Verfahren zur Herstellung von Verbindungen der Vitamin-AReihe
Es wurde gefunden, dass man Verbindungen der Vitamin-A-Reihe erhält, wenn man ein p-Jonyl- halogenid mit einem Phosphin, z. B. Triphenylphosphin, zum quartären Phosphoniumhalogenid umsetzt, dieses, zweckmässig in Dimethylformamidlösung, mit einer äquivalenten Menge eines Alkali- oder Erdalkalimetallalkoholats oder eines Alkaliacetylids in das p-Jonylidenphosphinylid umwandelt, auf dieses einen Aldehyd der Formel O -CH-CH=CH-C(CH3) = CHOR einwirken lässt, worin R eine Alkyl-, eine Carbonsäureester- oder eine gegebenenfalls verätherte oder veresterte CH2OH-Oruppe bedeutet, und das als Nebenprodukt entstandene Phosphinoxyd abtrennt.
Die Umsetzung lässt sich im Falle der Verwendung von fl-Jonylbromid, Triphenylphosphin, Natriumacetylid und 4-Methylhexadien-(2,4)-al-(1)-säure-(6)- äthylester wie folgt formulieren:

Das neue Verfahren erlaubt den Aufbau von Verbindungen der Vitamin-A-Reihe in einfacher Weise und liefert in guten Ausbeuten reine Endprodukte; es ist einfacher als die bisher bekannten Verfahren (vgl. z. B. Fortschritte der Chemie organischer Naturstoffe , Springer-Verlag, Wien 1952, Bd. IX, Seite 56ff.). Die nach dem erfindungsgemässen Verfahren erhältlichen Produkte, z. B. Vitamin A, Vitamin-A-Säureester, Vitamin-A-Äther und Axerophten, sind bekanntlich wertvolle pharmazeutische Produkte.
Die in den Beispielen genannten Teile sind Gewichtsteile.
Beispiel 1
In eine Lösung von 20 Teilen reinem ss-Jonol (vgl.
H. H. Inhoffen, F. Bohlmann und M. Bohlmann, Ann. 565, 1949, 35) in 150 Teilen abs. Äther wird bei +15 bis +20 C eine Lösung von 18 Teilen Phosphortribromid in 80 Teilen abs. Äther unter Rühren allmählich eingetragen. Nach 12 Stunden Rühren bei Zimmertemperatur wird das Gemisch auf Eis gegossen und die ätherische Schicht abgehoben und gut mit Wasser gewaschen. Die Ätherlösung wird 2 Stunden mit Calciumchlorid getrocknet und dann bei vermindertem Druck etwas eingeengt. Nach Zugabe von 22 Teilen Triphenylphosphin wird 5 Stunden bei Zimmertemperatur gerührt. Dann wird das Lösungsmittel abdestilliert und der Rückstand im Wasser strahlvakuum 30 Minuten auf 80 bis 90"C erhitzt.
Das z. T. kristalline Phosphoniumbromid wird in 150 Teilen Dimethylformamid gelöst und unter Rühren bei Zimmertemperatur mit 4,5 Teilen fein gepulvertem Natriumacetylid versetzt. Es tritt sofort Ylidbildung ein, die an der rötlichbraunen Verfärbung erkennbar ist. Nach 1 Stunde Rühren bei etwa 20"C wird eine Lösung von 5 Teilen 4-Methyl-hexadien (2,4)-al-(1)-säure-(6)-äthylester in 15 Teilen Dimethylformamid zugegeben. Es tritt geringe Temperaturerhöhung ein; man lässt noch 2 Stunden rühren, gibt auf ein Gemisch aus Eis und 10% der Phosphorsäure und extrahiert mit Petroläther. Die hellgelbe Petrol ätherlösung wird mit Wasser gewaschen, über Natriumsulfat getrocknet und 10 Stunden bei -5"C aufbewahrt. Dann wird vom ausgeschiedenen Triphenylphosphinoxyd filtriert.
