Abstract
1.- Proceso para la modificación de un sustrato, que comprende pasar el sustrato a través de una columna de lecho empacado de un volumen específico de enzima inmovilizada, caracterizado porque: a) el sustrato entra a la columna en o cerca de un extremo de la columna (el "extremo de entrada") y el sustrato modificado sale en o cerca del extremo opuesto de la columna (el "extremo de salida"), b) una porción del volumen de la enzima modificada es periódicamente removida en o cerca del extremo de entrada de la columna, y c) una porción de enzima inmovilizada es periódicamente agregada en o cerca del extremo de salida de la columna. 2.- Proceso de conformidad con la reivindicación 1, caracterizado porque la columna es sustancialmente vertical con su extremo de entrada en o cerca del fondo de la columna y su extremo de salida en o cerca de la parte superior de la columna. 3.- Proceso de conformidad con la reivindicación 1 o reivindicación 2, caracterizado porque es esencialmente un proceso continuo. 4.- Proceso de conformidad con cualquiera de las reivindicaciones precedentes, caracterizado porque la porción de la enzima inmovilizada la cual es removida de la columna, es menos de 35% del volumen total.5.- Proceso de conformidad con cualquiera de las reivindicaciones precedentes, caracterizado porque la porción de enzima inmovilizada la cual es removida de la columna, es sustancialmente equivalente en volumen a la porción de la enzima inmovilizada la cual es agregada.6.- Proceso de conformidad con cualquiera de las reivindicaciones precedentes, caracterizado porque las etapas de remover la enzima inmovilizada de y de agregar enzima inmovilizada a la columna, ocurren sustancialmente de manera concurrente.7.- Proceso de conformidad con cualquiera de las reivindicaciones precedentes, caracterizado porque la porción removida de enzima inmovilizada es usada para pre-tratar el sustrato antes de que entre a la columna.8.- Proceso de conformidad con la reivindicación 7, caracterizado porque la porción removida de la enzima inmovilizada es regenerada o reactivada antes de ser usada para pre-tratar el sustrato.
