技术领域
本发明涉及抗微生物和除臭组合物的领域。此外,本发明还涉及人体表面及其紧贴环境除臭效益的实现。所涉及的组合物和方法利用过渡金属螯合剂和特定的苯酚类或烯醇类化合物。当用于人体上时,本发明的组合物和方法当用于身体的最恶臭区域例如腋下区域或脚时一般具有最大效益。
背景技术
抗微生物和除臭剂组合物可以通过多种多样的方式发挥功效。当用于人体上时,这样的组合物可以通过要么减少出汗要么直接影响以皮肤为代表的人体表面上的微生物来显著减少微生物数目而发挥作用。正是有这后一种作用机理,本发明才受到很大关注。
大多数抗微生物和除臭组合物都使皮肤表面上活微生物的数目减少。众所周知的是,汗通常是无臭的,直至它被皮肤微生物群降解为止。典型的除臭剂包括乙醇和众所周知的抗微生物剂三氯生(2,4,4′-三氯-2′-羟基二苯醚)。然而,用这样的除臭剂所得到的除臭效果随着时间的流逝而逐渐消失,微生物群又渐进式地恢复其数目。因此,目前市场上仍然需要有效的和长时间持续的除臭组合物。因而,要解决的问题不只是减少人体表面的微生物数目;同等重要的是保持人体表面(尤其最恶臭区域,例如腋窝)上的低微生物数目(尤其低细菌数目)。
包含螯合剂的除臭组合物的描述见我们最近的英国专利申请GB0001133.8、GB 0001132.0、GB 0001131.2、GB 0001130.4、和GB0001129.6,这些全部列为本文参考文献。这些专利申请公开了如下假设:某些螯合剂能有效地抑制皮肤表面上的微生物对必需过渡金属离子营养物的摄取,从而使其生长降到最低。这些专利申请中给出了包含过渡金属螯合剂的除臭组合物的其它参考文献,以下详细说明其中最相关者。
US 4,356,190(Personal Products Co.)公开了所选择氨基多羧酸化合物用于抑制皮肤表面上棒状杆菌属(
Corvnebacterium)的短链脂肪酸生成的用途。
WO 97/02010(宝洁公司)公开了选自琥珀酸、戊二酸、和膦酸类别的螯合剂作为杀菌化合物的用途。
WO 97/44006(Ciba Speciality Chemicals Holding,Inc.)公开了含氮络合剂用于皮肤和纺织品纤维材料的抗微生物处理的用途。
WO 97/01360(Concat Ltd.)公开了使用特定的有取代多氮杂螯合剂化合物抑制细菌生长的方法。
在本调研的进程中,我们已经观察到,过渡金属螯合剂与苯酚类或烯醇类转铁蛋白离解促进剂的组合使用,维持了特别长时间持续的恶臭控制。
哺乳动物汗包括人体汗中转铁蛋白的存在是十分肯定的(见例如S.E.Lind,Corros,Sci.,1972,12(9),749)。也已经知道的是,某些细菌借助于包括铁载体和转铁蛋白的细胞表面受体的极复杂铁清除系统来利用与转铁蛋白结合的铁(E.Griffiths和P.Williams,The IronUptake Systems of Pathogenic Bacteria,Fungi和Protozoa inIron和Infection(editors:S.S.Bullen和E.Griffiths),2nd Edn.,1999,John Wiley和Sons,pp87-212)。除细菌性铁载体外,某些化学剂也能促进转铁蛋白中铁的释放,而且正是由于这种类型的特定转铁蛋白离解促进剂的使用,本发明才受到很大关注。具体地说,本发明涉及借助于与转铁蛋白结合的铁(III)还原成与转铁蛋白结合得不太强的铁(II)起作用的苯酚类或烯醇类转铁蛋白离解促进剂(见N.Kojima和G.W.Bates,J.Biol.Chem.,1979,254(18),8847)。
先有技术文献中,作为化妆组合物的防腐剂/抗氧剂系统,公开了某些过渡金属螯合剂和能用来作为转铁蛋白离解促进剂的某些药剂。然而,所公开的数量典型地是非常小的,此外,这些文献中公开的转铁蛋白离解促进剂也未被认识到如此程度。一个典型实例是EP 979,644(ITBR-N),它公开了一种包含0.05%丁基化羟基甲苯(BHT)和0.05%乙二胺四乙酸二钠(Na2EDTA)的除臭组合物。
