Description
- Die vorliegende Erfindung betrifft eine bisher unbekannte Verbindungsklasse, welche eine starke Aktivität bezüglich der Inhibierung unerwünschter Zellproliferation beispielsweise von Hautzellen und Krebszellen aufweist, des weiteren pharmazeutische Zubereitungen, die diese Verbindungen enthalten, Dosierungseinheiten solcher Zubereitungen und Verbindungen zur Verwendung bei der Behandlung und Prophylaxe von Erkrankungen, die durch eine abnormale Zelldifferenzierung und/oder Zellproliferation gekennzeichnet sind, wie Psoriasis und Krebs.
- Die erfindungsgemäßen Verbindungen entsprechen der allgemeinen Formel I oder tautomeren Formen davon, worin die Bindung an den Pyridinring in der 3- oder 4-Position vorliegt und R1 für einen oder mehrere Substituenten steht, die unabhängig voneinander ausgewählt sind unter: Wasserstoff, Halogen, Trifluormethyl, Carboxy, C1-C4-Alkyl, -Alkoxy oder -Alkoxycarbonyl, Nitro, Amino oder Cyano, Q für einen zweiwertigen C4-C20-Kohlenwasserstoffrest steht, der geradkettig, verzweigt, cyclisch, gesättigt oder ungesättigt sein kann, X für Carbonyl, Carbonylamino, Aminocarbonyl, Oxycarbonyloxy, Oxycarbonyl, Carbonyloxy, Aminocarbonyloxy, Aminothiocarbonyloxy, Oxycarbonylamino oder Oxythiocarbonylamino steht, Y für Benzylen oder Phenylen steht und R2 für einen oder mehrere Substituenten steht, die unabhängig voneinander ausgewählt sind unter: Wasserstoff, C1-C4-Alkyl, -Hydroxyalkyl oder -Alkoxy, Hydroxy, Halogen, Trifluormethyl, Cyano, Carboxamido, Sulfamoyl, Nitro, Amino, Carboxy, Alkoxycarbonyl oder Benzyloxy.
- Wenn die erfindungsgemäßen Verbindungen ein oder mehrere asymmetrische Kohlenstoffatome enthalten, so können sie optische Isomere oder Diastereoisomere bilden. Die vorliegende Erfindung umfasst auch derartige Isomere sowie Gemische davon.
- Die erfindungsgemäßen Salze der Verbindungen der Formel I können mit pharmazeutisch akzeptablen anorganischen oder organischen Säuren, wie Salzsäure, Bromwasserstoffsäure und Iodwasserstoffsäure, Phosphorsäure, Schwefelsäure, Salpetersäure, 4-Toluolsulfonsäure, Methansulfonsäure, Ameisensäure, Essigsäure, Propionsäure, Zitronensäure, Weinsäure, Bernsteinsäure, Benzoesäure und Maleinsäure, gebildet werden.
- Die erfindungsgemäßen Salze der Verbindungen der Formel I können auch mit pharmazeutisch akzeptablen anorganischen oder organischen Basen gebildet werden. Bei den mit pharmazeutisch akzeptablen nichttoxischen Basen gebildeten Salzen kann es sich um Alkalimetallsalze und Erdalkalimetallsalze, z. B. Lithium-, Natrium-, Kalium-, Magnesium- oder Calciumsalze handeln sowie um Salze mit Ammoniak und geeigneten nichttoxischen Aminen, wie C1-C6-Alkylamine, z. B. Triethylamin, C1-C6-Alkanolamine, z. B. Diethanolamin oder Triethanolamin, Procain, Cycloalkylamine, z. B. Dicyclohexylamin, Benzylamine, z. B. N-Methylbenzylamin, N-Ethylbenzylamin, N-Benzyl-2-phenylethylamin, N,N'-Dibenzylethylendiamin oder Dibenzylamin, sowie heterocyclische Amine, z. B. Morpholin, N-Ethylpiperidin und dergleichen.
