Claims (15)
1. Fremgangsmåde in vitro til modificering af et genom ved et genomisk locus af interesse i en muse-embryostam- (ES) celle, der omfatter: etablering af kontakt mellem muse-ES-cellen og et Cas9-protein, et CRISPR-RNA, der hybridiserer til en målsekvens ved det genomiske locus af interesse, et tracrRNA og en stor målvektor (LTVEC), der er mindst 10 kb i størrelse og omfatter en indsætningsnukleinsyre flankeret af: (i) en 5’-homologiarm, der er homolog med en 5’-målsekvens ved det genomiske locus af interesse; og (ii) en 3’-homologiarm, der er homolog med en 3’-målsekvens ved det genomiske locus af interesse, hvor, efter etablering af kontakt mellem muse-ES-cellen og Cas9-proteinet, CRISPR-RNA’et og tracrRNA’et i tilstedeværelse af LTVEC’et, genomet af muse-ES-cellen modificeres til at omfatte en målrettet genetisk modifikation, der omfatter deletion af et område af det genomiske locus af interesse, hvor deletionen er mindst 30 kb, og/eller indsætning af indsætningsnukleinsyren ved det genomiske locus af interesse, hvor indsætningen er mindst 30 kb.
2. Fremgangsmåde ifølge krav 1, hvor: (a) CRISPR-RNA’et og tracrRNA’et indføres som et enkelt transskript, der omfatter CRISPR-RNA’et og tracrRNA’et, og eventuelt hvor det enkelte transskript omfatter CRISPR-RNA’et og tracrRNA’et fusioneret i form af et enkelt guide-RNA (sgRNA); eller (b) CRISPR-RNA’et og tracrRNA’et indføres separat.
3. Fremgangsmåde ifølge et hvilket som helst foregående krav, hvor: (a) Cas9-proteinet indføres i form af et protein, et messenger-RNA (mRNA), der koder for Cas9-proteinet, eller et DNA, der koder for Cas9-proteinet; (b) CRISPR-RNA’et indføres i form af et RNA eller et DNA, der koder for CRISPR-RNA’et; og (c) tracrRNA’et indføres i form af et RNA eller et DNA, der koder for tracrRNA’et.
4. Fremgangsmåde ifølge krav 3, hvor Cas9-proteinet, CRISPR-RNA’et og tracrRNA’et indføres som et protein-RNA-kompleks.
5. Fremgangsmåde ifølge krav 3, hvor: (a) DNA’et, der koder for Cas9-proteinet, er i form af et første ekspressionskonstrukt, der omfatter en første promoter operativt bundet til en første nukleinsyre, der koder for Cas9-proteinet; (b) DNA’et, der koder for CRISPR-RNA’et, er i form af et andet ekspressionskonstrukt, der omfatter en anden promoter operativt bundet til en anden nukleinsyre, der koder for CRISPR-RNA’et; og (c) DNA’et, der koder for tracrRNA’et, er i form af et tredje ekspressionskonstrukt, der omfatter en tredje promoter operativt bundet til en tredje nukleinsyre, der koder for tracrRNA’et; hvor den første, anden og tredje promoter er aktive i muse-ES-cellen, og hvor det første, andet og tredje ekspressionskonstrukt er på et enkelt nukleinsyremolekyle eller på flere nukleinsyremolekyler.
6. Fremgangsmåde ifølge krav 3, hvor: (a) DNA’et, der koder for Cas9-proteinet, er i form af et første ekspressionskonstrukt, der omfatter en første promoter operativt bundet til en første nukleinsyre, der koder for Cas9-proteinet; og (b) DNA’et, der koder for CRISPR-RNA’et og DNA’et, der koder for tracrRNA’et er i form af et andet ekspressionskonstrukt, der omfatter en anden promoter operativt bundet til en anden nukleinsyre, der koder for et gRNA omfattende CRISPR-RNA’et og tracrRNA’et; hvor den første og den anden promoter er aktive i muse-ES-cellen, og hvor det første og det andet ekspressionskonstrukt er på et enkelt nukleinsyremolekyle eller på separate nukleinsyremolekyler.
7. Fremgangsmåde ifølge et hvilket som helst foregående krav, hvor (a) den målrettede genetiske modifikation omfatter simultan deletion af en endogen nukleinsyresekvens ved det genomiske locus af interesse og indsætning af indsætningsnukleinsyren ved det genomiske locus af interesse, eventuelt hvor den deleterede endogene nukleinsyresekvens er fra 30 kb til ca. 110 kb, og indsætningsnukleinsyren er fra ca. 40 kb til ca. 140 kb; (b) den målrettede genetiske modifikation er en biallel genetisk modifikation, og eventuelt hvor den biallelle genetiske modifikation omfatter deletion af en endogen nukleinsyresekvens og indsætning af indsætningsnukleinsyren ved det genomiske locus af interesse i to homologe kromosomer; eller (c) hvor den modificerede muse-ES-celle er forbundet heterozygot eller hemizygot ved det genomiske locus af interesse, og eventuelt hvor den målrettede genetiske modifikation ved det genomiske locus af interesse i ét kromosom omfatter deletion af en endogen nukleinsyresekvens og indsætning af indsætningsnukleinsyren, og eventuelt hvor den målrettede genetiske modifikation omfatter: (1) deletion af en endogen nukleinsyresekvens ved det genomiske locus af interesse i første og andet homologe kromosom; og (2) indsætning af indsætningsnukleinsyren i det genomiske locus af interesse i det første homologe kromosom og disruption af det genomiske locus af interesse i det andet homologe kromosom.
8. Fremgangsmåde ifølge et hvilket som helst foregående krav, hvor LTVEC er mindst 40 kb, mindst 50 kb, mindst 60 kb, mindst 70 kb, mindst 80 kb, mindst 90 kb, mindst 100 kb, mindst 150 kb eller mindst 200 kb.
9. Fremgangsmåde ifølge et hvilket som helst foregående krav, hvor målrettet genetisk modifikation omfatter indsætning af indsætningsnukleinsyren, hvor indsætningsnukleinsyren er mindst 40 kb, mindst 50 kb, mindst 60 kb, mindst 70 kb, mindst 80 kb, mindst 90 kb, mindst 100 kb, mindst 150 kb, mindst 200 kb, mindst 250 kb, mindst 300 kb eller fra ca. 40 kb til ca. 140 kb; eller hvor den målrettede genetiske modifikation omfatter indsætning af indsætningsnukleinsyren og deletion af et område af det genomiske locus af interesse, hvor deletionen er mindst 30 kb, og indsætningsnukleinsyren er mindst 10 kb, mindst 20 kb, mindst 30 kb, mindst 40 kb, mindst 50 kb, mindst 60 kb, mindst 70 kb, mindst 80 kb, mindst 90 kb, mindst 100 kb, mindst 150 kb, mindst 200 kb, mindst 250 kb eller mindst 300kb.
10. Fremgangsmåde ifølge et hvilket som helst foregående krav, hvor målsekvensen er umiddelbart flankeret af en PAM- (Protospacer Adjacent Motif) sekvens.

Fremgangsmåde ifølge et hvilket som helst foregående krav, hvor den målrettede genetiske modifikation omfatter deletion af et område af det genomiske locus af interesse, hvor deletionen er mindst 30 kb og indsætning af indsætningsnukleinsyren ved det genomiske locus af interesse, hvor indsætningen er mindst 30 kb.
