Verfahren zur Gewinnung eines den Blutkreislauf und die Herztätigkeit beeinflussenden Stoffes aus Pankreas In dem Patent 518 292 ist ein Verfahren zur Gewinnung eines den Blutkreislauf und die Herztätigkeit beeinflussenden Stoffes aus Harn beschrieben. Dieses Produkt ist dadurch charakterisiert, daß es eine Senkung des Blutdruckes bei gleichbleibendem und vermehrtem Schlagvolumen in Verbindung mit vermehrter Durchblutung von Lunge, Gehirn und Muskulatur bewirkt.Method for obtaining a substance from the pancreas that influences blood circulation and cardiac activity. This product is characterized by the fact that it brings about a lowering of the blood pressure with constant and increased stroke volume in connection with increased blood flow to the lungs, brain and muscles.
Es ist nun gefunden worden, daß das, gleiche Produkt auch in der Pankreasdrüse enthalten ist und .aus ihr gewonnen werden kann. Das Verfahren muß so geleitet werden, daß die übrigen bekannten Stoffe des Pankreas, besonders das Insulin, von der auf den Kreislauf wirkenden Substanz abgetrennt werden. Das gelingt durch die Löslichkeit des Insulins in nicht zu konzentrierten Mischungen von Wasser mit organischen Flüssigkeiten. Man versetzt dazu alkalische oder saure wäßrige Extrakte aus. der frischen oder der getrockneten Pankreasdrüse mit wasserlöslichen organischen Flüssigkeiten wie Alkohol, Aceton oder deren Homologen in dem Maße, daß die für Insulin bekannte Fällungskonzentration noch nicht erreicht wird. Dabei beträgt zweckmäßig die Konzentration an Alkohol etwa 7o bis 8o Volumprozent, die an Aceton etwa 70 Volumprozent. Man erhält so eine Fällung, die durch Umfällen oder Adsorption mit den üblichen Fällungs-oder Adsorptionsmitteln z. B. in der im Hauptpatent angegebenen Weise weiter gereinigt werden kann. Beispiel 1 kg getrockneter Pankreasdrüse wird mit 2o 1 Wasser 12 Stunden geschüttelt. Man filtriert vom Ungelösten ab und versetzt das Filtrat mit 8o 1 Äthylalkohol (9o Volumprozent). An Stelle von Äthylalkohol kann man z. B. auch etwa 7o bis 8o 1 Methylalkohol oder etwa, 6o 1 Aceton zur Fällung des wäßrigen Extraktes verwenden. Der gebildete Niederschlag wird abzentrifugiert und in to 1 Wasser gelöst. In die salzfrei dialysierte Lösung gießt man unter Umrühren den zwanzigsten Teil des Volumens gesättigte alkoholische Benzoesäurelösung ein, zentrifugiert und löst aus dem Niederschlag die Benzoesäure mit Alkohol und Äther heraus. Eine weitere Reinigung kann sowohl durch Fällungsmittel, wie Uranylacetat, als auch durch Adsorptionsmittel, wie Kaolin, Kohle u. dgl., nach den Beispielen des Hauptpatents 518a 292 erzielt werden.It has now been found that the same product is also contained in the pancreatic gland and can be obtained from it. This gives a precipitate which, by reprecipitation or adsorption with the usual precipitants or adsorbents, for. B. can be further cleaned in the manner indicated in the main patent. Example 1 kg of dried pancreatic gland is shaken with 2o 1 of water for 12 hours. The undissolved material is filtered off and 8o 1 of ethyl alcohol (90 percent by volume) are added to the filtrate. Instead of ethyl alcohol you can, for. B. also use about 7o to 8o 1 methyl alcohol or about 6o 1 acetone to precipitate the aqueous extract. The precipitate formed is centrifuged off and dissolved in 1 to 1 of water. One twentieth of the volume of saturated alcoholic benzoic acid solution is poured into the salt-free dialyzed solution with stirring, centrifuged and the benzoic acid is dissolved out of the precipitate with alcohol and ether. Further purification can be achieved both by precipitants, such as uranyl acetate, and by adsorbents, such as kaolin, coal and the like, according to the examples of main patent 518a 292 .
