Description
Die Erfindung betrifft Verfahren zur enantioselektiven
Synthese von pharmakologisch wirksamen Sesquiterpenen, wie sie
in den zugehörigen Ansprüchen 1-28 gekennzeichnet ist.
Pharmakologisch wirksame Sesquiterpene mit einem Eremophilan-
Grundgerüst sind als Inhaltsstoffe der Pestwurz bekannt. Als
Wirkstoffe wurden bislang die Verbindungen Petasin, Petasol,
Isopetasin und Isopetasol isoliert (a) A. Aebi, J. Büchi, T.
Waaler, E. Eichberger, Pharm. Acta Helv., 1955, 29, 277-278.
b) A. Stoll, R. Morf, A. Rheiner, J. Renz, Experientia, 1956,
12, 360-361.). Aus der EP-A 281 656 ist die Verwendung eines
Pestwurzextraktes zur Behandlung von gastrointestinalen
Erkrankungen und aus der DE-A 42 08 300 die Verwendung von
Oxopetasin zur Behandlung gastrointestinaler Erkrankungen
bekannt. Ebenfalls bekannt ist die Anwendung von Pestwurz
extrakten als Spasmolytikum und Analgetikum. Es ist bisher
jedoch kein Verfahren beschrieben worden, wodurch enantiose
lektiv die genannten Isomere herstellbar wären. Die aus C.A.
79, (1973), Nr. 105 427 d bekannte Dissertation berichtet
lediglich Mißerfolge bei dem Versuch, derartige Isomere
stereoselektiv zu synthetisieren.
Aufgrund des breiten Wirkungsspektrums, insbesondere aufgrund
der Fähigkeit in die Leukotrienbiosynthese einzugreifen, sind
die Inhaltsstoffe der Pestwurz von erheblichem medizinischem
und pharmakologischem Interesse.
Bisher wurden die Wirkstoffe in Form eines Extraktes aus der
Pestwurz als Pharmazeutikum eingesetzt. Dies ist mit einer
Reihe von Nachteilen verbunden, wobei insbesondere die geringe
Konzentration der Wirkstoffe in der Pflanze und die Verunrei
nigung des Extraktes durch medizinisch nicht unbedenkliche
Nebenprodukte zu nennen sind.
Auch die Reindarstellung der einzelnen Wirkstoffe aus der Pflanze
ist schwierig und ökonomisch nachteilig.
Eine Aufgabe der vorliegenden Erfindung ist es daher ein Verfahren
zur enantioselektiven Synthese der wirksamen Inhaltsstoffe der
Pestwurz zu schaffen, das die gewünschten Stoffe in hoher Reinheit,
hoher Ausbeute und ökonomisch sinnvoll herzustellen erlaubt.
Die Synthese von (±)-Isopetasol (4) ist bekannt (a) K.

