Claims (14)

1. Polypeptid med cellobiohydrolaseaktivitet, hvor polypeptidet omfatter en aminosyre-sekvens, der har mindst 85% sekvensidentitet med SEQ ID NO: 2, idet aminosyreresten i stilling Q1 i SEQ ID NO: 2 er modificeret ved erstatning eller udeladelse.
2. Polypeptid ifølge krav 1, hvor polypeptidet opretholder 50% af sin maksimale substratomdannelseskapacitet, når omdannelsen foretages i 60 minutter ved en temperatur på 60°C eller derover.
3. Polypeptid ifølge krav 1 eller 2, hvor polypeptidet omfatter en aminosyresekvens, der har mindst 90%, fortrinsvis mindst 95% og især mindst 99% sekvensidentitet med SEQ ID NO: 2.
4. Polypeptid ifølge et eller flere af de foregående krav, hvor polypeptidets aminosyresekvens har den ved SEQ ID NO: 2 definerede sekvens eller en sekvens, som defineret ved SEQ ID NO: 2, hvori 1 til 75 aminosyrerester, fortrinsvis 1 til 35 aminosyrerester, er erstattet, udeladt eller indsat.
5. Polypeptid ifølge krav 4, hvori yderligere én eller flere af følgende aminosyrerester i den ved SEQ ID NO: 2 definerede sekvens er modificeret ved erstatning eller udeladelse: stillingerne G4, A6, T15, Q28, W40, D64, E65, A72, S86, K92, V130, V152, Y155, K159, D181, E183, N194, D202, P224, T243, Y244, I277, K304, N310, S311, N318, D320, T335, T344, D346, Q349, A358, Y374, A375, T392, T393, D410, Y422, P442, N445, R446, T456, S460, P462, G463, H468 og/eller V482 blandt aminosyrerne 1 til 500 i SEQ ID NO: 2.
6. Polypeptid ifølge krav 5, hvor polypeptidet omfatter én eller flere af følgende foretrukne udvekslinger i forhold til den ved SEQ ID NO: 2 definerede sekvens:
7. Polypeptid ifølge et eller flere af de foregående krav, hvor polypeptidet har en aminosyresekvens valgt blandt listen over følgende mutationer i SEQ ID NO: 2:

Polypeptid ifølge et eller flere af de foregående krav, som udtrykkes og udskilles i et niveau på mere end 100 mg/l, fortrinsvis mere end 200 mg/l, særlig foretrukket mere end 500 mg/l og især mere end 1 g/l til centrifugatet efter indføring af en nukleinsyre, der koder for et polypeptid, som har en aminosyresekvens med mindst 85% sekvensidentitet med SEQ ID NO: 2, i en gær, hvori aminosyreresten i stilling Q1 i SEQ ID NO: 2 må være modificeret ved erstatning eller udeladelse.
9. Nukleinsyre, der koder for polypeptidet ifølge et eller flere af kravene 1 til 8, fortrinsvis med mindst 95% identitet med SEQ ID NO: 1.
10. Vektor, der omfatter nukleinsyren ifølge krav 9.
11. Værtscelle transformeret med en vektor ifølge krav 10.
12. Værtscelle ifølge krav 11, hvor værtscellen er afledt af gruppen, der består af Saccharomyces, Schizosaccharomyces, Kluyveromyces, Pichia, Pichia, Hansenula, Aspergillus, Trichoderma, Penicillium, Candida og Yarrowina.
13. Sammensætning omfattende polypeptidet ifølge et eller flere af kravene 1 til 9 og én eller flere endoglucanaser og/eller én eller flere β-glucosidaser og/eller én eller flere andre cellobiohydralaser og/eller én eller flere xylanaser.
14. Anvendelse af polypeptidet ifølge et eller flere af kravene 1 til 9 eller af sammensætningen ifølge krav 13 til enzymatisk nedbrydning af lignocellulosisk biomasse og/eller til tekstilforarbejdning og/eller som bestanddel i vaskemidler og/eller som bestanddel i levnedsmiddel- eller fodersammensætninger.
