Description
- Die Erfindung bezieht sich auf eine Durchlaufzentrifuge, vorzugsweise zur industriellen Produktion von Proteinen aus menschlichem Blutplasma, mit einer zylindrischen, mit mehreren konzentrisch angeordneten, im Bodenbereich untereinander verbundenen Kammern versehenen Schleudertrommel, in deren Innenbereich das Blutplasma oder dergleichen zugeführt und aus deren Außenbereich eine flüssige Phase kontinuierlich abgezogen wird.
- Es ist eine Durchlaufzentrifuge dieser Art bekannt (DE-C 2 423 319), bei der die Schleudertrommel zwei konzentrisch angeordnete Kammern aufweist, die im Bodenbereich durch Kanäle verbunden sind, durch die Blutplasma o.dgl. aus der Innenkammer in die Außenkammer fließen kann. Aus dieser Außenkammer fließt die flüssige Phase in eine Schälkammer und wird über eine Schälscheibe nach außen geführt. Die von der flüssigen Phase abgetrennten Plasmaproteine (Eiweiß) setzen sich in den beiden Kammern der Schleudertrommel ab. Nachdem diese Kammern gefüllt sind, muß das Eiweiß vom Bedienungspersonal manuell aus diesen Kammern her ausgeschabt werden. Hierbei ist zu beachten, daß weder die Plasmaproteine bei diesem Vorgang vom Bedienungspersonal infiziert werden dürfen, noch daß das Bedienungspersonal mit in den Plasmaproteinen enthaltenen Krankheitskeimen in Berührung kommen darf.
- Da die Schleudertrommel der bekannten Durchlaufzentrifuge sich ausschließlich aus hochbelastbaren Metallteilen zusammensetzt, kann diese Zentrifuge mit der maximalen Drehzahl betrieben werden, die das verwendete Zentrifugenmaterial erlaubt. Zur wirtschaftlichen Gewinnung der Plasmaproteine sind hochtourige Zentrifugen erforderlich.
- Der Erfindung liegt die Aufgabe zugrunde, eine Durchlaufzentrifuge, vorzugsweise zur industriellen Produktion von Proteinen aus menschlichem Blutplasma, der eingangs genannten Art so zu gestalten, daß unter Aufrechterhaltung eines wirtschaftlichen Betriebs mit maximalen Drehzahlen das Produkt vor Kontaminationen und das Bedienungspersonal von im Produkt enthaltenen Krankheitskeimen geschützt werden sowie der Arbeitsaufwand bei der Entnahme und Lagerung der abgeschleuderten Proteine verringert wird.
- Diese Aufgabe wird nach der Erfindung dadurch gelöst, daß in der Schleudertrommel konzentrisch zueinander angeordnete, im Bodenbereich durch mindestens einen Kanal miteinander verbundene ringförmige oder ringsegmentförmige Kammern bildende Kunststoffbehälter angeordnet sind, von denen der oder die der Drehachse der Schleudertrommel zugewandt liegenden Kunststoffbehälter einen Zulaufstutzen und der oder die im Außenbereich der Schleudertrommel vorgesehenen Kunststoffbehälter einen obenliegenden Ablaufkanal aufweisen,und daß die Außenflächen der Kunststoffbehälter, des oder der Verbindungskanäle, des oder der Ablaufkanäle und des oder der Zuflußstutzen sich an Innenflächen der Schleudertrommel und an die Schleudertrommel eingesetzten und demontierbaren Formteilen abstützen.
- Da den Außenflächen der in die Schleudertrommel eingesetzten Kunststoffbehältern über den gesamten Flächenbereich Stützflächen zugeordnet sind, die durch Teile der Schleudertrommel durch eingesetzte und demontierbare Formteile oder durch Wandungen benachbarter Kunststoffbehälter gebildet werden können, sind von den Wandungen der Kunststoffbehälter während des Zentrifugalbetriebs nur Kräfte auf Stützflächen zu übertragen, so daß auch bei hochtourigen Zentrifugen als Kunststoffbehälter Kunststoffbeutel verwendet werden können.