Bohlmann, W. Otto, Liebigs Ann. Chem., 1982, 186-190.), Mühle, H.
Neuenschwander, M. "Total synthesis of (±)-Petasol and (±)-
Isopetasol" Chimia 37(2), 41-44 (1983). Die Verbindung konnte
durch die beschriebenen Verfahren jedoch nur als Racemat erhalten
werden. Eine Trennung in die enantiomeren Formen erwies sich als
nicht praktikabel. Da die pharmakologische Wirkung der Substanzen
aber konfigurationsabhängig ist, konnten mit dem beschriebenen
Verfahren hergestellte Substanzen pharmakologisch nicht verwendet
werden.
Die absolute Konfiguration der Verbindungen gemäß Formel (I) ist
für die pharmakologische Wirkung von essentieller Bedeutung.
Daher ist eine weitere Aufgabe der Erfindung ein Verfahren zur
Herstellung von Verbindungen der allgemeinen Formel (I) in
enantiomerenreiner Form anzugeben.
Gegenstand der Erfindung ist somit ein Verfahren zur
Herstellung von Verbindungen der Formel (I):
worin R¹ eine Isopropenyl- oder Isopropylidengruppe, die
gegebenenfalls über die O-Brücke mit dem vorhandenen
Sauerstoffatom einen Ring bildet und R² Wasserstoff, Hydroxy,
ein verzweigter oder unverzweigter, gesättigter oder
ungesättigter Alkylester oder Arylester oder Alkyl- oder
Arylether mit 1 bis 20 C-Atomen, der gegebenenfalls substituiert sein
kann, bedeutet, bei dem man
- (a) vom bekannten enantiomerenreinen (1S,4R)-(-)-1-Acetoxycyclo hex-2-en-4-ol 5 ausgeht und nach Schützung des Alkohols sowie Esterspaltung und Oxidation in an sich bekannter Weise des OH- geschützte (4R)-(+)-Hydroxy-2-cyclohexen-1-on 8 herstellt (Figur 1);
- (b) die Verbindung 8 in einer Tandem-Michael-Addition in einer Kupfer-katalysierten Addition eines Methyl-Aniones in einem aprotischen Lösungsmittel oder einem Lösungsmittelgemisch bei einer Temperatur zwischen -95 und 0°C und Abfangen des intermediären Enolates mit einem Methyl-Elektrophil in Gegenwart eines polaren aprotischen Additives bei -95 bis +10°C zu dem Keton 9 umsetzt;
- (c) das Keton 9 über das Imin an C-2 mit einem Elektrophil bei 0 bis 150°C in einem aprotischen Lösungsmittel alkyliert;
- (d) den so erhaltenen Ester 10 zu der Keto-Carbonsäure 11 in an sich bekannter Weise spaltet;
- (e) die Keto-Carbonsäure 11 mit Triphenyl[(phenylimino)ethenyliden]phosphoran unter Zusatz eines Alkohols in einem aprotischen Lösungsmittel bei einer Temperatur von 20 bis 150°C zu der Verbindung 12 cyclisiert;
- (f) die Verbindung 12 in der 4-Position mit einer nicht nucleophilen Base in einem aprotischen Lösungsmittel bei einer Temperatur von -100 bis 0°C deprotoniert und mit einer Carbonylverbindung in einem aprotischen Lösungsmittel unter Zusatz einer Lewis-Säure bei -100 bis 0°C zu 13 alkyliert;
- (g) den so erhaltenen Alkohol 13 über einen intermediären Sulfonsäureester unter Zusatz einer Base bei -78 bis 50°C eliminiert und die Doppelbindung gegebenenfalls unter Säurekatalyse zu 14 isomerisiert;
- (h) die Alkohol-Schutzgruppe von der Verbindung 14 in an sich bekannter Weise entfernt und
- (i) den freien Alkohol in an sich bekannter Weise in den gewünschten Ester überführt.
Beispiele für die Alkylester sind Formyl-, Acetyl-, Propionyl-,
Butyryl-, Isobutyryl-, Pivalyl-, Angeloyl-, 3-Thio-methylacryl-,
Crotonyl-, Methacryl-, Tiglinyl-, Stearyl-, Palmityl-, Eicosyl
ester.
Beispiele für die Arylester sind Benzoylester, die gegebenenfalls
o, m und/oder p- durch Hydroxy, C1-4-Alkoxy, C1-4-Alkyl, C1-4-Acyl,
Acetoxy und/oder Halogen substituiert sind.
Beispiele für die Alkylether sind Methyl-, Ethyl-, Propyl-,
Isopropyl-, Butyl-, Isobutyl- und tert-Butylether.
Beispiele für die Arylether sind Phenyl- und Benzylether, die
gegebenenfalls o, m und/oder p- durch Hydroxy, C1-4-Alkoxy, C1-4-
Alkyl, C1-4-Acyl, Acetoxy und/oder Halogen substituiert sind.
Bevorzugt sind die Verbindungen, worin R² in der Formel (I)
Angeloyl bedeutet. Besonders bevorzugt sind die folgenden
Verbindungen.
Die Verbindungen 1 bis 4 sind als solche bekannt.
Den enantiomerenreinen Alkohol (1S,4R)-(-)-1-Acetoxycyclohex-2-
en-4-ol 5 erhält man nach R. J. Kazlauskas, et al., Org. Chem.,
1991, 58, 2656 bis 2665.
5 wird durch die Schützung des Alkohols sowie Esterspaltung und
Oxidation in das OH-geschützte (4R)-(+)-Hydroxy-2-cyclohexen-1-on
8 überführt (Figur 1).
Eine Tandem-Michael-Addition an 8 mit einem Methyl-Nucleophil und
Abfangen des Enolates mit einem Methyl-Elektrophil ergibt das
2,3-dimethylierte Keton 9.
Das Keton 9 wird in einer Michael-Addition in der 2-Position
alkyliert und in einer Robinson-artigen Anellierung zum
Dekalinenon 12 cyclisiert.
12 wird in 4-Position mit einer Base deprotoniert und mit einem
Elektrophil alkyliert.
Die Alkohol-Schutzgruppe wird entfernt und der freie Alkohol
verestert.
Die Schützung des Alkohols 5 erfolgt mit einem Alkyl- oder Silyl-
Ether, wie Benzyl, Tetrahydropyranyl, Methoxymethyl, tert-
Butyldiphenylsilyl, Triisopropylsilyl, Triethylsilyl, bevorzugt
dem tert-Butyldimethylsilylether. Die Esterspaltung, bevorzugt
mit K₂CO₃/MeOH, sowie die Oxidation, bevorzugt mit CrO₃/Pyridin,
zu 8 erfolgen in an sich bekannter Weise.
Die Michael-Addition an das Enon 8 erfolgt mit einer Kupfer
katalysierten Addition eines Methyl-Anions, wie MeLiCuCN,
MeMgCuBr₂, CuLi₂Me₃, bevorzugt mit CuLiMe₂ in einem aprotischen
Lösungsmittel oder einem Lösungsmittelgemisch, wie THF,
Dimethoxymethan, HMPTA (Hexamethyl-Phosphorsäuretrisamid),
Dioxan, tert-Butylmethylether, Dimethylsulfid, Tributylphosphin,
bevorzugt in Diethylether, bei einer Temperatur zwischen -95 und
0°C, bevorzugt bei -50 bis -30°C. Das intermediäre Enolat kann
als Silylenolether abgefangen werden oder mit 1 bis 10
Äquivalenten eines Methyl-Elektrophils, wie Methylbromid,
Methyltrifluormethansulfonat, Dimethylsulfat, bevorzugt mit
Methyliodid in Gegenwart von 0 bis 20 Äquivalenten eines
Additives wie Dimethoxyethan, Dimethylsulfid, N-Methyl-2-
pyrolidon, bevorzugt HMPTA, bei -95 bis +10°C, bevorzugt bei -50
bis -30°C zu dem Keton 9 reagieren.
Das Keton 9 wird nun über das Enolat, den Silylenolether, oder
bevorzugt das Imin, das durch azeotrope Destillation mit einem
primären Amin, bevorzugt dem (R)-Phenylethylamin, gewonnen wird,
an C-2 alkyliert. Die Alkylierung erfolgt mit einem Elektrophil,
wie Allylhalogenid, Brompropionsäureester, 2-Alkylacrylat, 3-
Buten-2-on, 3-Alkyl-3-Buten-2-on, 3-Trimethylsilyl-3-buten-2-on,
bevorzugt mit Acrylsäuremethylester bei 0 bis 150°C, bevorzugt
bei +80°C in 0 bis 100 Äquivalenten eines aprotischen
Lösungsmittels wie Toluol.
Die Spaltung des Esters 10 zu der Keto-Carbonsäure 11 erfolgt in
an sich bekannter Weise, bevorzugt mit LiOH/H₂O/THF bei
Raumtemperatur.
Die Cyclisierung der Keto-Carbonsäure 11 zu dem Decalinenon 12
erfolgt mit dem Reagenz
Triphenyl[(phenylimino)ethenyliden]phosphoran (H. J. Bestmann, M.

