichtungen können bevorzugt außerhalb, möglichst dicht am Chip, angebracht werden.
Die austretende Flüssigkeit muß nicht zusätzlich abgepumpt werden. Sie wird durch den Druck der injizierten Ersatzflüssigkeit hinreichend effektiv verdrängt.
Durch diese Art des Befüllens werden die Nachteile der elektroosmotischen Injektion vermieden, d.h. die Befüllung ist weitgehend unabhängig von Probenzusammensetzung, pH-Wert und dem Material des Analysensystems. Durch die vorhandenen Ventile oder dichtschließenden Pumpen wird jede störende Flüssigkeitsbewegung, wie beispielsweise durch hydrostatische Druckdifferenzen oder Elektroosmose, unterbunden.
Erfindungsgemäß werden alle Ventile, Pumpen bzw. Mikropumpen, dichtschließende Mikropumpen, Mikromischer oder sonstigen Anschlüsse der erfindungsgemäßen Vorrichtung, die zum Befüllen des Kanalsystems dienen, als Fluidikanschlüsse bezeichnet.
Die erfindungsgemäße Aufgabevorrichtung kann für jede Art von planarem miniaturisiertem Analysensystem eingesetzt werden. Es kann sich dabei um Systeme für die Analytik handeln oder auch um Systeme, die zusätzlich Trenn- oder Derivatisierungseinheiten enthalten. Dem Fachmann sind entsprechende miniaturisierte Systeme bekannt.
Viskosität und lonenstärke der Probenlösung oder der zu verdrängenden Lösung, d.h. z.B. eines Transportpuffers, haben nur geringen Einfluß auf die Dosierung oder die Einfüllgeschwindigkeit. Es ist möglich, Suspensionen, Emulsionen, partikel- und zellhaltige Flüssigkeiten einzufüllen. Genauso unterliegt die Wahl des Materials zum Aufbau der Analysenvorrichtung, d.h. besonders die Beschaffenheit der Wände des Kanalsystems der erfindungsgemäßen Vorrichtung zur Probenaufgabe keiner Einschränkung. Auch Druckschwankungen, Pulsationen, Anlaufoder Stopeffekte während des Einbringens der Probe haben auf die Dosiergenauigkeit keinen Einfluß.
Die erfindungsgemäße Vorrichtung besitzt bezüglich des Aufgabevolumens systembedingt weite Grenzen. Das Volumen der Probeflüssigkeit, die injiziert werden kann, wird allein durch das Volumen des Kanalabschnitts
bestimmt, der sich zwischen den Öffnungen befindet. Durch Variation der geometrischen Abmessungen dieses Abschnitts bei dem Design des Kanalsystems der Analysevorrichtung lassen sich vorab an das analytische Problem angepasste Probenvolumina festlegen. Genauso ist es möglich, verschieden große Abschnitte parallel und/oder in Reihe zu implementieren, so daß das Volumen des durch die Probenlösung zu verdrängenden Abschnitts variiert werden kann. Bevorzugterweise wird deshalb ein Analysesystem zur Nutzung der erfindungsgemäßen Vorrichtung mit mehreren Kanalabschnitten unterschiedlicher Dimension versehen, die über jeweils unabhängige Fluidikanschlüsse für die Probenaufgabe verwendet werden können. Dadurch können Probenvolumina zwischen 0,01 μl und 100 μl , bevorzugt zwischen 0,05 und 30 μl in unterschiedlicher Abstufung je nach Bedarf injiziert werden. Dabei werden üblicherweise Variationskoeffizienten bei der Aufgabe von Probenvolumina ab 1 μl von etwa 5%, typischerweise unter 2% erreicht.
Auf diese Weise können quantitativ reproduzierbare und leicht handhabbare repräsentative Probenmengen eines flüssigen Analyten in jedes mikrostrukturierte System eingebracht werden. Besonders bevorzugt ist der Einsatz der erfindungsgemäßen Vorrichtung für die ITP, da damit die
Möglichkeit gegeben ist, kleinste Mengen von Analyten aus großen Probenvomunina anzureichern und zu separieren.
