- 本发明属于生物技术发明领域,具体涉及用于基因转移、基因治疗和真核基因表达的一系列通用型腺病毒伴随病毒(adeno-associated virus,AAV)载体质粒。1.本发明构建的通用型AAV载体质粒pWAV-1、pWAV-2,其特征是含有2型AAV(AAV-2)基因组两端的ITR,在两个ITR之间依次为巨细胞病毒立早增强子和启动子、多克隆位点和polyA信号。
- 2.本发明构建的通用型AAV载体质粒pSNAV-1,其特征是含有AAV-2基因组两端的ITR,在两个ITR之间依次为巨细胞病毒立早增强子和启动子、多克隆位点和polyA信号,在ITR外侧具有新霉素抗性基因表达盒。
- 3.本发明构建的通用型AAV载体质粒pSNAV-2,其特征是含有AAV-2基因组两端的ITR,在两个ITR之间具有巨细胞病毒立早增强子和启动子、多克隆位点、polyA信号和新霉素抗性基因表达盒。
- 4.本发明构建的通用型AAV载体质粒用于携带外源基因,其特征是将外源基因插入通用型AAV载体质粒的多克隆位点。
- 5.本发明构建的通用型AAV载体质粒用于产生携带外源基因的重组AAV。
- 6.本发明构建的通用型AAV载体质粒用于建立稳定携带重组AAV载体的细胞株,其特征在于细胞株中含有稳定整合在染色体中的AAV ITRs及外源基因表达单位。
- 7.本发明构建的通用型AAV载体质粒用于真核细胞表达外源基因。
- 8.根据权利要求1、2、3,多克隆位点前为SV40早期增强子和启动子。
- 9.根据权利要求1、2、3,多克隆位点前为Rous肉瘤病毒基因组长末端重复区。
- 10.根据权利要求1、2、3,多克隆位点前为单纯疱疹病毒胸苷激酶启动子。
- 11.根据权利要求2、3,选择标记基因为潮霉素抗性基因。
- 12.根据权利要求2、3,选择标记基因为二氢叶酸还原酶基因。
- 13.根据权利要求2、3,选择标记基因为黄嘌呤一鸟嘌呤磷酸核糖转移酶基因。
- 14.根据权利要求2、3,选择标记基因为腺苷脱氨酶基因。