unter Zusatz von 1-10 Äquivalenten eines Alkoholes, bevorzugt
Ethanol, in einem aprotischen Lösungsmittel, wie Diethylether,
Benzol, THF, Petrolether, Xylol, bevorzugt in Toluol bei einer
Temperatur von +20 bis +150°C, bevorzugt bei +80 bis +110°C.
Die Deprotonierung des Dekalinenons 12 in der 4-Position erfolgt
mit 1 bis 10 Äqivalenten einer nicht-nukleophilen Base, wie
Natriumhydrid, Natrium-tert-Butanolat, einem Alkali-Amid, wie
Lithium- oder Natrium-Diisopropylamid, bevorzugt mit Lithium-
bis(trimethylsilyl)amid in einem aprotischen Lösungsmittel, wie
Diethylether, Hexan, Dioxan, Dimethoxyethan, bevorzugt in THF bei
einer Temperatur von -100 bis 0°C, bevorzugt bei -80 bis -70°C.
Das hierbei entstehende Enolat kann direkt mit einem
Alkylhalogenid, wie Methyliodid, Allylbromid oder Benzylbromid in
einem aprotischen Lösungsmittel alkyliert werden oder unter
Zusatz von 0 bis 10 Äquivalenten, bevorzugt 1.1 Äquivalenten,
einer Lewis-Säure, wie Titantetrachlorid, Zinntetrachlorid,
bevorzugt Zinkdichlorid mit einem Aldehyd, wie Acetaldehyd,
Propionaldehyd, Benzaldehyd, oder einem Keton, wie mit
Acetophenon, Ethylpyruvat, Butanon, bevorzugt Aceton bei -100 bis
0°C, bevorzugt bei -80 bis -70°C, in einer Aldol-Reaktion zu 13
reagieren.
Der bei der Aldol-Reaktion entstehende Alkohol 13 kann über einen
Sulfonsäureester, der aus 13 und Methansulfonylchlorid oder
Trifluormethansulfonsäureanhydrid gebildet wird, unter Zusatz
einer Base, wie Pyridin, DBU (Diazabicycloundecen), N,N-
Dimethylanilin, bevorzugt Triethylamin bei -78 bis +50°C,
bevorzugt bei 0°C, eliminiert werden.
Das entstehende Gemisch der Doppelbindungsisomeren kann unter
saurer Katalyse, wie HCl oder Toluolsulfonsäure in einem
aprotischen Lösungsmittel, wie Ether, THF, Toluol, Benzol,
bevorzugt mit saurem Aluminiumoxid in Ether bei +20°C, zu dem
stabileren 14 isomerisiert werden.
Der Silylether in 14 kann mit Säure, wie HCl in THF/H₂O, oder mit
Fluoriden, wie tetra-n-Butylammonium-Fluorid, KF mit 18-Krone-6,
bevorzugt mit 50% HF/H₂O unter Zusatz von einem organischen
Lösungsmittel, wie THF, Dioxan, DMF, Dimethoxyethan, bevorzugt
Acetonitril bei 0 bis +100°C, bevorzugt bei +20°C, abgespalten
werden, wodurch die Alkohole 15, insbesondere der Alkohol 4,
erhalten wird.
Die Alkohole 15 können mit verschiedenen Carbonsäuren nach einer
Aktivierung, wie über das Säurechlorid, aus der Säure unter
Zusatz von DCC (Dicyclohexylcarbodiimid), bevorzugt aus dem
gemischten Anhydrid mit 2,4,6-Trichlorbenzoesäure, in einem
aprotischen organischen Lösungsmittel, wie Diethylether, THF,
Benzol, bevorzugt Toluol bei 0 bis +110°C, bevorzugt bei +20°C,
zu den Verbindungen 16 verestert werden.
Bevorzugt sind die Ester 16 der Alkohole 15 mit den ungesättigten
Carbonsäuren, insbesondere der Ester 2 des Alkohols 4 mit der
Angelikasäure (Z-2-Methyl-2-butensäure).

R, R′ und R′′ besitzen die in der Figur 1 angegebenen
Bedeutungen.
Die Numerierung der Kohlenstoffatome entspricht der Numerierung
in der Verbindung 1 auf Seite 5.
Bei dem Alkohol 13 kann direkt, wie für 14 beschrieben, der
Silylether abgespalten werden (Figur 2). Das entstehende Diol 17
kann nun, analog den bei der Synthese von 16 aus 15, zu den
Verbindungen 18 verestert werden, wobei selektiv der sekundäre
Alkohol in Position 8 reagiert.
Der tertiäre Alkohol kann mit den bei der Synthese von 15 aus 14
beschriebenen Methoden eliminiert werden, wobei keine saure
Isomerisierung im Anschluß durchgeführt wird. Es entsteht
überwiegend das Hoffmann-Eliminierungsprodukt 19 sowie geringe
Mengen des Saytzeff-Produktes 16, das chromatographisch
abgetrennt werden kann.
Bevorzugt sind die Ester 19 mit den ungesättigten Carbonsäuren,
insbesondere der Ester 2 (R′ = Me, Petasin) mit der Angelikasäure
(Z-2-Methyl-2-butensäure).
Durch die Wahl eines enzymatisch leicht abspaltbaren Esters, wie
Acetyl, Dichloracetyl, bevorzugt Chloracetyl, läßt sich durch
Spaltung des Esters mit einer Esterase, bevorzugt CCL (Candida
Cylandrea Lipase), der Alkohol 20, insbesondere der Alkohol 3
(R′ = Me, Petasol), erhalten.

Zu einer Lösung von 16.2 g 5 (104 mmol) in 70 ml DMF werden 17.9 g
Imidazol (259 mmol, 2.50 eq) sowie 18.8 g tert-
Butyldimethylsilylchlorid (124 mmol, 1.20 eq) gegeben. Die Lösung
wird bei 37°C für 15 h gerührt, nach dem Abkühlen auf 400 ml
Wasser gegossen und die wäßrige Phase 3mal mit 400 ml Ether
extrahiert. Die gesammelten organischen Phasen werden 2mal mit
100 ml gesättigter NaCl-Lösung gewaschen, mit Na₂SO₄ getrocknet
und am RV eingeengt. KR-Destillation ergibt 23.3 g farbloses Öl
(86.2 mmol, 82% d. Th.).

