Abstract

Meetod ja testsüsteem B-kompleksi vitamiinide aktiivsuse samaaegseks kvantitatiivseks määramiseks on viie B-kompleksi vitamiini (B1, B2, B3, B5 ja B6) absoluutse aktiivsuse samaaegseks määramiseks toidus, toidu- ja biolisandites ning söödas, kasutades LC-MS-isotoop-sisestandardi-meetodit. Testsüsteemi lahendus baseerub ekstraktsiooniviaali lüofiliseeritud hüdrolüütilist aktiivsust omavate ensüümide ja isotooprikastusega B-kompleksi vitamiinide aktiivsust omavate vitameeride sisestandardite kasutamisel nii vastavalt proovis sisalduvate vitameeride ja kofaktorite viimiseks sisestandarditega samasse keemilisse vormi kui ka sellele järgnevaks kvantifitseerimiseks LC-MS- või LC-MS/MS-meetodil. Meetodi ja testsüsteemi kasutamine võimaldab minimeerida proovide ettevalmistamiseks vajalikku aega ja töömahtu ning analüüsida samaaegselt ja suure läbilaskvusega viie B-kompleksi vitamiini (B1, B2, B3, B5 ja B6) aktiivsust toidus ja toidulisandites, aga ka ravimpreparaatides ning muudes bioloogilistes proovides.