- Auch wenn die erfindungsgemäßen Verbindungen nach der enteralen Verabreichung gut absorbiert werden, kann es in manchen Fällen vorteilhaft sein, geeignete bioreversible Derivate der erfindungsgemäßen Verbindungen, d. h. so genannte Prodrugs, herzustellen; es handelt sich vorzugsweise um Derivate, deren physikalisch-chemischen Eigenschaften zu einer verbesserten Löslichkeit bei physiologischem pH und/oder einer verbesserten Absorption und/oder einer verbesserten Bioverfügbarkeit der betreffenden Verbindung führen.
- Bei diesen Derivaten handelt es sich beispielsweise um Pyridyl-N-oxid-Derivate der erfindungsgemäßen Verbindungen, wobei derartige Verbindungen durch die Oxidation des N-Atoms im Pyridylring mit einem geeigneten Oxidationsmittel, z. B. mit 3-Chlorperbenzoesäure in einem inerten Lösungsmittel, wie Dichlormethan, hergestellt werden.
- Es können auch andere geeignete Verfahren zur Verbesserung der physikalischchemischen Eigenschaften und/oder der Löslichkeit der betreffenden Verbindungen können angewandt werden.
- Die im britischen Patent Nr. 1,489,879 beschriebenen N-Alkyl-N'-cyano-N''-pyridylguanidine sind starke Kaliumkanalaktivatoren, welche eine ausgeprägte Wirkung als präkapillare Vasodilatatoren aufweisen und den totalen peripheren Widerstand beim Tier und beim Menschen herabsetzen und daher als Antihypertensiva geeignet sind. Wie in der internationalen Patentanmeldung PCT/DK 93/00291, Anmeldedatum 13. September 1993, Veröffentlichungsnummer WO 94/06770, behauptet wird, führt die Einführung Aryloxy-haltiger Reste in die aliphatischen Gruppen des oben genannten britischen Patents zu Strukturen, die im Vergleich zu der bekannten Wirkung von Verbindungen des oben genannten britischen Patents spezifischere pharmakologische Wirkungen auf isolierte Gewebe und Zellen aufweisen und außerdem keine oder eine nur vernachlässigbare Wirkung auf den 86Rb-Austritt aus Kaliumkanälen besitzen.
- Die erfindungsgemäßen Verbindungen inhibieren in Kultur die Proliferation unterschiedlicher Tumorzelllinien in niedrigeren Konzentrationen als die bekannten Verbindungen, wie nachstehende Tabelle 1 zeigt, was sie für die antineoplastische Chemotherapie potentiell geeignet macht.
- Die Inhibierung der Tumorzellenproliferation wurde unter Verwendung unterschiedlicher Typen von menschlichen Krebszelllinien untersucht. Bei den untersuchten Zelllinien handelte es sich um kleinzelliges Lungenkarzinom (NYH), nicht-kleinzelliges Lungenkarzinom (NCI-H460) und Brustkrebs (MCF-7), wobei folgendes allgemeine Verfahren angewandt wurde:
- Die Zellen wurden in Gegenwart der untersuchten Verbindung 24 Stunden in vitro kultiviert. Die DNA-Synthese wurde durch den Einbau von [3H]Thymidin bestimmt und die mittleren inhibierenden Konzentrationen (IC50) der Verbindungen wurden berechnet.

- Die Ergebnisse zeigen, dass die erfindungsgemäßen Verbindungen die Proliferation von Tumorzellen in vitro bei den gleichen oder niedrigeren Konzentrationen inhibieren als die Verbindungen A und B aus PCT/DK 93/00291.
- Die erfindungsgemäßen Verbindungen werden gut vertragen, sind nichttoxisch und üben die beschriebenen positiven Wirkungen ohne oder mit nur minimaler Auswirkung auf den systemischen Blutdruck aus. In der Regel können sie auf oralem, intravenösem, intraperitonealem, intranasalem oder transdermalem Weg verabreicht werden.
- Die vorliegende Erfindung betrifft auch Verfahren zur Herstellung der gewünschten Verbindungen der allgemeinen Formel 1. Die Verbindungen der Formel 1 können praktischerweise durch herkömmliche Verfahren des Standes der Technik hergestellt werden. Die Reaktionswege sind im nachfolgenden Reaktionsschema dargestellt. a) DCCD, NH2CN, Et3N, CH3CN, 2 Wochen (allgemeines Verfahren 1).
b) DMAP, Et3N, Pyridin, 55°C, 3 h (allgemeines Verfahren 2).
c) Pyridin, 0–5°C (allgemeines Verfahren 3). - Die erfindungsgemäßen Verbindungen sollen in pharmazeutischen Zusammensetzungen eingesetzt werden, die für die Behandlung der oben genannten Erkrankungen geeignet sind.
