Den foreliggende opfindelse angår en hidtil ukendt fremgangsmåde til fremstilling af et oligopeptid, som kan isoleres fra sunde hvide blodlegemer eller granulocyter, og som selektivt inhiberer formeringen af normale og leukæmiske 5 myeloide celler.
De ved fremgangsmåden fremstillede oligopeptider er af den type, som har den summariske aminosyresammensætning
Tau;L, Asxlf Ser2, Thr^, Glx3, Gly2, Ala^ (PO^i, 10 hvor Tau betyder en taurinrest, Asx betyder en L-asparagin-og/eller L-asparaginsyrerest, Ser betyder en L-serinrest,
Thr betyder en L-threoninrest, Glx betyder en L-glutamin-og/eller L-glutaminsyrerest, Gly betyder en glycocolrest, og 15 Ala betyder en L-alaninrest, som ved en pH-værdi på 6,5 udviser en negativ ladning og en relativ elektroforetisk mobilitet i forhold til asparaginsyre på fra -0,55 til -0,65, som ved en pH-værdi på 1,9 ingen ladning udviser, men som udviser en relativ mobilitet i forhold til e-(dini-20 trophenyl)-lysin på 0,26, og som reagerer positivt på chlor-toluidin.
Det er velkendt, at de cytostatiske stoffer, der anvendes i chemoterapien af ondartede tumorer, såsom alkyler ingsmidler, antimetaboliter og spindelgiftstoffer, al-25 mindeligvis er yderst toksiske forbindelser, som skader alle slags celler, som formerer sig ved deling. Da ondartede tumorer er pathologiske former for en given celletype, ville det være ønskværdigt at anvende selektive stoffer til inhi-bering af formeringen af disse celler [S. Eckhardt: Klinikai 30 Onkologia, Medicina, Budapest, 1977].
Til fastlæggelse af sådanne midler, som specifikt inhiberer væksten eller formeringen af tumorceller, er der tidligere gennemført en udbredt forskningsaktivitet, og der er bl.a. beskrevet ca. 25 celleekstrakter fremstillet ud 35 fra forskellige ensartede cellevæv, om hvilke det hævdes eller er påvist, at de indeholder naturligt forekommende 2

regulatorer for den selektive inhibering af formeringen af specifikke celletyper [A. Balåzs, I. Blazsek: Control of Cell Proliferatin by Endogenous Inhibitors, Akadémiai Konyv-kiado, Budapest og Elsevier - North Holland, 1979]. Der er 5 opnået særligt gunstige resultater med de rensede ekstrakter af hvide blodlegemer (granulocyter) [Rytomaa and Kiviniemi: Celle Tissue Kinet. 1, 341 (1968); Paukovits: Nat. Cancer. Inst. Monogr. 38/ 147; Paukovits et al; Oncology 34/ 187 (1977)].
10 Den biologiske aktivitet af den delvis rensede gra- nulocytekstrakt er også blevet påvist på leukæmipatienter [Rytomaa et al., Scand. J. Haematol., Suppl. 27, 5 (1976)]. Yderligere beskriver HU-patentskrift nr. 162.446 en fremgangsmåde til fremstilling af en delvis renset granulocyteks-15 trakt fra human- og kalveblod eller peritonealvæske. Den opnåede ekstrakt inhiberer selektivt formeringen af hvide blodlegemer.
De ovenfor nævnte ekstrakter er imidlertid kun delvis rensede præparater, som indeholder en væsentlig mængde uren-20 heder. Der kendes ikke nogen fremgangsmåde til fremstilling af rene, biologisk aktive stoffer, som er i stand til selektivt at inhibere væksten eller formeringen af specifikke celletyper.
Det har nu vist sig, at man ved en yderligere udvik-25 ling af de fremgangsmåder, der beskrives i HU-patentskrift nr. 162.446, kan løse problemet med fremstilling af et sådant, rent, aktivt stof. Mere specifikt har det vist sig, at man ved at underkaste egnede udgangsmaterialer, især ekstrakter af hvide blodlegemer isoleret fra hesteblod eller 30 kalvemilt, specielle fraktioneringsoperationer, kan opnå et kemisk rent, ensartet, godt karakteriserbart aktivt oligo-peptid, der inhiberer formeringen af leukæmiske myeloide celler, endog i yderst lave koncentrationer.
Den foreliggende opfindelse angår i overensstemmelse 35 hermed en hidtil ukendt fremgangsmåde til fremstilling af et biologisk aktivt oligopeptid af den ovenfor angivne type,