发明内容
我们已经令人惊讶地发现,通过有效数量的过渡金属螯合剂和特定苯酚类或烯醇类化合物的组合使用,可以实现优异的抗微生物效益和除臭效益。
因此,按照本发明的第一方面,提供一种实现抗微生物和除臭效益的方法,包含向人体或向可紧贴人体穿着的物品上施用一种抗微生物产品,该产品包含有效数量的一种过渡金属螯合剂和一种苯酚类或烯醇类化合物,即(a)借助于与转铁蛋白结合的铁(III)还原成铁(II)起作用的转铁蛋白离解促进剂,和/或(b)一种包含叔丁基苯酚基团的抗氧剂。
在本发明的上述方面中,“有效数量”系指对于在该抗微生物产品施用后24小时要观察的显著除臭效益来说足够的数量。“施用”涉及这两种有效成分在所处理基材上的沉积。
按照本发明的第二方面,提供一种能在人体上使用的抗微生物除臭组合物,包含至少0.35wt%过渡金属螯合剂和至少0.05wt%苯酚类或烯醇类化合物,即(a)借助于与转铁蛋白结合的铁(III)还原成铁(II)起作用的转铁蛋白离解促进剂,和/或(b)一种包含叔丁基苯酚基团的抗氧剂,其中,这些重量百分率不包括所存在的任何挥发性推进剂。
按照本发明的第三方面,提供一种可在人体上使用的除臭组合物的制造方法,包含形成如下的一种混合物:至少0.35wt%过渡金属螯合剂和至少0.05wt%苯酚类或烯醇类化合物,即(a)借助于与转铁蛋白结合的铁(III)还原成铁(II)起作用的转铁蛋白离解促进剂,和/或(b)一种包含叔丁基苯酚基团的抗氧剂,其中,这些重量百分率不包括所存在的任何挥发性推进剂。
具体实施方式
在本文中所述的一种过渡金属螯合剂和一种苯酚类或烯醇类化合物的组合使用时,发现了优异的抗微生物和除臭效益。不想受理论约束,假设该过渡金属螯合剂有助于通过抑制皮肤表面上的微生物对必需过渡金属离子营养物尤其铁(III)的摄取从而使其生长最小化所达到的效益。进一步的假设是,该苯酚类或烯醇类化合物通过干扰皮肤表面上的微生物从人体铁载体蛋白络合物转铁蛋白中夺取铁(III)的生物化学途径做出贡献。
在采用按照本发明的第一方面的方法时,并非必需的是,该过渡金属螯合剂和该苯酚类或烯醇类化合物是相同组合物的组成部分。本发明的使用所产生的抗微生物和除臭效益可以通过该螯合剂和该苯酚类或烯醇类化合物的独立施用获得。这样的施用可以是同时的也可以是相继的,只要所处理的基材同时经历这两种成分的存在即可。当这些成分从独立的组合物施用时,较好的是该产品也包含用来使这两种组合物施用到人体上的手段和/或说明书。
较好的是,该过渡金属螯合剂和该苯酚类或烯醇类化合物都是从同一种组合物施用的。
本发明所提供的恶臭控制方法是特别有用的,因为在该产品施用后该效益可以持续很多小时,例如10小时、24小时、或甚至更长。这可以代表一种长效除臭效益,就是说,能长时间抑制人体或密切相关物品上臭味的产生。
如以上所述,本发明也可以防止可紧贴人体穿着的物品产生恶臭。这样的物品包括任何紧贴皮肤穿着的服装,例如长统袜和短袜,以及鞋类和其它脚上穿着物品。
该过渡金属螯合剂和苯酚类或烯醇类化合物可以存在于呈任何形式的本发明的一种或多种组合物中。例如,这些药剂中任意一种或两种均既可以以净相使用,也可以用一种挥发性推进剂稀释并作为气雾剂使用,用另外一种液体稀释并作为例如一种裹身褡或挤压式喷雾产品使用,或者用一种载体液体和一种增稠剂或结构剂稀释并作为例如一种霜剂、凝胶剂、软固体剂、或固体棒状产品使用。
实现本发明的抗微生物和除臭效益的方法,当包含对人体施用该抗微生物产品时,是最具药效的。
本发明的抗微生物产品可以用任何手段施用列人体上。液体组合物的施用可以通过吸收到像纸、织物、或海绵这样的载体基体上并通过让所述载体基体与身体表面接触来施用。固体或半固体组合物可以通过直接接触来施用,也可以在施用前溶解或分散于一种液体介质中。施用也可以包含以上技术中任何两种或多种的组合。
过渡金属螯合剂