- Die Kunststoffbehälter werden steril angeliefert. Zur Aufrechterhaltung der Sterilität der Innenräume der Behälter werden die Zulauföffnungen und die Ablauföffnungen durch Stopfen verschlossen. Dieser Stopfenverschluß bleibt auch beim Einsetzen der Behälter in die Schleudertrommel und bei der Montage der Formteile und ggf. der Zwischenwände erhalten. Erst kurz vor dem Aufsetzen des Verteilers, der die Schälkammer für die abzuführende flüssige Phase begrenzenden Teile und des Trommeldeckels werden die Stopfen entfernt.
- Nachdem die Innenräume der Kunststoffbehälter mit Proteinen gefüllt sind, wird die Schleudertrommel angehalten und es werden nach dem Lösen des Trommeldeckels und dem Entfernen der Trommelteile im Zulauf- und Ablaufbereich sowie einiger Formteile und Zwischenwände die gefüllten Kunststoffbehälter aus der Schleudertrommel entfernt und ggf. gelagert. Für die Entnahme der Plasmaproteine aus den Kunststoffbehältern müssen die Kunststoffbehälter zerstört werden.
- Bei der erfindungsgemäßen Zentrifuge werden Produktverlustevermieden. Eine Reinigung der Trommeleinsätze zwischen den einzelnen Chargen ist nicht mehr erforderlich. Eine Trommelauskleidung aus Sondermaterialien, z.B. aus Titan, entfällt.
- Weitere Kennzeichen und Merkmale der Erfindung ergeben sich aus den Unteransprüchen und der folgenden Beschreibung eines Ausführungsbeispiels, das in den Zeichnungen dargestellt ist.
- Es zeigen:
- Fig. 1 die Schleudertrommel einer Durchlaufzentrifuge mit eingesetzten Kunststoffbehältern im Vertikalschnitt, und zwar im Bereich eines unteren Verbindungskanals und eines oberen Ablaufkanals,
- Fig. 2 eine Schleudertrommel mit eingesetzten Kunststoffbehältern im Vertikalschnitt, der versetzt zu dem Verbindungskanal und zu dem Ablaufkanal gelegt wurde,
- Fig. 3 einen Montagezwischenzustand,
- Fig. 4 einen Schnitt nach der Linie IV-IV in Fig. 3,
- Fig. 5 einen Schnitt nach der Linie V-V in Fig. 1 und
- Fig. 6 einen Schnitt nach der Linie VI-VI in Fig. 3,
- Fig. 7 einen Schnitt nach der Linie VII-VII in Fig. 3.
- Die Durchlaufzentrifuge weist eine Schleudertrommel 1 auf, die mit einem zylindrischen Unterteil 2, einem Deckel 3 und einem Verschlußring 4 versehen ist, durch den der Deckel gegenüber dem Unterteil festgelegt wird.
- In dem Unterteil 2 befindet sich ein Kühlmantel 5, der von Kühlflüssigkeit durchströmt wird, auf dessen Funktion hier aber nicht weiter eingegangen wird. In dem in Fig. 1 dargestellten Ausführungsbeispiel fluchtet seine Innenfläche 6 mit der Innenfläche 7 des Deckels.
- Der Füllraum der Schleudertrommel wird zur Drehachse 8 hin und nach unten durch ein Einsatzteil 9 begrenzt, dessen untere Bodenplatte 10 aus den einen stumpfen Winkel bildenden Plattenteilen 11 und 12 besteht.
- In der in der Fig. 3 dargestellten Montagezwischenstellung sind auf der Bodenplatte 10 des Einsatzteiles 9 die Kunststoffbehälter 13 und 14 angeordnet, die über einen Kanal 15 im Bodenbereich untereinander verbunden sind.
- Die konstruktive Ausbildung des Kanals 15 kann man aus der Fig. 4 entnehmen. Der Verbindungskanal 15 ist als Schlauch ausgebildet und über eine Rippe 16 mit dem Boden 17 des Kunststoffbehälters 13 verbunden. Der Kanal 15, die Rippe 16 und der Boden 17 des Behälters 13 sind einstückig ausgebildet. Der Kanal 15 ist ferner mit dem Kunststoffbehälter 14 verbunden, so daß die beiden konzentrisch angeordneten Behälter 13, 14 eine Baueinheit bilden.
