Claims (20)
1. Isoleret polypeptid med proteaseaktivitet, som er valgt fra gruppen bestående af: (a) et polypeptid, som udviser mindst 84% sekvensidentitet med polypeptidet ifølge SEQ ID NO: 5; (b) et polypeptid, der kodes for af et polynukleotid, som udviser mindst 84% sekvensidentitet med det modne polypeptids kodningssekvens ifølge SEQ ID NO: 1; (c) en variant af polypeptidet ifølge SEQ ID NO: 5, som omfatter en substitution, deletion og/eller insertion i en eller flere positioner, hvor det samlede antal aminosyresubstitutioner, -deletioner og/eller -insertioner ikke er over 20; og (d) et fragment af et polypeptid ifølge (a), (b) eller (c), som har proteaseaktivitet, hvor polypeptidet har forbedret proteaseaktivitet mellem pH 3 og 4 ved 25°C i sammenligning med proteasen med sekvensen ifølge SEQ ID NO: 8 for stammen NRRL 18262.
2. Polypeptid ifølge krav 1, som omfatter eller består af SEQ ID NO: 2, SEQ ID NO: 4, SEQ ID NO: 5 eller SEQ ID NO: 6.
3. Sammensætning, som omfatter polypeptidet ifølge et hvilket som helst af kravene 1-2.
4. Sammensætning ifølge krav 3, som foreligger i form af et granulat eller et mikrogranulat.
5. Isoleret polynukleotid, som koder for polypeptidet ifølge et hvilket som helst af kravene 1-2.
6. Nukleinsyrekonstruktion eller ekspressionsvektor, som omfatter polynukleotidet ifølge krav 5, hvilket polynukleotid er operativt forbundet til en eller flere kontrolsekvenser, der styrer fremstillingen af polypeptidet i en ekspressionsværtscelle.
7. Rekombinant ekspressionsværtscelle, som omfatter polynukleotidet ifølge krav 5, hvilket polynukleotid er operativt forbundet til en eller flere kontrolsekvenser, der styrer fremstillingen af polypeptidet, hvor værten er valgt fra gruppen bestående af Acremonium, Aspergillus, Aureobasidium, Bjerkandera, Ceriporiopsis, Chrysosporium, Coprinus, Coriolus, Cryptococcus, Filibasidium, Fusarium, Humicola, Magnaporthe, Mucor, Myceliophthora, Neocallimastix, Neurospora, Paecilomyces, Penicillium, Phanerochaete, Phlebia, Piromyces, Pleurotus, Schizophyllum, Talaromyces, Thermoascus, Thielavia, Tolypocladium, Trametes og Trichoderma, eller hvor værten er en gær, såsom Pichia eller Saccharomyces, eller en Bacillus, såsom Bacillus alkalophilus, Bacillus amyloliquefaciens, Bacillus brevis, Bacillus circulans, Bacillus clausii, Bacillus coagulans, Bacillus firmus, Geobacillus stearothermophilus, Bacillus lautus, Bacillus lentus, Bacillus licheniformis, Bacillus megaterium, Bacillus pumilus, Bacillus stearothermophilus, Bacillus subtilis eller Bacillus thuringiensis.
8. Værtscelle ifølge krav 7, hvor værten er en Aspergillus, såsom Aspergillus awamori, Aspergillus foetidus, Aspergillus japonicus, Aspergillus nidulans, Aspergillus niger eller Aspergillus oryzae.
9. Fremgangsmåde til fremstilling af polypeptidet ifølge et hvilket som helst af kravene 1 -2, hvilken fremgangsmåde omfatter: (a) dyrkning af en celle, som i sin vildtypeform fremstiller polypeptidet, under betingelser, som er befordrende for fremstilling af polypeptidet; og (b) indvinding af polypeptidet.
10. Fremgangsmåde til fremstilling af polypeptidet ifølge et hvilket som helst af kravene 1-2, hvilken fremgangsmåde omfatter: (a) dyrkning af en værtscelle ifølge et hvilket som helst af kravene 7-8 under betingelser, som er befordrende for fremstilling af polypeptidet; og (b) indvinding af polypeptidet.
11. Dyrefodersammensætning, som omfatter polypeptidet ifølge et hvilket som helst af kravene 1 -2.
12. Dyrefodersammensætning ifølge krav 11, som endvidere omfatter en eller flere amylaser, phytaser, xylanaser, galactanaser, alfa-galactosidaser, proteaser, phospholipaser, beta-glucanaser eller en hvilken som helst blanding deraf.
13. Dyrefodersammensætning ifølge krav 12, som endvidere omfatter mindst ét probiotikum eller en mikrobe til direkte tilførsel (DFM - direct fed microbial).
14. Anvendelse af mindst ét polypeptid ifølge et hvilket som helst af kravene 1-2 i dyrefoder; i dyrefoderadditiver; ved fremstilling af en sammensætning til anvendelse i dyrefoder; til forbedring af et dyrefoders næringsværdi; til forøgelse af fordøjeligt og/eller opløseligt protein i dyrefoder; til forøgelse af hydrolysegraden for proteiner i dyrefoder og/eller til behandling af proteiner.
15. Fremgangsmåde til forbedring af et dyrefoders næringsværdi, hvor mindst ét polypeptid ifølge et hvilket som helst af kravene 1-2 tilsættes til foderet.
16. Dyrefoderadditiv, som omfatter mindst ét polypeptid ifølge et hvilket som helst af kravene 1-2; og mindst ét fedtopløseligt vitamin; og/eller mindst ét vandopløseligt vitamin og/eller mindst ét spormineral.
17. Dyrefoderadditiv ifølge krav 16, som endvidere omfatter en eller flere amylaser, phytaser, xylanaser, galactanaser, alfa-galactosidaser, proteaser, phospholipaser, beta-glucanaser eller en hvilken som helst blanding deraf.
18. Dyrefoder, som har et råproteinindhold på 50 til 800 g/kg og omfatter mindst ét polypeptid ifølge et hvilket som helst af kravene 1-2.
19. Fremgangsmåde til behandling af proteiner, hvilken fremgangsmåde omfatter trinet til tilsætning af mindst ét polypeptid ifølge et hvilket som helst af kravene 1-2 til mindst ét/én protein eller proteinkilde.
20. Fremgangsmåde ifølge krav 19, hvor sojabønne eller sojabønnemel er indbefattet blandt den mindst ene proteinkilde.