Aus dem Filtrat destilliert man unter 0,001 mm Druck alle unterhalb 100"C übergehenden Anteile ab. Der Rückstand besteht aus Vitamin-A-Säureäthylester (max. = 351-353m, u, e = 28500 (in Methanol), aus dem man durch alkalische Verseifung 4,8 Teile Vitamin-A-Säure als gelbe Nadeln vom Fp. 181-182"C erhalten kann.
Beispiel 2
Zu einer wie im Beispiel 1 aus 20 Teilen ss-Ionol hergestellten Lösung von ss-Jonyliden-triphenylphos- phin-ylid in Dimethylformamid gibt man eine Lösung von 6 Teilen y-Methylsorbinaldehyd in 20 Teilen Dimethylformamid. Unter Temperaturerhöhung tritt Reaktion ein, die nach 2 Stunden Rühren ohne äussere Wärmezufuhr beendet ist. Nun wird auf ein Gemisch aus Eis und 10% iger Phosphorsäure gegossen und mit Petroläther extrahiert. Die Petrol ätherlösung wird mit Wasser neutral gewaschen, über Natriumsulfat getrocknet und 12 Stunden bei -5"C aufbewahrt.
Dann wird durch eine kurze Aluminiumoxydsäule filtriert und das Filtrat destilliert, wobei 4,3 Teile Axerophten vom Kp.,,,, = 140-143"C als gelbes zähes Ö1 erhalten werden [Ärnas. = 322-324m, u, 8 = 38000 (in Hexan) j. Mit Antimonchlorid erhält man eine rotviolette Farbreaktion.
Beispiel 3
In eine Lösung von 40 Teilen ss-Jonol in 250 Teilen abs. Äther trägt man bei +10 C unter Rühren eine Lösung von 40 Teilen Phosphortribromid in 60 Teilen abs. Äther allmählich ein. Nach 6 Stunden Rühren wird auf Eis gegossen und gut mit Wasser gewaschen.
Die gewaschene ätherische Lösung von ss-Jonylbromid wird mit 20 Teilen Calciumchlorid 20 Minuten geschüttelt, filtriert und bei vermindertem Druck bei höchstens 35"C Badtemperatur vom Lösungsmittel befreit. Das so erhaltene rohe ss-Jonylbromid wird in 45 Teile geschmolzenes Triphenylphosphin eingerührt, wobei unter Selbsterwärmung bis auf etwa 90"C das ss-Jonyltriphenylphosphoniumbromid entsteht. Die Ausbeute beträgt etwa 100 Teile.
Die Lösung dieses Phosphoniumsalzes in 200 Teilen Dimethylformamid wird allmählich mit einer l0%igen methanolischen Natriummethylatlösung versetzt, bis ein pEr-Wert von 7,9 erreicht ist. Zu der so erhaltenen dunkelvioletten Lösung von ss-Jonylidentriphenyl- phosphin-ylid lässt man allmählich unter Rühren eine Lösung von 30 Teilen 4-Methyl-hexadien-(2,4)-al-(1)säure-(6)-äthylester in 30 Teilen Dimethylformamid zulaufen. Nach 60 Minuten Erwärmen auf 70"C wird das Methanol im Vakuum abdestilliert und die Dimethylformamidlösung unter Rühren auf ein Gemisch aus Eis und 10% iger Schwefelsäure gegossen.
Dann wird mit Petroläther extrahiert; die mit Wasser neutral gewaschenen Extrakte werden mit Natriumsulfat 5 Stunden bei -5"C getrocknet. Nach Verjagen des Lösungsmittels verbleiben 50 Teile Rückstand, die im wesentlichen aus cis-Vitamin-A-Säureäthylester bestehen.