Abbildung 1 zeigt beispielhaft eine mögliche Anordnung des Kanalsystems der erfindungsgemäßen Aufgabevorrichtung. Das Kanalsystem ist in zwei Kanalabschnitte 1A und 1 B mit unterschiedlichem Volumen unterteilt. Daran angrenzend befindet sich der Trennkanal 1 C. Über die Fluidikanschlüsse 11 , 12 und 13 kann entweder der Kanalabschnitt 1A (Bei Öffnen der Anschlüsse 11 und 12) oder der Kanalabschnitt 1 B (Bei Befüllen über die Anschlüsse 12 und 13) oder die beiden Kanalabschnitte zusammen (bei Befüllen über die Anschlüsse 11 und 13) mit der Proben-
lösung gefüllt werden. Nach Befüllen der Aufgabeabschnitte wird durch Anlegen einer Spannung die Probe in Abschnitt 1 C aufgetrennt. Falls nur Abschnitt 1A mit der Probe befüllt wurde, kann auch Abschnitt 1 B als Trennstrecke genutzt werden, so daß die Trennstrecke bei Bedarf verlängert werden kann.
Abbildung 2 zeigt eine mögliche Vorgehensweise bei der Befüllung eines miniaturisierten Analysensystems. Dargestellt ist ein Kanalsystem bestehend aus drei Reservoiren R1 bis R3, den Kanalabschnitten K1 bis K4, den Fluidikanschlüssen F1 bis F6 und einer Verzweigungsstelle Vz.
Das in der Abbildung gezeigte System besitzt einen Kanalabschnitt K1 zur Probenaufgabe. Die Trennung kann entlang des Kanalabschnitts K2 und K3 oder K2 und K4 erfolgen. Zur Durchführung einer isotachophoretischen Trennung muß das System mit einer Probe und entsprechenden Puffern befüllt werden. Dabei muß das Probevolumen an einer Seite in Richtung der Trennstrecke mit einem Puffer (Leading-Puffer) und an der anderen Seite mit einem anderen Puffer (Terminating-Puffer) in Kontakt sein. Durch die Verzweigung Vz des Kanalsystems besteht die Möglichkeit, über die Reservoire R2 und R3 unterschiedliche Leading-Puffer einzufüllen. Das Ausschleusen von aufgetrennten Komponenten aus der Probe kann über den Fluidikanschluß F3 erfolgen.
Um die gewünschte Anordnung von Probe und Puffern im Kanalsystem zu erreichen, werden zunächst, wie unter A in der Abbildung schematisch dargestellt, die Fluidikanschlüsse F2 (Auslaß), F4, F5 und F6 (Einlasse) geöffnet und das Kanalsystem über die drei Reservoirs mit den beiden Leading-Puffern (über R2 und R3, schräg schraffiert bzw. gepunktet dargestellt) und dem Terminating-Puffer (über R1 , senkrecht gestreift dargestellt) gefüllt. Überschüssiger Puffer kann durch den Fluidikanschluß F2 austreten. Auf diese Weise füllt sich Kanalabschnitt K1 mit Terminating-
Puffer, der Abschnitt K3 mit Leading-Puffer (LE2) über R2, Abschnitt K4 mit Leading-Puffer (LE1 ) über R3 und Kanalabschnitt K2 enthält eine Mischung
der beiden Leading-Puffer. Die Fluidikanschlüsse F1 und F3 bleiben bei diesem Schritt geschlossen.
