Description
Method for the preparation of # solutions with low or low cholesterol the active ovarian substance In the study of the nature of the active substance A distinction is made today between the placenta and the ovary in two directions.
W. 1925, Nr. 25; D o d ds und D i ck e n s, Bio. JOurn. 1925, ig S.853; H a r t m a n n, Bio. 175, 47), während andere Forscher (Laqueur, Deutsche rned. Wochensch. 1926, 1 u. 2; Zondek, Klin. Wochensch. Due to the fact that it is possible to produce effective extracts which, on the one hand, have lipoid character and, on the other hand, show a certain amount of water solubility, a number of researchers consider the lipoid-like character of the active substance to be essential (F aust, Schw. Med. W. 1925, No. 25; D o d ds and D i ck ens, Bio. JOurn. 1925, ig p.853; H artmann, Bio. 175, 47), while other researchers (Laqueur, Deutsche rned. Wochensch. 1926, 1 and 2; Zondek, Klin. Wochensch. 1926, No. 27) consider the active substance to be soluble in water and attribute the lipoid properties to still existing dietary fiber.
Wochensch. ig?6, Nr. 33, 1521).In accordance with these notions, two types of solutions of ovarian secretion have been used: an oil solution and an aqueous solution. The former4 has the advantage of greater potency, while tolerability is insufficient '(Zondek, Klin. Wochensch. 1926, No. 27, p. 1219); the latter, conversely, shows good tolerance, while the effectiveness is very limited, namely only 4 to 10 U per cubic centimeter. It has by no means been proven that there are true solutions in the aqueous extracts, since such commercial products usually cause turbidity with water (Zondek, Klin. Wochensch. Ig? 6, No. 33, 1521).
For this reason there is a need for an ovarian preparation that the greater effectiveness of the previously oil-soluble with the good tolerance of the aqueous preparations combined.
It has now been found that a preparation with the advantages of aqueous and oil- soluble preparations can be obtained by using water-soluble ethers and esters of polyhydric alcohols as solvents. These solvents, by virtue of their solubility in water and their lipid-dissolving power, make it possible to produce extracts of high concentration of the active substance. The use of these solvents offers another important advantage. It has been found that by treating the crude extract obtained after saponification of the fats and cholesterol esters with the solvents mentioned, with the addition of certain amounts of water, almost all of the cholesterol which is insoluble in these solvents is separated and can be easily removed by filtration . In this way it is possible, solutions of the active substance at any concentration and display of good compatibility, be diluted with water can 'without precipitations or turbidity to - show.
The crude alkaline extract obtained by saponification of the fats and cholesterol esters according to the usual methods is taken up with water and extracted with ether. The dried ether extract is concentrated and the residue is taken up with 40 to 60 cc of diethyline per kilo of ovary. After adding 30 to 50 cc of water (depending on the amount of cholesterol present in the extract), all of the cholesterol precipitates in crystalline, pure form and is filtered at o '. The filtrate contains the active substance in purified form and can be used in any concentration.
