Description
Verfahren zur Darstellung von gereinigten Tuberkulinpräparaten Die für die Therapie und Diagnose der Tuberkulose verwendeten Tuberkulinpräparate enthalten mehr oder weniger große Mengen Eiweiß. sowie Eiweißabbauprodukte, Salze und andere Verunreinigungen, welche den Bakterien oder den für die Züchtung der Bakterien verwendeten Nährlösungen entstammen. Diese Stoffe, welche an sich als unspezifisch anzusehen sind, können bekanntlich die bei der Tuberkulose angewandte Cutanreaktion in positivem Sinne begünstigen oder vortäuschen, wodurch der Wert dieser Reaktion naturgemäß sehr in Frage gestellt ist, außerdem bei der Therapie störende Nebenwirkungen hervorrufen, welche die gewünschte reine Tuberkulinwirkung in ungünstigem Sinne beeinflussen. Es wurde nun gefunden, daß Tuberkulinpräparate beliebiger Herkunft durch Behandlung mit den üblichen Enteiweißungsmhteln, wie Eisendialysat, Bleisalzen und anderen, nichts oder fast nichts von ihrer Wirksamkeit einbüßen, daß vielmehr lediglich die Eiweißstoffe und deren Spaltprodukte gefällt werden. Die so behandelten Lösungen enthalten die wirksamen Bestandteile der Tuberkuline und zeigen keine Biuretreaktion mehr. Weiter wurde gefunden, daß man die in den enteiweißten Lösungen vorhandenen wirksamen Bestandteile dadurch einer weiteren Reinigung unterziehen bzw. die wirksamen Substanzen zur Abscheidung bringen kann, daß man die Lösungen mit Aceton, Alkoholen oder anderen Lösungsmitteln, in denen die wirksamen Substanzen unlöslich sind, behandelt oder daß man die wirksamen Stoffe zunächst mit Fällungsmitteln der Alkaloidreagentiengruppe abscheidet und die erhaltenen Fällungen durch nachträgliches Zerlegen vom Fällungsmittel befreit. Beispiele i. zog humanes Trockentuberkulin, hergestellt aus Kochbouillon durch Alkoholfällung, werden in der Sofachen Menge destilliertem Wasser gelöst und die Lösung so lange mit Eisendialysat versetzt, bis im Filtrat keine .Eiweiß- und Biuretreaktion mehr feststellbar ist. Aus dem eingeengten Filtrat werden die wirksamen Substanzen mit wäßriger Pikrinsäurelösung gefällt und die entstandenen Tuberkulinfällungen abgesaugt und mit salzsäurehaltigem Wasser zerlegt. Aus den entstandenen Lösungen werden die wirksamen Stoffe mit Aceton gefällt. Statt von Trockentuberkulin kann man auch von der entsprechenden Menge Alttuberkulin »Koch« ausgehen. 2. 20 g bovine Tuberkelsäure, hergestellt durch Extraktion entfetteter und entwachster boviner Tuberkelbazillen werden in der Sofachen Menge destilliertem Wasser _ gelöst und die Lösung mit neutralem Bleiacetat enteiweißt.
