Abstract
本发明公开了一种人多巴胺能神经元样细胞分化的方法及分化的细胞和应用。向人羊膜上皮干细胞中加入含抗坏血酸,成纤维细胞生长因子‑2和noggin的诱导组合物‑1和含有成纤维细胞生长因子‑8,音猬因子,脑源性神经营养因子,胶质细胞源性神经营养因子,CHIR99021和二丁酰环磷腺苷的诱导组合物‑2,分别诱导分化后,诱导产生的细胞相较于人羊膜上皮干细胞,其神经元及多巴胺能神经元的特异性标记物的表达水平显著上升,此外,其多巴胺的分泌水平也显著提高。且诱导得到的人多巴胺能神经元样细胞免疫原性低,细胞活力好,表明其很适合作为帕金森病细胞治疗的移植物。

技术领域
本发明属于生物技术领域,具体涉及一种人羊膜上皮干细胞诱导分化出人多巴胺能神经元样细胞的方法及分化的细胞和在治疗帕金森病中的应用。
技术背景
帕金森病(Parkinson’s disease,PD)是目前最常见的神经退行性疾病之一,在世界范围内影响超过1%的60岁以上人群。帕金森病的两大主要病理特征分别是中脑多巴胺能神经元的丢失和神经元中α-突触核蛋白累积进而形成路易小体。随着帕金森病病情的发展,病人大脑中的多条多巴胺通络会受到影响,从而导致多种运动症状(如静止性震颤、行动缓慢)和非运动症状的出现。目前,临床上对于帕金森病的治疗主要分为药物治疗和手术治疗两大方面。其中药物治疗的金标准疗法是多巴胺补充疗法,以左旋多巴为代表,这些药物通过增加病人中枢神经系统中的多巴胺量,达到缓解帕金森症状从而改善生活质量的效果。然而,长期服用这些药物也会产生许多副作用,包括胃肠道功能障碍和精神障碍等。如前所述,由于中脑多巴胺能神经元的退化是帕金森病的一个重要病理特征,细胞替代疗法被认为是一种很有前景的帕金森治疗方式。根据以往研究,向帕金森模型动物移植体外诱导的多巴胺能组细胞/多巴胺能神经元,能够成功重建帕金森模型动物的脑内靶区域,并在不同程度上恢复功能。目前报道的体外诱导的多巴胺能组细胞/多巴胺能神经元主要来源于人胚胎干细胞(human embryonic stem cells,hESCs)和人诱导多能干细胞(humaninduced pluripotent stem cells,hiPSCs),尽管已经取得了一些振奋人心的结果,但作为帕金森病治疗的细胞来源,hESCs和hiPSCs存在缺陷,如伦理限制和安全性问题。
由于胎盘是一种生产后通常会被废弃的组织,所以相较于其他人体组织,来源于胎盘的围产期干细胞,具有更少的伦理问题。此外,胎盘属于新生儿附属组织,具有较少的DNA损伤,是再生医学的理想选择。人羊膜上皮干细胞(human amniotic epithelial stemcells,hAESCs),作为人围产期干细胞之一,可以从废弃的胎盘中获得。从发育学的角度,hAESCs和hESCs都来源于第8天受精卵的囊胚内细胞团,因此hAESCs保留了多向分化潜能等干细胞的特性。另一方面,通过实验也证明了人羊膜上皮干细胞确实能表达多种多能干细胞的标记物,如阶段特异性胚胎抗原-4(stage specific embryonic antigen-4,SSEA-4),性别决定区Y框转录因子-2(sex determining region Y-box transcription factor-2,sox-2)等。此外,由于hAESCs的低免疫原性和非致瘤性,其生物安全性也已被证实。因此,hAESCs有希望成为帕金森病治疗中的多巴胺能神经元再生的理想细胞来源。
基于以上所述hAESCs的优势属性,本发明开发了一种不添加动物血清,更符合临床标准的方案,尝试将废弃胎盘来源的hAESCs诱导分化成人多巴胺能神经元样细胞,并在帕金森模型动物中验证了其对于帕金森病的治疗效果。本发明实现了生物废弃物的再利用,且hAESCs具有无伦理问题,生物安全性高等优点,本法将在帕金森病的临床应用上具有广泛的前景。
