Verfahren zur Herstellung von 5-Mercapto-1,2,4-thiodiazolderivaten
Es wurde gefunden, dass man Thiodiazolderivate der allgemeinen Formel
in der R eine gegebenenfalls substituierte Alkylgruppe bedeutet, vorteilhaft erhält,
wenn man a) Isothioharnstoffverbindungen der Formel
in der R die oben genannte Bedeutung hat, in Gegenwart basisch wirkender Stoffe,
oder b) die entsprechenden Thiuroniumsalze von Verbindungen der Formel II in Gegenwart
von mindestens stöchiometrischen Mengen basisch wirkender Stoffe in organischen
Lösungsmitteln bei Temperaturen bis 100°C mit 1 bis 2 Äquivalenten Schwefel und
1 bis 10 Äquivalenten Schwefelkohlenstoff umsetzt.
In den S-Alkylisothioharnstoffverbindungen
der Formel II steht der Alkylrest für eine Methylgruppe oder einen Alkylrest, der
bis zu 4 Kohlenstoffatomen enthält. Die Alkylreste können ihrerseits substituiert
sein, z.B. durch Alkoxyreste, wie Methoxy-, Äthoxy- oder Butoxygruppen, oder Arylreste,
wie Phenyl-, Naphthyl-oder substituierte Phenylgruppen. Beispielsweise seien folgende
S-Alkylisothioharnstoffderivate genannt: S-Methylisothioharnstoff, S-Benzylisothioharnstoff
und S-(p-Chlorbenzyl)-thioharnstoff. Als basisch wirkende Stoffe seien beispielsweise
Alkoholate, Oxyde von Alkali- oder Erdalkalimetallen oder tertiäre Amine genannt.
Besonders vorteilhaft sind Alkoholate, wie Natriummethylat, Natriumäthylat, Natriumisopropylat
und Natrium-t-butylat. Als organische Lösungsmittel eignen sich alle gegenüber den
Reaktionspartnern inerten Lösungsmittel, wie Tetrahydrofuran, Dioxan, Alkohole,
wie Methanol, Isopropanol, Isobutanol, Benzol, Toluol, Xylol, Chlorbenzol, Dichlorbenzol,
Dimethylformamid und N-Methylpyrrolidon. Man kann auch Mischungen dieser Lösungsmittel
verwenden. Das neue Verfahren wird z.B. durchgeführt, indem man die Isothioharnstoffverbindungen
in einem organischen Lösungsmittel und in Gegenwart von bis zu 2,5 Äquivalenten,
bezogen auf-Isothioharnstoff, an basischen Stoffen der genannten Art, mit 1
bis 2, vorteilhaft etwa 1 Äquivalent Schwefel und 1 bis 10, vorteilhaft 2 bis 6
Äquivalenten Schwefelkohlenstoff, jeweils bezogen auf Iaothioharnstoff, bei Temperaturen
bis 100 °C umsetzt.
Geht man nicht von den freien
Isothioharnstoffverbindungen, son-, dern von den entsprechenden Isothiuroniumsalzen,
wie den Chlori-. den, Sulfaten oder Toluolsulfaten, aus, so ist
für die Umsetzung
die Gegenwart von einem äquivalent, bezogen auf Thiuroniumsalz,
an basisch wirkenden Stoffen der oben genannten Art erforderlich.
Zweckmässig
verwendet man 1 bis 3,5 Äquivalente, vorteilhaft etwa
3 Äquivalente
Hase auf eingesetztes Thiuroniumsalz. Bei dieser
Ausführungsform
des erfindungsgemässen Verfahrens ist besonders
zweckmässig,
wenn man zum Umsetzungsgemisch zunächst erst die fUr die Umsetzung
der Thinroniumsalze zu den freien Thioharnstoffen erforderliche
Menge des basisch wirkenden Stoffes gibt, und die
Zugabe der restlichen
Menge des basisch wirkenden Stoffes erst
nach Zugabe aller anderen
Reaktionspartner vornimmt. Nach dem erfindungsgemässen Verfahren erhält
man die 5-Mercapto-1,2,4-thiodiazolderivate in guten Ausbeuten. Dieses vorteilhafte
Ergebnis
ist überraschend, da aus Liebings Annalen 192 (1878),
Seite 1, bekannt
ist, dass Amidine mit Schwefelkohlenstoff zu
Thioamiden und Amidiniumrhodaniden
reagieren.
Die nach dem erfindungsgemässen Verfahren erhgtlichen
Verbindungen
sind zum Teil gegen Pflanzenschädlinge wirksam
und können auch
als Zwischenprodukte fUr Schädlingsbekämpfungsmittel,
Pharmazeu-
tika und Farbstoffe verwendet werden.
Die in dem
Beispiel genannten Teile und Prozentangaben beziehen
sich auf Gewichtseinheiten.
Beispiel
Zu 83,4 Teilen S-Methylisothiuroniumsulfat und 75 Teilen Dimethylformamid werden
16.2 Teile Natriummethylat gegeben. Nach einstündigem Rühren werden 39 Teile Schwefelkohlenstoff,
10,6 Teile Schwefelpulver sowie weitere 32,4 Teile Natriummethylat zugegeben. Man
erwärmt das Gemisch 6 Stunden auf 60 bis 65°C, gibt dann 250 Teile Wasser hinzu,
filtriert mit Tierkohle, säuert mit Essigsäure an und saugt nach dem Abkühlen auf
OOC ab. Das Produkt wird aus Eisessig umkristallisiert. Man erhält 21 Teile 3-Methylmercapto-5-mercapto-1,2,4-thiodiazol
(43 % der Theorie).
