Abstract
Die vorliegende Erfindung betrifft neue polymergebundene Phosphoniumessigsäure Aktivester der Formel (I), Verfahren zu ihrer Darstellung, sowie deren Verwendung zur Synthese von Heterocyclen und Chalkonen. DOLLAR F1
Description
- Die vorliegende Erfindung betrifft neue polymergebundene Phosphoniumessigsäure Aktivester, Verfahren zu Ihrer Darstellung, sowie deren Verwendung zur Synthese von Heterocyclen und Chalkonen.
- Die Verbindung Triphenylphosphinoketen wurde bereits in der Literatur beschrieben und zur Darstellung von Heterocyclen und Chalkonen benützt (J. Westman, K. Orling, Combinatorial Chemistry and High Throughput Screening 7, 2002, 571-574 und darin zitierte Literatur). Es stellte sich jedoch als nachteilig heraus, daß bei der Reaktion entstandene Triphenylphosphinoxid vom gewünschten Produkt abzutrennen wie Westmann und Orling beschreiben. Dieser Nachteil kann umgangen werden, wenn man harzgebundene Phosphoniumessigsäure Aktivester benützt. Das nach erfolgter Reaktion gebildete Phosphinoxid verbleibt am Harz und kann so durch einen einfachen Filtrationsschritt abgetrennt werden.
- Die Aufgabe der vorliegenden Erfindung war es, neue polymergebundene Phosphoniumessigsäure Aktivester, die insbesondere für die Synthese von Heterocyclen und Chalkonen geeignet sind, bereitzustellen.
- Die vorliegende Erfindung betrifft Verbindungen der allgemeinen Formel (I): worin:
Pol ein Träger ist;
L ein Linker oder eine Bindung ist;
R1 und R2 unabhängig voneinander ein Alkyl-, Aryl-, Cycloalkyl-, oder ein Aralkylrest sind,
R3 ein Aktivesterrest und
X eine Abgangsgruppe wie z.B. F, Cl, Br, I, NR1R2, OR3 oder SOnR4 ist, wobei R1, R2, R3 und R4 unabhängig voneinander ein Wasserstoffatom, ein Alkyl-, Alkenyl-, Alkinyl-, Heteroalkyl-, Aryl-, Heteroaryl-, Cycloalkyl-, Heterocycloalkyl-, Alkylcycloalkyl-, Heteroalkylcycloalkyl-, Aralkyl- oder ein Heteroaralkylrest sind oder R1 und R2 zusammen Teil eines gegebenenfalls substituierten Heterocycloalkylringsystems sind und n = 0, 1, 2 oder 3 ist. - Der Ausdruck Alkyl bezieht sich auf eine gesättigte, geradkettige oder verzweigte Kohlenwasserstoffgruppe, die 1 bis 20 Kohlenstoffatome, vorzugsweise 1 bis 12 Kohlenstoffatome, besonders bevorzugt 1 bis 6 Kohlenstoffatome aufweist, z.B. die Methyl-, Ethyl-, Propyl-, Isopropyl-, Isobutyl-, tert-Butyl, n-Hexyl-, 2,2-Dimethylbutyl- oder n-Octyl-Gruppe.
- Die Ausdrücke Alkenyl und Alkinyl beziehen sich auf zumindest teilweise ungesättigte, geradkettige oder verzweigte Kohlenwasserstoffgruppen, die 2 bis 20 Kohlenstoffatome, vorzugsweise 2 bis 12 Kohlenstoffatome, besonders bevorzugt 2 bis 6 Kohlenstoffatome aufweisen, z. B. die Ethenyl-, Allyl-, Acetylenyl-, Propargyl-, Isoprenyl- oder Hex-2-enyl-Gruppe.