发明内容
针对当前帕金森病的细胞治疗中尚且存在的问题,本发明提供了一种人羊膜上皮干细胞诱导分化出人多巴胺能神经元样细胞的方法及分化的细胞和在治疗帕金森病中的应用,并且在帕金森动物模型中验证了其应用潜力,为临床上对于帕金森病的治疗提供了一种新的思路。
为了达到上述目的,本发明首先提供了一种诱导人羊膜上皮干细胞向人多巴胺能神经元样细胞分化的方法,所述方法包括如下的步骤:
(1)从废弃的胎盘分离人羊膜上皮干细胞,在合适条件下贴壁培养24-48小时;
(2)将细胞培养液更换为诱导组合物-1,培养6~8(优选为7)天,诱导人羊膜上皮干细胞分化为人神经元样细胞;
(3)7天后,将诱导组合物-1更换为诱导组合物-2,继续培养12~16(优选为14)天,进一步诱导细胞分化为人多巴胺能神经元样细胞。
所述方法步骤(1)中的人羊膜上皮干细胞为小于P2代次的人羊膜上皮干细胞。
所述方法步骤(2)中的诱导组合物-1为含有0.2-5mM抗坏血酸,1-20ng/ml成纤维细胞生长因子-2和10-500ng/ml noggin的细胞培养基。
所述方法步骤(3)中的诱导组合物-2为含有5-100ng/ml成纤维细胞生长因子-8,10-500ng/ml音猬因子,2-50ng/ml脑源性神经营养因子,5-50ng/ml胶质细胞源性神经营养因子,0.3-10uM CHIR99021和10-500uM二丁酰环磷腺苷的细胞培养基。
所述方法中的人羊膜上皮干细胞通过包括以下步骤的方法制备:
(1)从废弃的胎盘组织剥离获取新鲜羊膜;
(2)仔细清洗羊膜后,依次经过酶消化、离心等步骤,收获人羊膜上皮干细胞。
所述方法中,羊膜上皮干细胞来源于人类,指人羊膜上皮干细胞。
所述方法中,新鲜羊膜采集自健康、足月的剖腹产产妇,且已获得所有产妇的书面知情同意。所有捐献者都经过梅毒螺旋体,艾滋病毒,甲型、乙型、丙型和丁型肝炎的检测,且结果均为阴性。
基于以上结果,本发明进一步公开了人羊膜上皮干细胞起源的人多巴胺能神经元样细胞在治疗/改善帕金森病中的作用,所述方法包括如下的步骤:
(1)给大鼠右侧大脑半球注射6-羟基多巴胺(6-Hydroxydopamine,6-OHDA),构建单侧损毁的大鼠帕金森模型;
(2)挑选模型构建成功的大鼠,向其右脑纹状体移植人羊膜上皮干细胞起源的人多巴胺能神经元样细胞;
(3)从动物行为学,组织学染色等方面评价治疗效果。
本发明采取的技术方案之一为:一种从废弃胎盘的羊膜组织中分离得到人羊膜上皮干细胞的方法,所述方法包括以下步骤:
步骤1:从人废弃胎盘内表面机械剥离,获取羊膜。以上过程获得所有产妇的书面知情同意,且通过伦理审查。羊膜须经冷链运输快速转移至生物安全柜进行下一步操作;
步骤2:用添加抗生素的磷酸盐平衡生理盐水(Phosphate buffered saline,PBS)多次清洗羊膜,以去除羊膜表面附着的血块和杂质,后将羊膜剪成小块组织,加入0.25%胰蛋白酶,于37℃水浴消化,终止消化后,经离心获得人羊膜上皮干细胞沉淀,使用培养液重悬;
步骤3:将适量人羊膜上皮干细胞接种于培养皿内,置于37℃,5%CO2培养箱中培养。待细胞贴壁后,每两天更换一次培养液;
步骤4:待细胞长满培养皿后,使用0.25%胰蛋白酶消化,收集消化得到的细胞,离心,使用冻存液重悬细胞于冻存管,经-80℃冰箱缓慢冷冻后,转移至液氮中保存。
所述方法步骤1中通常将羊膜至于装有100-200ml预冷培养液的样品收集瓶中,全程冷链快速运输至生物安全柜。
所述方法步骤2中的清洗液指含1%(v/v)双抗(青霉素和链霉素)的磷酸盐平衡生理盐水,一般视具体情况清洗羊膜3次及以上。
