Erfindungsgem:.ß kann das Verfahren des Hauptpatents auf Grun.d der überraschenden Beobachtung, daß die Wanderungsgeschwindigkeit der Vitamine der Bl2-Gruppe an der Zellulosepulver-Säule während der Entwicklung mit cyamd- und wasserhaltigem n-Butanol stark beeinflußt werden kann, wenn der Entwickler geringe Mengen an Salzen enthält, wesentlich verbessert werden. Derartige Salze können außerdem die Trennschärfe, d. h. die Breite der einzelnen Zonen, beeinflussen. Viele anorganische Salze verursachen diffuse Zonen und erschweren somit die Trennung. Andere Salze jedoch bewirken eine Verschärfung der Zonen. Gew.isse Salze der Perchlorsäure, so z. B. das Kaliumperchlorat, besitzen völlig überraschend die Eigenschaft, in sehr geringer Konzentration auf die zu entwickelnden Zonen bremsend zu. wirken, und zwar in dem Sinne, daß die sich langsam auf der Chromatographiersäule bewegenden Zonen der Cobaiamine, wie z. B. der Cobalamincarbonsäuren, sowie des Vitamin B12-Faktors III, stark abgebremst werden. Die schnellen Zonen der Cobalamine, wie z. B. des Ätiocohalamins, werden dagegen weniger gebremst. Außerdem verursacht das im Entwickler gelöste Kaliumperchlorat eine beträchtliche Verschärfung (verschmälerung) der Zonen. Der Endeffekt ist der, daß bei Verwendung von Kaliumperchlorat sich das Auflösungsvermögen der Zellulosepuiver-Säule beträchtlich erhöht.