较好的过渡金属螯合剂是其阴离子对铁(III)有亲和力、较好对铁(III)有高亲和力的酸或盐,就是说,对铁(III)的结合常数大于1010,或对于最佳性能而言,大于1026。以上提到的“铁(III)结合常数”是该螯合剂-铁(III)络合物的绝对稳定性常数。这样的值与pH无关,而只考虑该螯合剂的最阴离子的、充分去质子的形式。可以用电位差法以及很多其它方法进行测量。适用方法的充分细节可参阅“Determination和Use of Stability Constants”,A.E.Martell和R.J.Motekaitis(VCH,New York,1989)。适用数值表可查阅许多资料源,例如“Critical Stability Constants”,R.M.Smith和A.E.Martell(Plenum Pub.Corp.,1977)。
本发明中使用的螯合剂较好有带至少两个可离解酸基团的酸形式。该酸基团较好是羧酸的和/或膦酸的,但可以是磺酸的或次膦酸的,或这些基团的任何一种混合。
有膦酸基团的较好螯合剂是呈酸形式的二亚乙基三胺五(甲基膦酸)(DTPMP)、乙烷羟基二膦酸(EHDP)、乙二胺四(亚甲基膦酸)(EDTMP)、和六亚甲基二胺四(亚甲基膦酸)(HMDTMP)。
有羧酸基团的较好螯合剂是氨基多羧酸化合物。较好的氨基多羧酸化合物的酸形式包括乙二胺四乙酸(EDTA)、反式-1,2-二氨基环己烷-N,N,N′,N′-四乙酸(CDTA)、和乙二胺二琥珀酸(EDDS)。更好的氨基多羧酸螯合剂有酸形式的二亚乙基三胺五乙酸(DTPA)、三亚乙基四胺六乙酸(TTHA)、和乙二〔2-(2-羟基苯基)甘氨酸〕(EDDHA)。
特别好的螯合剂是当以3×10-6mol.dm-3或更小的浓度存在时能显著抑制表皮葡萄球菌(
Staphylococcus epidermidis)这种微生物的生长。当能使支持培养基上表皮葡萄球菌(
S.epidermidis)的生长减少至少30%、较好至少45%时,就认为抑制是显著的。实施例中给出了适用于测定表皮葡萄球菌(
S.epidermidis)生长抑制程度的试验。DTPA和TTHA是能实现显著抑制的螯合剂;然而,EDTA和CDTA不是。
该螯合剂较好只有适度分子量,这指的是呈其酸形式的螯合剂具有1000以下、更好200~800、最好290~580的分子量,呈其盐形式的螯合剂具有2000以下、更好300~1400、最好500~1000的分子量。
与该螯合剂阴离子缔合的阳离子的性质,就组合物均匀性和稳定性而言,是非常重要的。较好,该螯合剂具有有机阳离子。该螯合剂盐可以是该螯合剂酸基团用一种有机碱完全中和或部分中和的结果。也包括该螯合剂酸基团用一种有机碱部分中和以及用一种无机碱部分中和而得到的盐,尽管较好的是这样的盐占其用该有机碱中和的有效酸基团的至少40%。
适用的有机阳离子是质子化的或季碱化的胺。用脂肪族胺生成的盐优于从芳香族胺生成的盐。进一步较好的是,用来生成该螯合剂盐的胺是相对疏水性的,具有至少一个包含C1~C10末端烃基基团的N-取代基。
这里的烃基基团是只包含碳原子和氢原子的基团。
该螯合剂盐的较好质子化或季碱化胺阳离子有式R1R2R3R4N(+),式中R1是H或CH3,R2、R3、和R4各自独立地是H或一个含有0~3个羟基基团、任选地有醚、胺、酯、或酰胺等官能团插入和/或取代的脂肪族或芳香族取代基,先决条件是,R2、R3、或R4中至少一个包含一个C1~C10末端烃基基团,任选地R2和R3一起形成一个作为该末端烃基基团的环。
以上提到的,式R1R2R3R4N(+)的较好过渡金属螯合剂中,特别好的是其阳离子的特征在于R2、R3、或R4中至少一个包含一个直接连接到N原子或O原子上的H原子的过渡金属螯合剂。R2、R3、或R4中至少一个中,一个直接连接到O原子上的H原子(即一个羟基)的存在是特别好的,且可多达以上所述每个N-取代基3个羟基的极限。
其它特别好的过渡金属螯合剂盐的阳离子包含总体上含有合计0~3个羟基基团、较好0~2个羟基基团的N-取代基(按照该式的R1、R2、R3、和R4)。
在很多理想的螯合剂盐中,每个N-取代基(按照该式的R1、R2、R3、和R4)含有不多于一个羟基基团。