- Der Ausdruck Heteroalkyl bezieht sich auf eine Alkyl-, eine Alkenyl- oder eine Alkinyl-Gruppe, in der ein oder mehrere (bevorzugt 1, 2 oder 3) Kohlenstoffatome durch ein Sauerstoff-, Stickstoff-, Phosphor- oder Schwefelatom ersetzt sind (bevorzugt Sauerstoff oder Stickstoff), z.B. eine Alkyloxy-Gruppe wie z.B. Methoxy oder Ethoxy, oder eine Methoxymethyl-, Nitril-, Methylcarboxyalkylester-, Carboxyalkylester- oder 2,3-Dioxyethyl-Gruppe. Der Ausdruck Heteroalkyl bezieht sich des weiteren auf eine Carbonsäure oder eine von einer Carbonsäure abgeleitete Gruppe wie z.B. Acyl, Acyloxy, Carboxyalkyl, Carboxyalkylester z.B. Methylcarboxyalkylester, Carboxyalkylamid, Alkoxycarbonyl oder Alkoxycarbonyloxy.
- Der Ausdruck Cycloalkyl bzw. Cyclo- bezieht sich auf eine gesättigte oder teilweise ungesättigte cyclische Gruppe, die einen oder mehrere Ringe aufweist, die ein Gerüst bilden, welches 3 bis 14 Kohlenstoffatome, vorzugsweise 3 bis 10 Kohlenstoffatome enthält, z.B. die Cyclopropyl-, Cyclohexyl-, Tetralin- oder Cyclohex-2-enyl-Gruppe.
- Der Ausdruck Heterocycloalkyl bzw. Heterocyclo- bezieht sich auf eine Cycloalkylgruppe wie oben definiert, in der ein oder mehrere (bevorzugt 1, 2 oder 3) Kohlenstoffatome durch ein Sauerstoff-, Stickstoff-, Phosphor- oder Schwefelatom ersetzt sind und kann beispielsweise für die Piperidin-, Morpholin-, N-Methylpiperazin- oder N-Phenylpiperazin-Gruppe stehen.
- Die Ausdrücke Alkylcycloalkyl bzw. Heteroalkylcycloalkyl beziehen sich auf Gruppen, die entsprechend den obigen Definitionen sowohl Cycloalkyl- bzw. Heterocycloalkyl- wie auch Alkyl-, Alkenyl-, Alkinyl- und/oder Heteroalkyl gruppen enthalten.
- Der Ausdruck Aryl bzw. Ar bezieht sich auf eine aromatische Gruppe, die einen oder mehrere Ringe hat, und durch ein Gerüst gebildet wird, das 5 oder 6 bis 14 Kohlenstoffatome, vorzugsweise 5 oder 6 bis 10 Kohlenstoffatome enthält z.B. eine Phenyl-, Naphthyl-, Biphenyl-, 2-, 3- oder 4-Methoxyphenyl-, 2-, 3- oder 4-Ethoxyphenyl-, 4-Carboxyphenylalkyl- oder 4-Hydroxyphenyl-Gruppe.
- Der Ausdruck Heteroaryl bezieht sich auf eine Aryl-Gruppe, in der ein oder mehrere (bevorzugt 1, 2 oder 3) Kohlenstoffatome durch ein Sauerstoff-, Stickstoff-, Phosphor- oder Schwefelatom ersetzt sind, z.B. die 4-Pyridyl-, 2-Imidazolyl-, 3-Pyrazolyl- und Isochinolinyl-Gruppe.
- Die Ausdrücke Aralkyl bzw. Heteroaralkyl beziehen sich auf Gruppen, die entsprechend den obigen Definitionen sowohl Aryl- bzw. Heteroaryl- wie auch Alkyl-, Alkenyl-, Alkinyl- und/oder Heteroalkyl- und/oder Cycloalkyl- und/oder Heterocycloalkylgruppen enthalten, z.B. die Tetrahydroisochinolinyl-, Benzyl-, 2- oder 3-Ethylindolyl- oder 4-Methylpyridino-Gruppe.
- Die Ausdrücke Alkyl, Alkenyl, Alkenyl, Heteroalkyl, Cycloalkyl, Heterocycloalkyl, Alkylcycloalkyl, Heteroalkylcycloalkyl, Aryl, Heteroaryl, Aralkyl und Heteroaralkyl beziehen sich auch auf Gruppen, in denen ein oder mehrere Wasserstoffatome solcher Gruppen durch Fluor-, Chlor-, Brom- oder Jodatome oder OH, SH, NH2 oder NO2-Gruppen ersetzt sind. Diese Ausdrücke beziehen sich weiterhin auf Gruppen, die mit unsubstituierten Alkyl-, Alkenyl-, Alkinyl-, Heteroalkyl-, Cycloalkyl-, Heterocyclo-alkyl-, Aryl-, Heteroaryl-, Aralkyl- oder Heteroaralkyl-Gruppen substituiert sind.