3 hvilken fremgangsmåde er ejendommelig ved, at man a) homogeniserer sunde hvide blodlegemer isoleret fra animalsk eller humant blod, især fra hesteblod, i en pufferopløsning med en pH-værdi på fra 7 til 8, centrifugerer det 5 flydende homogenisat, adskiller de opløste bestanddele, der har en molekylvægt på mindre end 10.000, fra den ovenstående væske, underkaster denne fraktion en yderligere fraktionering, fortrinsvis ved gelchromatografi, fraskiller og, om ønsket, lyophiliserer den fraktion, som fraskilles ved vo-10 lumenværdier Ve/vQ på fra 1,15 til 1,45, eller b) homogeniserer animalske organer, der indeholder granulo-cyter, fortrinsvis kalvemilt, i et vandblandbart organisk opløsningsmiddel, adskiller faststofferne fra den opnåede flydende suspension, ekstraherer dem med et organisk opløs- 15 ningsmiddel, der er i stand til at opløse fedtstoffer, ekstraherer de således fremkomne faststoffer med vand, fraskiller den uopløselige del fra den således fremkomne, flydende suspension, adskiller de opløste bestanddele, som har en molekylvægt på mindre end 10.000, fra den flydende 20 fase, underkaster denne fraktion en yderligere fraktionering, fortrinsvis ved gelchromatografi, fraskiller og, om ønsket, lyophilisrer den fraktion, som fraskilles ved volumenværdier ve/vo På fra i*3 til 2,5, 25 og renser koncentratet af aktivt stof fremstillet ved fremgangsmådevariant a) eller b) ved en ionbytterchromatografi, som fraktionerer på basis af ionladning og partikelstørrelse, og/eller ved papirelektrophorese, fraskiller og eventuelt lyophiliserer et oligopeptid, som har en negativ lad-30 ning ved en pH-værdi på 6,5 og en relativ mobilitet på -0,55 til -0,65 i forhold til asparaginsyre, som ikke har nogen ladning ved en pH-værdi på 1,9, men har en mobilitet på ca. 0,26 i forhold til e-dinitrophenyl-lysin, og som giver en positiv chlortoluidinreaktion.

4 gende "fraktion GI-3". Slutproduktet betegnes kort "GP", hvilket betyder "granulopeptid".
Volumenværdien Vq/Vq refererer til, at den aktive og specifikke fraktion kan påvises mellem elueringsmaksimum 5 for 3H-thymidin anvendt som mærkestof og vQ.
Ved fremgangsmådevariant a) foretrækkes, at man, inden man gennemfører de for fremgangsmåden ejendommelige foranstaltninger, udvinder et granulocytbundfald, som anvendes som udgangsmateriale i variant a). Dette bundfald fås, 10 ved at et granulocytberiget bundfald suspenderes i pufferopløsningen (pH 7-8) og centrifugeres. Granulocytberigelsen gennemføres med fordel, ved at animalsk eller humant blod behandles med heparin, hvorefter det på hvide blodlegemer (leukocyter) rige lag efter sedimentering skilles fra bund-15 faldet ved centrifugering.
Eventuelt kan også fraktionen indeholdende bestanddele med en molekylvægt mindre end 10.000 lyophiliseres, før den fraktioneres yderligere.
Ved fremgangsmådevariant b) kan de animalske organer, 20 som er homogeniseret med det vandblandbare organiske opløsningsmiddel fermenteres, før faststofferne fraskilles.
Ved den her omhandlede fremgangsmåde foretrækkes det, at 1) der som pufferopløsning anvendes en phosphatpufferopløs- 25 ning, f.eks. ud fra dinatriumhydrogenphosphat og kalium- dihydrogenphosphat, 2) fraskillelsen af fraktionen med en molekylvægt på mindre end 10.000 gennemføres ved molekylfiltrering, 3) acetone anvendes som det vandblandbare organiske opløs- 30 ningsmiddel, og 4) et chlorcarbonhydrid anvendes som fedtopløsende uorganisk opløsningsmiddel.
Det ved den her omhandlede fremgangsmåde fremstillede oligopeptid GP har en selektiv aktivitet. Det inhiberer 35 især formeringen af myeloide celler (granuloide celler hos mus og rotter, transplantable animalske myeloide leukæmicel- 5

