Description
Procedure for the extraction of placental tissue raw materials It is known per se that the placental tissue extracts obtained by the previous processes not suitable for further use in a weakly alkaline environment. The numerous protein-like components with NH and NH groups already prevent the use of a weakly acidic medium in the production of the extracts. On the other hand, for fundamental considerations, it cannot be justified during the extraction the more alkali-sensitive components of the placenta Fabric raw materials are simply negligible and quite alkaline from the start Use extraction components.
Surprisingly, these difficulties can now be eliminated by extracting raw placental tissue materials gradually starting at p $ 7 and then progressing from weakly alkaline to more alkaline pH values, the alkaline pH value being adjusted with alkylolamines (mono to tri ) is set. According to this new procedure, extracts are obtained which are very suitable for further use in a weakly alkaline medium. In addition, the step-by-step extraction achieves a significantly higher yield of the placental active ingredients, the phosphatase values completely countering those of the blood serum and in particular the group of the amino acid complex quantitatively coming to the fore. Furthermore, a higher proportion of 17-ketosteroids is found in the extracts produced in this way. It is already known to isolate a certain antigen from the thin fetal layer of the placenta, detached from the mother and carrier layers, by isolating this fetal layer extracted using a strongly alkaline solution (p. value approx. 13) and the extract then neutralized using hydrochloric acid and a buffer substance up to a pH value of approx. 6.9. However, alkyl-sensitive substances in the placenta cannot be obtained in this way.
Furthermore, the chromatographic separation of placenta extracts has been described, preferably using organic solvents as the extraction liquid have been received. The chromatogram is performed using eluents different degrees of hydrophylicity washed out, so that metabolism-inhibiting and metabolism-promoting Substances can be collected separately as eluate.
The procedure according to the invention can be further improved by surface-active extraction components are also used. Suitable for this purpose In particular, the organic phosphoric acid esters, as these do not precipitate protein Possess properties and also with regard to the later use of the extracts need not be severed. For example, a secondary Stearyl phosphoric acid esters with two ethylene oxide groups in the molecule are used. Even the natural phospholipids, such as lecithin and cephalin, are suitable as extraction aids, although these compounds are not as durable as the synthetically produced organic ones Phosphoric acid ester.
To improve the osmotic diffusion of the extraction material you can expediently already 0.59 / o surface-active to the first extraction liquid (pg 7) Substances are added.
As an alkalizing agent in the step-by-step extraction according to the Invention are mainly alkylolamines such. B. mono- and diethanolamine used. The placental extracts according to the invention are narrowed in a manner known per se Way at the lowest possible temperatures.
The extraction treatment is preferably carried out in the p11 range from 7 to 10 carried out.
The invention is explained in more detail by the following example. Example 1 The amount of 10 kg of placental crude substance is extracted with 5 kg of double-distilled water (pH 7.0) with constant movement of the entire extraction mass. Subsequently, 2.5 kg of a 0.5% aqueous solution of the commercially available sodium salt of the secondary stearyl phosphoric acid ester with two ethylene oxide groups in the molecule, which is adjusted to pH 8.0 with monoethanolamine, are added. A further 2.5 kg of the same 0.5% aqueous solution are then added, but this time after a pH value of 9.0 has been set by means of diethanolamine. It is then extracted in the usual way and the finished extract is concentrated at + 30 ° C.
Example 2 10 kg of placental crude substance are extracted with 5 kg of the purest water under sterile conditions - with constant movement of the extraction material. After separation of the crude substance and 5 kg of extraction liquid, 2.5 kg of a 0.5% aqueous solution of a tertiary phosphoric acid oleyl ester, which is adjusted to pH 8.0 with monoethanolamine, are added. After this extract has been isolated, a further 2.5 kg of the same 0.5% aqueous solution are added, but this time after a pH value of 9.0 has been set using diethanolamine. It is then extracted in the usual way and the total extract is concentrated at + 30 ° C. Example 3 10 kg of placental crude substance are extracted with 5 kg of double-distilled water (p11 value 7) and 5 kg of extraction liquid are separated off.
This is retained during the recovery of the total extract the placenta raw material with 2.5 kg of a 1 o / o aqueous solution of the ether Coconut fatty alcohols with ten ethylene oxide groups are extracted at pH 8. The pH value is adjusted with monoethanolamine.
After 2.5 kg of extraction liquid has been separated off, the process is repeated the extraction process with another 2.5 kg of the same solution, but with the pH value 9 (discontinuation with diethanolamia). After combining the three alkaline solutions concentration at low temperature (30 ° C) is necessary.