- Verbindungen der Formel (I) können aufgrund ihrer Substitution ein oder mehrere Chiralitätszentren enthalten. Die vorliegende Erfindung umfasst daher sowohl alle reinen Enantiomere und alle reinen Diastereomere, als auch deren Gemische in jedem Mischungsverhältnis. Des weiteren sind von der vorliegenden Erfindung auch alle cis/trans-Isomeren der Verbindungen der allgemeinen Formel (I) sowie Gemische davon umfasst.
- Der Ausdruck Linker bezieht sich auf eine chemische Gruppe, die dazu geeignet ist, Moleküle mit dem Träger zu verbinden. Der Linker ist eine chemische Struktureinheit, die einerseits kovalent an den festen Träger und andererseits kovalent an das Phosphoratom gebunden ist. Aufgrund des Linkers kann das Zielmolekül nach einer vollendeten Synthese unter schonenden Bedingungen wieder vom Träger gespalten werden. Je nach Synthesestrategie ist es möglich, dass bei der Abspaltung ein Teil des Linkers Teil der Zielverbindung wird. Ein Linker kann eine Alkylen-, Heteroalkylen-, Arylen-, Heteroarylen-, Cycloalkylen-, Heterocycloalkylen-, Aralkylen- oder eine Heteroaralkylengruppe sein. Beispiele für Linker sind in P. Seneci, Solid-Phase Synthesis and Combinatorial Technologies, John Wiley & Sons, New York, 2000; D. Obrecht und J. M. Villalgordo, Solid-Supported Combinatorial and Parallel Synthesis of Small-Molecular-Weight Compound Libraries, Tetrahedron Organic-Chemistry Series Volume 17, Elsevier Science Ltd, Oxford, 1998 sowie in F. Z. Dörwald, Organic Synthesis on Solid Phase, Wiley-VCH, Weinheim, 2000 beschrieben. Beispielhaft seien hier der Trityllinker (gegebenenfalls in 2-Position durch ein Chloratom oder in 4-Position durch eine Methyl- oder eine Methoxygruppe substituiert), der Wang-Linker, der SASRINTM-Linker, der Rink-Säure-Linker, der Benzhydrylalkohol-Linker, der HMBA-Linker sowie polymeres Benzylhalogenid (Linker des Merrifield Harzes) sowie der PAM-Linker genannt.
- Der Ausdruck Träger bezieht sich auf ein Polymer (bevorzugt ein chemisch inertes Polymer), das für die organische Festphasensynthese modifiziert wurde, so dass chemische Substanzen kovalent daran gebunden werden können. Beispiele für polymere Träger sind in P. Seneci, Solid-Phase Synthesis and Combinatorial Technologies, John Wiley & Sons, New York, 2000; D. Obrecht und J. M. Villalgordo, Solid-Supported Combinatorial and Parallel Synthesis of Small-Molecular-Weight Compound Libraries, Tetrahedron Organic-Chemistry Series Volume 17, Elsevier Science Ltd, Oxford, 1998 sowie in F. Z. Dörwald, Organic Synthesis on Solid Phase, Wiley-VCH, Weinheim, 2000 beschrieben. Es wird zwischen löslichen und unlöslichen polymeren Trägern unterschieden. Beispiele für lösliche polymere Träger sind die Polyethylenglycole (PEG). Beispiele für unlösliche polymere Träger sind Polymere auf Polystyrol-Basis (vernetzt mit Divinylbenzol) wie z.B. das Merrifield Harz, das 2-Clorotrityl Harz, Tentakel-Polymere wie z.B. Tentagel®, Kieselgel, Controlled Pore Glass (CPG), Cellulose, Copolymere und Gold. Polymere Träger sind bei einer Reihe von Firmen wie z.B. Merck Biosciences AG, Läufelfingen, Schweiz, Bachem AG, Bubendorf, Schweiz, Rapp Polymere GmbH, Tübingen, Deutschland, Advanced ChemTech, Louisville, KY, USA sowie Shearwater Corporation, Huntsville, Alabama, USA kommerziell erhältlich. Bevorzugte Träger sind Polystyrrol (quervernetzt mit bis zu 3% Divinylbenzol (bevorzugt 1-2% DVB), Polyethylenglycolderivate und Tentakel Polymere (insbesondere Tentagel®).
