Abstract
本发明公开了一株节杆菌soilh01及其在降解黄曲霉毒素和赭曲霉毒素中的应用,属于微生物技术领域。本发明所述的节杆菌soilh01保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏编号为CGMCC No.28894。该菌对黄曲霉毒素B1和赭曲霉毒素A均具有较好的降解作用,从而在食品、饲料及农产品生物脱毒应用方面具有实质性的应用价值和重大意义。
Description
技术领域
本发明属于微生物技术领域,具体涉及一株节杆菌soilh01及其在降解黄曲霉毒素和赭曲霉毒素中的应用。
背景技术
真菌毒素是由曲霉属真菌或镰刀属真菌产生的次级代谢产物,主要包括黄曲霉毒素(AFT)、玉米赤霉烯酮(ZEN)、和赭曲霉毒素(OTA)等,其中AFT毒性最强,污染也最为严重。据联合国粮食及农业组织统计,全世界每年约有25%的农产品因真菌及真菌毒素污染,造成巨大的经济损失,而且真菌毒素有致畸、致癌等作用,严重威胁人类的健康。
黄曲霉毒素(Aflatoxin,AFT)是由黄曲霉(Aspergllus flavus)、黑曲霉(Aspergillus Niger)、寄生曲霉(Aspergllu.Parasiticus)等多种真菌在生长过程中产生并分泌的一类毒性极强的次生代谢产物。AFT的分子量范围为312至346,基本结构为双呋喃环和香豆素,其中双呋喃环是基本毒性结构,香豆素是致癌结构。目前黄曲霉毒素已经确定和表征了20余种AFT类似物,比较常见的是黄曲霉毒素B1(AFB1),黄曲霉毒素B2(AFB2),黄曲霉毒素G1(AFG1)和黄曲霉毒素G2(AFG2),还有AFB1和AFB2被动物摄入后经羟化作用产生的代谢物黄曲霉毒素M1(AFM1)和M2(AFM2)。在这几种常见的黄曲霉毒素中,AFB1毒性最强,其毒性是氰化钾的10倍,砒霜的68倍,能引起人和动物急性中毒死亡。AFB1具有极强的肝毒性,已经被国际癌症研究机构(IARC)列为一级致癌物。
赭曲霉毒素(Ochratoxin,OT)是一种主要由曲霉属(Aspergillus spp.)和青霉属(Penicillium spp.)等丝状真菌产生的次生代谢产物,被国际癌症研究机构列为“2B”类致癌物,其包含多种衍生物,根据其化学结构主要可分为:赭曲霉毒素α(OTα)、赭曲霉毒素β(OTβ)、赭曲霉毒素A(OTA)、赭曲霉毒素B(OTB)、赭曲霉毒素C(OTC)、(4-R)和(4-S)-羟基赭曲霉毒素A(4R-OH-OTA和4S-OH-OTA)、(4-R)和(4-S)-羟基赭曲霉毒素B(4R-OH-OTB和4S-OH-OTB)和10-羟基赭曲霉毒素A(10-OH-OTA),赭曲霉毒素中又以OTA毒性最大,在农业和食品工业中污染最广泛。赭曲霉毒素A化学名称为7-(L-β-苯基丙氨基-羰基)-羧基-5-氯代-8-羟基-3,4-二氢化-3R-甲基异氧杂奈邻酮,通过酰胺键将异香豆素与L-β-苯丙氨酸相连形成。
目前,真菌毒素的防控问题是一个世界性难题,开发新型绿色安全的真菌毒素消减技术对于农产品加工行业具有重要意义。
发明内容
本发明提供了一种节杆菌(Achromobacter gandavensis)soilh01,所述菌种保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏编号为CGMCC No.28894。
上述节杆菌soilh01对黄曲霉毒素B1和赭曲霉毒素A均具有较好的降解作用,基于此,本发明提供了上述节杆菌soilh01在降解黄曲霉毒素B1和/或赭曲霉毒素A中的应用。
本发明还提供了上述节杆菌soilh01在制备降解黄曲霉毒素B1和/或赭曲霉毒素A的微生物制剂中的应用。
本发明提供了一种降解黄曲霉毒素B1和/或赭曲霉毒素A的微生物制剂,该制剂含有上述节杆菌soilh01。
所述微生物制剂优选为节杆菌soilh01发酵液或节杆菌soilh01发酵液经离心后的上清液。
本发明提供了上述发酵液的制备方法,步骤如下:将节杆菌soilh01接种于发酵培养基中,37℃,160rpm震荡培养24h,得到发酵菌液。
所述发酵培养基的配方如下:蛋白胨10g/L,牛肉膏3g/L,氯化钠8.5g/L,磷酸二氢钾1g/L,葡萄糖1g/L,pH 7.0。
在本发明中,所述上清液中的蛋白含量为2~6mg/mL。
本发明提供了一种黄曲霉毒素B1和/或赭曲霉毒素A脱毒方法,步骤如下:将上述微生物制剂喷洒于物料表面,室温静置,以降解物料中的黄曲霉毒素B1和/或赭曲霉毒素A。
上述物料可优选自食品、饲料及农产品。
上述微生物制剂的施用量为:每5g物料加入1~10mL微生物制剂。由于不同的原料,毒素污染程度不一样,可以根据原料毒素含量适当调整微生物制剂的加入量。
本发明的有益效果为:
本发明提供了一种节杆菌soilh01,该菌对黄曲霉毒素B1和赭曲霉毒素A均具有较好的降解作用,从而在食品、饲料及农产品生物脱毒应用方面具有实质性的应用价值和重大意义。