特别好的螯合剂盐的阳离子是诸如下列的质子化脂肪族胺盐:异丙醇胺、2-氨基-2-乙基-1,3-丙二醇、2-(N,N-二甲基氨基)-2-甲基-1-丙醇和N,N-二甲基氨基乙醇。尤其好的是2-氨基-2-甲基-1-丙醇(AMP)、二异丙醇胺、2-氨基丁-1-醇、和环己基胺等的盐。
该过渡金属螯合剂较好以它是其组成部分的、不包括也存在的任何挥发性推进剂的组合物的至少0.35wt%的水平使用。具体地说,该螯合剂是以它存在于其中的、不包括也存在的任何挥发性推进剂的组合物的0.45wt%~7wt%的水平、尤其以0.65wt%~5wt%的水平存在。螯合剂的混合物也可以使用。
苯酚类或烯醇类化合物
苯酚类或烯醇类化合物是(a)借助于使与转铁蛋白结合的铁(III)还原成铁(II)起作用的转铁蛋白离解促进剂和/或(b)一种包含叔丁基苯酚基团的抗氧剂。以下试验合格的材料是(a)类化合物:
2.7g.l-1人体二高铁转铁蛋白(购自Sigma Chemicals公司)在用氢氧化钠调整到pH6.5的50mM HEPES(N-〔2-羟基乙基〕哌嗪-N′-〔2-乙磺酸〕)缓冲剂中,连同0.115g.l-1 FerroZine(购自Sigma Chemicals公司;3-(2-吡啶基)-5,6-二(4-苯基磺酸)-1,2,4-三嗪单钠盐)和10mmol.l-1试验材料(当水溶性时)或2.5g.l-1试验材料(当不溶于水时作为精细研磨的粉末添加)一起在37℃培养。
24小时后,用肉眼或分光光度计法估计转铁蛋白离解程度。转铁蛋白离解促进剂导致紫色着色;具体地说,它们导致在562nm处0.15或更大的吸收。这种紫色着色是铁(II)三(FerroZine)络合物产生的结果。这种络合物的摩尔浓度等于从该转铁蛋白离解的铁的摩尔浓度。
按照本发明的较好水溶性转铁蛋白离解促进剂是:抗坏血酸(及其盐),磷酸抗坏血酸酯钠,托可索仑,原儿荼酸(及其盐),水杨酸(及其盐),Tiron(4,5-二羟基-1,3-苯二磺酸)。较好的水不溶性材料是棕榈酸抗坏血酸酯-6,丁子香酚,阿魏酸(及其盐),百里酚,Trolox(6-羟基-2,5,7,8-四甲基苯并二氢吡喃-2-羧酸)(及其盐),生育酚,BHA(丁基化羟基茴香醚)和BHT(丁基化羟基甲苯)。按照本发明特别好的转铁蛋白离解促进剂包含一个叔丁基苯酚基团。
包含叔丁基苯酚基团的较好苯酚类或烯醇类化合物是包含一种有两个叔丁基取代基的苯酚基团的化合物,例如BHT、2,2′-偏亚乙基二(4,6-二叔丁基苯酚),和季戊四醇四(3-(3,5-二叔丁基-4-羟基苯基)丙酸酯)。
特别好的苯酚类或烯醇类化合物是抗坏血酸(及其盐)、棕榈酸抗坏血酸酯-6、BHT、2,2′-偏亚乙基二(4,6-二叔丁基苯酚),和季戊四醇四(3-(3,5-二叔丁基-4-羟基苯基)丙酸酯)。尤其好的是BHT。
该苯酚类或烯醇类化合物较好以它是其组成部分的、不包括也存在的任何挥发性推进剂的组合物的至少0.05wt%的水平使用。也较好的是,该使用水平是它是其组成部分的、不包括也存在的任何挥发性推进剂的组合物的5wt%以下。具体地说,该苯酚类或烯醇类化合物是以它是其组成部分的、不包括也存在的任何挥发性推进剂的组合物的0.075wt%~2.5wt%的水平、尤其以0.1wt%~1wt%的水平使用。苯酚类或烯醇类化合物的混合物也可以使用。
苯酚类或烯醇类化合物与过渡金属螯合剂的重量比较好是1∶20~2∶1、尤其1∶10~1∶1。
任选的附加成分
载体材料是本发明产品的高度希望的附加成分。该载体材料可以是疏水的或亲水的、固体的或液体的。较好的载体材料是液体。适合于与本发明的螯合剂盐一起使用的疏水液体包括液体硅酮,就是说,液体聚有机硅氧烷。这样的材料可以是环状的或线型的,实例包括DowCorning硅酮液344、345、244、245、246、556、和200系列;联合碳化物公司硅酮7207和7158;以及通用电气硅酮SF1202。替而代之、或加之,非硅酮疏水液体也可以使用。这样的材料包括矿物油,加氢聚异丁烯、聚癸烯,至少10个碳原子的烷属烃、异烷属烃,脂肪族或芳香族酯油(例如肉豆蔻酸异丙酯、肉豆蔻酸月桂酯、棕榈酸异丙酯、壬二酸二异丙酯、己二酸二异丙酯、或苯甲酸C8~C18烷酯),和聚二醇醚,例如聚二醇丁醇醚。
