Abstract

一种用于斑点酶免疫测定的特制固相物,以硝酸纤维素滤膜(NC)为载体,用于各种抗原抗体快速检测。其特征是将NC制成带梳齿的梳状薄膜,梳齿上包被有微量的供检测样本用的已知抗原抗体,检测时配用含有小孔(当小试管用)的塑料板,本发明可同时检测多份标本,操作简化,灵敏度高,为在NC上采用斑点酶联免疫吸附法(Dot-ELISA)并制成成本低的试剂盒创造了条件。

Description

本实用新型属于免疫学技术领域,特别是一种以特制硝酸纤维素滤膜(NC)为载体的以斑点酶联免疫吸附法(Dot-ELISA)进行各种抗原、抗体系统方面快速检测用的固相物。
目前,国内外采用酶联免疫吸附技术进行抗原、抗体的检测时,大多以表面密排着若干小孔的聚苯乙烯塑料板作为固相载体,并制成各种商品化试剂盒出售,使用这种载体的塑料板因载体的反应表面积较大,尚存在难以微量化,且操作费时,检测速度慢等缺点。近几年,虽已在实验室建立了采用以硝酸纤维素滤膜(NC)为固相载体的斑点酶联免疫吸附测定法(Dot-ELISA),但目前在国内由于没有特殊加工处理的、专供抗原抗体检测的商品化NC,致使应用NC进行Dot-ELISA检测时,操作仍不方便,工作效率低,无法制成商品化的试剂盒产品。
本实用新型的任务是提供一种能在NC上采用Dot-ELISA法进行抗原、抗体检测的使操作简化、省时省料又实用的特制固相物,并在测定中配用带有若干小孔(起微型试管作用)的塑料板,使得Dot-ELISA法易转化制成商品化的试剂盒产品。
本实用新型主要是采用带梳状的硝酸纤维素滤膜(NC)来实现,即将NC滤膜加工成带有若干梳齿的梳状薄膜,然后在梳齿尾部包被上微量的供检测样本用的已知抗原或抗体,另配用可供插入这些梳齿的带有若干小孔的塑料板(小孔起微型试管的作用),使抗原或抗体在微量情况下在这些小孔中进行反应。
使用本实用新型固相物可同时检测多份标本,且检测时间快速(在1~2小时内),灵敏度高(如对乙肝病毒可检出0、4~10ul的抗原或抗体),操作方便,又能避免标本彼此之间的相互混淆。由于可用于多种抗原抗体系统的检测,有利于微生物学快速检测法的建立,为Dot-ELISA法转化制成成本低的试剂盒创造了条件。

附图中:1、梳状薄膜,2、梳齿,3、抗原或抗体,4、塑料板,5、小孔。
参照附图,每相邻两个梳齿2之间的距离(齿距)应与小孔5之间的相互距离(孔距)相同,每个梳齿2本身的宽度应小于小孔5的直径,这样,在进行检测操作时才便于将梳状薄膜1上的梳齿2插入小孔5内。起微型试管作用的小孔5整齐地排列在塑料板4上。
应用本实用新型检测的实例:
为了要知道某甲和某乙有否受到乙型肝炎病毒感染,可利用本实用新型所制成的乙肝实验诊断试剂盒,快速检测待测人血清中有无乙肝病毒表面抗原(HBsAg)的存在。具体检测操作如下:在塑料板4的小孔5中分别加入待测对象的血清和酶标单克隆抗-HBs各40ul,同时在另外两个小孔5中分别作已知阳性和阴性的血清对照;取出梳状薄膜1(靠近梳齿2的尾部已包被有抗-HBs的薄膜),将梳齿2插入到小孔5中,让它们在37℃温箱内反应1小时左右;然后取出梳状薄膜1,经洗涤后,以相应底物显色观察结果,即可判定出待测标本为阳性或是阴性(阳性者显色,表示血清中有HBsAg,阴性者不显色,表示血清中未测到HBsAg)。

Claims (2)

1、一种可用于多种抗原、抗体系统检测的斑点酶免疫测定用的固相物,该固相物以硝酸纤维素滤膜(NC)为载体,检测中配用带有小孔5的塑料板4,其特征是作为固相物的硝酸纤维素滤膜(NC)被制成带有梳齿2的梳状薄膜1,在梳齿2尾部包被有微量的供检测样本用的已知抗原、抗体。
2、按权利要求1所述的固相物,其特征是梳齿2之间的相互距离(齿距)与呈整齐排列的小孔5之间的相互距离(孔距)相同,梳齿2的宽度小于小孔5的直径。