具体实施方式
菌种的筛选与鉴定:
1、筛选
以自然界的土壤、枯枝败叶为细菌来源,取10g土壤样本(来源于青岛花生种植基地),将其加入至100mL无菌水中,用振荡器充分混匀得到细菌悬浊液,静置30s。取细菌悬浊液以10%接种量接种于LB肉汤培养基(胰蛋白胨10g/L,氯化钠10g/L,胰蛋白胨5g/L,pH7.4),37℃,160rpm培养12-16h,得到细菌富集液。将细菌富集液以5%的接种量接种于以香豆素为唯一碳源的初筛培养基(磷酸二氢钾0.25g/L,七水硫酸镁0.25g/L,硝酸钾0.5g/L,硫酸铵0.5g/L,氯化钙0.005g/L,六水三氯化铁0.003g/L,pH 7.0,121℃高压灭菌20min后加入除菌的香豆素溶液至1g/L),37℃,160rpm,培养10天,进行初筛。10天后,用无菌PBS对得到的细菌培养液进行梯度稀释至10-2、10-3、10-4、10-5。吸取稀释液20μL涂布于LB固体培养基中,37℃培养24h。选取生长良好的单菌株,在LB固体培养基中连续3次划线,纯化,获得一株菌,编号为菌株soilh01。用20%的甘油保存菌株,置于-20℃冰箱中保存。
菌株soilh01在LB固体培养基中的形态如图1所示。
2、鉴定
按照“伯杰细菌鉴定手册”(第八版)中描述的方法,对菌株soilh01进行形态特征和基因学特征鉴定,具体结果如下:
(1)形态学特征
如图1所示,菌株soilh01菌落直径为0.1~0.2mm,菌落表面光滑粘稠,不透明,容易挑起菌落质地均匀,菌落正反面,边缘、中央部位颜色一致,呈黄色。
(2)基因学特征
将纯化的单菌株soilh01接种于LB肉汤培养基中,37℃,160rpm培养12h,再取2%接种量的菌液接种于新鲜的LB肉汤培养基中,37℃,160rpm培养12h后测16sRNA序列。利用16S rRNA通用引物提取菌株soilh01的基因组DNA作为模板,进行PCR扩增,并对扩增产物进行测序。