- Die zur Erzielung einer therapeutische Wirkung erforderliche Menge einer Verbindung der Formel I (welche im Folgenden als Wirkstoff bezeichnet wird) hängt selbstverständlich sowohl von der einzelnen Verbindung als auch vom Verabreichungsweg und vom zu behandelnden Säuger ab. Eine für die systemische Behandlung geeignete Dosis einer Verbindung der Formel I beträgt 0,1 bis 400 mg pro Kilogramm Körpergewicht, wobei eine Dosis 1,0 bis 100 mg pro Kilogramm Körpergewicht des Säugers, z. B. 5 bis 20 mg/kg, besonders bevorzugt ist und wobei die Dosis einmal oder mehrmals am Tag verabreicht werden kann.
- Eine Tagesdosis (für Erwachsene) kann 1 bis 10000 mg, vorzugsweise 70 bis 5000 mg betragen; entsprechendes gilt in der Veterinärmedizin, wobei die tägliche Dosis 0,1 bis 400 mg/kg Körpergewicht beträgt.
- Auch wenn der Wirkstoff allein in Rohform verabreicht werden kann, ist es dennoch bevorzugt, ihn als pharmazeutische Formulierung bereitzustellen. Geeigneterweise liegt der Wirkstoff in einer Menge von 0,1 bis 99 Gew.-%, bezogen auf die Formulierung, vor. Geeigneterweise enthalten Dosierungseinheiten einer Formulierung 0,5 mg bis 1 g Wirkstoff. Bei der topischen Verabreichung liegt der Wirkstoff in einer Menge von vorzugsweise 1 bis 20 Gew.-%, bezogen auf die Formulierung, vor, wobei der Wirkstoff auch bis zu 50 Gew.-% ausmachen kann. Für die nasale oder buccale Verabreichung geeignete Formulierungen können 0,1 bis 20 Gew.-%, z. B. etwa 2 Gew.-%, Wirkstoff enthalten.
- Der Begriff „Dosierungseinheit" bezeichnet eine standardisierte Dosis, d. h. eine Einzeldosis, die einem Patienten verabreicht werden kann, die außerdem leicht handhabbar ist und verpackt werden kann und dabei eine physikalisch und chemisch stabile Einheitsdosis bleibt, welche entweder den Wirkstoff als solchen oder im Gemisch mit festen oder flüssigen pharmazeutischen Verdünnungsmitteln oder Trägern enthält.
- Die erfindungsgemäßen Formulierungen umfassen sowohl bei der Anwendung im Veterinärbereich als auch in der Humanmedizin einen Wirkstoff in Kombination mit einem pharmazeutisch akzeptablen Träger sowie gegebenenfalls weitere therapeutische Bestandteile. Der/die Träger müssen dahingehend „akzeptabel" sein, dass sie mit den anderen Formulierungsbestandteilen kompatibel und für den Rezipienten nicht schädlich sind.
- Die Formulierungen umfassen für die orale, rektale und parenterale (einschließlich subkutane, intramuskuläre, intravenöse und intraperitoneale) Verabreichung geeignete Formen.
- Die Formulierungen können geeigneterweise in Form von Dosierungseinheiten bereitgestellt werden und können durch alle auf dem Gebiet der Pharmazie bekannten Verfahren hergestellt werden. Bei allen Verfahren wird der Wirkstoff mit dem Träger, welcher einen oder mehrere Zusatzbestandteile darstellt, in Kontakt gebracht. In der Regel werden die Formulierungen hergestellt, indem man dem Wirkstoff mit einem flüssigen Träger oder mit einem fein zerteilten festen Träger oder mit beiden einheitlich und innig in Kontakt bringt und anschließend, wenn erforderlich, das Produkt zu der gewünschten Formulierung formt.