亲水液体载体材料例如水也可以采用。
特别好的液体载体材料包含有机溶剂。为有助于该螯合剂与该有机溶剂之间的兼容性,特别好的有机溶剂是相对亲水的,其c.logP为2以下、特别是-2~1、尤其-0.5~0.5。c.logP是辛醇:水分配系数的以10为底的对数计算值;这样的值的一种计算方法可参阅“Computer-assisted computation of partition coefficients from molecularstructures using fragment constants”,J.Chou和P.Jurs,J.Chem.Inf.Comput.Sci.,19,172(1979)。此外,较好的有机溶剂有低于10℃、较好低于5℃的熔点;这对于低温贮存稳定性和制造容易性都会有好处。一类较好的有机溶剂是脂肪族醇类(较好有2~8个碳原子的一元醇或多元醇)和聚二醇醚,较好只有2~5个重复单元的低聚二醇醚。实例包括二聚丙二醇、甘油(c.logP-1.538)、丙二醇(c.logP-1.06)、丁二醇(c.logP-0.728)、乙醇(c.logP 0.235)、丙醇(c.logP 0.294)、异丙醇(c.logP-0.074),和工业甲基化酒精。最好的有机溶剂是脂肪族醇类,尤其那些有2~3个碳原子者,特别是乙醇和异丙醇。
载体材料的混合物也可以使用。所采用载体材料的数量较好是不包括存在的任何挥发性推进剂的组合物的至少5wt%、更好30wt%~99wt%、最好60wt%~98wt%。
当有机溶剂存在于该组合物中时,它较好以该组合物的液体成分总重量的30wt%~98wt%存在;更好,该有机溶剂占所存在总液体的60wt%~97wt%。
对于一些组合物、尤其裹身褡和挤压喷雾组合物来说,理想的是使用含水乙醇载体材料。为了易于实现产品均匀性,较好的是,在这样的组合物中乙醇与水的重量比是1∶1~2.5∶1、尤其1.5∶1~2∶1。
对于一些应用来说,希望的是,不足50wt%水是作为该组合物的液体成分的一部分存在的,更好的是不足10wt%。对于一些较好的组合物来说,其它液体成分与水的重量比是95∶5~99∶1之间。在这样的组合物中,有有机阳离子的螯合剂盐具有特别的溶解度和兼容性优势。
有有机溶剂的较好组合物包含该螯合剂在所述有机溶剂中的溶液。这样的溶液较好是均匀的,使用一台Pharmacia Biotech Ultrospec200分光光度计或类似仅器测定(在600nm,1cm光程长)时,相对于溶剂而言的吸收较好为0.2以下、尤其在0.1以下。
在本发明的方法和产品中,也可以有利地采用惯常的有机抗微生物剂。掺入水平较好是它们存在于其中的、不包括也存在的任何挥发性推进剂的组合物的0.01wt%~3wt%、更好0.03wt%~0.5wt%。业内常用的药剂类别大多数都可以利用。较好的额外有机抗微生物剂是杀菌剂,例如季铵化合物,如鲸蜡基三甲铵盐;双氯苯双胍己烷(洗必太)及其盐;和二聚甘油一癸酸酯、二聚甘油一月桂酸酯、甘油一月桂酸酯、和类似材料,如“Deodorant Ingredients”,S.A.Makin和M.R.Lowry,in“Antiperspirants和Deodorants”Ed.K.Laden(1999,Marcel Dekker,New York)中所述。供本发明组合物中使用的更好的额外抗微生物剂是聚六亚甲基双缩胍盐;2′,4,4′-三氯-2-羟基二苯醚(三氯生);和3,7,11-三甲基十二碳-2,6,10-三烯醇(金合欢醇)。
