Das neue Verfahren erlaubt den Aufbau von Verbindungen der Vitamin Α-Reihe in einfacher Weise und liefert in guten Ausbeuten reine Endprodukte; es ist einfacher als die bisher bekannten Verfahren (vgl. z. B. »Fortschritte der Chemie organischer Naturstoffe«, Springer-Verlag, Wien 1952, Bd. IX, S. 56 ff). Die nach dem Verfahren erhältlichen Produkte, wie Vitamin A, Vitamin Α-säure, Vitamin A-äther und Axerophten, sind bekanntlich wertvolle pharmazeutische Produkte.The new process allows the construction of compounds of the vitamin Α series in a simple manner and delivers pure end products in good yields; it is simpler than the previously known procedures (cf.e.g. "Advances in the chemistry of organic natural products", Springer-Verlag, Vienna 1952, vol. In eine Lösung von 20 Teilen reinem /?-Ionol (vgl. H. H. Inhoffen, F. Bohlmann und M. Bohlmann, Lieb. Ann. d. Chem., Bd. 565, 1949, S. 35) in 150 Teilen absolutem Äther wird bei +15 bis + 20° eine Lösung von 18 Teilen Phosphortribromid in 80 Teilen absolutem Äther unter Rühren allmählich eingetragen. Nach 12 Stunden Rühren bei Zimmertemperatur wird das Gemisch auf Eis gegossen und die ätherische Schicht abgehoben und gut mit Wasser gewaschen. Die Ätherlösung wird 2 Stunden mit Calciumchlorid getrocknet und dann bei vermindertem Druck etwas eingeengt. Nach Zugabe von 22 Teilen Triphenylphosphin wird 5 Stunden bei Zimmertemperatür gerührt. Dann wird das Lösungsmittel abdestilliert und der Rückstand im Wasserstrahlvakuum 30 Minuten auf 80 bis 90° erhitzt. Das zum Teil kristalline Phosphoniumbromid wird in 150 Teilen Dimethylformamid gelöst und unter Rühren bei Zimmertemperatur mit 4,5 Teilen fein gepulvertem Natriumacetylid versetzt. Es tritt sofort Ylidbildung ein, die an der rötlichbraunen Verfärbung erkennbar ist. Nach lstündigem Rühren bei etwa 20° wird eine Lösung von 5 Teilen 4-Methyl-hexadien-(2, 4)-al-(l)-säure-(6)-äthylester in 15 Teilen Dimethylformamid zugegeben. Es tritt geringe Temperaturerhöhung ein; man rührt das Gemisch noch 2 Stunden, gibt es auf ein Gemisch aus Eis und 10°/0iger Phosphorsäure und extrahiert es mit Petroläther. Die hellgelbe Petrolätherlösung wird mit Wasser gewaschen, über Natriumsulfat getrocknet und 10 Stunden bei — 5° aufbewahrt. Dann wird das ausgeschiedene Triphenylphosphinoxyd abfiltriert. Aus dem Filtrat destilliert man unter einem Druck von 0,001 mm Hg alle unterhalb .100° übergehenden Anteile ab. Der Rückstand (etwa 8 Teile) besteht aus Vitamin A-säureäthylester (Xmax = 351 bis 353 ηιμ, ε = 28 500 [in Methanol]), aus dem man durch alkalische Verseifung 4,8 Teile Vitamin Α-säure als gelbe Nadeln vom F. = 181 bis 182° erhalten kann.In a solution of 20 parts of pure /? - ionol (cf. Unter Temperaturerhöhung tritt Reaktion ein, die nach 2 Stunden Rühren ohne äußere Wärmezufuhr beendet ist. Nun wird das Reaktionsprodukt auf ein Gemisch aus Eis und 10°/0iger Phosphorsäure gegossen und mit Petroläther extrahiert. Die Petrolätherlösung wird mit Wasser neutral gewaschen, über Natriumsulfat getrocknet und 12 Stunden bei — 5° aufbewahrt.
