Claims (5)
1. Fremgangsmåde ex vivo til spaltning af et dobbeltstrenget DNA, hvilken fremgangsmåde omfatter: (a) identificering i DNA’et af mindst ét genkendelsessted for en rationelt designet ICrel-afledt meganuklease med ændret specificitet i forhold til I-Crel, hvor genkendelsesstedet ikke spaltes af en naturligt forekommende I-Crel, hvor genkendelsesstedet identificeres baseret på tilstedeværelse af en central sekvens med fire basepar udvalgt fra gruppen bestående af TTGT, TT AT, TTTC, TTCC, TTAG, TTAC og TTGC; (b) tilvejebringelse af den rationelt designede megenuklease og (c) etablering af kontakt mellem DNA’et og den rationelt designede megenuklease; hvorved den rationelt designede megenuklease spalter DNA’et.
2. Fremgangsmåde ex vivo til spaltning af et dobbeltstrenget DNA, hvilken fremgangsmåde omfatter: (a) inkorporering af en central sekvens med fire basepar udvalgt fra gruppen bestående af TTGT, TT AT, TTTC, TTCC, TTAG, TTAC og TTGC i et genkendelsessted for en rationelt designet I-Crel-afledt meganuklease med ændret specificitet i forhold til I-Crel, hvor genkendelsesstedet ikke spaltes af en naturligt forekommende I-Crel, (b) indføring af genkendelsesstedet i DNA’et, (c) tilvejebringelse af den rationelt designede megenuklease og (d) etablering af kontakt mellem DNA’et og den rationelt designede megenuklease; hvorved den rationelt designede megenuklease spalter DNA’et.
3. Fremgangsmåde ex vivo ifølge et hvilket som helst af kravene 1 -2, hvor DNA-spaltningen sker in vitro, eventuelt hvor DNA’et er udvalgt fra gruppen bestående af et PCR-produkt; et kunstigt kromosom; genomisk DNA isoleret fra bakterier, svampe, planter eller dyreceller og viralt DNA.
4. Fremgangsmåde ex vivo ifølge et hvilket som helst af kravene 1-3, der endvidere omfatter rationelt design af den I-Crel-afledte meganuklease for at genkende genkendelsesstedet.
5. Fremgangsmåde ex vivo ifølge et hvilket som helst af kravene 1 -4, der endvidere omfatter frembringelse af den rationelt designede I-Crel-afledte meganuklease.