本发明的组合物中也可以使用无机抗微生物剂。这样的材料当以适用浓度存在时往往能作为止汗剂有效成分起作用。实例通常选自收敛药活性盐,具体地包括铝盐、锆盐、和铝/锆混合盐,包括无机盐、与有机阴离子的盐、和络合物。较好的止汗剂盐包括铝、锆和铝/锆卤化物和卤水合物(halohydrate)盐,例如氯水合物(chlorohydrate)。当存在时,较好的掺入水平是该组合物的0.5~60wt%、尤其5wt%~30wt%或40wt%、特别是5wt%或10wt%~30wt%或35wt%。特别好的铝卤水合物盐,已知为活性铝氯水合物,详见EP 6,739(Unilever PLC和NV)。锆·铝氯水合物活性物也是较好的材料,所谓的ZAG(锆-铝-甘氨酸)络合物也是如此,例如US 3,792,068(宝洁公司)中公开的那些。苯酚磺酸锌也可以使用,且较好占该组合物的可多达3wt%。
结构剂和乳化剂是在某些产品形式上高度理想的本发明组合物的进一步附加成分。当采用时,结构剂较好以该组合物的1wt%~30wt%存在,而乳化剂较好以该组合物的0.1wt%~10wt%存在。适用的结构剂包括纤维素增稠剂例如羟丙基纤维素和羟乙基纤维素,和二偏亚苄基山梨糖醇。其它适用的结构剂包括硬脂酸钠、硬脂醇、鲸蜡醇、加氢蓖麻油、合成蜡、石蜡、羟基硬脂酸、二丁基月桂酰戊二酰胺、烷基硅酮蜡、quaternium-18膨润土、qaternium-18锂蒙脱石、硅石、和碳酸丙二醇酯。适用的乳化剂包括steareth-2、steareth-20、steareth-21、ceteareth-20、硬脂酸甘油酯、鲸蜡醇、鲸蜡-硬脂醇、PEG-20硬脂酸酯、共聚二甲基硅酮多醇、和poloxamines。
本发明某些组合物中理想的进一步乳化剂/结构剂是香料增溶剂和洗去剂。前者的实例包括PEG-加氢蓖麻油,可在Cremaphor RH和CO范围内购自BASF公司,较好以可多达1.5wt%、更好0.3~0.7wt%存在。后者的实例包括聚氧乙烯醚。某些感官改性剂是本发明组合物中进一步理想的成分。这样的材料较好以该组合物的可多达20wt%的水平使用。润肤剂、湿润剂、挥发油、非挥发油、和能赋予润滑性的颗粒状固体,全都是适用的感官改性剂类别。这样的材料的实例包括环状甲基硅酮、二甲基硅酮、二甲基硅酮醇、肉豆蔻酸异丙酯、棕榈酸异丙酯、滑石、微细硅石(例如Aerosil 200)、聚乙烯(例如AcumistB18)、多糖、玉米淀粉、苯甲酸C12~C15醇酯、PPG-3肉豆蔻基醚、辛基十二烷醇、C7~C14异链烷烃、己二酸二异丙酯、月桂酸异山梨醇酯、PPG-14丁基醚、甘油、加氢聚异丁烯、聚癸烯、二氧化钛、苯基三甲基硅酮、己二酸二辛酯、和六甲基二硅氧烷。
香料也是本发明组合物中理想的附加成分。适用的材料包括惯常的芳香剂例如芳香油,而且也包括所谓除臭芳香剂,如EP 545,556及其它出版物中所述。这后一类材料也可以有资格作为附加的有机抗微生物剂。掺入水平较好可多达4wt%、尤其0.1~2wt%、特别是0.7~1.7wt%。在本发明的基本成分与某些香料成分之间会存在着协同作用-其结果是长时间持续的臭味控制。
应当说明的是,组合物的某些成分执行不止一种功能。这样的成分是特别好的附加成分,它们的使用往往既省钱又省配方空间。这样的成分的实例包括乙醇、肉豆蔻酸异丙酯、和很多既能充当结构剂又能充当感光改良剂的成分,例如硅石。也可以包括的进一步附加成分是着色剂和防腐剂例如对羟基苯甲酸C1~C3烷酯。
当本发明涉及气雾剂组合物的使用时,挥发性推进剂是这样的组合物的一种基本成分。该挥发性推进剂的掺入水平典型地是30~99重量份、尤其50~95重量份。较好的是非氯代挥发性推进剂,具体地说,是沸点在10℃以下的液化烃类或卤代烃气体(尤其氟代烃类,例如1,1-二氟乙烷和/或1,1,1-三氟-2-氟乙烷),特别是那些沸点在0℃以下者。特别好的是采用液化烃气体,尤其C3~C6烃类,包括丙烷、异丙烷、丁烷、异丁烷、戊烷和异戊烷、及其两种或多种的混合物。较好的推进剂是异丁烷,异丁烷/异丙烷、异丁烷/丙烷,以及异丙烷、异丁烷和丁烷的混合物。
可以期待的其它推进剂包括烷基醚例如二甲醚,或者压缩的非反应性气体例如空气、氮气或二氧化碳。
制造方法
相关制造方法的细节取决于所涉及的产品形式。然而,对于按照本发明的某些较好的组合物来说,该制造方法涉及形成下列的一种混合物:至少0.35wt%一种过渡金属螯合剂和至少0.05wt%一种苯酚类或烯醇类化合物,即(a)借助于与转铁蛋白结合的铁(III)还原成铁(II)起作用的转铁蛋白离解促进剂,和/或(b)一种包含叔丁基苯酚基团的抗氧剂,其中,重量百分率不包括所存在的任何挥发性推进剂。
