Claims (13)

1. Fremgangsmåde til fremstilling af isopren, hvilken fremgangsmåde omfatter: a) at dyrke celler omfattende en eller flere heterologe nukleinsyrer, der koder for isopren-syntase-polypeptid under egnede forhold til fremstilling af isopren, hvor en gas tilsættes ved en gasoverrislingsrate (gas sparging rate) mellem 0,01 vvm og 2 vvm; og b) at fremstille isopren, hvor cellerne producerer mere end 400 nmol/gwcm/time af isopren, og hvor kuldioxid-udviklingshastigheden af cellerne er større end 1 x 10'18 mmol/L/time.
2. Fremgangsmåden ifølge krav 1, (i) hvor fremgangsmåden yderligere omfatter at genvinde isoprenet, eller (ii) hvor cellerne yderligere omfatter en eller flere heterologe nukleinsyrer eller en eller flere yderligere kopier af en endogen nukleinsyre, der koder for et isopren-syntase-polypeptid, eller (iii) hvor cellerne yderligere omfatter en eller flere heterologe nukleinsyrer eller en eller flere yderligere kopier af en endogen nukleinsyre, der koder for et IDI-polypeptid, et MVA-omsætningsvej-enzym, eller et DXP-omsætningsvej-enzym, eller (iv) hvor isopren-syntase-polypeptidet er fra Pueraria eller Populus eller en hybrid, Populus alba x Populus tremula.
3. Fremgangsmåden ifølge krav 2(iii), hvor MVA-omsætningsvej-enzymet er mevalonat-kinase.
4. Fremgangsmåden ifølge krav 2(iv), hvor isopren-syntase-polypeptidet vælges fra gruppen bestående af Pueraria montana eller Pueraria lobata, Populus tremuloides, Populus alba, Populus nigra, og Populus trichocarpa.
5. Fremgangsmåden ifølge krav 1, hvor cellerne er grampositive bakterieceller, Streptomyces-celler, gramnegative bakterieceller, Escherichia-ceUer, Pantoea-celler, svampeceller, filamentøse svampeceller, Trichoderma-ceUer, Aspergillus-celler, eller gærceller.
6. Fremgangsmåden ifølge krav 5, hvor cellerne vælges fra gruppen bestående af Bacillus subtilis, Streptomyces lividans, Streptomyces coelicolor, Streptomyces griseus, Escherichia coli, Pantoea citrea, Trichoderma reesei, Aspergillus oryzae, Aspergillus niger, Saccharomyces cerevisiae og Yarrowia lipolytica.
7. Fremgangsmåde til fremstilling af isopren, hvilken fremgangsmåde omfatter: a) at dyrke celler omfattende en eller flere heterologe nukleinsyrer, der koder for isopren-syntase-polypeptid under egnede forhold til fremstilling af isopren, hvor en gas tilsættes ved en gasoverrislingsrate (gas sparging rate) mellem 0,01 vvm og 2 vvm; og b) at fremstille isopren, hvor isopren-koncentrationen i den flydende fase er mindre end 1 g/L og cellerne producerer mere end 400 nmol/gwcm/time af isopren.
8. Fremgangsmåden ifølge krav 7, (i) hvor fremgangsmåden yderligere omfatter at genvinde isoprenet, eller (ii) hvor cellerne yderligere omfatter en eller flere heterologe nukleinsyrer eller en eller flere yderligere kopier af en endogen nukleinsyre, der koder for et isopren-syntase-polypeptid, eller (iii) hvor cellerne yderligere omfatter en eller flere heterologe nukleinsyrer eller en eller flere yderligere kopier af en endogen nukleinsyre, der koder for et IDI-polypeptid, et MVA-omsætningsvej-enzym, eller et DXP-omsætningsvej-enzym, eller (iv) hvor isopren-syntase-polypeptidet er fra Pueraria eller Populus eller en hybrid, Populus alba x Populus tremula, eller (v) hvor cellerne er grampositive bakterieceller, Streptomyces-ceWer, gramnegative bakterieceller, Escherichia-ceUer, Pantoea-celler, svampeceller, filamentøse svampeceller, Trichoderma-ceUer, Aspergillus-celler, eller gærceller.
9. Fremgangsmåden ifølge krav 8(iii) hvor MVA-omsætningsvej-enzymet er mevalonat-kinase.
10. Fremgangsmåde til fremstilling af en forbindelse, hvor forbindelsen har et eller flere karakteristika valgt fra gruppen bestående af (a) en Henrys lovkoefficient på mindre end 250 M/atm og (b) en opløselighed i vand på mindre end 100 g/L, hvilken fremgangsmåde omfatter: a) at dyrke celler omfattende en eller flere heterologe nukleinsyrer, der koder for isopren-syntase-polypeptid under egnede forhold til fremstilling af forbindelsen, hvor en gas tilsættes ved en gasoverrislingsrate (gas sparging rate) mellem 0,01 vvm og 2 vvm; og b) at fremstille forbindelsen.
11. Fremgangsmåden ifølge krav 10, (i) hvor fremgangsmåden yderligere omfatter at genvinde forbindelsen, eller (ii) hvor den fremstillede forbindelse er ethylen, eller (iii) hvor den fremstillede forbindelse er isopren.
