Den foreliggende opfindelse angår monoklonale antistoffer, der genkender et kerneprotein fra det lymfadenopati-associerede virus (LAV), hybridomer, der udskiller disse antistoffer, og en fremgangsmåde til påvisning eller rensning af et polypeptid indeholdende en epitop, som gen-5 kendes af et af de monoklonale antistoffer.
De omhandlede monoklonale antistoffer genkender såvel glycosylerede som ikke-glycosylerede polypeptider kodet for af genomt RNA fra LAV eller klonet DNA afledt deraf.
I ansøgerens internationale patentansøgning, der er indleveret d.
10 18. oktober 1985 under nummeret PCT/EP85/00548, beskrives monoklonale antistoffer, der specifikt genkender et protein, polypeptid eller glycoprotein fra omhyl ningen af LAV-vi rus. Der er ingen beskrivelser af antistoffer, som genkender kerneproteiner fra LAV-virus. Den pågældende ansøgning, der er videreført i Danmark under nr. 2849/86, var ikke ti 1 -15 gængelig for offentligheden på det tidspunkt, hvor den foreliggende ansøgning skal anses for indleveret.
Både den molekylære kloning af cDNA og af integrerede provirale former af LAV er blevet rapporteret tidligere. Rekombinantfagklonerne i soleredes fra et genombibliotek af LAV-inficeret humant T-lymfocyt DNA 20 nedbrudt delvis med Hind III. Rekombinant-fag AJ19-indskuddet genereredes ved Hind III-spaltning inden for R-elementet i den lange terminalgentagelse (LTR). Hver ekstremitet af indskuddet indeholder således en del af LTR. Muligheden for klynger af Hind III-steder i R er blevet elimineret ved sekvensbestemmelse af en del af en LAV dDNA-klon, pLAV 75, 25 svarende til dette område. Den totale sekvensinformation for LAV-genomet kan således hidrøre fra AJ19-klonen.
Under anvendelse af M13 "shotgun" klonings- og dideoxykædetermine-ringsmetoden (Sanger et al, 1977) er AJ19-indskuddets nukleotidsekvens blevet bestemt. Det rekonstruerede virusgenom med to kopier af R-sekven-30 sen er 9193 nukleotider langt. Nummereringssystemet begynder ved viruspartikel RNA'ets cap-punkt. Hele sekvensen er vist i figur la-le i den internationale ansøgning, der også er inkluderet i den foreliggende ansøgning.
Den foreliggende opfindelse har til hensigt at muliggøre mere nøj-35 agtig identifikation af betydningsfulde LAV-proteiner eller glycoprotei-ner og også at tilvejebringe monoklonale antistoffer mod proteiner og polypeptider med signifikante immunogene steder eller epitoper af LAV-virusproteinerne eller glycoproteinerne.

Opfindelsen angår nærmere bestemt monoklonale antistoffer, der genkender LAV-kerneproteiner eller fragmenter deraf, specielt pl3-, pl8-, p25- og p55-proteinerne.
Specielt angår opfindelsen monoklonale antistoffer, der henholdsvis 5 genkender: et p55-protein, der menes at blive kodet for af DNA-sekvensen, der strækker sig ca. fra nukleotid 336 op til ca. nukleotid 1650 i LAV cDNA, hvilket p55-protein anses for at indeholde aminosyresekvenser svarende til sekvenserne i kerneproteinerne pl8 og p25 i LAV-viruset; 10 - et p25-protein, der menes at blive kodet for af DNA-sekvensen, der strækker sig ca. fra nukleotid 732 op til ca. nukleotid 1300 i LAV cDNA; et pl3-protein, der menes at blive kodet for af DNA-sekvensen, der strækker sig ca. fra nukleotid 1371 op til ca. nukleotid 1650; et pl8-protein, der menes at blive kodet for af DNA-sekvensen, der 15 strækker sig ca. fra nukleotid 336 op til ca. nukleotid 611.