- Erfindungsgemäße Formulierungen, die für die orale Verabreichung geeignet sind, können in Form diskreter Einheiten, wie Kapseln, Sachets, Tabletten oder Pastillen, die jeweils eine vorbestimmte Wirkstoffmenge enthalten, in Form eines Pulvers oder Granulats, in Form einer Lösung oder Suspension in einer wässrigen oder nicht wässrigen Flüssigkeit oder in Form einer Öl-in-Wasser-Emulsion oder Wasser-in-Öl-Emulsion, vorliegen. Der Wirkstoff kann auch in Form eines Bolus, eines Electuariums oder einer Paste verabreicht werden.
- Formulierungen für die rektale Verabreichung können in Form eines Zäpfchens, welches den Wirkstoff und einen Träger, wie Kakaobutter, enthält, oder in Form eines Klistiers vorliegen.
- Formulierungen, die für die parenterale Verabreichung geeignet sind, umfassen geeigneterweise eine sterile ölige oderwässrige Wirkstoffzubereitung, welche vorzugsweise mit dem Blut des Rezipienten isotonisch ist.
- Neben den zuvor genannten Bestandteilen können die erfindungsgemäßen Formulierungen wenigstens einen weiteren Bestandteil, beispielsweise Verdünnungsmittel, Puffer, Aromastoffe, Bindemittel, oberflächenaktive Stoffe, Verdicker, Gleitmittel, Konservierungsmittel, z. B. Methylhydroxybenzoat (einschließlich Antioxidantien), Emulgatoren und dergleichen, enthalten.
- Die Zusammensetzungen können außerdem weitere therapeutisch wirksame Verbindungen, die üblicherweise bei der Behandlung der zuvor genannten pathologischen Zustände eingesetzt werden, enthalten, z. B. antineoplastische Mittel, die zu einer synergistischen Wirkung auf Tumorzellen führen können.
- Die Erfindung wird durch die folgenden allgemeinen Verfahren, Zubereitungen und Beispiele näher beschrieben.
- Die in den Beispielen verwendeten Verbindungen I sind in Tabelle 2 aufgeführt.
- Alle Schmelzpunkte sind nicht korrigiert. Bei den 1H- und 13C-Kernresonanzspektren (NMR) (300 MHz) sind die Werte der chemischen Verschiebung (δ) von Deuterochloroform- und Hexadeuterodimethylsulfoxidlösungen gegenüber Chloroform (1H-NMR δ 7,25; 13C-NMR δ 76,81) oder Tetramethylsilan (δ 0,00) als internem Standard angegeben, sofern nichts Gegenteiliges vermerkt ist. Der Wert eines definierten (Dublett (d), Triplett (t) oder Quartett (q)) oder nicht definierten Multipletts ist als ungefährer Mittelpunktswert angegeben, es sei denn, es ist ein Bereich angegeben (s Singulett, b breit). Die Chromatographie erfolgte über Kieselgel. Es werden die folgenden Abkürzungen und Formeln verwendet: DCCD (N,N'-Dicyclohexylcarbodümid), NH2CN. (Cyanamid), Et3N (Triethylamin), CH3CN (Acetonitril), DMAP (Dimethylaminopyridin), MeOH (Methanol), CH2Cl2 (Methylenchlorid), NH3 (Ammoniak), CDCl3 (Deuterochloroform) und DMSO-d6 (Hexadeuterodimethylsulfoxid).
- Allgemeines Verfahren 1: Überführung von Verbindungen der allgemeinen Formel II in Verbindungen der allgemeinen Formel I
- Zu einer in Acetonitril (12 ml) suspendierten Verbindung der allgemeinen Formel II (5 mmol) gab man Dicyclohexylcarbodümid (10 mmol), Cyanamid (10 mmol) und Triethylamin (0,07 ml). Das Reaktionsgemisch wurde 2 Wochen bei Raumtemperatur gerührt.
- Das Reaktionsgemisch wurde filtriert und mit Acetonitril gewaschen. Der weiße Feststoff, der das Produkt und Dicyclohexylthioharnstoff enthielt, wurde mit Chloroform (20 ml) über Nacht verrieben, wobei man nach der Filtration das Produkt der allge- meinen Formel I in Form weißer Kristalle erhielt.