23.3 g 6 (86.2 mmol) werden in 50 ml Methanol gelöst, zu einer
Lösung von 35.7 g K₂CO₃(258 mmol, 3.00 eq) in 300 ml Methanol
gegeben und 2 h bei RT gerührt. Das K₂CO₃ wird abfiltriert und
das Filtrat mit wenig Methanol gewaschen. Die vereinigten
Methanollösungen werden am RV einrotiert und in 400 ml Ether
aufgenommen. Die etherische Lösung wird mit je 2mal 200 ml
gesättigter NH₄Cl- und NaCl-Lösung extrahiert und mit Na₂SO₄
getrocknet. Einrotieren ergibt ein farbloses Öl, das durch
Säulenchromatographie (ca. 500 g SiO₂, 6 : 1→3 : 1→1 : 1 Hex/EE)
gereinigt wird. Man erhält 17.0 g einer farblosen Flüssigkeit
(74.4 mmol, 86% d. Th.). Die analytische Probe wird durch KR-
Destillation gereinigt.

43.6 g CrO₃ (436 mmol, 6.00 eq) werden unter Rühren zu einer
Lösung von 70.4 ml Pyridin (68.8 g, 871 mmol, 12.0 eq) in 1000 ml
CH₂Cl₂ gegeben und 30 min bei RT gerührt. Zu der rot-braunen
Lösung werden 16.6 g 7 (72.6 mmol) in 100 ml CH₂Cl₂ gegeben. Die
entstehende Suspension wird 15 h bei RT gerührt und zur
Vermeidung der Emulsionsbildung auf gerührtes Na₂SO₄ gegossen.
Der verbleibende Rückstand wird mit 2mal 100 ml gesättigter
NaHCO₃ aufgenommen und ebenfalls zu dem Na₂SO₄ gegeben. Der
Kristallbrei wird mit 4mal 300 ml CH₂Cl₂ gewaschen. Die
vereinigten organischen Phasen werden mit je 2mal 200 ml
gesättigter NaHCO₃₁ 2% H₂SO₄, NaHCO₃- und NaCl-Lösung
extrahiert, mit Na₂SO₄ getrocknet und einrotiert. Der Rückstand
wird mit Säulenchromatographie (ca. 300 g SiO₂₁ 6 : 1 Hex/EE)
gereinigt. Man erhält 13.4 g (59.2 mmol, 81% d. Th.) einer
farblosen Flüssigkeit. Die analytische Probe wurde mit KR-
Destillation gereinigt.

7.07 g CuI (37.1 mmol, 1.05 eq) werden in 150 ml Ether suspendiert
und im Eisbad auf 0°C gekühlt. Zu dieser gerührten Suspension
werden 46.4 ml einer 1.6 M Methyllithium-Lösung in Ether (74.2 mmol,
2.10 eq) aus einer frisch geöffneten Flasche (Fluka) mit
einer Transfernadel zugetropft. Nach der Beendigung des
Zutropfens wird 1 h bei 0°C gerührt und dann in einem
Methanol/CO₂-Bad auf -60°C IT gekühlt. Zu der entstandenen,
leicht weißlich-trüben Lösung wird nun mit einer Transfernadel
die auf 0°C vorgekühlte Lösung von 8.00 g 8 (35.3 mmol) in 50 ml
Ether so zugegeben, daß die IT nicht über -55°C steigt (ca. 30
min). Die nun intensiv gelbe Suspension wird durch Wechsel des
Kühlbades mit einem vorbereiteten, -35°C kalten Methanol/CO₂-Bad
auf diese Temperatur erwärmt und 100 min bei einer IT von -30 bis -35°C
gerührt (DC-Kontrolle!). Dann wird auf -70°C IT gekühlt
und unter starkem Rühren der gelben Suspension und des Kühlbades
über eine Transfernadel 17.6 ml Methyliodid (40.1 g, 282 mmol,
8.0 eq) in 50 ml HMPTA schnell zugegeben (5 min). Hierbei erwärmt
sich die Lösung auf eine IT von ca. -30°C, bei der sie durch
Wechsel des Kühlbades gehalten wird. Der gelbe Niederschlag löst
sich unter gleichzeitiger Bildung eines weißen Niederschlages
auf. Die nun weiße Suspension wird bei -30 bis -35°C für 3 h
gerührt (DC-Kontrolle!). Die Reaktionslösung wird durch die
Zugabe von 20 ml gesättigter NH₄Cl/NH₃, 1 : 1 gequencht und aus dem
Kühlbad genommen. Es wird mit 500 ml Ether/Hexane 1 : 1 verdünnt und
mit je 2mal 200 ml NH₃/NH₄Cl, H₂O und NaCl-Lösung extrahiert.
Einrotieren ergibt ein farbloses Öl, das durch
Säulenchromatographie (ca. 200 g SiO₂, Hex/EE 10 : 1 → 6 : 1) gereinigt
wird. Man erhält 6.34 g farbloses Öl (24.7 mmol, 70% d. Th.), das
die beiden C-2-Epimeren im Verhältnis 9 : 1 (GC) enthält. Die
analytischen Proben der beiden Epimere werden durch KR-
Destillation und nochmalige Chromatographie (SiO₂, Hex/EE 10 : 1)
gewonnen.

