Description
Die vorliegende Erfindung betrifft ein neues Verfahren zur Herstellung von
3,7-Dialkylxanthinen der allgemeinen Formel 1
in der R¹ und R² C₁-C₄-Alkylgruppen bedeuten.
3,7-Dialkylxanthine waren bislang nur durch aufwendige Totalsynthesen oder
durch die Isolierung aus pflanzlichen Produkten wie Kakaobohnen oder
Teeabfällen zugänglich. Eine Ausnahme bildet die Partialsynthese von
Theobromin (3,7-Dimethylxanthin) nach H. Biltz et al. (Chem. Ber., 1931,
Band 64, Seite 1970), bei der Chlorwasserstoff bei 160°C in eine Schmelze
aus Coffeidin und Methyl- oder Ethylharnstoff eingeleitet wird. Die
Nachteile dieses Verfahrens bestehen in der schwierigen Handhabung der
sehr korrosiven Schmelze und in der aufwendigen Reinigung des Theobromins.
Weiterhin ist es aus der Arbeit von Biltz et al. bekannt (Chem. Ber.,
(1928), Band 61, Seite 1409), daß bei der Umsetzung von Coffeidin mit
Kaliumcyanat in wäßriger Lösung Coffein entsteht. Den experimentellen
Angaben zufolge findet diese Reaktion im schwach sauren bis neutralen
Milieu im pH-Bereich von etwa 5,5 bis 7,5 statt.
Der Erfindung lagen neue Verfahren zur Darstellung von 3,7-Dialkyl
xanthinen als Aufgabe zugrunde.
Demgemäß wurde ein Verfahren zur Herstellung von 3,7-Dialkylxanthinen der
allgemeinen Formel I
in der R¹ und R² C₁-C₄-Alkylreste bedeuten, gefunden, welches dadurch
gekennzeichnet ist, daß man ein 1-Alkyl-4-alkylamino-5-alkylaminocarbonyl
imidazol der allgemeinen Formel II
in der R³ für C₁-C₄-Alkylgruppen steht, in wäßriger Lösung bei pH 3 bis 5
mit einem Alkalimetallcyanat umsetzt und das hierbei erhältliche Harn
stoffderivat bei pH 8 bis 14 zu den entsprechenden 3,7-Dialkylxanthinen
(I) cyclisiert.
Daß diese Reaktion unter Abspaltung des Amins R³-NH₂ verläuft, ist im Hin
blick auf die Arbeit von Biltz et al. loc. cit. über die Coffeinsynthese
als überraschend zu bezeichnen.
Die Ausgangsverbindungen II, unter denen das Coffeidin (R¹, R² und R³=Methyl)
die größte Bedeutung hat, sind bekannt oder in bekannter Weise er
hältlich. Vorzugsweise stellt man sie durch alkalische Hydrolyse der ent
sprechenden 1,3,7-Trialkylxanthine III
her. Weitere wichtige Ausgangsverbindungen sind solche, bei denen R¹ und
R² für Ethylgruppen oder Isopropylgruppen stehen.
Bei dem erfindungsgemäßen Verfahren bringt man die wäßrigen Lösungen der
Verbindungen II mit einer Säure auf einen pH-Wert von 3 bis 5.
Als Säuren eignen sich Mineralsäuren wie Salzsäure, Schwefelsäure und
Phosphorsäure oder organische Säuren wie Essigsäure, Ameisensäure und
Oxalsäure, und da die Reaktion pH empfindlich ist, können zur Einhaltung
bestimmter pH-Werte auch Puffersysteme wie Natriumacetat/Essigsäure oder
Natriumdihydrogenphosphat/Dinatriumhydrogenphosphat eingesetzt werden.
Im Falle der Herstellung von Theobromin erzielt man im pH-Bereich von
4,3 bis 4,7 Ausbeuten von über 60%. Geht man von Coffeidin-Homologen aus,
kann der optimale pH-Bereich durch einige Vorversuche leicht ermittelt
werden.
Als Alkalimetallcyanate werden Natriumcyanat und Kaliumcyanat bevorzugt,
und zwar in Mengen von 1 bis 5 mol, insbesondere 1 bis 2 mol pro Mol II.
Größere Überschüsse von Alkalimetallcyanat sind möglich, aber nicht
erforderlich. Die Konzentration der Verbindung II in der wäßrigen Lösung
ist nicht kritisch und liegt in der Regel zwischen 0,2 und 4 mol pro
Liter. Im Falle des Theobromin erzielt man die besten Ergebnisse bei
Konzentrationen von 0,5 bis 2 mol Coffeidin pro Liter. Die Reaktion
verläuft im Bereich von 0 bis 50°C befriedigend. Höhere Temperaturen sind
möglich, aber es kommt zu einem höheren Verbrauch an Alkalimetallcyanat.
Die so hergestellten Harnstoffderivate IV
kann man isolieren oder direkt weiter zu den 3,7-Dialkylxanthinen um
setzen. Die Cyclisierung zu den Verbindungen I wird im alkalischen Milieu
bei erhöhten Temperaturen durchgeführt. Der pH-Bereich von 11 bis 13 ist
besonders bevorzugt, wobei sich im Falle des Theobromin der pH-Bereich von
11,6 bis 12,0 besonders empfiehlt.
Zur Aufarbeitung kann man die 3,7-Dialkylxanthine durch Einstellen des
pH-Wertes auf den isoelektrischen Punkt ausfällen und abfiltrieren oder
mit einem geeigneten Lösungsmittel extrahieren. Die Verfahrensprodukte,
darunter vor allem Theobromin und seine Derivate haben bekanntermaßen
große Bedeutung als pharmakologische Wirkstoffe.
Eine Lösung von 84 g (0,5 mol) Coffeidin in 450 ml Wasser wurde mit Eis
essig auf pH 4,5 eingestellt und bei 25°C im Verlauf von 4,5 Stunden por
tionsweise mit 41,6 g (0,66 mol) Natriumcyanat versetzt. Nach 12stündigem
Stehenlassen wurde der ausgefallene Feststoff mit 100 ml Wasser wieder
aufgelöst, und die Lösung wurde mit 100 ml 50%iger Natronlauge auf pH 11,8
eingestellt. Anschließend wurde die Reaktionslösung 1,5 Stunden zum Sieden
erhitzt-
Zur Aufarbeitung wurde die Lösung mit Eisessig auf pH 6,1 eingestellt.
Beim Abkühlen fiel Theobromin aus. Das Theobromin wurde abfiltriert, mit
Wasser und wenig Aceton gewaschen und nochmals mit heißem Wasser
gewaschen.

Claims (1)
- Verfahren zur Herstellung von 3,7-Dialkylxanthinen der allgemeinen Formel I in der R¹ und R² C₁-C₄-Alkylreste bedeuten, dadurch gekennzeichnet, daß man ein 1-Alkyl-4-alkylamino-5-alkylaminocarbonyl-imidazol der allgemeinen Formel II in der R³ für C₁-C₄-Alkylgruppen steht, in wäßriger Lösung bei pH 3 bis 5 mit einem Alkalimetallcyanat umsetzt und das hierbei erhältliche Harn stoffderivat bei pH 8 bis 14 zu den entsprechenden 3,7-Dialkylxanthinen (I) cyclisiert.