Der Kanalabschnitt K2 kann wahlweise mit Leading-Puffern über R2 oder R3 gefüllt werden. K2 stellt den ersten Abschnitt der Trennstrecke dar
In Teil B der Abbildung wird gezeigt, wie die Probe in den Kanalabschnitt K1 eingebracht wird und der Kanalabschnitt K2 mit einem Leading-Puffer über R3 gefüllt wird. Die Fluidikanschlüsse F5 und F6 sind geschlossen und es wird kein weiterer Terminating-Puffer über R1 sowie kein weiterer Leading-Puffer (LE2) über R2 gepumpt. Fluidikanschluß F4 ist geöffnet und Kanalabschnitt K2 wird mit Leading-Puffer (LE1) über R3 gefüllt. Gleichzeitig ist der Fluidikanschluß F1 geöffnet und die Probe wird über F1 zugeführt (dargestellt als Wellenlinien). Über den geöffneten Fluidik- anschluß F2 kann überschüssige Probe und überschüssiger Leading-Puffer (LE1 ) austreten. Dadurch, daß der Leading-Puffer (LE1) und das Probevolumen simultan gegeneinander gepumpt werden, wird eine besonders präzise Füllung der Kanalabschnitte K1 und K2 erreicht. Auf diese Weise ist es möglich auch mit Pumpen, die eine geringe Pulsation besitzen, eine exakte Befüllung vorzunehmen.
Nach Beendigung des Füllvorganges werden die Fluidikanschlüsse geschlossen. Man erhält so ein abgeschlossenes System ohne hydrodynamischen Fluß, in dem die Trennung reproduzierbar durchgeführt werden kann. Die Probe kann ganz oder in Fraktionen über die
Kanalabschnitte K2 und K3 oder über die Kanalabschnitte K2 und K4 getrennt werden. Sobald die Probe oder eine gewählte Fraktion durch den Kanalabschnitt K2 gewandert ist und an der Verzweigung Vz angekommen ist, kann entschieden werden, ob die Trennung weiter in Richtung K4 oder K3 geführt werden soll. Dies geschieht durch dauerndes oder zeitweises Umschalten der Anodenpotentials von F4 auf F6.
Die folgende Tabelle zeigt nochmals im Überblick die Schaltung der Fluidikanschlüsse während der einzelnen Schritte der Probenaufgabe:
Nach Beendigung des Füllvorganges werden die Fluidikanschlüsse (F1-F6) geschlossen.

Abbildung 3 zeigt eine Möglichkeit zur elektrokinetischen Probenaufgabe in miniaturisierten Analysensystemen nach dem Stand der Technik. Die Bilder A, B, C und D zeigen die einzelnen Schritte der Probenaufgabe. Bild A zeigt schematisch eine gekreuzte Kanalstruktur. An den Enden der Kanäle befinden sich die Elektroden E1 bis E4. Zunächst wird, wie in Bild B verdeutlicht, durch Anlegen einer Spannung zwischen Elektrode E1 (0 V) und E2 (+ 500 V) ein Kanal mit Probe befüllt. Anschließend werden, wie in Bild C gezeigt, die Elektroden an dem befüllten Kanal auf gleiches Potential geschaltet (z.B. E1 und E2 beide auf + 400 V) und eine Spannung an dem senkrecht dazu liegenden Trennkanalsystem angelegt (E3 = 0 V und E4 = + 2,5 kV). Auf diese Weise wird das Probenvolumen, das sich an der Schnittstelle der beiden Kanalsysteme befindet, in das Trennkanalsystem transportiert (Bild D). Das so erzeugte Probenvolumen liegt im Bereich einiger Nanoliter oder weniger.