1.80 g Epimerengemisch 9 (70.2 mmol) werden in 20 ml Toluol
gelöst und mit 1.09 ml (R)-(+)-1-Phenylethylamin versetzt. Die
farblose Lösung wird für 3 Tage unter RF am Wasserabscheider
erhitzt und anschließend das Toluol im ÖV (Ölpumpenvakuum)
abgezogen. Das zurückbleibende-Öl wird mit 6.30 ml Methylacrylat
(6.02 g, 70.2 mmol, 10.0 eq) versetzt und 5 Tage auf 80°C
erhitzt. Das überschüssige Methylacrylat wird im ÖV abgezogen,
das zurückbleibende Öl mit 3 ml 20%-iger Essigsäure und 5 ml
Methanol versetzt und 48 h bei RT gerührt. Es wird mit 200 ml
Ether verdünnt, mit je 2mal 50 ml 2%-iger H₂SO₄, gesättigter
NaHCO₃- und NaCl-Lösung extrahiert und die etherische Phase mit
Na₂SO₄ getrocknet. Der Ether wird abrotiert und der Rückstand
einer Säulenchromatographie (ca. 100 g SiO₂; 6 : 1 Hex/EE)
unterworfen. Man erhält 1.86 g farbloses Öl (5.42 mmol, 77%),
das noch geringe Mengen (<5%) des C-2-epimeren enthält.

15 h bei RT gerührt (DC-Kontrolle) und anschließend das THF am RV
abgezogen. Man gibt 100 ml Ether zu und säuert mit 50 ml 2%-iger
H₂SO₄ an. Die organische Phase wird mit 20 ml gesättigter NaCl-
Lösung gewaschen und mit Na₂SO₄ getrocknet. Das Lösungsmittel
wird abgezogen und der Rückstand über eine kurze Säule
chromatographiert (ca. 30 g SiO₂, 20 : 1 CH₂Cl₂/MeOH). Man erhält
nach dem Einengen farblose Kristalle, die aus Hexan
umkristallisiert werden. 1.58 g diastereomerenreines 11 als
farblose Nadeln (4.81 mmol, 91% d. Th.).

werden in 60 ml Toluol gelöst und 5 h bei 80°C gerührt. Man gibt
3 ml Ethanol zu und erhitzt 15 h unter RF. Die Lösung wird nach
dem Abkühlen mit 200 ml Ether verdünnt und mit 2mal 50 ml
gesättigter NaCl-Lösung extrahiert. Die Lösung wird einrotiert
und der Rückstand durch 2fache Säulenchromatographie (ca. 150 g
SiO₂, 6 : 1 Hex/EE) gereinigt. Man erhält 1.33 g (4.29 mmol, 67%
d. Th.) farbloses Öl, das beim Kühlen zu farblosen, weichen
Kristallen erstarrt. Es wird aus wenig Hexan umkristallisiert.

6.77 ml Hexamethyldisilazan (5.18 g, 32.1 mmol, 5.1 eq) werden in
60 ml THF gelöst und auf -60°C gekühlt. Hierzu werden 12.6 ml
2.5 N n-Butyllithium-Lösung in Hexan (Janssen) (31.5 mmol, 5.0 eq)
gegeben und es wird 30 min gerührt. Die farblose Lösung wird
auf -78°C mit einem Methanol/CO₂-Bad gekühlt und es werden 1.94 g
12 (6.29 mmol) in 20 ml THF langsam zugegeben (ca. 15 min). Man
rührt 1 h bei dieser Temperatur und gibt 3.15 ml einer 2.2 N
ZnCl₂-Lösung in CH₂Cl₂ (Fluka) (6.93 mmol, 1.10 eq) zu. Man rührt
10 min und gibt 3.70 ml Aceton (2.93 g, 50.4 mmol, 8.00 eq) zu.
Die Lösung wird 15 min bei -78°C gerührt und durch die Zugabe
von 20 ml 5%-iger Weinsäure gequencht. Man läßt auf RT erwärmen,
verdünnt mit 200 ml Hex/Ether 1 : 1 und extrahiert mit je 2mal 20 ml
5%-iger Weinsäure- und gesättigter NaCl-Lösung. Trocknen mit
Na₂SO₄ und Einrotieren ergibt ein Öl, das durch
Säulenchromatographie (ca. 50 g SiO₂, 3 : 2 Hex/EE) gereinigt wird.
Man erhält 2.08 g farbloses Öl (5.66 mmol, 90% d. Th.).

900 mg 13 (2.45 mmol) werden in 15 ml CH₂Cl₂ gelöst und auf 0°C
gekühlt. Es werden 4.27 ml Triethylamin (3.10 g, 30.6 mmol, 12.5 eq)
sowie 1.14 ml Methansulfonsäurechlorid (1.68 g, 14.7 mmol,
6.00 eq) zugegeben. Die sich bildende weißliche Suspension wird 2
h gerührt (DC-Kontrolle) und anschließend mit 50 ml CH₂Cl₂
verdünnt. Man extrahiert mit je 2mal 10 ml Wasser, 2%-iger
H₂SO₄ und NaCl-Lösung und trocknet anschließend mit Na₂SO₄. Die
Lösung wird über eine kurze Säule aus saurem Al₂O₃ (WOELM, ca. 20
g) mit CH₂Cl₂ chromatographiert und das CH₂Cl₂ abrotiert.
Nochmalige Säulenchromatographie (ca. 50 g SiO₂₁ 6 : 1 Hex/EE)
ergibt 638 mg farbloses Öl (1.83 mmol, 75% d. Th.).

Zu 256 mg Angelikasäure (2.56 mmol, 2.0 eq) in 4 ml Toluol werden
bei RT 0.400 ml 2,4,6-Trichlorbenzoylchlorid (625 mg, 2.56 mmol,
2.0 eq) und 0.357 ml Triethylamin (259 mg, 2.56 mmol, 2.0 eq)
gegeben. Die entstehende weiße Suspension wird 2 h bei RT
gerührt. Es werden 300 mg Isopetasol (1.28 mmol) in 4 ml Toluol
zugegeben und für 24 h auf 80°C erhitzt (DC-Kontrolle). Man läßt
auf RT abkühlen und verdünnt mit 20 ml Ether. Es wird über 2 cm
SiO₂ filtriert mit Ether gewaschen und einrotiert. Das Produkt
wird durch Flash-Chromatographie (ca. 40 g SiO₂, 10 : 1 Hex/EE)
gereinigt. Man erhält nach dem Umkristallisieren (Ether/Hexan)
336 mg Isopetasin als farblose Kristalle (1.06 mmol, 83% d.
Th.).