在NCBI网站进行BLAST比对,结果显示,根据Gen-Bank序列同源性比较,菌株soilh01与Achromobacter gandavensis ICMP 20898(2018)(GenBank登录号MG786417.1)同源性为100%。结合其形态学特征,将该菌鉴定为节杆菌(Achromobacter gandavensis)。该菌已于2023年11月07日保藏于中国微生物菌种保藏管理委员会普通微生物中心(简称CGMCC,地址:北京市朝阳区北辰西路1号院3号,中国科学院微生物研究所,邮编100101),保藏编号为CGMCC No.28894。
本发明所采用的其它材料,如无特殊声明,均可通过市售渠道获得。本发明所使用的其它术语,除非有另外说明,一般具有本领域普通技术人员通常理解的含义。下面结合具体实施例,并参照数据进一步详细的描述本发明。以下实施例只是为了举例说明本发明,而非以任何方式限制本发明的范围。
实施例1
黄曲霉毒素B1降解试验:
1、发酵液
将纯化的单菌株接种于发酵培养基(蛋白胨10g/L,牛肉膏3g/L,氯化钠8.5g/L,磷酸二氢钾1g/L,葡萄糖1g/L,pH 7.0),37℃,160rpm震荡培养24h得到发酵菌液。取发酵菌液400μL加入到200μL浓度为500ppb的AFB1溶液中,另加400μL新鲜发酵培养基,混匀,得到终浓度为100ppb的AFB1的混合菌液,37℃,160rpm避光孵育72h。分别在24h,48h,72h时,利用EILSA试剂盒检测AFB1残留量。对照组用800μL无菌发酵培养基与200μL浓度为500ppb的AFB1溶液混合。
AFB1降解率(%)=(对照组残留AFB1量-实验组残留AFB1量)/对照组残留AFB1量×100%。

2、发酵上清液
将纯化的单菌株接种于发酵培养基(蛋白胨10g/L,牛肉膏3g/L,氯化钠8.5g/L,磷酸二氢钾1g/L,葡萄糖1g/L,pH 7.0),37℃,160rpm震荡培养24h得到发酵菌液。取发酵菌液1mL进行离心,10000rpm离心2min,得到上清液。取200μL浓度为500ppb的AFB1溶液加入到灭菌的2mL离心管中,另加800μL上清液,混匀,得到终浓度为100ppb的AFB1的混合菌液,37℃,160rpm避光孵育72h。分别在24h,48h,72h时,利用EILSA试剂盒检测AFB1残留量。对照组用800μL无菌发酵培养基与200μL浓度为500ppb的AFB1溶液混合。
AFB1降解率(%)=(对照组残留AFB1量-实验组残留AFB1量)/对照组残留AFB1量×100%。

实施例2
赭曲霉毒素A降解试验:
1、发酵液
将纯化的单菌株接种于发酵培养基(蛋白胨10g/L,牛肉膏3g/L,氯化钠8.5g/L,磷酸二氢钾1g/L,葡萄糖1g/L,pH 7.0),37℃,160rpm震荡培养24h得到发酵菌液。取发酵菌液400μL加入到200μL浓度为500ppb的OTA溶液中,另加400μL新鲜发酵培养基,混匀,得到终浓度为100ppb的OTA的混合菌液,37℃,160rpm避光孵育72h。分别在24h,48h,72h时,利用EILSA试剂盒检测OTA残留量。对照组用800μL无菌发酵培养基与200μL浓度为500ppb的OTA溶液混合。
OTA降解率(%)=(对照组残留OTA量-实验组残留OTA量)/对照组残留OTA量×100%。

2、发酵上清液
将纯化的单菌株接种于发酵培养基(蛋白胨10g/L,牛肉膏3g/L,氯化钠8.5g/L,磷酸二氢钾1g/L,葡萄糖1g/L,pH 7.0),37℃,160rpm震荡培养24h得到发酵菌液。取发酵菌液1mL进行离心,10000rpm离心2min,得到上清液。取200μL浓度为500ppb的OTA溶液加入到灭菌的2mL离心管中,另加800μL上清液,混匀,得到终浓度为100ppb的OTA的混合菌液,37℃,160rpm避光孵育72h。分别在24h,48h,72h时,利用EILSA试剂盒检测OTA残留量。对照组用800μL无菌发酵培养基与200μL的OTA溶液混合。
OTA降解率(%)=(对照组残留OTA量-实验组残留OTA量)/对照组残留OTA量×100%。