- Allgemeines Verfahren 2: Kupplung von Verbindungen der allgemeinen Formel III mit Verbindungen der allgemeinen Formel IV zu Verbindungen der allgemeinen Formel I
- Eine Verbindung der allgemeinen Formel III (4 mmol), eine Verbindung der allgemeinen Formel IV (5 mmol), Triethylamin (0,12 ml) und 4-Dimethylaminopyridin (15 mg) wurden in Pyridin (4 ml) gelöst. Das Reaktionsgemisch wurde 3 Stunden bei 55°C gerührt und dann auf Raumtemperatur gekühlt.
- Das Produkt wurde mit Ether ausgefällt. Nach der Filtration erhielt man das reine Produkt der allgemeinen Formel I in Form weißer Kristalle.
- Allgemeines Verfahren 3: Kupplung von Verbindungen der allgemeinen Formel V mit Verbindungen der allgemeinen Formel VI zu Verbindungen der allgemeinen Formel I
- In einem Zweihalskolben mit Dreiwegehahn und Septum wurde eine Verbindung der allgemeinen Formel V (2,0 mmol) in trockenem Pyridin (4 ml) suspendiert. Die Luft wurde über den Dreiwegehahn durch Argon ersetzt. Der Kolben wurde auf Eis gekühlt und eine Verbindung der allgemeinen Formel VI (3,0 mmol) wurde über das Septum hinzugefügt. Das Reaktionsgemisch wurde 2 Stunden bei 0°C und 5 Tage bei 5°C gerührt.
- Das Produkt wurde mittels Flash-Chromatographie (Eluierungsmittel: 1% NH3(aq) und 0–20% MeOH in CH2Cl2) gereinigt und aus Ether kristallisiert.
- Beispiel 1: N-Cyano-N'-(5-phenylaminocarbonylpentyl)-N''-4-pyridylguanidin (Verbindung 101)

13C-NMR (CDCl3) δ: 157,7, 153,5, 150,7, 145,0, 149,6, 131,4, 129,6, 122,4, 117,1, 116,0, 68,8, 42,4, 29,0, 28,4, 26,3, 25,3. - Beispiel 6: N-(6-(4-Chlorphenoxycarbonyl)pentyl)-N'-cyano-N''-4-pyridylguanidin (Verbindung 106)
- N-(5-Carboxypentyl)-N'-cyano-N''-4-pyridylguanidin (2,0 g), 4-Chlorphenol (0,94 g), Triethylamin (2,0 ml) und Brom-tris-pyrrolidinophosphoniumhexafluorophosphat (3,4 g) sowie trockenes Dichlormethan (100 ml) wurden gemischt und über Nacht bei Raumtemperatur gerührt. Man fügte weiteres Brom-tris-pyrrolidinophosphoniumhexafluorophosphat (0,68 g) und Triethylamin (0,2 ml) hinzu und rührte das Reaktionsgemisch über Nacht.
- Man dampfte die Lösungsmittel ab und reinigte das Reaktionsgemisch mittels Flash-Chromatographie (Eluierungsmittel: 0,5% NH3(aq) und 0–17% MeOH in CH2Cl2). Die Kristallisation aus Ether ergab weiße Kristalle, die sowohl das Produkt als auch ein Triethylammoniumsalz enthielten. Das Triethylammoniumsalz wurde durch Extraktion mit Wasser entfernt, wobei man das reine Produkt erhielt.
13C-NMR (DMSO-d6) δ: 171,5, 157,1, 150,1, 149,1, 145,7, 129,8, 129,3, 123,7, 116,4, 114,6, 41,5, 33,2, 28,2, 25,5, 23,8. - Beispiel 7: N-(6-(4-Chlorphenylaminothiocarbonyloxy)hexyl)-N'-cyano-N''-4-pyridylguanidin (Verbindung 107)
- In einem Zweihalskolben mit Dreiwegehahn und Septum wurden N-Cyano-N'-(6-hydroxyhexyl)-N''-4-pyridylguanidin (0,5 g), 4-Chlorphenylisothiocyanat (0,4 g) und Diisopropylethylamin (0,32 ml) gemischt. Die Luft wurde über den Dreiwegehahn durch Argon ersetzt. Trockenes Pyridin (10 ml) wurde über das Septum hinzugefügt. Das Reaktionsgemisch wurde 5 Tage bei Raumtemperatur gerührt.
