According to the application, on the basis of CN117297996A, the characteristic that xanthan gum has better solubility in hot and cold water is utilized, the xanthan gum can be dissolved in glycerin in the mask, the hirudin has the characteristic of thermal stability in a moisture-free environment, other components are creatively freeze-dried to prepare a semi-finished mask 1, the hirudin is uniformly dispersed in a moisture-free glycerin and xanthan gum system and then sprayed on the semi-finished mask 1, and the semi-finished mask is freeze-dried for the second time to prepare the skin care mask containing the hirudin, so that the problem that the hirudin is dissolved in water firstly in the patent CN117297996A, namely, the hirudin is generally deactivated in the freeze-drying sublimation stage in the analysis of the environment which is larger than room temperature is solved, and the problem that the hirudin is difficult to dissolve out in the use of the xanthan gum is avoided. Specific embodiments of the present invention are described below with reference to the accompanying drawings. The experimental methods used in the implementation examples are all conventional methods unless otherwise specified; materials, reagents and the like used, unless otherwise indicated, are all commercially available.

2) The hirudin is respectively added into the astragalus extract, the angelica extract, the ginseng extract, the white atractylodes rhizome extract, the angelica extract, the bletilla striata extract, the white stiff silkworm extract, the white poria extract and the white peony root extract, and the mass ratio of the extract to the hirudin is 2:1, so that the traditional Chinese medicine extract and the hirudin composition is obtained.
Referring to the measurement of DPPH free radical scavenging ability, hydroxyl free radical scavenging ability, tyrosinase activity in B16 cells and melanin content in B16 cells (concentration of the combination of traditional Chinese medicine extract and hirudin is 2.5 mg/ml) in patent CN116270287A, DPPH scavenging rate, hydroxyl free radical scavenging rate, tyrosinase activity inhibition rate and melanin synthesis inhibition rate are measured, and specific results are shown in figures 1-4.
As can be seen from fig. 1-4, after the extracts of astragalus root, angelica sinensis, ginseng, bighead atractylodes rhizome, angelica dahurica, bletilla striata, silkworm larva, white poria and white paeony root are respectively combined with hirudin, the DPPH free radical elimination rate and the hydroxyl free radical elimination rate of the ginseng group (i.e. the combination of the ginseng extract and the hirudin) and the bletilla striata group (i.e. the combination of the bletilla striata extract and the hirudin) are obviously higher than those of other groups, wherein the DPPH free radical elimination rate of the ginseng group is slightly higher than that of the bletilla striata group, and the hydroxyl free radical elimination rate of the bletilla striata group is slightly higher than that of the ginseng group;
The inhibition rate of tyrosinase activity and melanin synthesis of the ginseng group (namely the combination of the ginseng extract and the hirudin) and the bletilla striata group (namely the combination of the bletilla striata extract and the hirudin) are also obviously superior to those of other Chinese herbal medicines and the combination of the hirudin, and the inhibition rate of tyrosinase activity and melanin synthesis of the ginseng group are slightly higher than those of the bletilla striata group.
The DPPH free radical elimination rate, the hydroxyl free radical elimination rate, the tyrosinase activity inhibition rate and the melanin synthesis inhibition rate are all common measurement indexes for beautifying and protecting skin, so that the combination of the ginseng, the bletilla striata and the hirudin has good beautifying and skin-protecting effects, and therefore, the combination of the ginseng, the bletilla striata and the hirudin is selected to strengthen the beautifying and skin-protecting effects of the hirudin.
Example 2 preparation method of skin-caring facial mask containing hirudin

