Description

Process for the preparation of purified tuberculin preparations The Contain tuberculin preparations used for the therapy and diagnosis of tuberculosis more or less large amounts of protein. as well as protein breakdown products, salts and others Impurities used by the bacteria or that used to grow the bacteria Nutrient solutions come from. These substances, which in themselves are to be regarded as unspecific are, as is well known, the cutane reaction used in tuberculosis can be positive Senses favor or pretend, making the value of this response natural is very questionable, also cause disruptive side effects during therapy, which influence the desired pure tuberculin effect in an unfavorable sense. It is therefore of great diagnostic and therapeutic value to use tuberculin preparations to be used, which are largely freed from these unspecific substances.

It has now been found that tuberculin preparations of any origin by treatment with the usual protein removal agents, such as iron dialysate, lead salts and others, lose nothing or almost nothing of their effectiveness, that rather only the proteins and their breakdown products are precipitated. Those treated like that Solutions contain the active components of the tuberculins and do not show any biuret reaction more. It has also been found that those present in the deproteinized solutions can be used subject the active components to further cleaning or the active components Substances can be brought to the separation that one the solutions with acetone, alcohols or other solvents in which the active substances are insoluble or that the active substances are first treated with precipitants from the alkaloid reagent group separates and the precipitates obtained by subsequent decomposition of the precipitant freed. Examples i. pulled human dry tuberculin, made from cooking broth by alcohol precipitation, are dissolved in twice the amount of distilled water and iron dialysate is added to the solution until there is no protein and protein in the filtrate Biuret reaction is more noticeable. The concentrated filtrate becomes the effective Substances precipitated with aqueous picric acid solution and the resulting tuberculin precipitations sucked off and decomposed with hydrochloric acid-containing water. From the resulting solutions the active substances are precipitated with acetone. Instead of dry tuberculin can one can also start from the corresponding amount of old tuberculin "Koch". 2. 20 g of bovine tubercular acid, produced by extraction of defatted and dewaxed bovine tubercle bacilli are dissolved in twice the amount of distilled water and deproteinize the solution with neutral lead acetate. The filtrate is used for removal defleaded from still existing small amounts of lead salt with hydrogen sulfide and the filtrate is precipitated with acetone to separate the active components.

@o g killed tubercle bacilli are extracted with alcohol and the alcoholic solution evaporated in vacuo. The residue that is next to the Tubercle fat is still rich in tuberculin, is extracted with water and the aqueous solution, as indicated in Example 2, freed from dietary fiber.