所述方法步骤2中的详细消化过程如下:向含有羊膜组织块的离心管中加入没过组织量的0.25%胰蛋白酶,于37℃水浴锅消化30min,其间每10min摇晃几下离心管以促进消化。使用10%胎牛血清(Fetal bovine serum,FBS)中止消化。
所述方法步骤2中经离心收集人羊膜上皮干细胞的具体过程如下:室温条件下,于500xg,离心6min,弃上清,收集细胞沉淀。
本发明采取的技术方案之二为:将人羊膜上皮干细胞诱导分化为人多巴胺能神经元样细胞,所述方法包括以下步骤:
步骤1:冻存的人羊膜上皮干细胞的复苏。从液氮罐中取出冻存的人羊膜上皮干细胞,置于37℃水浴锅中化冻,离心后使用人羊膜上皮干细胞培养液铺板,培养24-48小时使其贴壁;
步骤2:配制诱导组合物-1和诱导组合物-2;
步骤3:将人羊膜上皮干细胞培养液更换为诱导组合物-1,于细胞培养箱中培养7天,期间每2天换液一次;
步骤4:将诱导组合物-1更换为诱导组合物-2,于培养箱中继续培养14天,期间每2天换液一次。
所述方法步骤1中,从液氮中取出细胞后,须快速将细胞转移至37℃水浴锅进行化冻。
本发明采取的技术方案之三为:检测诱导所得人羊膜上皮干细胞起源的人多巴胺能神经元样细胞的细胞活力和免疫原性,所述方法包括以下步骤:
步骤1:使用细胞活/死染色试剂盒对分化得到的人多巴胺能神经元样细胞进行细胞活力检测,实验步骤遵循试剂盒说明书;
步骤2:使用免疫荧光法和流式细胞分析法对分化得到的的人多巴胺能神经元样细胞进行免疫原性检测。
所述方法步骤2中免疫荧光法检测的指标为人类白细胞抗原-A(Human leukocyteantigen-A,HLA-A)和人类白细胞抗原-B(Human leukocyte antigen-B,HLA-B)。
所述方法步骤2中流式细胞分析法检测的指标为人类白细胞抗原-DQ(Humanleukocyte antigen-DQ,HLA-DQ)和人类白细胞抗原-DR(Human leukocyte antigen-DR,HLA-DR)。
本发明采取的技术方案之四为:构建单侧损毁的大鼠帕金森模型,使用该模型验证分化得到的人多巴胺能神经元样细胞对于帕金森病的治疗/改善效果,所述方法包括以下步骤:
步骤1:从上海斯莱克公司购买雌性Sprague Dawley品系成年大鼠,雌性,体重为220-240g;
步骤2:将实验动物饲养在通风环境中,给予每天12小时光照/12小时黑暗,并提供充足的食物和饮用水;
步骤3:所有动物被随机分为2组:对照组和细胞治疗组。
步骤4:使用脑立体定位注射仪搭配微量注射器,向大鼠右脑的内侧前脑束(medial forebrain bundle,MFB)注射6-羟基多巴胺,以造成其右脑黑质-纹状体通路的多巴胺能神经元的损伤。给6-羟基多巴胺4周后,通过行为学评估,挑选造模成功的大鼠,用于后续实验;
步骤5:细胞治疗组:使用脑立体定位注射仪搭配微量注射器,向大鼠右脑纹状体注射人多巴胺能神经元样细胞(重悬于重悬培养基);对照组:使用脑立体定位注射仪搭配微量注射器,向大鼠右脑纹状体注射相同体积的重悬培养液;
步骤6:术后给每只大鼠肌肉注射兽用青霉素,预防术后感染,同时使用加热垫子为动物进行保温。
步骤7:于细胞移植后第4,8,12,16周进行阿朴吗啡诱导的旋转试验和步态试验两项行为学实验,并进行统计。
步骤8:所有行为学实验结束后,处死动物,取材,进行后续组织学染色分析。
所述方法步骤4和5中,使用丙泊酚对大鼠进行术前麻醉。
所述方法步骤4和5中,6-羟基多巴胺/细胞/培养液注射的速度均为0.4ul/min,且注射完成后均停针10min,后缓慢退针。