ler, spontane humane akutte myeloide leukæmiceller) og er ineffektivt over for normale thymocyter, phytohæmagglutinin-stimulerede lymphocyter, subakutte lymphoide leukæmithymocy-ter, humane lymphoide leukæmiblodceller og humane HeLa-tu-5 morceller.
Fraktionen GI-3 opnået som en mellemproduktfraktion ved den her omhandlede fremgangsmåde indeholder naturligvis også det aktive stof GP, sammen med mindre aktive bestanddele og et stort antal inaktive urenheder. GI-3-fraktionen viser 10 også en specifik inhiberingsaktivitet over for formeringen af myeloide celler, til en vis udstrækning svarende til dens indhold af den aktive bestanddel GP. Ved forsøg in vitro andrager den minimale effektive dosis (MED) af GI-3-fraktionen, ved inhibering af inkorporeringen af 3H-thymidin 15 i syre-uopløselig DNA, 110 Mg/ml, medens MED i tilfælde af kapillære kolonier dannet i agargel er 8 jug/ml. De tilsvarende ED5g-værdier er 430 og 90 /jg/ml.
De fysisk-kemiske og celle-biologiske egenskaber af det aktive oligopeptid GP er som følger: 20 Varighed af virkning i suspensionskulturer: 8 timer i gelkolonier: 1 uge
Det aktive stof er ikke toksisk, idet det ikke reducerer cellevitaliteten målt ved 51Cr-frigørelsesforsøg.
Dets angrebspunkt er G1-fasen af cellens cyclus, dvs. den 25 fase, som ligger mellem fasen med DNA-syntese og fasen med celledeling.
Thermostabilitet: Stoffet mister ikke sin aktivitet og heller ikke den specifikke karakter af aktiviteten efter en behandling ved 80°C i 60 minutter. I opløsning i et tids-30 rum på 72 timer konstateres der ikke nogen sænkning i aktivitet. Stoffet kan opbevares som et lyophilisat ved -20°C i mere end 7 uger, i form af et pulver udfældet fra acetone eller chloroform ved +4°C i mere end 18 måneder.
Resultaterne af forsøg in vitro udført for at sammen-35 ligne aktiviteten af det aktive stof GP isoleret ved ionbyt-terchromatografi eller papirelektroforese og igen undersøgt 6

Eksempel 1
Fremstilling af GI-3 fraktion fra hesteblod 10 liter hesteblod behandles med 3500 E/liter heparin. Blodet sedimenterer derpå ved stuetemperatur i 30 minutter, 10 hvorpå det øvre lag, som er rigt på leukocyter, fraskilles og centrifugeres ved 800 G. Den fraskilte cellepopulation, der indeholder 80% granulocyter, vaskes i 200 ml af en 0,06 M phosphatpufferopløsning (pH = 7,4) under centrifugering ved 800 G. Sammensætningen af phosphatpufferen er: 15 9,500 g dinatriumhydrogenphosphat, 2H20 og 1,815 g kalium- dihydrogenphosphat i 1 liter destilleret vand. Det opnåede granulocytbundfald suspenderes i en phosphatpuffer med den ovenfor nævnte sammensætning i en tæthed på 4xl09 celler/ml i en Potter-homogenisator indstillet på 10 slag pr. minut.
20 Det opnåede homogenisat centrifugeres i 30 minutter ved 1500 G. Den adskilte ovenstående væske underkastes ul-trafiltrering under anvendelse af en "Amicon PM 10"-membran under et tryk på 3 atm. Det opnåede filtrat lyophiliseres, og det opnåede lyophilisat underkastes gelchromatografi på 25 en "Sephadex G-10"-sø]le under anvendelse af en 0,05 M ammoniumhydrogencarbonatpufferopløsning (pH = 7,9). Fraktionen svarende til ve/v0 = 1,15 til 1,45 fraskilles og lyophiliseres. Der opnås således 65 mg af et aktivt faststofkoncentrat (fraktion GI-3).

Eksempel 2
Fremstilling af fraktion GI-3 fra kalvemilt Til 1000 g hakket kalvemilt, frigjort fra dens fibrøse lag, sættes acetone ved 4°C, indtil rumfanget er 4 liter.
35 Blandingen homogeniseres ved denne temperatur, inkuberes i 60 minutter, centrifugeres, og det fraskilte bundfald vaskes 8

med acetone og tørres ved 20°C i vakuum.
Tørstoffet ekstraheres i 30 minutter med 2 liter chloroform, den faste fase skilles fra, tørres ved stuetemperatur og opløses i 3 liter to gange destilleret vand under 5 omrøring i 16 timer. Opløsningen centrifugeres ved 20.000 G
i én time. Den ovenstående flydende fase filtreres gennem © © "Amicon XM 50" og derefter på en "Amicon PM 10"-membran under et tryk på 3 atm. Filtratet lyophiliseres. Det opnåede © lyophilisat underkastes gelchromatografi på en "Sephadex 10 G-15"-søjle under anvendelse af en 0,05 M ammoniumhydrogen-carbonatpufferopløsning (pH=7,9). Fraktionen svarende til ve/v0 = 1/3 til 2,5 fraskilles og lyophiliseres. Der opnås således 3,54 g af et aktivt faststofkoncentrat (fraktion GI-3).