Mere specielt angår opfindelsen monoklonale antistoffer, der genkender polypeptider med peptidsekvenser, som er identiske med eller ækvivalente til de sekvenser, der kodes for af DNA-sekvenserne, der strækker sig ca. mellem følgende nukleotidpositioner: 20 336 til 1650 (p55) 336 til 611 (pl8) - 1371 til 1650 (pl3) 656 til 1300 (p25) 25
Det bør nævnes, at pl3, pl8 og p25 alle synes at hidrøre fra samme prækursor, dvs. p55.

De betegnelser, der anvendes til identifikation af hybridomerne, svarer til de ved standardrensningsprocedurer ud fra lysater af LAV-vi-rus opnåede rensede peptider, som indledningsvis anvendtes til immunise-5 ring af dyrene, fra hvilke de til fremstilling af de tilsvarende hybri-domer anvendte milt-celler opnåedes. Rensede peptider til brug ved fremstilling af de monoklonale antistoffer kan også opnås ved immunisering af dyr med de tilsvarende rensede ekspressionsprodukter af de i ovennævnte PCT-ansøgning omhandlede DNA-rekombinanter. Den generelle frem-10 gangsmåde til fremstilling af disse hybridomer vil blive beskrevet senere.
En generel fremgangsmåde anvendt til fremstilling af hvert af ovennævnte monoklonale antistoffer vil blive beskrevet i det følgende.
15 Immunisering af mus
Der anvendtes grupper af 6-8 uger gamle Balb/c-mus. De forskellige grupper modtog respektivt de ovenfor nævnte forskellige proteiner. Immuniseringsforskriften, der er identisk for alle grupper, omfattede injektioner tre gange ad intraperitoneal vej efterfulgt af en gang ad intra-20 venøs vej, hver gang af 10 μg af det antigeni ske præparat i nærvær af komplet Freund-adjuvans ved dag 0 og af ukomplet Freund-adjuvans ved dag 14 og uden adjuvans ved dag 28 og 42.
Fusion og dyrkning af hybriderne 25 Den azaguanin-resistente og ikke-afsondrende variant 6.53 af myelom P3 x 63 Ag8, som selv stammede fra MPOC-21-cellelinien anvendtes. Fusionen med splenocyter fra de immuniserede mus udførtes i nærvær af poly-ethylen-glycol 4000 i henhold til Faseka de St-Groth og Scheidegger's teknik på dag 45. Udvælgelsen af hybriderne i RPMI 16-40 "HAT”-medium 30 udførtes i henhold til samme dyrkningsteknik i plader med 24 brønde (Costar).
Hybridomerne, som producerede de specifikke antistoffer klonedes derefter i plader med 96 brønde i nærvær af et fødel ag af syngene thymo-cyter. De udvalgte afsondrende kloner straktes derefter i plader med 24 35 brønde stadig i nærvær af thymocyter. Når sammenflydning viste sig i en af brøndene, injiceredes klonen intraperitonealt i Balb/c-mus, som 8 dage tidligere havde modtaget en injektion af Pristane, og/eller holdtes i væskekultur.
DK 168667 B1 4 Påvisning af anti-LAV-antistofferne
Fem forskellige teknikker har tilladt karakterisering af kloner, der producerer antistofferne med ønsket specificitet. I et første trin 5 påvistes de hybrider, der udskilte de ønskede antistoffer ved en ELISA-analyse, som afslørede museimmunoglobul inerne i supernatanten. Udgående fra denne første udvælgelse blev de supernatanter, som indeholdt antistoffer orienteret mod de søgte viruskomponenter, screenet ved hjælp af en ELISA-analyse eller ved en immunofluorescensanalyse på humane celler, 10 der producerede viruset. Endelig analyseredes supernatanterne ved radio-immunopræcipitation af virus mærket med 35S-cystein og ved Westernblot teknikken på et viruspræparat, hvorved anti-LAV-antistoffernes specificiteter bestemtes.