所述方法步骤5中,细胞分别移植至大鼠纹状体的两处,两处坐标分别为前囟前,1.2mm;中线旁开,-2.6mm;硬膜下,-4.4mm;前囟前,0.5mm;中线旁开,-3.0mm;硬膜下,-4.0mm,两处分别注射3ul和2ul细胞悬液,细胞悬液的浓度为150000个细胞/ul。
所述方法步骤5中的对照组,给予重悬培养液的坐标和体积与细胞治疗组相同。
所述方法步骤8中的组织学染色包括脑片的免疫组织化学染色和免疫荧光染色。
与现有技术相比,本发明具有如下优点:
(1)本发明开发了一种不添加动物血清,更符合临床标准的能够稳定将人羊膜上皮干细胞诱导分化为人多巴胺能神经元样细胞的方法,基于人羊膜上皮干细胞的易获取、生物安全性、低免疫原性等优势属性,相较于现有技术,本发明更具有临床上应用于治疗帕金森病的广泛前景。
(2)本发明中分化得到的人多巴胺能神经元样细胞,移植后能够改善帕金森大鼠的行为学,增加其黑质-纹状体通路的多巴胺能神经元数量。且组织学染色分析表明,移植的人多巴胺能神经元样细胞能在移植区域存活,并整合进大鼠的纹状体中。以上结果为本发明的临床转化进一步奠定了基础。

具体实施方式
下面将结合附图,对本发明的优选实施例进行详细的描述。实施例中未注明具体条件的实验条件及方法,通常按照常规条件或按照制造厂商所建议的条件。
本发明的目的是开发一套无动物血清,且化学成分明确的分化体系,将胎盘来源的人羊膜上皮干细胞诱导分化成多巴胺能神经元样细胞,并且在帕金森动物模型中进一步验证其对于帕金森病的治疗作用。
为达到上述目标,本发明采取的技术方案如下:
1.原代人羊膜上皮干细胞的分离培养;
2.无血清、化学成分明确的分化体系的构建;
3.在帕金森大鼠动物模型中进一步验证其治疗作用。
实施例1人羊膜上皮干细胞培养液、冻存液的配制
1、人羊膜上皮干细胞培养液的配制:向DMEM/F12培养基中添加15%KSR,1%非必需氨基酸(non-essential amino acid,NEAA),1%L-谷氨酰胺(L-glutamine),1%丙酮酸钠(sodium pyruvate),1%双抗(青霉素和链霉素,penicillin and streptomycin),使用前加入终浓度为10ng/ml的人表皮生长因子(human epithelial growth factor,hEGF);
2、人羊膜上皮干细胞冻存液的配制:90%胎牛血清(FBS)+10%二甲基亚砜(Dimethyl sulfoxide,DMSO)。
实施例2人羊膜上皮干细胞的分离和冻存
1、人羊膜组织的获取:在获得产妇的书面知情同意后,取剖腹产后的胎盘组织,从胎盘内表面进行机械剥离,得到整张羊膜。提前对所有捐献者进行梅毒螺旋体,艾滋病毒,甲型、乙型、丙型和丁型肝炎病毒检测,且结果均为阴性。将新鲜的人羊膜组织至于装有100-200ml预冷培养液的样品收集瓶中,全程冷链快速运输至生物安全柜;
2、人羊膜上皮干细胞的分离:使用添加1%双抗的磷酸盐平衡生理盐水漂洗羊膜数次,以去除羊膜表面附着的血块和杂质。后将羊膜剪碎成小块组织,转移至50ml离心管中。向含有羊膜组织块的离心管中加入约20ml 0.25%胰蛋白酶,于37℃水浴锅消化30min,期间每10min摇晃几下离心管以促进消化,使用10%胎牛血清中止消化。后于500xg,离心6min,去上清,使用20ml培养基重悬细胞,依次过200目和400目细胞筛,得到人羊膜上皮干细胞的细胞悬液;
3、人羊膜上皮干细胞的培养:取少量细胞悬液进行计数,后以1×107个细胞/15cm培养皿的密度进行接种,置于37℃,5%CO2培养箱中培养。待细胞贴壁后,每两天更换一次培养液;