5) Pushing the semi-finished product 2 swing disc into a freeze dryer, and performing secondary freeze drying to obtain a secondary freeze-dried mask; freeze drying at-50deg.C for 4 hr, sublimation analysis at 35deg.C for 12 hr;
6) Rapidly packaging the freeze-dried semi-finished product 2 into a facial mask packaging bag, performing cobalt radiation sterilization treatment on the facial mask packaging bag, vacuumizing the facial mask packaging bag in a nitrogen environment, and sealing to obtain the cosmetic skin care facial mask containing hirudin.
When in use, the facial mask is soaked or sprayed with water for 1-8 min to dissolve xanthan gum, and then used.
Comparative example 1: the mask was prepared using the method of CN117297996a example 1.
Comparative example 2: the same as in example 2, except that gelatin was substituted for xanthan gum.
Comparative example 3: the same as in example 2, except that xanthan gum in step 4 was omitted.
The anticoagulant activity of the masks of example 2 and comparative examples 1-3 was measured with reference to the CN117297996A example 14 stability long-term test, and is shown in Table 1 below.
As can be seen from example 2 and comparative example 1 in Table 1, in example 2, hirudin is uniformly dispersed in an anhydrous glycerin and xanthan gum system by utilizing the heat stability of hirudin in an anhydrous environment, and the hirudin is subjected to secondary freeze drying to prepare a mask, so that the anticoagulation activity of hirudin can be greatly maintained, the anticoagulation activity of hirudin is smaller than that of comparative example 1 due to the encapsulation of xanthan gum and glycerin in the early stage of example 2, the protection effect is lost due to the long-term absorption of environmental moisture by xanthan gum, and the anticoagulation activity of hirudin is rapidly reduced after 18 months.
Because gelatin is not good in cold water solubility, compared with example 2, the effect of comparative example 2 is better than that of example 2, but when the mask of comparative example 2 is used, namely the mask is soaked, the problem that hirudin is not easy to dissolve exists, so that hirudin in the mask of comparative example 2 is not easy to enter the skin surface layer, and the skin care and beauty effects are affected.
Comparative example 3, which uses only glycerin and secondary freeze drying, can better retain hirudin anticoagulation activity when prepared, but the hirudin anticoagulation activity is also reduced rapidly when left in a moist environment for a long time.

6) Rapidly packaging the freeze-dried semi-finished product 2 into a facial mask packaging bag, performing cobalt radiation sterilization treatment on the facial mask packaging bag, vacuumizing the facial mask packaging bag in a nitrogen environment, and sealing to obtain the cosmetic skin care facial mask containing hirudin.

Claims (10)
1. A preparation method of a cosmetic skin care mask containing hirudin is characterized by comprising the following steps: the mask is prepared from active ingredients and auxiliary materials, wherein the active ingredients comprise 0.2-10 parts of hirudin, 0.4-25 parts of ginseng extract and 0.4-25 parts of bletilla striata extract, and the auxiliary materials comprise 1-10 parts of xanthan gum, 40-120 parts of deionized water, 1-10 parts of sodium hyaluronate, 3-20 parts of glycerol, 2-10 parts of trehalose and 0.05-0.5 part of polyglycerol-2 oleate;
the preparation method of the mask comprises the following steps: 1) Dissolving auxiliary materials such as sodium hyaluronate, half glycerol, trehalose, polyglycerol-2 oleate and active ingredient ginseng extract and bletilla striata extract in deionized water to obtain solution 1;
2) Sterilizing the mask cloth, folding, immersing in the solution 1, standing, fully absorbing the solution 1 by the mask cloth to obtain a semi-finished product 1, pushing a tray of the semi-finished product 1 into a freeze dryer, and performing first freeze drying to obtain a first freeze-dried mask; freeze drying and pre-freezing at-30- (-80) deg.C for 2-5 hours, sublimation and resolution at 30-40 deg.C for 10-15 hours;
3) Heating the rest half of glycerol to 65deg.C, adding xanthan gum, and stirring to dissolve to obtain xanthan gum glycerol composition;
4) Adding hirudin into the xanthan gum glycerol composition, stirring uniformly, and rapidly spraying onto the surface of a once freeze-dried mask to obtain a semi-finished product 2;
5) Pushing the semi-finished product 2 swing disc into a freeze dryer, and performing secondary freeze drying to obtain a secondary freeze-dried mask; freeze drying and pre-freezing at-30- (-80) deg.C for 2-5 hours, sublimation and resolution at 30-40 deg.C for 10-15 hours;
6) Rapidly packaging the freeze-dried semi-finished product 2 into a facial mask packaging bag, performing cobalt radiation sterilization treatment on the facial mask packaging bag, vacuumizing the facial mask packaging bag in a nitrogen environment, and sealing to obtain the cosmetic skin care facial mask containing hirudin.