15 Resultater
De ud fra forskellige fusioner opnåede celler dyrkedes derefter i 648 brønde. Den mikroskopiske undersøgelse har vist, at de fleste af disse brønde indeholdt en enkelt hybridklon, som var i stand til at vokse i det selektive "HAT"-medium. Mere end 50% af klonerne producerede 20 antistoffer, som gav en positiv reaktion ved antivirus-ELISA-analysen.
De mest repræsentative fusioner testedes ved Western-blot teknikken, og flere hybridomer fra hver gruppe subklonedes under hensyntagen til deres specificitet, deres reaktivitet ved antivirus-ELISA-analysen og deres udviklingshastighed i kulturer. Hybrider, der producerede antistoffer, 25 som mere specifikt genkendte de proteiner eller polypeptider, der fra start var blevet anvendt til immunisering af musene, blev bibeholdt. Alle opnåede subkloner vistes at udskille antistoffer, som efter ekspression injiceredes i syngene mus. Analysen af antistofspecificiteter i de forskellige opnåede ascites-væsker bekræftede specificiteterne af de i 30 hver af asciterne dannede antistoffer med hensyn til de tilsvarende proteiner.
Hybridomerne, som er deponeret ved CNCM, og som identificeredes ovenfor som repræsentative for de hybridomer, der kan opnås under anvendelse af ovenstående proteiner, udgør også en del af opfindelsen.
35 De opnåede monoklonale antistoffer kan i sig selv bringes til anvendelse til rensning af proteiner eller polypeptider, som har et anti-genisk sted fælles med de proteiner, der fra start anvendtes til fremstilling af hybridomerne. Opfindelsen angår således også en rensnings- DK 168667 Bl 5 fremgangsmåde som angivet i krav 10. En sådan fremgangsmåde anvendes med fordel til behandling af lysater af LAV, af inficerede T-lymfocyter eller af vilkårlige andre celler, som er i stand til at producere LAV eller et analogt virus. Fremgangsmåden kan også anvendes til identifika-5 tion af de proteiner, der fremstilles af celler, som er fremstillet genteknisk med rekombinante DNAér som ovenfor defineret og indeholdende en DNA-sekvens, der koder for den relevante epitop. De monoklonale antistoffer, der anvendes ved denne fremgangsmåde, er med fordel immobilise-ret på en fast bærer, f.eks. på en bærer, som er egnet til affinitets-10 kromatografioperationer, såsom et tre-dimensionelt tværbundet agarose-gitter, som er bragt i handelen under varemærket Sepharose af de svenske firma Pharmacia A.G., f.eks. ved cyanogenbromidmetoden.
Som et særligt aspekt angår opfindelsen anvendelsen af et monoklo-nalt antistof ifølge opfindelsen til in vitro påvisning af det eller de 15 tilsvarende polypeptider i en biologisk prøve eller af det eller de tilsvarende ekspressionsprodukter fra LAV-inficerede lymfocyter.
En udførelsesform af fremgangsmåden ifølge opfindelsen omfatter således trin, der indbefatter, at man bringer opløsningen indeholdende po-lypeptidet i kontakt med en affinitetssøjle, som bærer de monoklonale 20 antistoffer for selektivt at tilbageholde polypeptiderne, derefter udvinder polypeptidet efter dissociation af antigen-antistof-komplekset ved hjælp af en passende puffer, f.eks. en saltopløsning af passende ionstyrke, f.eks. ved pH 2-4. Et egnet salt til dannelse af sådanne puffere udgøres af ammoniumacetat.
25 Det af hybridomen deponeret ved CNCM under nr. 1-355 producerede monoklonale antistof er af særlig betydning. Nærmere bestemt genkender det monoklonale antistof både pl8 og p55. Det følger heraf, at den epitop, som genkendes mere signifikant af dette antistof, forbliver umodi-ficeret, når p55 (prækursor for pl8 og p25) spaltes i de forskellige 30 komponenter.
