Claims (12)
1. Diagnosticeringsfremgangsmåde baseret på Next Generation Sekventering omfattende at ekstrahere nukleinsyremateriale fra en prøve og mindst en kontrolprøve, hvor kontrolprøven forberedes inden ekstraktion, endvidere omfattende at underkaste det ekstraherede nukleinsyremateriale for en Next Generation Sekventerings-fremgangsmåde, hvor kontrolprøven omfatter prøvemateriale suppleret med en nukleinsyrekontrol, hvor nukleinsyrekontrollen omfatter mindst en nukleinsyrekontrolsekvens er der forskellig fra målnukleinsyresekvensen, og hvor kontrollen omfatter mindst en vildtype-nukleinsyresekvens og mindst en mutant af vildtype-nukleinsyresekvensen.
2. Diagnosticeringsfremgangsmåde baseret på Next Generation Sekventering ifølge krav 1, hvor nukleinsyrekontrolsekvensen er valgt fra ikke-humane afledte nukleinsyrer, eller valgt fra virale nukleinsyrer.
3. Diagnosticeringsfremgangsmåde baseret på Next Generation Sekventering ifølge et hvilket som helst af kravene 1 og 2, hvor nukleinsyrekontrolsekvensen er valgt fra nukleinsyrer, der er afledt fra tobakmosaikvirus.
4. Diagnosticeringsfremgangsmåde baseret på Next Generation Sekventering ifølge et hvilket som helst af kravene 1 til 3, omfattende mindst to nukleinsyrekontroller som defineret i et hvilket som helst af kravene 1 til 3 i et forudbestemt forhold.
5. Diagnosticeringsfremgangsmåde baseret på Next Generation Sekventering ifølge et hvilket som helst af kravene 1 til 4, hvor fremgangsmåden er egnet til detekteringen af tilstedeværelse eller fraværet af sekvenser afledt fra et infektiøs agens eller af et onkogen.
6. Diagnosticeringsfremgangsmåde baseret på Next Generation Sekventering ifølge et hvilket som helst af kravene 1 til 5, hvor tilstedeværelsen eller fraværet af en målsekvens afledt fra et infektiøs agens eller af et onkogen og tilstedeværelsen af mindst en nukleinsyrekontrolsekvens efter sekventeringsreaktionen er tegn på fuldført nukleinsyreekstraktion fra prøverne.
7. Diagnosticeringsfremgangsmåde baseret på Next Generation Sekventering ifølge et hvilket som helst af kravene 1 til 6, endvidere omfattende en kontamineringskontrol.
8. Diagnosticeringsfremgangsmåde baseret på Next Generation Sekventering ifølge et hvilket helst af kravene 1 til 7, hvor kontamineringskontrollen omfatter mindst en nukleinsyrekontrol som defineret i kravene 1 til 3, hvor kontamineringskontrollen ikke omfatter prøvemateriale.
9. Diagnosticeringsfremgangsmåde baseret på Next Generation Sekventering ifølge et hvilket som helst af kravene 1 til 8, omfattende at underkaste kontamineringskontrollen for en målnukleinsyre-specifik sekventeringsreaktion.
10. Diagnosticeringsfremgangsmåde baseret på Next Generation Sekventering ifølge et hvilket som helst af kravene 1 til 9, omfattende at underkaste kontamineringskontrollen for en målnukleinsyre-specifik sekventeringsreaktion, hvor fraværet af detektering af en målnukleinsyre-specifik sekvens i kontamineringskontrollen og detekteringen af tilstedeværelsen afen kontrolnukleinsyre-specifik sekvens er tegn på fraværet af kontaminering.
11. Diagnosticeringsfremgangsmåde baseret på Next Generation Sekventering eller målnukleinsyredetekterings-fremgangsmåde baseret på Next Generation Sekventering ifølge et hvilket som helst af kravene 1 til 10, hvor kontrolnukleinsyren er til stede i en mængde på 0,1 til 10 fg efter ekstraktion af nukleinsyrerne fra prøverne.
12. Diagnosticeringsfremgangsmåde baseret på Next Generation Sekventering eller målnukleinsyredetekterings-fremgangsmåde baseret på Next Generation Sekventering ifølge et hvilket som helst af kravene 1 til 11, hvor prøven er en klinisk prøve afledt fra et menneske, og hvor kontrolnukleinsyren er et plasmid omfattende vildtype- og/eller mutantsekvenser af en ikke-human virus, valgt fra dyrevirusser, der ikke smitter mennesker, plantevirusser, eller hvor kontrolnukleinsyren er blevet kunstigt fremstillet og ikke har nogen naturlig ækvivalent.
