Claims (9)

1. Fremgangsmåde til at bestemme et eller flere interaktionsparametre for interaktionen mellem en analyt og en ligand ved anvendelse af en biosensor, som omfatter trinnene at: A: at tilvejebringe en sensoroverflade som har liganden immobiliseret derpå, B: at bringe sensoroverfladen i kontakt med en styringsanalyt, C: at registrere sensorresponsen fra binding af styringsanalytten til ligandens bindingssteder, D: at bestemme styremæthedsresponsen (Rmaxc) for interaktionen mellem styringsanalytten og liganden, E: at transformere styremæthedsresponsen (Rmaxc) til en analytmæthedsrespons (RmaxA) ved anvendelse af det relative molære responsbidrag af analytten og styringsanalytten. F: at bringe i kontakt sensoroverfladen med en eller flere prøversom indeholder henholdsvis en koncentration eller forskellige koncentrationer af analytten, G: at registrere sensorresponsen fra binding af analytten til bindingsstederne, og H: at tilpasse det registrerede sensorrespons til en forudbestemt interaktionsmodel ved anvendelse af analytmæthedsresponsen (R max A) og analytkoncentrationen eller koncentrationer for at bestemme interaktionsparametre.
2. Fremgangsmåde ifølge krav 1, hvor styringsanalytten er tilvejebragt med en koncentration således at den er i stand til at optage alle bindingssteder for at tilvejebringe en direkte bestemmelse af styremæthedsresponsen (Rmaxc) fra en interaktionshændelse.
3. Fremgangsmåde ifølge krav 1, hvor styremæthedsresponsen (Rmaxc) bestemmes ud fra en ustabil tilstandsinteraktion mellem styringsanalytten og liganden.
4. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 3, hvor det relative molære responsbidrag af analytten og styringsanalytten i trin E tilnærmes af molvægtforholdet mellem analytten og styringsanalytten.
5. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 4, hvor trinnene E til H gentages for en flerhed af analytter.
6. Fremgangsmåde ifølge krav 5, hvor trinnene B til D gentages efter trinnene E til H er blevet udført for et forudbestemt antal analytter.
7. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 6, omfattende trinnene: I: at udvælge en fremgangsmåde til at bestemme Rmax for hver analyt, fra global bestemmelse af RmaxGiobai fra registreret sensorrespons med forskellige koncentrationer af analytten, og at bestemme RmaxA ved anvendelse af en styringsanalyt og den associerede Rmaxc J: at evaluere bindingsopførslen af analyt-ligand-interaktionen med eller flere bindingsopførselskriterie valgt fra gruppen bestående af: a) . Høj regist reret R b) Forhøjet basislinje efter indsprøjtning c) Forøgende respons under indsprøjtning d) Forhold mellem RmaxGiobai Og RmaxA K: Baseret på evalueringen i trin J at foreslå bindingsmodel som skal anvendes til udregning af interaktionsparameteret af hver analyt.
8. Analytisk system til at detektere molekylære bindingsinteraktioner, omfattende: (i) en biosensor omfattende mindst en sensoroverflade, detektionsorgan til at detektere molekylære bindingsinteraktioner ved den mindst ene sensoroverflade, og organ til at producere detektionsdata som repræsenterer bindingskurver, hvor hver kurve repræsenterer fremgangen af en bindingsinteraktion over tid, og (ii) databehandlingsorgan til at udføre trinnene A til H ifølge krav 1 eller som defineret endvidere i et hvilket som helst af kravene 2 til 7.
9. Computerprogram omfattende programkodeorgan til at bestemme et eller flere interaktionsparametre for interaktionen mellem en analyt og en ligand ifølge et hvilket som helst af kravene 1 til 7 når programmet køres på en computer.