- Das Produkt wurde mittels Flash-Chromatographie (Eluierungsmittel: 1% NH3(aq) und 0–10% MeOH in CH2Cl2) gereinigt und aus Ether kristallisiert.
1H-NMR (DMSO-d6) δ: 11,16 (bs, 1H), 9,37 (bs, 1H), 8,38 (bd, 2H), 7,84 (bt, 1H), 7,40 (m, 4H), 7,22 (bd, 2H), 4,46 (m, 2H), 3,26 (q, 2H), 1,72 (m, 2H), 1,54 (m, 2H), 1,36 (m, 4H). - Beispiel 8: N-(6-(4-Chlorbenzoylamino)hexyl)-N'-cyano-N''-4-pyridylguanidin (Verbindung 108)

- Ausgangsverbindung III: S-Methyl-N-cyano-N'-4-pyridylisothioharnstoft.
- Ausgangsverbindung IV: 4-Chlorbenzoylhexylamin.
- Reinigung: Nach dem allgemeinen Verfahren. Beispiel 14: Kapseln:
1 Kapsel enthält: N-(6-(4-Chlorbenzoyloxy)hexyl)-N'-cyano-N''-4-pyridylguanidin (Wirkstoff) 100 mg Polyethylenglykol 962 mg Gelatinekapsel Nr. 00 Gelatine 122 mg - I wird in einem stark scherenden Mischer innig vermischt, mit II angefeuchtet und zu einer feuchten Masse granuliert. Das feuchte Granulat wird in einem Fließbetttrockner mit einer Lufteinlasstemperatur von 60°C getrocknet, bis das getrocknete Granulat eine Wasseraktivität von 0,3–0,4 (= im Gleichgewicht mit Luft einer relativen Feuchte von 30–40%) aufweist.
- Das getrocknete Granulat wird durch ein Sieb mit Mesh-Öffnungen von 850 μm gestrichen.
- Das gesiebte Granulat wird schließlich mit III in einem Kegelmischer gemischt.
- Das fertige Granulat wird zu Tabletten der Masse 1071 mg und ausreichender Härte komprimiert.
Claims (8)
- Verbindung der Formel I oder ihre tautomeren Formen, worin die Bindung an den Pyridinring in der 3-oder 4-Position vorliegt und R1 für einen oder mehrere Substituenten steht, die unabhängig voneinander ausgewählt sind unter: Wasserstoff, Halogen, Trifluoromethyl, Carboxy, C1-C4-Alkyl, -Alkoxy oder -Alkoxycarbonyl, Nitro, Amino oder Cyano, Q für einen zweiwertigen C4-C20-Kohlenwasserstoffrest steht, der geradkettig, verzweigt, cyclisch, gesättigt oder ungesättigt sein kann, X für Carbonyl, Carbonylamino, Aminocarbonyl, Oxycarbonyloxy, Oxycarbonyl, Carbonyloxy, Aminocarbonyloxy, Aminothiocarbonyloxy, Oxycarbonylamino oder Oxythiocarbonylamino steht, Y für Benzylen oder Phenylen steht und R2 für einen oder mehrere Substituenten steht, die unabhängig voneinander ausgewählt sind unter: Wasserstoff, C1-C4-Alkyl, -Hydroxyalkyl oder -Alkoxy, Hydroxy, Halogen, Trifluormethyl, Cyano, Carboxamido, Sulfamoyl, Nitro, Amino, Carboxy, Alkoxycarbonyl oder Benzyloxy; und pharmazeutisch akzeptable nichttoxische Salze und N-Oxide davon.