4、人羊膜上皮干细胞的冻存:待细胞长满培养皿后,向每个15cm培养皿加入5-7ml0.25%胰蛋白酶,后于37℃消化约7min,当在显微镜下观察到细胞形态变圆,轻轻敲击培养皿时细胞呈流沙样脱落,即可加入10%胎牛血清中止消化。收集消化得到的人羊膜上皮干细胞,离心,弃上清后,使用冻存液重悬细胞于冻存管,经-80℃冰箱缓慢冷冻后,转移至液氮中保存。
实施例3人多巴胺能神经元样细胞的诱导分化方案
1、诱导组合物-1的配制:向含有15%KSR,1%非必需氨基酸,1%L-谷氨酰胺,1%丙酮酸钠,1%双抗和10ng/ml人表皮生长因子的DMEM/F12培养基中加入终浓度为1%N-2补充剂,2%B-27补充剂,0.2-5mM抗坏血酸(ascorbic acid),1-20ng/ml成纤维细胞生长因子-2(fibroblast growth factor-2,FGF-2)和10-500ng/ml noggin即得到组合物-1。
2、诱导组合物-2的配制:向含有15%KSR,1%非必需氨基酸,1%L-谷氨酰胺,1%丙酮酸钠,1%双抗,10ng/ml人表皮生长因子,1%N-2补充剂,2%B-27补充剂,2mM抗坏血酸(ascorbic acid)和10ng/ml成纤维细胞生长因子-2(fibroblast growth factor-2,FGF-2)的DMEM/F12培养基中加入终浓度为5-100ng/ml成纤维细胞生长因子-8(fibroblastgrowth factor-8,FGF-8),10-500ng/ml音猬因子(sonic hedgehog,SHH),2-50ng/ml脑源性神经营养因子(Brain-derived neurotrophic factor,BDNF),5-50ng/ml胶质细胞源性神经营养因子(Glia-derived neurotrophic factor,GDNF),0.3-10uM CHIR99021和10-500uM二丁酰环磷腺苷(dibutyryl cAMP)即得到组合物-2。
3、诱导人羊膜上皮干细胞向人多巴胺能神经元样细胞分化的方法:取小于P2的人羊膜上皮干细胞,以合适密度接种于6孔板/12孔板内。24-48小时后弃去培养液,每孔添加2ml诱导组合物-1,置于细胞培养箱中,隔天换液,培养7天。7天后,弃去诱导组合物-1,每孔添加2ml诱导组合物-2,继续培养14天,隔天换液。于第21天收获人多巴胺能神经元样细胞。
实施例4免疫细胞化学(Immunocytochemistry,ICC)
1、从培养箱中取出12孔板,弃培养液,每孔使用磷酸盐平衡生理盐水洗涤3次,后每孔加入1ml 4%多聚甲醛(paraformaldehyde,PFA),固定细胞15min;
2、弃固定液,每孔使用磷酸盐平衡生理盐水洗涤3次,每次5min;
3、每孔加入1ml含0.3%Triton-X-100的磷酸盐平衡生理盐水溶液,透化5-10min(视具体染色指标确定);
4、弃透化液,每孔使用磷酸盐平衡生理盐水洗涤3次,每次5min;
5、每孔加入1ml封闭液,于室温封闭2小时。封闭液的配方如下:5%山羊血清和1%牛血清蛋白(bovine serum albumin,BSA)溶于磷酸盐平衡生理盐水;
6、将一抗以说明书标注的稀释比例稀释于封闭液,将铺有细胞的爬片倒扣于一抗稀释液上,于4℃敷育过夜;
7、弃一抗稀释液,将爬片放回12孔板对应孔中,每孔使用磷酸盐平衡生理盐水洗涤3次,每次5min;
8、将带荧光标签的二抗以说明书标注的稀释比稀释于磷酸盐平衡生理盐水,12孔板每孔加入500ul二抗稀释液,室温避光敷育1小时;
9、弃二抗稀释液,每孔使用磷酸盐平衡生理盐水洗涤3次,每次10min;
10、从孔板中取出爬片,使用含DAPI的封片剂封片;
11、使用共聚焦显微镜FV3000采集图片,使用ImageJ软件对图片进行处理。