Zur Isolierung von cis-Vitamin-A-Säure resp. Umwandlung in trans-Vitamin-A-Säureester kann man wie folgt verfahren:
Cis-Vitamin-A-Säure: 25 Teile des oben erhaltenen cis-Esters werden in 150 Teilen Alkohol gelöst und mit 5 Teilen Kaliumhydroxyd in 10 Teilen Wasser 30 Minuten unter Rückfluss gekocht. Der Alkohol und nicht verseifbare niedermolekulare Anteile werden mit Wasserdampf abgetrieben. Nach Ansäuern mit 10% iger Phosphorsäure, Aufnehmen der ausfallenden Säure mit Äther und Verdampfen des Äthers werden 11 Teile hellgelbe Kristalle erhalten, die nach Umkristallisieren aus Methanol bei 146 C schmelzen [Amax. 330ru, E = 42000 (in Methanol)]; sie bestehen demnach aus cis-Vitamin-A-Säure.
Trans- Vitamin-A-Säureäthylester: 25 Teile des oben erhaltenen rohen cis-Esters werden im Hochvakuum destilliert, wobei unter 0,005 mm Druck bei 145-150 C der trans-Vitamin-A-Säureäthylester [Amaxt = 351m, ± = 38000 (in Methanol)] in einer Ausbeute von 13 Teilen übergeht. Durch Verseifung kann man daraus in üblicher Weise die trans-Vitamin-A-Säure vom Fp. 181-182"C erhalten.

The new process allows the construction of compounds of the vitamin A series in a simple manner and gives pure end products in good yields; it is simpler than the previously known processes (see, for example, advances in the chemistry of organic natural substances, Springer-Verlag, Vienna 1952, vol. IX, page 56ff.). The products obtainable by the process of the invention, e.g. B. Vitamin A, vitamin A acid esters, vitamin A ethers and axerophtes are known to be valuable pharmaceutical products.
The parts mentioned in the examples are parts by weight.
example 1
In a solution of 20 parts of pure ss-ionol (cf.
H. H. Inhoffen, F. Bohlmann and M. Bohlmann, Ann. 565, 1949, 35) in 150 parts abs. At +15 to +20 C, ether becomes a solution of 18 parts of phosphorus tribromide in 80 parts of abs. Example 2
A solution of 6 parts of γ-methylsorbinaldehyde in 20 parts of dimethylformamide is added to a solution of β-ionylidene-triphenylphosphin-ylide in dimethylformamide, prepared from 20 parts of β-ionol as in Example 1. With a rise in temperature, a reaction occurs which is ended after stirring for 2 hours without external heat input. It is then poured onto a mixture of ice and 10% phosphoric acid and extracted with petroleum ether. The petroleum ether solution is washed neutral with water, dried over sodium sulfate and stored at -5 ° C. for 12 hours.

Example 3
In a solution of 40 parts of ss-ionol in 250 parts of abs. Ether is carried at +10 C with stirring, a solution of 40 parts of phosphorus tribromide in 60 parts of abs. Ether gradually one. After stirring for 6 hours, it is poured onto ice and washed well with water.
The washed ethereal solution of ss-ionyl bromide is shaken with 20 parts of calcium chloride for 20 minutes, filtered and freed from the solvent under reduced pressure at a maximum bath temperature of 35 "C. The crude ss-ionyl bromide thus obtained is stirred into 45 parts of molten triphenylphosphine, with self-heating ss-Jonyltriphenylphosphoniumbromid is formed up to about 90 "C. A 10% strength methanolic sodium methylate solution is gradually added to the solution of this phosphonium salt in 200 parts of dimethylformamide until a pEr value of 7.9 is reached. Cis-vitamin A acid: 25 parts of the cis-ester obtained above are dissolved in 150 parts of alcohol and refluxed with 5 parts of potassium hydroxide in 10 parts of water for 30 minutes. The alcohol and unsaponifiable low molecular weight fractions are driven off with steam. After acidification with 10% phosphoric acid, absorption of the precipitating acid with ether and evaporation of the ether, 11 parts of light yellow crystals are obtained which, after recrystallization from methanol, melt at 146 ° C.