Eksempel 3
Fremstilling af rent aktivt stof (GP) 300 mg af et aktivt faststofkoncentrat GI-3 fremstil- © let ifølge eksempel 1 eller eksempel 2 påføres et "Whatman 20 3MM" chromatografisk papir, idet der begyndes fra midten af papiret, i en længde på 30 cm. Der gennemføres elektroforese i en 90:4:900-blanding af pyridin, eddikesyre og vand (puffer, pH = 6,5) i et vandret udstyr, med en spændingsgradient på 50 V/cm i 2 timer. Stillingen for den sure ninhydrin-25 negative og chlor-positive bestanddel bestemmes ved farvning med chlortoluidin. Den opnåede aktive bestanddel elueres fra papiret med en 8:2:90-blanding af eddikesyre, myresyre og vand og underkastes yderligere elektroforese i denne pufferblanding (pH = 1,9) ved 80 V/cm i 90 minutter. Stil-30 lingen for den uladede bestanddel (det aktive stof GP) bestemmes ved en chlortoluidinreaktion. Det opnåede stof elueres fra papiret med den ovenfor beskrevne sure pufferblanding, og eluatet lyophiliseres. Der opnås således 8 μg af det rene stof GP.
Claims (5)
1. Fremgangsmåde til fremstilling af et oligopeptid, som kan isoleres fra sunde hvide blodlegemer eller granulo-cyter, som selektivt inhiberer formeringen af normale og 5 leukæmiske myeloide celler, som har den summariske aminosy-resammensætning Taux, Asxlf Ser2, Thrlf Glx3, Gly2, Alalr (PO^, 10 hvor Tau betyder en taurinrest, Asx betyder en L-asparagin-og/eller L-asparaginsyrerest, Ser betyder en L-serinrest, Thr betyder en L-threoninrest, Glx betyder en L-glutamin-og/eller L-glutaminsyrerest, Gly betyder en glycocolrest, og Ala betyder en L-alaninrest, som ved en pH-værdi på 6,5 15 udviser en negativ ladning og en relativ elektroforetisk mobilitet i forhold til asparaginsyre på fra -0,55 til -0,65, som ved en pH-værdi på 1,9 ingen ladning udviser, men som udviser en relativ mobilitet i forhold til e-(dini-trophenyl)-lysin på 0,26, og som reagerer positivt på chlor-20 toluidin, kendetegnet ved, at man a) homogeniserer sunde hvide blodlegemer isoleret fra animalsk eller humant blod, især fra hesteblod, i en pufferopløsning med en pH-værdi på fra 7 til 8, centrifugerer det flydende homogenisat, adskiller de opløste bestand- 25 dele, der har en molekylvægt på mindre end 10.000, fra den ovenstående væske, underkaster denne fraktion en yderligere fraktionering, fortrinsvis ved gelchromatografi, fraskiller og, om ønsket, lyophiliserer olen fraktion, som fraskilles ved volumenværdier Ve/VQ på fra 1,15 til 30 1,45, eller b) homogeniserer animalske organer, der indeholder granulo-cyter, fortrinsvis kalvemilt, i et vandblandbart organisk opløsningsmiddel, adskiller faststofferne fra den opnåede flydende suspension, ekstraherer dem med et organisk 35 opløsningsmiddel, der er i stand til at opløse fedtstoffer, ekstraherer de således fremkomne faststoffer med DK 155712 B vand, fraskiller den uopløselige del fra den således fremkomne, flydende suspension, adskiller de opløste bestanddele, som har en molekylvægt på mindre end 10.000, fra den flydende fase, underkaster denne fraktion en 5 yderligere fraktionering, fortrinsvis ved gelchromatogra-fi, fraskiller og, om ønsket, lyophilisrer den fraktion, som fraskilles ved volumenværdier ve/vQ på fra 1,3 til 2,5, 10 og renser koncentratet af aktivt stof fremstillet ved fremgangsmådevariant a) eller b) ved en ionbytterchromatografi, som fraktionerer på basis af ionladning og partikelstørrelse, og/eller ved papirelektrophorese, fraskiller og eventuelt lyophiliserer et oligopeptid, som har en negativ lad- 15 ning ved en pH-værdi på 6,5 og en relativ mobilitet på -0,55 til -0,65 i forhold til asparaginsyre, som ikke har nogen ladning ved en pH-værdi på 1,9, men har en mobilitet på ca. 0,26 i forhold til e-dinitrophenyl-lysin, og som giver en positiv chlortoluidinreaktion.
2. Fremgangsmåde ifølge krav 1, kendetegnet ved, at bestanddelene med en molekylvægt på mindre end 10.000 fraskilles ved molekylfiltrering.
3. Fremgangsmåde ifølge krav 1, kendetegnet ved, at der som vandblandbart organisk opløsningsmiddel 25 anvendes acetone.
4. Fremgangsmåde ifølge krav 1,kendetegnet ved, at fraskillelsen af de faststoffer eller uopløselige dele, som findes i de flydende suspensioner, sker ved centrifugering.
5. Fremgangsmåde ifølge krav 1, kendeteg net ved, at der som organisk opløsningsmiddel, der er i stand til at opløse fedtstoffer, anvendes chlorerede carbon-hydrider.