500 mg 13 (1.36 mmol) werden in 20 ml Acetonitril gelöst und 2 ml
ca. 50%-ige HF zugegeben. Die Lösung wird bei RT 2 h gerührt
(DC-Kontrolle), dann mit 100 ml Ether verdünnt und mit je 2mal
20 ml gesättigter NaHCO₃- und NaCl-Lösung extrahiert. Es wird mit
Na₂SO₄ getrocknet und nach dem Einengen chromatographiert (ca. 20 g
SiO₂, 1 : 2 Hex/EE). Man erhält nach dem Umkristallisieren aus
Ether/Hexan 323 mg farblose Nadeln (1.28 mmol, 94% d. Th.).

2.0 eq) gegeben. Die entstehende weiße Suspension wird 2 h bei RT
gerührt. Es werden 200 mg 17 (0.793 mmol) in 4 ml Toluol
zugegeben und 24 h auf 80°C erhitzt (DC-Kontrolle). Man läßt auf
RT abkühlen und verdünnt mit 20 ml Ether. Es wird über 2 cm SiO₂
filtriert mit Ether gewaschen und einrotiert. Das Produkt wird
durch Flash-Chromatographie (ca. 40 g SiO₂, 6 : 1 → 1 : 1 Hex/EE)
gereinigt. Man erhält als erste Fraktion 50 mg Isopetasin (0.158 mmol,
20% d. Th), dann das gewünschte Produkt und anschließend
50 mg Edukt (0.198 mmol). Umkristallisation des Produktes aus
Hex/Ether ergibt 104 mg farblose Kristalle (0.311 mmol, 39% d. Th.,
52% bezogen auf den Umsatz).

120 mg 18 (0.359 mmol) werden in 5 ml CH₂Cl₂ gelöst und im
Methanol/CO₂-bad auf -78°C gekühlt. Zu dieser Lösung werden
0.188 ml Diisopropylethylamin (140 mg, 1.08 mmol, 3.00 eq) und
0.091 ml Trifluormethansulfonsäureanhydrid (152 mg, 0.539 mmol,
1.50 eq) gegeben und 60 min bei -78°C gerührt (DC-Kontrolle). Es
werden 2 ml gesättigte NH₄Cl-Lösung zugegeben und auf
Raumtemperatur erwärmt. Verdünnen mit 20 ml CH₂Cl₂ und Extraktion
mit gesättigter NH₄Cl-Lösung, sowie Trocknung mit Na₂SO₄ und
Einrotieren ergibt ein Öl, das durch Flash-Chromatographie (ca.
20 g SiO₂, 10 : 1 → 3 : 1 Hex/EE) gereinigt wird. Man erhält als erste
Fraktion 19 mg Isopetasin (0.060 mmol, 17% d. Th.), dann 55 mg
Petasin (174 mmol, 48% d. Th., 69% bezogen auf den Umsatz),
sowie 36 mg Edukt (108 mmol).

Drehwerte: [α]D = +34.1° (c = 0.7 in CHCl₃).
30 mg 17 (0.12 mmol) werden in 2 ml Ether gelöst und 0.5 ml
Triethylamin sowie ca. 5 mg Dimethylaminopyridin zugegeben. Die
Lösung wird auf 0°C gekühlt und langsam 20 mg 2-Chloracetylchlorid
(0.18 mmol, 1.5 eq) in 1 ml Ether zugegeben. Die
entstehende Suspension wird 12 h bei RT gerührt (DC-Kontrolle!).
Die Suspension wird wieder auf 0°C gekühlt und 137 mg
Methansulfonylchlorid (1.2 mmol, 10 eq) zugegeben. Es wird 4 h
bei RT gerührt (DC-Kontrolle!) und bei 0°C durch die Zugabe von
2 ml Wasser gequencht. Extraktion mit 2mal 5 ml gesättigter
NaHCO₃-Lösung, Trocknen mit Na₂SO₄ sowie Einrotieren ergibt ein
Öl (Petasyl- : Isopetasylester = 3 : 1), das durch
Säulenchromatographie (ca. 5 g SiO₂₁ 6 : 1 Hex/EE) gereinigt wird.
Man erhält 18 mg farbloses Öl (49% d. Th.).

15 mg 19 (0.048 mmol) werden in 0.5 ml Ether gelöst und 2 ml
Phosphatpuffer pH 7.0 zugegeben. Zu dieser gerührten Suspension
werden 50 mg Lipase von Candida cylandrea (CCL, SIGMA) gegeben
und 2 Tage bei RT gerührt (DC-Kontrolle). Es werden 5 g Na₂SO₄
zugegeben und der Kristallbrei mit 5mal 10 ml Ether gewaschen.
Die gesammelten Etherphasen werden einrotiert und der Rückstand
chromatographiert (ca. 5 g SiO₂, 1 : 1 Hex/EE). Man erhält 5 mg
farbloses Öl (45% d. Th.), das nach H-NMR-Untersuchungen ca. 8%
Isopetasol 4 enthält.

