und ihrer Salze mit pharmazeutisch annehmbaren Säuren.and their salts with pharmaceutically acceptable acids.
(Kiso to Rinsho, 8, 1 (1974); Arzneimitt. Compound I is known in the literature as "YC-93 11 and it is particularly nt in terms of their pharmacological properties interess a from T. Takenaka et coil (Kiso to Rinsho, 8, 1 (1974);.. Two methods are known for the preparation of the compound I which are essentially complementary to one another:
The first of these methods is described in BE-PS 811 324, for example. Here you use 3-nitrobenzaldehyde II with 3-aminocrotonic acid methyl ester III and the 2- (N-benzyl-N-methylamlno) ethyl ester of acetoacetic acid IV according to the following reaction equation:

miteinander um.with each other.
The second method, which is also described in this patent specification, is analogous to this and it consists in that the 3-nitrobenzaldehyde II with the methyl ester of acetoacetic acid V and the 2- (N-benzyl-N-methylamino) ethyl ester the 3-aminocrotonic acid converts.

The two methods described can be considered equivalent, taking into account the fact that the The tendency of enamines to undergo transamination reactions is known. In practice it is of relatively little importance which of the two ingredients, namely the enamine form or the carbonyl form, is used.
Yields between 19 and kk% are described in the literature.
The disadvantages of the synthetic routes described are evident. As with all reactions in which two analogous compounds are reacted with each other, they lead to a mixture of products: In fact, it is with the named ones Process possible that the 3-nitrobenzaldehyde reacts similarly and continues to react in the desired sense with 2 Hol of the compound mentioned. In this case, as by-products the symmetrical esters of 2,6-dimethyl-4- (3-nitrophenyl) -1,4-dihydropyridine-3,5-dicarboxylic acid obtain. Experiments by the applicant have shown that the by-products mentioned are always available in an amount that can amount to up to 20 MoI-Jo of the gross reaction product.
Considering its quantity and the pharmaceutical use of the Taking into account the final product, then these contaminants are absolutely intolerable and they have to be complicated through IMcristallization process or by column chromatography Cleaning methods are removed. In the former case, large material losses are inevitable, while in the second case the procedure turns out to be complicated and time-consuming because of the need for repeated rechromatography Treatment of the product designed.

It is known and complies with the above-described statements consistent that the symmetrical ester of 2,6-dimethyl-4- (3-nitrophenyl) -1,4-dihydropyridine-3,5-dicarboxylic acid can be obtained easily and in high yields. For example, U.S. Patent 3,485,847 describes the preparation of the dimethyl ester 2,6-dimethyl-4- (3-nitrophenyl) -1,4-dihydropyridine-3,5-dicarboxylic acid VII described.
It is therefore a process to strive for a single the methoxyl radicals of the ester VII through the 2- (N-benzyl-N-methyl) aminoethoxyl radical to replace in order to obtain the compound I contemplated according to the invention.
It has now been found that the implementation of the dirnethyl ester of 2,6-dimethyl-4- (3-nitrophenyl ^ "! ^ - dihydropyridine, 5-dicarboxylic acid VII with N- (2-hydroxyethyl) -N-methylbenzylamine VIII the desired product, namely 2- (N-benzyl-N-methylamino ) ethyl ester of 2,6-dimethyl-4- (3-nitrophenyl ) -3-methoxycarbonyl-1,4-dihydropyridine-5-carboxylic acid I. according to the following reaction equation:

Han carries out the reaction in such a way that one gets a suspension or a solution of the starting product VII in a suitable solvent with the amino alcohol VIII and the Catalyst is added and the mixture is heated while the methanol that is released in the reaction gradually distilled off.
Suitable solvents for the reaction described are all solvents that are present under the conditions used are inert, such as aliphatic and aromatic hydrocarbons, chlorinated aromatic hydrocarbons, ethers and ketones.
It is preferred to use aromatic hydrocarbons in the reaction mixture where they azeotrope with methanol form. In particular, toluene is preferred.
The reaction is carried out at an elevated temperature. In view of the need to remove the released methanol, the reaction temperature should not be below 70 ° C. On the other hand, it is not advantageous to use temperatures in excess of 180 to 200 ° C.

The preferred temperature range is 100 to 130 0 C, with the reflux temperature of the solvent used is particularly advantageous.
As already stated, the reaction should be carried out in the presence of a transesterification catalyst. The alkoxides of earth metals are particularly suitable for this purpose, in particular the aluminum alkoxides of the general formula Al (OR) ,, where R is a straight-chain or branched alkyl radical having 1 to 5 carbon atoms. In particular aluminum isopropoxide (R * = i-C ^ H ™) is preferred.
The transesterification catalyst is used in an amount of 1 to 10 MoI-Ji, based on the dimethyl ester VII, added.
As is well known in all transesterification reactions the reaction according to the invention is also an equilibrium reaction. The final reaction product therefore always contains three products: the starting compound III and the product VIII according to the invention and are also found in the gross reaction mixture a certain amount of bis [2- (N-benzyl-N-methylamino) ethyl ester] the 2,6-dimethyl-4- (3-nitrophenyl) -1, ^ dihydropyridines, 5-dicarboxylic acid IX, which apparently forms as an intermediate product of a double Ifa decomposition of the transesterification.
Nevertheless, with the appropriate selection of the reaction conditions, the amount of the by-product IX can be varied within acceptable limits Keep boundaries.
Es ist daher ausreichend, wenn man das rohe Reaktionsprodukt mit einerThe separation of the reaction mixture can be carried out without difficulty take place. The starting compound has acidic properties while the two products are basic. It is therefore sufficient if the crude reaction product is treated with a

Die organische Phase enthält die Ausgangsverbindung VII, die in praktisch quantitativer Menge in die lÄnesterungsreaktion zurückgeführt werden kann.aqueous solution of a mineral acid, e.g. with hydrochloric or sulfuric acid, treated and the aqueous phase containing the products, separated. The organic phase contains the starting compound VII, which can be returned to the esterification reaction in a practically quantitative amount.
Fractionation on an ion exchange resin in the H + form is particularly suitable for separating the compound I from the by-product IX. This resin only holds back the basic compounds.
By eluting with water it is possible to completely remove the last traces of compound VII which are still present in the product to remove. The invention is separated with high purity by successive fractionated elution intended product I and the by-product IX from each other.
The fractional elution can be carried out by using a mineral acid such as hydrochloric acid in two different concentrations to elute a first fraction containing compound I and a second fraction containing compound IX. When using 0.1N hydrochloric acid to elute compound I and when using 1N hydrochloric acid to elute compound IX, good results have been obtained. Satisfactory results are also obtained if a buffer solution with a pH between 3 f 5 and 4.5 is used for the elution of the first fraction, which contains the compound I. Optimal results are obtained with a 0.1 N acetate buffer with a pH of 4.0.
All resin acids are present as ion exchange resins made of crosslinked polystyrene polymers containing sulfonic groups, suitable.