Abbildung 4 zeigt eine Möglichkeit zur Probenaufgabe in makroskopischen Analysensystemen, wie beispielsweise dem Isotachophoresegerät ItaChrom® EA 101 der Firma I + M, Analytische Meß- und Regeltechnik, Deutschland. Die Bilder A1/A2, B1/B2 und C1/C2 zeigen die unterschiedlichen Stufen der Probenaufgabe, wobei die Bilder A1 , B1 und C1 eine Seitenansicht der Aufgabevorrichtung zeigen, die Bilder A2, B2 und C2 eine Ansicht von oben. Diese mechanische Vorrichtung zur Probenaufgabe besteht aus einem Hahn K, der von einer Ummantelung U umgeben ist. Sowohl die Ummantelung U, wie auch der Hahn K sind mehrfach von Kanälen durchbrochen. Der Hahn K kann in der Ummantelung U derart gedreht werden, daß jeweils bestimmte Kanäle in Hahn und Ummantelung verbunden werden und so Flüssigkeiten aus Vorratsgefäßen durch die gezeigte Vorrichtung definiert in das angeschlossene Isotachophoresegerät gelangen. Vorratsbehälter und das ITP-Gerät sind in der Abbildung nicht gezeigt, sondern lediglich durch Pfeile angedeutet. In den Abbildungen A1/A2 ist der Hahn derart gedreht, daß eine Verbindung der Kanalstücke 3,
4 und 5, sowie 2 und 6 besteht. Dadurch wird Kanalstück 5 im inneren des Hahns mit Probelösung aus einem Vorratsgefäß gefüllt, das mit Kanal 3 verbunden ist. Außerdem wird über ein Vorratsgefäß an Kanal 2 das Kanalsystem des Isotachophoresegeräts mit einem der beiden für eine ITP notwendigen Trennpuffer (Puffer 1) gefüllt.
In einem zweiten Schritt (Bild B1/B2) wird der Hahn K so gedreht, daß die in Bild A1/A2 bestandenen Kanalverbindungen unterbrochen werden. Statt dessen wird eine Verbindung der Kanalstücke 1 und 7 hergestellt. Auf diese Weise wird das hinter der Aufgabevorrichtung liegende Kanalsystem mit einem zweiten Puffer (Puffer 2) gefüllt. In Bild C1/C2 wird schließlich der Hahn K erneut gedreht, so daß eine Verbindung der Kanalstücke 1 , 5 und 2 entsteht. Kanal 2 ist mit Puffer 1 gefüllt, Kanal 5 mit der Probenlösung und Kanal 1 mit Puffer 2. Auf diese Weise ist ein durch die Abmessungen von Kanal 5 definiertes Volumen der Probenlösung zwischen den für die ITP notwendigen zwei Puffern eingebettet. Durch Anlegen einer Spannung kann nun die Trennung begonnen werden.
Auch ohne weitere Ausführungen wird davon ausgegangen, daß ein Fachmann die obige Beschreibung im weitesten Umfang nutzen kann. Die bevorzugten Ausführungsformen und Beispiele sind deswegen lediglich als beschreibende, keineswegs als in irgendeiner Weise limitierende Offenbarung aufzufassen.
Die vollständige Offenbarung aller vor- und nachstehend aufgeführten Anmeldungen, Patente und Veröffentlichungen, sowie der korrespondierenden Anmeldung DE 199 27 534, eingereicht am 16.06.1999, ist durch Bezugnahme in diese Anmeldung eingeführt.
Claims
1. Vorrichtung zur Aufgabe definierter Probenvolumina über 0,01 μl für miniaturisierte Analysensysteme umfassend mindestens einen Kanalabschnitt, an dessen Enden jeweils mindestens ein
Fluidikanschluß vorhanden ist.
2. Vorrichtung nach Anspruch 1 , dadurch gekennzeichnet, daß das
Probenvolumen zwischen 0,05 und 30 μl beträgt.
3. Vorrichtung nach Anspruch 1 oder 2, dadurch gekennzeichnet, daß das Kanalsystem mindestens zwei hintereinanderliegende Kanalabschnitte enthält, die jeweils von Fluidikanschlüssen begrenzt werden.
4. Vorrichtung nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, daß das Kanalsystem mindestens zwei parallele Kanalabschnitte enthält, die unabhängig voneinander von Fluidikanschlüssen begrenzt werden.
5. Vorrichtung nach einem der Ansprüche 1 bis 4, dadurch gekennzeichnet, daß als Fluidikanschlüsse dichtschließende Mikropumpen dienen.
6. Vorrichtung nach einem der Ansprüche 1 bis 4, dadurch gekennzeichnet, daß als Fluidikanschlüsse Mikromischer, Ventile und
Mikropumpen dienen.