- Der Ausdruck Aktivester bzw. Aktivesterrest ist dem Fachmann bekannt und z.B. in: Methoden der organischen Chemie (Houben-Weyl) Vol. E 22a. Synthesis of peptides and peptidomimetics, Thieme, Stuttgart, New York, 2002 oder in: John Jones, The chemical Synthesis of Peptides, Clarendon Press, Oxford, 1991 beschrieben. Beispiele für Aktivesterreste sind die folgenden Gruppen:
- Bevorzugt ist Pol ein mit bis zu 3% DVB (Divinylbenzol) (bevorzugt 1-2%) quervernetztes Polystyrol.
- Weiter bevorzugt ist L eine Bindung, ein Sauerstoffatom oder eine Gruppe der Formel -O-(CO)- oder -N-(CO)-.
- Wiederum bevorzugt sind R1 und R2 Benzyl-, Cyclohexyl-, C1-C6 Alkyl- oder Phenylgruppen (besonders bevorzugt Butyl- oder Phenylgruppen).
- Des weiteren bevorzugt ist R3 eine Su oder ein Np Gruppe.
- Wiederum bevorzugt ist X ein Bromatom.
- Die Verbindungen der allgemeinen Formel (I) können durch Umsetzung von Verbindungen der Formeln (II) und (III) hergestellt werden.
- Durch Umsetzung von Verbindung (I) mit einer Base (z.B. einem Trialkylamin wie z.B. Triethylamin) entsteht eine Ylen/Ylid Verbindung der Formel (IV), die z.B. mit Aminosäureestern oder Hydroxycarbonylverbindungen zu den entsprechenden Heterocyclen oder mit Aminen und Carbonylverbindungen zu den entsprechenden Chalkonen umgesetzt werden kann. Dabei kann das Ylen über das offenkettige Ylid in seiner cis- und trans-Form vorliegen.
- 1.) Beladung von Triphenylphosphin-Harz mit Bromessigsäure-N-hydroxy-succinimidester
- 10 g (15.5 mmol) Triphenylphosphin-Harz (Polymer Laboratories) werden in 70 ml abs. Toluol gequollen und dann 7.317 g (31 mmol) Bromessigsäure-N-hydroxysuccinimidester zugesetzt. Diese Mischung wird 16 Stunden geschüttelt. Es wird abgesaugt und dann das Harz dreimal mit Toluol, Dichlormethan und Diethylether gewaschen. Zum Schluß wird das Harz im Hochvakuum getrocknet. Auswaage: 13.52 g (entspricht 1.2 mmol/g laut Gewichtszunahme).
- 2.) Umwandlung des Harzes zum Ylid
- 13.52 g (16.224 mmol) Harz aus Punkt 1. werden in 80 ml Dichlormethan/Triethylamin (1:1) gequollen und 16 Stunden geschüttelt. Dann wird abgesaugt und das Harz zweimal mit Dichlormethan/Triethylamin (1:1) gewaschen. Zum Schluß wird es je dreimal mit Dichlormethan und Diethylether gewaschen und am Hochvakuum getrocknet. Auswaage: 12.2 g (entspricht 1.1 mmol/g nach Gewichtsabnahme).
- 3.) Beladung von Diphenylphosphinomethylpolystyrrol-Harz mit Bromessigsäure-N-hydroxy-succinimidester
- 1 g (1.1 mmol) Diphenylphosphinomethylpolystrrol-Harz (Novabiochem) wird in 5 ml absolutem Toluol gequollen und dann 519.3 mg (2.2 mmol) Bromessigsäure-N-hydroxy- succinimidester zugesetzt. Diese Mischung wird 16 Stunden geschüttelt. Es wird abgesaugt und dann das Harz je dreimal mit Toluol, Dichlormethan und Diethylether gewaschen. Zum Schluß wird das Harz im Hochvakuum getrocknet. Auswaage: 1.121 g (entspricht 0.46 mmol/g nach Gewichtszunahme).
