Description
Process for the production of natural sennoside salts containing Active ingredients from senna drugs The active ingredients of the senna drug are because of their laxative Known effect. The invention relates to a simple method for obtaining Active ingredients that contain the natural sennoside salts from senna leaves and Sennessehoten. The German patents 646 651 and 648 378 describe the Extraction of senna leaves with weakly alkaline water, after which the extracts evaporated to dryness and the residues to remove the impurities with anhydrous methanol or
Ethanol are treated, the active ingredients remain undissolved. The Swiss patent specification 225 885 describes the isolation of crystallized Sennosides by extracting the drug, pre-purified with benzene, using methanol, Precipitation of the sennosides as alkaline earth salts and separation of them by acidification sennosides obtained due to their different solubilities.
F ai rb ai rn und M i chael s (J. Pharm. F ai rb ai rn and Michael s (J. Pharm. Pharmacol., 2 [1950] p. 825) describe the production of senna extracts with the help of 700% methanol, Ethanol or acetone. The extracts have a high content of active ingredients, the possibility their further processing is not mentioned.
The present method assumes that first an extraction the drug with hydrous lower alcohols or ketones, preferably with hydrous Methanol takes place, whereupon then according to the invention the organic solvent is evaporated in vacuo and the sennoside salts from the remaining aqueous solution be precipitated. Surprisingly, it has now been shown that to this precipitation water-soluble alcohols, ketones or water-soluble cyclic ethers are suitable, although the same water soluble solvents are used to extract the drug will. However, the extraction is carried out with hydrous alcohols or ketones. Methanol or ethanol has proven to be most suitable for the precipitation, namely in such an amount that the resulting solutions 65 to 900 / o methanol or Contain ethanol, the most favorable ratio depending on the nature of the product the drug, which is subject to great fluctuations in its composition is. For better separation, the use of one in the work-up has been found Precipitation auxiliaries commonly used in plants, such as talc or kieselguhr, have been tried and tested.
The separated product can be cleaned in a little water dissolved and - as Isen described - precipitated with one of the solvents mentioned above be, this being the same or different to that used for the first precipitation can be.
The end product is a light brown powder, that from the natural Sennoside salts, inorganic salts and physiologically completely ineffective impurities, such as polysaccharides.
Compared to the method of Swiss patent specification 225 885 this procedure means a significant simplification; a pre-cleaning of the Drugs that require large amounts of benzene or chloroform can be omitted. the Extraction requires a far smaller amount of solvent, which also helps the active ingredient losses that occur when the solvent is distilled off as a result of the thermolability the sennosides always arise, reduce to a minimum. Is beneficial above all, that the natural sennoside salts are unchanged in the end product are and have not experienced any change during the processing; the procedure for Isolation of the active ingredients consists only in one or more repairs by means of one of the solvents mentioned. the yield of sennoside salts is about same drug better than that described in the above patent.
Both folia sennae (senna leaves) as well as Folliculi Sennae (Sennesschotten) can be used. For the former it has proven to be cheapest extraction agent methanol or ethanol with a water content of 10 up to 300/0, as this water content provides the optimal amount of active ingredient and only a small amount of mucilage, which makes processing difficult, dissolved will.
For the extraction of Folliculi Sennae, which contain significantly less mucilage contain, more dilute alcohols, such as 500% methanol, can also be used will. Extraction with water alone is disadvantageous. The precipitation of the sennosides will be in this Fall through the considerable amount of mucilage that has been dissolved very difficult.
Example 1 1 kg of Folia Sennae pulv.subt. and 8 1 70/0 methanol are stirred for 3 hours at room temperature. Receives by squeezing the drug product one 7 l of a 700% methanol extract; these are in a vacuum at -40 ° C, the outside temperature is reduced to 350 ccm. The aqueous concentrate is shaken with about 10 g of talc to remove the precipitated chlorophyll. To the filtered drug extract solution, which now along with other plant ingredients also contains the native sennoside salts, 11 l of 950% ethanol are slowly allowed pour in a thin stream while stirring.
This gives an approximately 700% ethereal solution, in which settles a greasy brown precipitate on the bottom. After several hours The supernatant solution is decanted and the greasy precipitate, which contains the native sennoside salts, dissolved in 150 cc of water. To this solution 850 to 900 cc of methanol are allowed to flow in slowly with stirring. It separates a granular powder that can be easily washed dry with methanol and ether leaves. This gives 18 to 24 g of a light brown salt mixture, in addition to inorganic Salts contains the native sennoside salts. The salt mixture is different depending on the drug Contains 25 to 50% sennoside.
Example 2 1 kg of Folliculi Sennae subt. and 8 1,500% methanol are stirred for 3 hours at room temperature.
Then - as described in Example 1, an aqueous chlorophyll-free Drug concentrate extract produced. In this concentrate 20 to 25 g of kieselguhr suspended as a customary filter aid, then slowly 1.0 Pour 1 ethanol in a thin stream while stirring, whereby a -brown precipitate, which contains the native sennoside salts, is deposited on the kieselguhr. This precipitate is separated off, treated with 150 ccm of water and the solution is filtered off from the kieselguhr. As described, 850 cc of methanol are again allowed to flow into the filtrate. Included a granular powder separates, which can be washed dry with methanol and ether leaves. This gives 35 to 45 g of a salt mixture, depending on the drug. the 12 to 18ovo contains sennoside salts.