- Der Behälter 13 ist mit einem Zulaufstutzen 18 ausgerüstet. Der Kunststoffbehälter 14 weist einen obenliegenden Ablaufkanal 19 auf, der mit einem Ablaufstutzen 20 ausgerüstet ist. Der Zulaufstutzen 18 und der Ablaufstutzen 20 sind bei der Anlieferung und bei dem Einbau der Kunststoffbehälter in die Schleudertrommel durch Stopfen 21,22 verschlossen. Hierdurch wird verhindert, daß irgendwelche Verunreinigungen in den Innenraum der Kunststoffbehälter gelangen.
- Bei dem in den Zeichnungen dargestellen Ausführungsbeispiel ist zwischen dem Kunststoffbehälter 13 und dem Kunststoffbehälter 14 ein Ringmantel 23 angeordnet, der Stützflächen für die benachbart liegenden Wandungen der genannten Behälter bildet. Der Ringmantel 23 besitzt Ausnehmungen, durch die der Verbindungskanal 15 und der Ablaufkanal 19 hindurchführen.
- Unterhalb des Bodens 17 des Innenbehälters 13 bzw. der Innenbehälter ist ein Formteil 24, vorzugsweise aus Kunststoff gefertigt, angeordnet, das sich an der Bodenplatte 10 des Einsatzteils 9 abstützt und den Raum zwischen dem Boden 17 des oder der Innenbehälter und der Bodenplatte 10 ausfüllt. Dieses Formteil weist eine oder mehrere Ausnehmungen zur Aufnahme jeweils eines Verbindungskanals 15 auf. Damit die gesamte Außenfläche des durch einen Schlauch gebildeten Verbindungskanals 15 sich an Formteilflächen während des Zentrifugenbetriebes abstützen kann, werden in die Ausnehmung 25 des Formteils 24 weitere Formteile 26 eingesetzt.
- Aus der Fig. 4 kann man entnehmen, daß die gesamte Oberfläche des Verbindungskanals 15 in Formteilen eingebettet ist.
- Auch der Ablaufkanal 19 bzw. die Ablaufkanäle 19 werden in Formteilen 27,28 eingebettet, die zwisehen dem Oberboden 29 des innenliegenden Kunststoffbehälters 13 bzw. zwischen den Oberböden 29 der innenliegenden Kunststoffbehälter und dem Deckel 3 angeordnet werden.
- Die Zuordnung dieser Formteile 27,28 zu dem Ablaufkanal 19 ergeben sich aus den Fig. 1 und 5.
- Aus der Fig. 6 ergibt sich, daß die konzentrisch angeordneten Kunststoffbehälter 13 und 14 ringsegmentförmig ausgebildet sein können, so daß durch eine Reihe dieser Kunststoffbehälter der Innenraum der Schleudertrommel ausgefüllt wird. Durch die Verwendung kleinerer, ringsegmentförmiger Kunststoffbehälter 13 und 14 wird die Herstellung der Kunststoffbehälter im Blasverfahren ermöglicht.
- Bei dem Ausführungsbeispiel nach der Fig. 6 stützen sich während des Betriebs benachbarte, radial verlaufende Wandungen der Kunststoffbehälter aneinander ab.
- Die Durchlaufzentrifuge ist mit einem Verteilerraum 30 ausgerüstet, dem das Blutplasma über ein ortsfestes Einlaufrohr 31 zugeführt wird. Dem Verteilerraum 30 ist eine Sammelkammer 32 und ein Füllstutzen 33 zugeordnet, die in die Zulaufstutzen 18 der Kunststoffbehälter 13 eingreifen. In den Ablaufstutzen 20 des Ablaufkanals 19 greift ein Stutzen 34 ein, durch den die abgezogene flüssige Phase in eine Schälkammer 35 gelangt.
- In dem dargestellten Ausführungsbeispiel (Fig. 2) ist die Bodenplatte 10 mit einem Ringsteg 36 versehen, auf dem sich der Ringmantel 23 abstützt. Der Ringsteg 36 weist im Bereich der Verbindungskanäle 15 Ausnehmungen 37 auf, die mit Ausnehmungen 38 in dem Ringmantel 23 fluchten. Die Ausnehmungen sind mit einem Formteil 39 ausgefüllt, welches eine Baueinheit mit den Kunststoffbehältern 13,14 und dem Verbindungskanal 15 bildet.