图1结果显示相较于人羊膜上皮干细胞,分化得到的人多巴胺能神经元样细胞呈现出明显的神经细胞形态,且神经元特异性标记物MAP-2和多巴胺能神经元特异性标记物TH,DAT的表达显著上升,说明分化得到的细胞从形态到蛋白表达均符合人多巴胺能神经元细胞的基本特征。
图2结果显示,从人羊膜上皮干细胞分化得到的人多巴胺能神经元样细胞,在有/无IFN-γ的刺激下,均基本不表达HLA-A和HLA-B。
实施例5流式细胞分析(Flow cytometry analysis,FCM)
1、配制清洗缓冲液:含2%牛血清蛋白的磷酸盐平衡生理盐水溶液;
2、向贴壁生长的细胞培养皿中加入0.25%胰蛋白酶,于37℃培养箱消化约5min,中止消化后,收集细胞于离心管中,于4℃,1000rpm离心5min;
3、弃上清,每管加入1ml清洗缓冲液重悬细胞,于4℃,1000rpm离心3分钟;
4、重复步骤3一次;
5、弃上清,每管加入50ul清洗缓冲液,重悬细胞后,每管加入5ul带荧光标签的抗体,再次重悬后,于4℃,避光敷育30min;
6、于4℃,1000rpm离心3min,弃上清,每管加入1ml清洗缓冲液重悬细胞,再次离心弃上清。重复以上步骤2次;
7、每管加入500ul清洗缓冲液,重悬细胞后,将细胞悬液转移至流式管中上机检测。使用FlowJo分析软件对结果进行分析。
图2显示,从人羊膜上皮干细胞分化得到的人多巴胺能神经元样细胞,在有/无IFN-γ的刺激下,HLA-DR和HLA-DQ均基本为阴性,而HLA-G为阳性,表明分化后的细胞依然保持低免疫原性的优势,适合用于体内移植。
实施例6定量实时聚合酶链式反应(Quantitative RT-PCR,qRT-PCR)
1、从培养箱中取出6孔板,弃培养液,每孔使用磷酸盐平衡生理盐水洗涤2次,后每孔加入1ml Trizol,平放1min,后使用移液枪吹打使细胞裂解完全,后收集细胞裂解液;
2、使用RNA提取试剂盒提取细胞总RNA,步骤遵循试剂盒说明书;
3、使用反转录试剂盒将RNA反转录成cDNA,步骤遵循试剂盒说明书;
4、将反转录后得到的cDNA稀释至合适浓度,按下列体系进行实时定量PCR反应:
2x All-in-one qPCR Mix 10ul
PCR前引物(10uM)0.5ul
PCR后引物(10uM)0.5ul
cDNA 5ul
ddH2O.4ul
反应程序如下:95℃,10min;(95℃,10s;60℃,20s;72℃,15s)x40次;
5、反应结束后确认扩增曲线和溶解曲线,使用excel软件处理数据,使用Graphpad软件作图。
图1中的qRT-PCR结果显示,相较于人羊膜上皮干细胞,分化得到的人多巴胺能神经元样细胞的神经元特异性标记物MAP-2,OTX-2和TUJ1,多巴胺能神经元特异性标记物TH,DAT,NURR1,Pitx3和FOXA2的表达量均明显上升。
实施例7酶联免疫吸附试验(Enzyme-linked immunosorbent assay,ELISA)
1、收集细胞培养48小时上清,4℃,300xg,离心5min,去除细胞碎片,吸取上清备用;
2、在提取板每孔加入500ul标准品溶液或检测样品,然后每孔加入25ul TE缓冲液,盖上薄膜后,室温,100rpm,振荡孵育60min;
3、拿掉薄膜,弃去孔板中的液体。用移液枪向每孔加入1ml洗涤缓冲液,室温,100rpm,振荡5min,然后弃去洗涤液。重复洗涤2次;
4、向所有孔中依次加入150ul的酰化缓冲液和25ul的酰化剂,室温,100rpm,振荡孵育20min;
5、弃去孔板中的液体,用移液枪向每孔加入1ml洗涤缓冲液,室温,100rpm,振荡5min,然后弃去洗涤液。重复洗涤2次;