Dette antistof er således af speciel interesse til rensning af både pl8 og p55. De andre komponenter indeholdt i p55, specielt p25 kan så fremstilles på grundlag af det rensede p55. Det har yderligere vist sig, at dette antistof er i stand til at genkende LAV-viruset i præparater 35 indeholdende dette.
Dette antistof er således af speciel interesse til påvisning af viruspartikler i en biologisk prøve, navnlig et serum opnået fra patienter, som skal diagnosticeres for AIDS eller LAS, DK 168667 B1 6 påvisning af inficerede lymfocyter, påvisning i en biologisk prøve som ovenfor nævnt eller i en kultur af inficerede lymfocyter, og behandling af celler, som udtrykker viruset, med antistof.
5 Opfindelsen angår desuden et hvilket som helst andet monoklonalt antistof, som genkender en vilkårlig af de epitoper, der mere specifikt genkendes af de monoklonale antistoffer, som udskilles af hybridomerne, der er blevet deponeret ved CNCM og følgelig 10 - de hybridomer, som udskiller de andre monoklonale antistoffer.
Claims (9)
1. Monoklonale antistoffer, KENDETEGNET ved, at de genkender et kerneprotein fra det lymfadenopati-associerede virus (LAV) og er produ- 5 ceret af de hybridomer, der er deponeret ved CNCM under numrene 1-355, 1-356, 1-357, 1-358, 1-359 og 1-360, eller er produceret af en hvilken som helst hybridom af en art fremstillet af en hvilken som helst af ovennævnte deponerede hybridomer og genkender samme epitop som den, der genkendes af den tilsvarende deponerede hybridom. 10
2. De monoklonale antistoffer ifølge krav 1, som genkender et pl3-protein fra LAV.
3. De monoklonale antistoffer ifølge krav 1, som genkender et 15 pl8-protein fra LAV.
4. De monoklonale antistoffer ifølge krav 1, som genkender et p25-protein fra LAV.
5. De monoklonale antistoffer ifølge krav 1, som genkender et p55-protein fra LAV.
6. De monoklonale antistoffer ifølge krav 1, som både genkender et pl3-protein og et p25-protein fra LAV. 25
7. De hybridomer, der udskiller de monoklonale antistoffer ifølge et hvilket som helst af kravene 1-6.
8. Anvendelse af et monoklonalt antistof ifølge et hvilket som 30 helst af kravene 1-6 til in vitro påvisning af et polypeptid, som er identisk eller ækvivalent med et polypeptid med en peptidsekvens, som er identisk eller ækvivalent med et polypeptid kodet for af en DNA-sekvens udvalgt blandt dem, der strækker sig ca. mellem følgende nukleotidposi-tioner: 336 til 1650 (p55) 336 til 611 (pl8) - 1371 til 1650 (pl3) 35 5 8 DK 168667 B1 656 til 1300 (p25) i nukleotidsekvensen for et cDNA opnået ud fra det genome RNA for et lymfadenopati-associeret virus (LAV) og vist i fig. la til le.
9. Fremgangsmåde til rensning af et polypeptid udvalgt blandt de i krav 1 definerede polypeptider og indeholdt i opløsel iggjort form i en biologisk prøve, hvilket polypeptid indeholder en epitop, som genkendes af et af de monoklonale antistoffer ifølge et hvilket som helst af kra-10 vene 1-6, KENDETEGNET ved, at den omfatter, at man bringer det biologiske medium i kontakt med det tilsvarende monoklonale antistof fæstnet til eller immobiliseret på en uopløseliggjort bærer for at fremkalde fiksering af polypeptidet på det immobil i serede monoklonale antistof, hvorved et polypeptid-antistofkompleks dannes, fraskil!er de ikke-fikse-15 rede polypeptider og udvinder det fikserede polypeptid ved dissociering af polypepti d-anti stofkomplekset. 20 25 30 35 t ---------- ---