应用例1
将纯化的菌株soilh01接种于发酵培养基(蛋白胨10g/L,牛肉膏3g/L,氯化钠8.5g/L,磷酸二氢钾1g/L,葡萄糖1g/L,pH 7.0),37℃,160rpm震荡培养24h得到发酵菌液。
1、花生粕脱毒
取花生饼粕粉碎,65℃烘干过夜,充分干燥后,室温干燥保存。取5g花生粕粉末于50mL锥形瓶,121℃高压灭菌20min。灭菌后,加入1mL浓度为100ppb的AFB1溶液,混匀后65℃过夜烘干。取4mL发酵菌液在4℃下以10000rpm离心2min,将上清液用无菌PBS缓冲液稀释至蛋白含量为6mg/mL,然后经无菌的0.22μm过滤器过滤后转移至新的无菌EP管,静置待用。将上清液加入含AFB1花生粕粉末中,于37℃,160rpm避光培养5d,将花生发酵物于5000rpm离心15min,65℃烘干后称重。对照组用4mL发酵培养基与5g含AFB1花生粕粉末混合,同样进行上述处理。利用EILSA试剂盒测量发酵花生粕中AFB1降解率为82.80%。
2、饲料脱毒
取饲料65℃烘干过夜,充分干燥后,用研磨机磨碎过20目筛,室温干燥保存。取5g饲料于50mL锥形瓶,121℃高压灭菌20min。灭菌后,加入1mL浓度为100ppb的OTA溶液,混匀后65℃过夜烘干。取4mL发酵菌液在4℃下以10000rpm离心2min,将上清液用无菌PBS缓冲液稀释至蛋白含量为6mg/mL,然后经无菌的0.22μm过滤器过滤后转移至新的无菌EP管,静置待用。将上清液加入到含OTA干燥饲料中,于37℃,160rpm避光培养5d,将饲料发酵物于5000rpm离心15min,65℃烘干后称重。对照组用4mL发酵培养基与5g含OTA干燥饲料混合,同样进行上述处理。利用EILSA试剂盒测量发酵饲料中OTA降解率为76.60%。使用高效液相色谱法复测菌株soilh01的发酵上清液对OTA的降解率为77.18%。
综上所述,本发明提供的节杆菌soilh01可以同时降解AFB1和OTA两种真菌毒素,从而在食品、饲料及农产品生物脱毒应用方面具有实质性的应用价值和重大意义。

1.一种节杆菌soilh01,其特征在于,所述菌种保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏编号为CGMCC No.28894。
2.权利要求1所述节杆菌soilh01在降解黄曲霉毒素B1和/或赭曲霉毒素A中的应用。
3.权利要求1所述节杆菌soilh01在制备降解黄曲霉毒素B1和/或赭曲霉毒素A的微生物制剂中的应用。
4.一种降解黄曲霉毒素B1和/或赭曲霉毒素A的微生物制剂,其特征在于,所述制剂含有权利要求1所述的节杆菌soilh01。
5.根据权利要求4所述的微生物制剂,其特征在于,所述微生物制剂为节杆菌soilh01的发酵液。
6.根据权利要求5所述的微生物制剂,其特征在于,所述发酵液的制备步骤如下:将节杆菌soilh01接种于发酵培养基中,37℃条件下,160 rpm震荡培养24 h,得到发酵液。
7.根据权利要求6所述的微生物制剂,其特征在于,所述发酵培养基的配方如下:蛋白胨10 g/L、牛肉膏3 g/L、氯化钠8.5 g/L、磷酸二氢钾1 g/L和葡萄糖1 g/L;所述发酵培养基的pH为7.0。
8.一种对物料进行黄曲霉毒素B1和/或赭曲霉毒素A脱毒的方法,其特征在于,步骤如下:将权利要求4~7任一项所述的微生物制剂喷洒于物料表面,室温静置,以降解物料中的黄曲霉毒素B1和/或赭曲霉毒素A。
9.根据权利要求8所述的脱毒方法,其特征在于,所述物料为饲料。
