Description
Patentkrav: 1. Fremgangsmåde til fremstilling af ikke-patogene, infektiøse pankreasenekrose-virus, hvilken fremgangsmåde omfatter følgende trin: fremstilling af et cDNA, som indeholder genom-segmenterne A og B fra infektiøs pankreasnekrose-virus, hvor cDNAet af segment A er modificeret for at forhindre ekspression af NS-protein, transkribering af cDNAet til frembringelse af syntetiske RNA-transkripter, transfektion af værtsceller med de syntetiske RNA-transkripter, inkubering af værtscellerne i et dyrkningsmedium og isolering af levende, ikke-patogene, infektiøse pankreasenekrose-virus fra dyrkningsmediet. 2. Fremgangsmåde ifølge krav l, hvorved værtscellerne er valgt fra gruppen bestående af kongelakseembryo (CHSE)-celler, regnbueørredgonade (RTG-2)-celler, Bluegill-fiske (BF-2)-celler, Brown Bullhead (BB)-celler og Fathead Minnow (FHM)-celler. 3. Fremgangsmåde ifølge krav i, hvorved cDNAet stammer fra mere end én stamme af infektiøs pankreasnekrose-virus. 4. Fremgangsmåde ifølge krav 1, hvorved cDNAet for segment A er modificeret ved hjælp af mutation af initieringskodonen for NS-genet til en stopkodon. 5. Fremgangsmåde til frembringelse af en vaccine af levende, ikke-patogen, infektiøs pankreasnekrose-virus, hvilken fremgangsmåde omfatter trinene: fremstilling af et uforkortet cDNA, som indeholder genomsegmenterne A og B fra infektiøs pankreasnekrose-virus, hvor segment A er modificeret for at forhindre ekspression af NS-protein, transkribering af cDNAet til frembringelse af syntetiske RNA-transkripter, transfektion af værtsceller med RNA-transkripterne, inkubering af værtscellerne i et dyrkningsmedium, isolering af levende, infektiøse, ikke-patogene pankreasnekrose-virus fra dyrkningsmediet, og blanding af de levende, ikke-patogene, infektiøse pankreasnekrose-virus med et farmaceutisk acceptabelt bæremedium til frembringelse af en vaccine med levende, ikke-patogene, infektiøse pankreasnekrose-virus. 11. Fremgangsmåde til frembringelse af et ikke-patogent, kimært gen, hvilken fremgangsmåde omfatter følgende trin: fremstilling af et cDNA, som indeholder genomsegmenterne A og B fra infektiøs pankreasnekrose-virus, hvor cDNAet af segment A er modificeret for at forhindre ekspression af NS-protein, og cDNAet af segment A koder epitopiske determinanter fra mindst to forskellige stammer af infektiøs pankreasnekrose-virus, transkribering af cDNAet til frembringelse af syntetiske RNA-transkripter, transfektion af værtsceller med de syntetiske RNA- transkripter, inkubering af værtscellerne i et dyrkningsmedium, og isolering af levende, ikke-patogene, kimære, infek-tiøse pankreasnekrose-virus fra dyrkningsmediet. 12. Ikke-patogent, kimært, akvatisk Birnavirus, hvor viruset er NS-proteindefekt og indeholder epitopiske determinanter fra mindst to forskellige Birnavirusstammer. 13. Virus ifølge krav 12, hvor det akvatiske Birnavirus er infektiøst pankreasnekrose-virus. 14. Virus ifølge krav 13, hvor de infektiøse pankreasnekrose-virusstammer er valgt fra gruppen bestående af West Buxton, Jasper, NI og SP. 15. Vaccine, som omfatter det ikke-patogene, kimære, akvatiske Birnavirus ifølge krav 12 kombineret med et farmaceutisk acceptabelt bæremedium. 16. Vaccine ifølge krav 15, hvori det akvatiske

Vaccine ifølge krav 17, hvori de bakterielle antigener er fra bakterier, der er valgt fra gruppen bestående af Aeroinonas salmonicida, Vibrio anguillarum, Vibrio salmonicida, Vibrio viscosus, Yersinia ruckri, Piscirickettsia salmonis, Renibacterium salmoninarum, Pasturella piscicida, Flavobacterium columnare, Flavobacterium psychrophilum og Lactococcus garvieae. 22.