Von den aufgezeigten Verbindungen ist besonders die
enantiomerenreine Verbindung 13 (R = SiMe₂t-Bu, R′ = CH₃) bevorzugt.
Sie stellt eine Schlüsselverbindung bei der enantioselektiven
Synthese der Petasine dar und wurde bisher noch nicht
beschrieben, Formel III.
Mit den erfindungsgemäßen Verfahren ist es möglich, die aus der
Pestwurz bekannten Wirkstoffe in enantiomerenreiner Form ohne
nachteilige Nebenprodukte und ökonomisch vorteilhaft zur
Verfügung zu stellen.
Die erfindungsgemäß herstellbaren Verbindungen sind zur Beeinflussung der Leukotrienbiosynthese
einsetzbar und daher insbesondere zur Behandlung entzündlicher
Erkrankungen verwendbar. Beispielsweise können die Verbindungen
zur Behandlung von entzündlichen Erkrankungen der Atemwege sowie
akuten und chronischen Bronchitis, Asthma bronchiale und
entzündlichen Erkrankungen des Magen- und Darmtraktes wie Magen- und
Darmulkus, Colitis ulcerosa und Morbus Crohn sowie
entzündlichen Erkrankungen der Haut- und Schleimhaut verwendet
werden.
Weitere Beispiele für die Verwendung der erfindungsgemäß herstellbaren Verbindungen sind die
spasmolytische Behandlung bei krampfartigen Beschwerden des
Magen-Darmtraktes, der Harnwege, bei Dysmenorrhoe und Spannungs
kopfschmerzen.
Claims (28)
1. Verfahren zur enantioselektiven Synthese von (4S,6R,7R,8R)-
(+)-6,7-Dimethylbicyclo[4.4.0]dec-1-en-3-onen) (Verbindungen
der Formel (I))
worin
R¹ eine Isopropenyl- oder Isopropylidengruppe und
R² Wasserstoff, Hydroxy, ein verzweigter oder unverzweigter, gesättigter oder ungesättigter Alkylester oder Arylester oder Alkyl- oder Arylether mit 1 bis 20 C-Atomen, der gegebenenfalls substituiert sein kann, bedeutet,
bei dem man
R¹ eine Isopropenyl- oder Isopropylidengruppe und
R² Wasserstoff, Hydroxy, ein verzweigter oder unverzweigter, gesättigter oder ungesättigter Alkylester oder Arylester oder Alkyl- oder Arylether mit 1 bis 20 C-Atomen, der gegebenenfalls substituiert sein kann, bedeutet,
bei dem man
- (a) vom bekannten enantiomerenreinen (1S,4R)-(-)-1- Acetoxycyclohex-2-en-4-ol 5 ausgeht und nach Schützung des Alkohols sowie Esterspaltung und Oxidation in an sich bekannter Weise das OH-geschützte (4R)-(+)-Hydroxy-2- cyclohexen-1-on 8 herstellt (Figur 1);
- (b) die Verbindung 8 in einer Tandem-Michael-Addition in einer Kupfer-katalysierten Addition eines Methyl-Anions in einem aprotischen Lösungsmittel oder einem Lösungsmittelgemisch bei einer Temperatur zwischen -95 und 0°C und Abfangen des intermediären Enolates mit einem Methyl-Elektrophil in Gegenwart eines polaren aprotischen Additives bei -95 bis +10°C zu dem Keton 9 umsetzt;
- (c) das Keton 9 über das Imin an C-2 mit einem Elektrophil bei 0 bis 150°C in einem aprotischen Lösungsmittel alkyliert;
- (d) den so erhaltenen Ester 10 zu der Keto-Carbonsäure 11 in an sich bekannter Weise spaltet;
- (e) die Keto-Carbonsäure 11 mit Triphenyl[(phenylimino)- ethenyliden]-phosphoran unter Zusatz eines Alkoholes in einem aprotischen Lösungsmittel bei einer Temperatur von 20 bis 150°C zu der Verbindung 12 cyclisiert;
- (f) die Verbindung 12 in der 4-Position mit einer nicht- nucleophilen Base in einem aprotischen Lösungsmittel bei einer Temperatur von -100 bis 0°C deprotoniert und mit einer Carbonylverbindung in einem aprotischen Lösungsmittel unter Zusatz einer Lewis-Säure bei -100 bis 0°C zu 13 alkyliert;
- (g) den so erhaltenen Alkohol 13 über einen intermediären Sulfonsäureester unter Zusatz einer Base bei -78 bis 50°C eliminiert und die exocyclische Doppelbindung gegebenenfalls unter Säurekatalyse zu 14 isomerisiert;
- (h) die Alkohol-Schutzgruppe von der Verbindung 14 in an sich bekannter Weise entfernt und
- (i) den freien Alkohol in an sich bekannter Weise in den gewünschten Ester überführt.
2. Verfahren nach Anspruch 1, bei dem R² in der Formel (I)
Formyl, Acetyl, Propionyl, Butyryl, Isobutyryl, Succinyl,
Aspartyl, 2,2-Dimethylbutyryl, Pivalyl, Angeloyl, 3-
Thiomethylacryl, Crotonyl, Methacryl, Tiglinyl, Stearyl oder
Eicosyl bedeutet.
3. Verfahren nach Anspruch 1, bei dem R² in der Formel (I) einen
substituierten Benzoylrest bedeutet.
4. Verfahren nach Anspruch 3, bei dem der Benzoylrest in o-, m-
und/oder p-Stellung durch Hydroxy-, C₁- bis C₄-Alkoxy, C₁- bis
C₄-Alkyl-, C₁- bis C₄-Acyl-, -Acetoxy- und/oder Halogen
substituiert ist.
5. Verfahren nach Anspruch 1, bei dem in der Formel (I) R² C₁-
bis C₄-Alkyloxy, Allyloxy, 2-Methylallyloxy oder Benzyloxy
bedeutet.
6. Verfahren nach einem der vorhergehenden Ansprüche, bei dem
in der Formel (I) R¹ Isopropyliden und R² Angeloyl bedeutet.
7. Verfahren nach einem der Ansprüche 1 bis 5, bei dem in der
Formel (I) R¹ Isopropyliden und R² 2,2-Dimethylbutyryl
bedeutet.
8. Verfahren nach einem der Ansprüche 1 bis 5, bei dem in der