- 4.) Umwandlung des Harzes zum Ylid
- 1.121 g (0.52 mmol) Harz aus Punkt 3. werden in 8 ml Dichlormethan/Triethylamin (1:1) gequollen und 16 Stunden geschüttelt. Dann wird abgesaugt und das Harz zweimal mit Dichlormethan/Triethylamin (1:1) gewaschen. Zum Schluß wird es je dreimal mit Dichlormethan und Diethylether gewaschen und im Hochvakuum getrocknet. Auswaage: 1.07 g (entspricht 0.48 mmol/g nach Gewichtsabnahme).
- 5.) Beladung von Dibutylphenylphosphinpolystyrrol-Harz mit Bromessigsäure-N-hydroxy-succinimidester
- 1 g (0.72 mmol) Dibutylphenylphosphinpolystyrrol-Harz (Novabiochem) wird in 5 ml absolutem Toluol gequollen und dann 519.3 mg (2.2 mmol) Bromessigsäure-N-hydroxy-succinimidester zugesetzt. Diese Mischung wird 16 Stunden geschüttelt. Es wird abgesaugt und dann das Harz je dreimal mit Toluol, Dichlormethan und Diethylether gewaschen. Zum Schluß wird das Harz im Hochvakuum getrocknet. Auswaage: 1.156 g (entspricht 0.57 mmol/g nach Gewichtszunahme).
- 6.) Umwandlung des Harzes zum Ylid
- 1.156 g Harz aus Punkt 5. werden in 8 ml Dichlormethan/Triethylamin (1:1) gequollen und 16 Stunden geschüttelt. Dann wird abgesaugt und das Harz zweimal mit Dichlormethan/Triethylamin (1:1) gewaschen. Zum Schluß wird es je dreimal mit Dichlormethan und Diethylether gewaschen und am Hochvakuum getrocknet. Auswaage: 1.088 g (entspricht 0.60 mmol/g nach Gewichtsabnahme).
- 7.) Beladung von Triphenylphosphin-Harz mit Bromessigsäure-4-nitrophenylester
- 3 g (4.65 mmol) Triphenylphosphin-Harz (Polymer Laboratories) werden in 15 ml abs. Toluol gequollen und dann 2.42 g (9.3 mmol) Bromessigsäure-4-nitrophenylester zugesetzt. Diese Mischung wird 16 Stunden geschüttelt. Es wird abgesaugt und dann das Harz je dreimal mit Toluol, Dichlormethan und Diethylether gewaschen. Zum Schluß wird das Harz im Hochvakuum getrocknet. Auswaage: 4.2 g (entspricht 1.11 mmol/g laut Gewichtszunahme).
- 8.) Umwandlung des Harzes zum Ylid
- 4.2 g (4.662 mmol) Harz aus Punkt 7. werden in 20 ml Dichlormethan/Triethylamin (1:1) gequollen und 16 Stunden geschüttelt. Dann wird abgesaugt und das Harz zweimal mit Dichlormethan/Triethylamin (1:1) gewaschen. Zum Schluß wird es je dreimal mit Dichlormethan und Diethylether gewaschen und im Hochvakuum getrocknet. Auswaage: 3.8 g (entspricht 1.22 mmol/g nach Gewichtsabnahme).
- 9.) Darstellung von 4-Methoxy-1H-chinolin-2-on
- 182 mg Harz (0.2 mmol) aus Punkt 2. werden in ein Smith Process Glasreaktionsgefäß gegeben und in 1 ml absolutem Dioxan gequollen. Dann werden 0.039 ml Anthranilsäuremethylester (0.3 mmol) sowie ein Magnetrührstäbchen zugesetzt und das Reaktionsgefäß verschlossen. Diese Mischung wird mit der Mikrowellensystem Smith Creator von der Firma Personal Chemistry 15 Minuten auf 180°C erhitzt. Nach dem Abkühlen wird das Reaktionsgefäß geöffnet und abfiltriert. Harz wird mehrmals mit Dioxan nachgewaschen. Es wird zur Trockne einrotiert und der Rückstand mittels präparativer HPLC gereinigt. (Reversed Phase-C18-Phase, Eluent Methanol + 0.5% Essigsäure/Wasser + 0.5% Essigsäure). C10H9NO2 (175,1887); MS (ESI): 176 [M+H].