- Verbindung der Formel 1 nach Anspruch 1, worin die Bindung an den Pyridinring in der 4-Position vorliegt und R1 für Wasserstoff steht, Q für einen zweiwertigen C4-C12-Kohlenwasserstoffrest, der geradkettig, verzweigt, gesättigt oder ungesättigt sein kann, X für Carbonyl, Carbonylamino, Aminocarbonyl, Oxycarbonyloxy, Oxycarbonyl, Carbonyloxy, Aminocarbonyloxy, Aminothiocarbonyloxy, Oxycarbonylamino oder Oxythiocarbonylamino steht, Y für Benzylen oder Phenylen steht, und R2 für einen oder mehrere Substituenten steht, die unabhängig voneinander ausgewählt sind unter: Wasserstoff, C1-C4-Alkyl oder -Alkoxy, Halogen, Trifluormethyl oder Cyano; und pharmazeutisch akzeptable nichttoxische Salze und N-Oxide davon.
- Salz nach Anspruch 1, wobei das Salz ausgewählt ist unter mit Salzsäure, Bromwasserstoffsäure, Jodwasserstoffsäure, Phosphorsäure, Schwefelsäure, Salpetersäure, p-Toluolsulfonsäure, Methansulfonsäure, Ameisensäure, Essigsäure, Propionsäure, Zitronensäure, Weinsäure und Äpfelsäure gebildeten Salzen und den Lithium-, Natrium-, Kalium-, Magnesium- und Calciumsalzen sowie den Salzen mit Ammoniak, C1-C6-Alkylaminen, C1-C6-Alkanolaminen, Procain, Cycloalkylaminen, Benzylaminen und heterocyclischen Aminen.
- Verbindung nach Anspruch 1, ausgewählt unter: N-(6-Benzoylaminohexyl)-N'-cyano-N''-4-pyridylguanidin; N-(6-(4-Chlorbenzoyloxy)hexyl)-N'-cyano-N''-4-pyridylguanidin; N-(5-(4-Chlorbenzoyl)pentyl)-N'-cyano-N''-4-pyridylguanidin; N-(7-(4-Chlorbenzoyl)heptyl)-N'-cyano-N''-4-pyridylguanidin; und ihre Salze und reinen enantiomeren Formen.
- Pharmazeutische Zubereitung, enthaltend eine Verbindung nach einem der Ansprüche 1 bis 4 alleine oder zusammen mit den erforderlichen Hilfsstoffen.
- Wirksame Menge einer oder mehrerer Verbindungen nach einem der Ansprüche 1 bis 5, gegebenenfalls zusammen mit oder begleitet von einem oder mehreren anderen Wirkstoffen, zur Verwendung zur Behandlung und Prophylaxe von Erkrankungen, die durch unerwünschte Zellproliferation z. B. in Hautzellen oder Krebszellen charakterisiert sind.
- Verfahren zur Herstellung einer Verbindung der Formel 1 nach Anspruch 1, bei dem man a) eine Verbindung der allgemeinen Formel II worin R1, Q, X, Y und R2 die oben angegebene Bedeutung haben, mit Dicyclohexylcarbodümid und Cyanamid in Gegenwart von Triethylamin oder einem anderen tertiären Amin in Acetonitril oder einem anderen inerten Lösungsmittel bei Raumtemperatur oder darüber umsetzt, oder b) eine Verbindung der allgemeinen Formel III worin R1 die oben angegebene Bedeutung hat, mit einer Verbindung der allgemeinen Formel IV worin Q, X, Y und R2 die oben angegebene Bedeutung haben, in Gegenwart von Triethylamin oder einem andern tertiären Amin und 4-Dimethylaminopyridin in Pyridin oder einem inerten Lösungsmittel bei Raumtemperatur oder darüber umsetzt; oder c) eine Verbindung der allgemeinen Formel V worin R1 und Q die oben angegebene Bedeutung haben und A für Sauerstoff, Schwefel oder durch Wasserstoff substituierten Stickstoff steht, mit einer Verbindung der allgemeinen Formel VI worin Y und R2 die oben angegebene Bedeutung haben und B für Carbonyl oder Carbonyloxy steht, in Pyridin oder einem inerten Lösungsmittel bei 0°C oder darüber umsetzt.
- Verwendung einer Verbindung nach Anspruch 1 zur Herstellung eines Arzneimittels zur Behandlung und Prophylaxe verschiedener Erkrankungen, die durch unerwünschte Zellproliferation z. B. in Hautzellen und Krebszellen gekennzeichnet sind.