Formel (I) R¹ Isopropyliden und R² Stearyl bedeutet.
9. Verfahren nach einem der Ansprüche 1 bis 5, bei dem in der
Formel (I) R¹ Isopropyliden und R² Acetylsalicoyl bedeutet.
10. Verfahren nach einem der Ansprüche 1 bis 5, bei dem in der
Formel (I) R¹ Isopropyliden und R² Succinyl bedeuet.
11. Verfahren nach einem der Ansprüche 1 bis 5, bei dem in der
Formel (I) R¹ Isopropyliden und R² n-Butyryl bedeutet.
12. Verfahren nach einem der Ansprüche 1 bis 5, bei dem in der
Formel (I) R¹ Isopropyliden und R² 2-Methylallyloxy bedeutet.
13. Verfahren nach einem der Ansprüche 1 bis 5, bei dem in der
Formel (I) R¹ Isopropenyl und R² Angeloyl bedeutet.
14. Verfahren nach einem der vorhergehenden Ansprüche, bei dem
man die Kupfer-katalysierte Tandem-Addition in Schritt b) mit
CuLiMe2 in Diethyhlether bei -50 bis -30°C sowie die Alkylie
rung mit Methyliodid unter Zusatz von Hexamethylphosphor
säuretrisamid, oder N-Methyl-2-pyrrolidon bei -50 bis -30°C
durchführt.
15. Verfahren nach Anspruch 14, bei dem man in Schritt c) das
Keton 9 mit R-Phenylethylamin in das Imin überführt und mit
Acrylsäuremethylester alkyliert.
16. Verfahren nach Anspruch 15, bei dem man die Reaktion bei
80°C durchführt.
17. Verfahren nach einem der Ansprüche 14 bis 16, bei dem man
in Schritt e) die Cyclisierung mit Triphenyl[(phenylimino)
ethenyliden]-phosphoran unter Zusatz von Ethanol in Toluol bei
80 bis 110°C durchführt.
18. Verfahren nach einem der Ansprüche 14 bis 17, bei dem man
in Schritt f) die Verbindung 12 mit Lithium-Hexamethylsilazid
in THF bei -80 bis -70°C durchführt und das entstehende Enolat
mit Aceton bei -80 bis -70°C alkyliert.
19. Verfahren nach einem der Ansprüche 14 bis 18, bei dem man
in Schritt g) den Alkohol mit Methansulfonylchlorid unter
Zusatz von Triethylamin bei 0 bis +30°C eliminiert.
20. Verfahren nach Anspruch 19, bei dem man das Doppelbin
dungsisomerengemisch mit saurem Aluminiumoxid isomerisiert.
21. Verfahren nach einem der Ansprüche 14 bis 20, bei dem man
in Schritt (i) die Veresterung des freien Alkohols mit Angeli
kasäure nach vorheriger Aktivierung der Säure mit 2,4,6-
Trichlorbenzoylchlorid und Triethylamin in Toluol durchführt.
22. (4S,6R,7R,8R-(+)-8-(tert-Butyldimethylsiloxy)-4-(1-
hydroxy-1-methyl-ethyl)-6,7-dimethylbicyclo[4.4.0]dec-1-en-3-
on entsprechend der Formel (III)
als Zwischenprodukt.
23. Verfahren zur Herstellung der Verbindung nach Anspruch 22,
bei dem man
- (a) vom bekannten enantiomerenreinen (1S,4R)-(-)-1- Acetoxycyclohex-2-en-4-ol 5 ausgeht und nach Schützung des Alkohols mit tert- Butyldimethylsilylchlorid und Imidazol sowie Esterspaltung und Oxidation in an sich bekannter Weise das OH-geschützte (4R)-(+)-Hydroxy-2-cyclohexen-1-on 17 herstellt (Figur 11);
- (b) die Verbindung 17 in einer Tandem-Michael-Addition in einer Kupfer-katalysierten Addition eines Methyl-Aniones in einem aprotischen Lösungsmittel oder einem Lösungsmittelge misch bei einer Temperatur zwischen -95 und 0°C und Abfangen des intermediären Enolates mit einem Methyl-Elektrophil in Gegenwart eines polaren aprotischen Additives bei -95 bis +10°C zu dem Keton 18 umsetzt;
- (c) das Keton 18 über das Imin an C-2 mit einem Elektrophil bei 0 bis 150°C in einem aprotischen Lösungsmittels alkyliert;
- (d) den so erhaltenen Ester 19 zu der Keto-Carbonsäure 20 in in sich bekannter Weise spaltet;
- (e) die Keto-Carbonsäure 21 mit Triphenyl[(phenylimino)ethenyliden]phosphoran unter Zusatz eines Alkoholes in einem aprotischen Lösungsmittel bei einer Temperatur von 20 bis 150°C zu der Verbindung 22 cyclisiert;
- (f) die Verbindung 22 in der 4-Position mit einer nicht- nucleophilen Base in einem aprotischen Lösungsmittel bei einer Temperatur von -100 bis 0°C deprotoniert und mit einer Aceton in einem aprotischen Lösungsmittel unter Zusatz einer Lewis- Säure bei -100 bis 0°C zu 23 alkyliert.
24. Verfahren nach Anspruch 23, bei dem man nach Schritt b)
die Kupfer-katalysierte Tandem-Addition mit CuLiMe₂ in
Diethyhlether bei -50 bis -30°C sowie die Alkylierung mit
Methyliodid unter Zusatz von Hexamethylphosphorsäuretrisamid,
oder N-Methyl-2-Pyrrolidon bei -50 bis -30°C durchführt.
25. Verfahren nach Anspruch 23, bei dem man in Schritt c) das
Keton 19 mit R-Phenylethylamin in das Imin überführt und mit
Acrylsäure-methylester alkyliert.
26. Verfahren nach Anspruch 23, bei dem man in Schritt 3 die
Reaktion bei 80°C durchführt.
27. Verfahren nach Anspruch 23, bei dem man in Schritt d) die
Cyclisierung mit Triphenyl[(phenylimino)ethenyliden]phosphoran
unter Zusatz von Ethanol in Toluol bei 80 bis 110°C durch
führt.
28. Verfahren nach Anspruch 23, bei dem man in Schritt f) die
Verbindung 22 mit Lithium-Hexamethylsilazid in THF bei -80
bis -70°C durchführt und das entstehende Enolat mit Aceton bei
-80 bis -70°C alkyliert.