- 10.) Darstellung von 5-Benzyl-4-methoxy-1,5-dihydropyrrol-2-on
- Vorschrift siehe Punkt 9.. Zugabe von 68.9 mg Phenylalaninethylester Hydrochlorid (0.3 mmol) und 0.042 ml Triethylamin (0.3 mmol). C13H15NO2 (217,2700); MS (ESI) 218 [M+H].
- 11.) Darstellung von 4-Styrylchromen-2-on
- Vorschrift siehe Punkt 9.. Zugabe von 67,3 mg 2-Hydroxychalcon (0,3 mmol). C17H12NO2 (248;2840); MS (ESI) 249 [M+H].
- 12.) Darstellung von 6-Bromochromen-2-on

- 100 mg Harz (0.048 mmol) aus Punkt 4. werden in ein Smith Process Glasreaktionsgefäß gegeben und in 1 ml absolutem Dioxan gequollen. Dann werden 0.0094 ml Anthranilsäuremethylester (0.072 mmol) sowie ein Magnetrührstäbchen zugesetzt und das Reaktionsgefäß verschlossen. Diese Mischung wird mit der Mikrowellensystem Smith Creator von der Firma Personal Chemistry 15 Minuten auf 180°C erhitzt. Nach dem Abkühlen wird das Reaktionsgefäß geöffnet und abfiltriert. Harz wird mehrmals mit Dioxan nachgewaschen. Es wird zur Trockne einrotiert und der Rückstand mittels präparativer HPLC gereinigt. (Reversed Phase-C18-Phase, Eluent Methanol + 0.5% Essigsäure/Wasser + 0.5% Essigsäure). C10H9NO2 (175,1887); MS (ESI): 176 [M+H].
- 22.) Darstellung von 4-Methoxy-1H-chinolin-2-on
- 100 mg Harz (0.06 mmol) aus Punkt 6. werden in ein Smith Process Glasreaktionsgefäß gegeben und in 1 ml absolutem Dioxan gequollen. Claims (6)
- Verbindungen der Formel (I), worin: Pol ein Träger ist; L ein Linker oder eine Bindung ist; R1 und R2 unabhängig voneinander ein Alkyl-, Aryl-, Cycloalkyl-, oder ein Aralkylrest sind, R3 ein Aktivesterrest und X eine Abgangsgruppe wie z.B. F, Cl, Br, I, NR1R2, OR3 oder SOnR4 ist, wobei R1, R2, R3 und R4 unabhängig voneinander ein Wasserstoffatom, ein Alkyl-, Alkenyl-, Alkinyl-, Heteroalkyl-, Aryl-, Heteroaryl-, Cycloalkyl-, Heterocycloalkyl-, Alkylcycloalkyl-, Heteroalkylcyclo-alkyl-, Aralkyl- oder ein Heteroaralkylrest sind oder R1 und R2 zusammen Teil eines gegebenenfalls substituierten Heterocycloalkylringsystems sind und n = 0, 1, 2 oder 3 ist.
- Verbindungen nach Anspruch 1, wobei Pol ein mit bis zu 3% DVB (Divinylbenzol) quervernetztes Polystyrol ist.
- Verbindungen nach Anspruch 1 oder 2, wobei L eine Bindung, ein Sauerstoffatom oder eine Gruppe der Formel -O-(CO)- oder -N-(CO)- ist.
- Verbindungen nach einem der Ansprüche 1 bis 3, wobei R1 und R2 Benzyl-, Cyclohexyl-, C1-C6 Alkyl- oder Phenylgruppen sind.
- Verbindungen nach einem der Ansprüche 1 bis 4, wobei R3 eine Su oder ein Np Gruppe ist.
- Verbindungen nach einem der Ansprüche 1 bis 5, wobei X ein Bromatom ist.