The method for preparing the skin care facial mask containing the water-hirudin according to claim 1, wherein the effective components comprise 0.2 part of hirudin, 0.4 part of ginseng extract and 0.4 part of bletilla striata extract, and the auxiliary materials comprise 1 part of xanthan gum, 40 parts of water, 1 part of sodium hyaluronate, 3 parts of glycerol, 2 parts of trehalose and 0.05 part of polyglycerol-2 oleate.
3. The method for preparing the skin care facial mask containing the hirudin according to claim 1, wherein the effective components comprise 10 parts of hirudin, 25 parts of ginseng extract and 25 parts of bletilla striata extract, and the auxiliary materials comprise 10 parts of xanthan gum, 120 parts of water, 10 parts of sodium hyaluronate, 20 parts of glycerol, 10 parts of trehalose and 0.5 part of polyglycerol-2 oleate.

The method for preparing the skin care facial mask containing the hirudin according to claim 1, wherein the effective components comprise 5 parts of hirudin, 12 parts of ginseng extract and 12 parts of bletilla striata extract, and the auxiliary materials comprise 5 parts of xanthan gum, 70 parts of water, 5 parts of sodium hyaluronate, 10 parts of glycerol, 5 parts of trehalose and 0.2 part of polyglycerol-2 oleate.

The method for preparing the skin care facial mask containing the hirudin according to claim 1, wherein the preparation method of the ginseng extract and the bletilla striata extract is as follows: taking ginseng and bletilla striata, respectively superfine pulverizing, sieving, respectively adding 20 times of ethanol solution with concentration of 60% of the ginseng and the bletilla striata, ultrasonically extracting for 1.5h under the power of 480W, respectively filtering the extracting solution to obtain filtrate and filter residue, respectively adding 10 times of water of the filter residue, ultrasonically extracting for 2h under the power of 480W, respectively filtering the extracting solution to obtain filtrate and filter residue, respectively combining the 2 times of filtrate, and concentrating to 1/4 of the original volume to obtain the ginseng extract and the bletilla striata extract.
6. The method for preparing the skin care facial mask containing the hirudin according to claim 1, wherein the pre-freezing temperature of the freeze drying in the step 2) and the step 5) is-30 ℃ for 5 hours, and the sublimation-analysis drying temperature is 40 ℃ for 10 hours.
7. The method for preparing the skin care facial mask containing the hirudin according to claim 1, wherein the pre-freezing temperature of the freeze drying in the step 2) and the step 5) is-80 ℃ for 2 hours, and the sublimation-analysis drying temperature is 30 ℃ for 15 hours.
8. The cosmetic skin care mask comprising hirudin prepared by the process of any one of claims 1 to 7.
9. A preparation method of a cosmetic skin care mask containing hirudin is characterized by comprising the following steps: the mask is prepared from active ingredients and auxiliary materials, wherein the active ingredients comprise 0.2 part of hirudin, 0.8 part of ginseng extract and 0.8 part of bletilla striata extract, and the auxiliary materials comprise 1 part of xanthan gum, 40 parts of deionized water, 1 part of sodium hyaluronate, 3 parts of glycerol, 2 parts of trehalose and 0.05 part of polyglycerol-2 oleate;
The preparation method of the mask comprises the following steps: 1) Dissolving adjuvants such as sodium hyaluronate, half glycerol, trehalose, polyglycerol-2 oleate, half Ginseng radix extract, and half rhizoma Bletillae extract in appropriate amount of deionized water to obtain solution 1;

6) Pushing the semi-finished product 2 swing disc into a freeze dryer, and performing secondary freeze drying to obtain a secondary freeze-dried mask; freeze drying at-50deg.C for 4 hr, sublimation analysis at 35deg.C for 12 hr;
7) Rapidly packaging the freeze-dried semi-finished product 2 into a facial mask packaging bag, adding a bag 1, performing cobalt radiation sterilization treatment on the facial mask packaging bag, vacuumizing the facial mask packaging bag in a nitrogen environment, and sealing to obtain the cosmetic skin care facial mask containing hirudin.
10. The cosmetic skin care mask comprising hirudin prepared by the method of claim 9.