实施例

气雾剂除臭剂组合物的制备
按以下方式制备表1中详细列出的组合物。
向96%(w/w)乙醇约64g(确切数量见表1)中添加粉末状DTPA0.50g。向此混合物中添加(边搅拌边滴加)AMP 0.38g。所得到的混合物在温和加热(50℃)下搅拌30分钟。向所得到的溶液中添加肉豆蔻酸异丙酯0.33g,并在实施例1的情况下与BHT 0.10g、在实施例2的情况下与BHT 0.05g一起混合。所得到的混合物密封到一个有阀门接口的惯常铝质除臭剂听中,用一个聚乙烯转移器件经由阀门从一个推进剂“转移听“向该听中导入液化推进剂(CAP 40,Calor公司)35g(±0.2g)。最后,给该听装配一个适用调节器,以便能有效地喷雾施用该产品。
除臭试验1
按照所述方法,制备了按照本发明的抗微生物组合物(实施例1和2-见表1)和一种对照组合物(比较例A-见表1)。按照以下试验方案测试这些组合物的除臭表现。表1中所列的结果说明从使用按照本发明的组合物得到的除臭效益。
除臭试验方案
采用的试用组包含在该试验之前那一周期间接受过使用对照乙醇类除臭剂产品的指导的50位个人。该试验开始时,试用组成员用无香料的肥皂洗涤,并将不同产品(气雾剂剂量为1.20g,裹身褡剂量为0.30g)施用到每个腋窝。(考虑到任何左/右偏性,产品施用是随机进行的)。指导试用组成员在该试验持续期间不要消费香辣食品或酒类,而且不要洗涤他们自己的腋窝。在施用后设定的时间(选自5小时、10小时和24小时),至少三位专家评价员测定腋窝臭味的强度,按1~5的尺度给该强度评分。每24小时评价之后,该试用组成员像以上那样重新洗涤、重新施用产品。这一程序重复4次。试验结束时,用标准统计技术分析这些数据。

要说明的是,在施用了实施例1的整个腋窝试验期内未观察到臭味增加。也值得注意的是,实施例C(包含BHT)并未表现得显著优于对照(实施例D)。
裹身褡除臭剂组合物的制备
实施例E(对照)
向水约25g中添加DTPA(以游离酸计)1.0g。滴加AMP 0.76g,使pH调整到约7.0。向乙醇70g中,独立地添加羟丙基纤维素(KlucelM,Aqualon公司制)0.65g和PEG 40加氢蓖麻油(Cremaphor RH 40,BASF公司制)0.5g,同时在一台Silverson L4RT混合机(Silverson公司制,Chesham,Bucks)上以大约8000rpm的速度剪切直至得到一种均匀溶液。让这种乙醇溶液冷却、与DTPA/AMP的水溶液混合,用水将重量调整到100g。
实施例4

抗微生物表现试验
以下试验证实DTPA和TTHA的微摩尔级活性性质。
表皮葡萄球菌(
S.epidermidis)的腋窝分离物在胰化胨大豆肉汤(TSB,Oxoid公司)100ml中生长过夜。取这种培养物10ml进行离心分离。分离的细胞重新悬浮于磷酸盐缓冲食盐水10ml中,重复该离心分离程序。洗涤的细胞重新悬浮于磷酸盐缓冲食盐水10ml中,给出接种物。向事先用高压釜在121℃灭菌20分钟的、含有(NH4)2SO4(0.066g)、MgSO4·7H2O(0.012g)、KCl(0.1g)、KH2PO4(0.27g)、Na2HPO4(1.43g)、硫胺素(0.1mg)、生物素(0.05mg)、胨P(0.05g)、和葡萄糖(2.0mmole)的半合成培养基(SSM)100ml中添加该接种物100μl。该SSM的pH在灭菌后、添加接种物之前用HCl调整到6.7。在离体抑制研究中全都利用这种对照培养基。以类似的方式制备含有螯合剂的试验培养基,该螯合剂在用HCl调整pH之前以3×10-6mol.dm-3的浓度导入。
将表皮葡萄球菌(
S.epidermidis)接种物100μl导入该对照培养基中和导入含有表2中所指出螯合剂的试验培养基中。这些培养物在37℃(以200rpm搅拌)培养16小时,在600nm测定该培养物的光密度以确定细菌生长的程度。通过该培养物在螯合剂的存在下的光密度与该对照组的光密度的比较,确定了生长的百分率抑制。(光密度测定是该培养物用0.9%(w/v)食盐水以1比4稀释、用1cm光程长比色杯、在一台Pharmacia Biotech Ultrospec 200分光光度计上进行的。)

转铁蛋白离解促进试验
2.7g.l-1人体二高铁转铁蛋白(购自Sigma Chemicals公司)在用氢氧化钠调整到pH6.5的50mM HEPES(N-〔2-羟基乙基〕哌嗪-N′-〔2-乙磺酸〕)缓冲剂中,连同0.115g.l-1 FerroZine(购自Sigma Chemicals公司;3-(2-吡啶基)-5,6-二(4-苯基磺酸)-1,2,4-三嗪单钠盐)和10mmol.l-1试验材料一起在37℃培养。
24小时后,通过测定562nm的吸收(参见上文),以分光光度法估计转铁蛋白离解程度。该数据(见表5)说明在所指出试验材料的存在下发生的强化转铁蛋白离解。

Claims (21)
1.一种实现抗微生物和除臭效益的方法,包含向人体或向可紧贴人体穿着的物品上施用一种抗微生物产品,该产品包含有效数量的一种过渡金属螯合剂和一种苯酚类或烯醇类化合物,即(a)借助于与转铁蛋白结合的铁(III)还原成铁(II)起作用的转铁蛋白离解促进剂,和/或(b)一种包含叔丁基苯酚基团的抗氧剂。
2.按照权利要求1的方法,包含向人体上或向可紧贴人体穿着的物品上施用一种包含至少0.35wt%一种过渡金属螯合剂和至少0.05wt%一种苯酚类或烯醇类化合物的组合物,其中,该重量百分率不包括所存在的任何挥发性推进剂。
3.按照权利要求1或2的方法,其特征在于该转铁蛋白离解促进剂能通过以下试验:

6.按照以上任何一项权利要求的方法,其特征在于该过渡金属螯合剂选自二亚乙基三胺五乙酸(DTPA)、三亚乙基四胺六乙酸(TTHA)、DTPA的盐、TTHA的盐、或这些过渡金属螯合剂的任何混合物。
7.按照以上任何一项权利要求的方法,其特征在于该过渡金属螯合剂是一种含有有机阳离子的盐。
8.按照以上任何一项权利要求的方法,其特征在于利用了一种附加的惯常有机抗微生物剂。
9.一种用于人体的抗微生物除臭组合物,包含至少0.35wt%过渡金属螯合剂和至少0.05wt%苯酚类或烯醇类化合物,即(a)借助于与转铁蛋白结合的铁(III)还原成铁(II)起作用的转铁蛋白离解促进剂,和/或(b)一种包含叔丁基苯酚基团的抗氧剂,其中,这些重量百分率不包括所存在的任何挥发性推进剂。
10.按照权利要求9的除臭组合物,其特征在于该苯酚类或烯醇类化合物是一种能通过以下试验的转铁蛋白离解促进剂:

14.按照权利要求9~13中任何一项的除臭组合物,还包含一种附加的惯常有机抗微生物剂。
15.按照权利要求9~14中任何一项的除臭组合物,其特征在于该过渡金属螯合剂选自二亚乙基三胺五乙酸(DTPA)、三亚乙基四胺六乙酸(TTHA)、DTPA的盐、TTHA的盐、或这些过渡金属螯合剂的任何混合物。
16.按照权利要求9~14中任何一项的除臭组合物,其特征在于该过渡金属螯合剂对铁(III)有大于1026的结合系数。
17.按照权利要求12~16中任何一项的除臭组合物,其特征在于该苯酚类或烯醇类化合物是BHT。
18.按照权利要求9~17中任何一项的除臭组合物,它是一种包含挥发性推进剂的气雾剂组合物。
19.按照权利要求9~17中任何一项的除臭组合物,它是一种以1∶1~2.5∶1的重量比包含乙醇和水的裹身褡或挤压喷雾组合物。
20.按照权利要求9~17中任何一项的除臭组合物,它是一种包含增稠剂或结构剂的霜剂、凝胶剂、软固体剂、或固体棒状组合物。
21.一种用于人体的除臭组合物的制造方法,包括形成含有下列组分的一种混合物:至少0.35wt%过渡金属螯合剂和至少0.05wt%苯酚类或烯醇类化合物,即(a)借助于与转铁蛋白结合的铁(III)还原成铁(II)起作用的转铁蛋白离解促进剂,和/或(b)一种包含叔丁基苯酚基团的抗氧剂,其中,这些重量百分率不包括所存在的任何挥发性推进剂。