6、用移液枪向每个孔中加入100ul盐酸,盖上薄膜,室温,100rpm,振荡孵育10min;
7、从提取板的每孔中移取90ul液体,转移到酶反应板的相应孔中,加入25ul酶溶液。盖上薄膜,室温,100rpm,振荡1min混匀,37℃,继续孵育2小时;
8、从酶反应板中移取100ul液体,转移到多巴胺检测板的相应孔中,并在每孔中加入50ul多巴胺抗体。盖上薄膜,室温,100rpm,振荡1min混匀,后于4℃孵育过夜;
9、拿掉薄膜,弃去孔中液体,使用300ul洗涤缓冲液洗涤4次,洗涤方法同上;
10、向每孔加入100ul酶偶联物,室温,100rpm,振荡孵育30min;
11、拿掉薄膜,弃去孔中液体,使用300ul洗涤缓冲液洗涤4次,洗涤方法同上;
12、每孔加入100ul显色底物,室温,100rpm,避光振荡孵育30min;
13、每孔加入100ul反应中止液,后在10min内读取每孔在450nm和630nm波长下的吸光度;
14、收集数据,绘制标准曲线,使用excel软件处理数据,使用Graph pad软件作图。
图1中的ELISA检测结果显示,相较于人羊膜上皮干细胞,人多巴胺能神经元样细胞的多巴胺分泌量显著增加,说明分化得到的细胞在生理学特性上符合人多巴胺能神经元细胞的特征。
实施例8细胞活/死染色(Live/dead assay)
1、使用1x检测缓冲液轻轻洗涤贴壁细胞2次;
2、每孔加入含有2ul钙黄绿素-乙酰甲氧基甲酯的1ml 1x检测缓冲液,于37℃,避光孵育20min;
3、取4ul碘化丙啶染色液加入上述溶液中,室温,避光染色5min;
4、弃染色液,使用磷酸盐平衡生理盐水轻轻洗涤2次;
5、使用共聚焦显微镜FV3000采集图片,使用ImageJ软件对图片进行处理。
图2结果显示,分化得到的人多巴胺能神经元样细胞保持很高的细胞活力(约为99.94%),适于进行细胞移植。
实施例9构建单侧损毁的大鼠帕金森模型
1、从上海斯莱克公司购买雌性Sprague Dawley品系成年大鼠,雌性,体重为220-240g。将大鼠饲养在通风环境中,给予每天12小时光照/12小时黑暗,并提供充足的食物和饮用水;
2、所有大鼠被随机分为2组:对照组和细胞治疗组;
3、腹腔注射丙泊酚,麻醉大鼠,每只鼠根据体重注射4-5ml左右;
4、使用剃毛器给大鼠的头部去毛,碘伏消毒后,固定于脑立体定位仪上。皮下注射2ml生理盐水以防止术中脱水,且手术全程使用保温垫子保暖;
5、手术刀沿大鼠头部正中划开头皮,暴露出颅骨,找到大鼠脑部前囟位置,它是脑内相对固定的位置,用记号笔在此处做一标记,以它作为原点;
6、按照Paxinos&Watson脑图谱(第6版)确定右脑内侧前脑束的位置,其坐标为:前囟后4.4mm,中线旁开1.2mm,硬膜下7.8mm,用记号笔在此处做一标记;
7、使用牙科钻小心钻开MFB定位处的颅骨,5ul微量注射器缓慢进针,到达相对位置后注射3.66ul 6-OHDA溶液(6mg/ml,溶解于含0.2%抗坏血酸的生理盐水),注射速度为0.4ul/min,注射完成后在原处停针10min,后缓慢退针;
8、缝合大鼠头部皮肤,使用碘伏在伤口及附近区域消毒。此外,给每只大鼠肌肉注射120000单位青霉素以防止术后感染。将大鼠置于保温垫子上,直至苏醒。术后每天称量大鼠体重,对体重进行检测,并密切关注其状态,如进食情况等。
实施例10移植前的细胞准备
1、从培养箱中取出6孔板,弃诱导组合物-2,每孔使用杜氏磷酸盐缓冲液(Dulbecco’s phosphate-buffered saline,DPBS)洗涤3次,弃洗涤液,每孔加入1mlAccutase酶,于37℃消化15min;
2、收集消化得到的人多巴胺能神经元样细胞于离心管中,离心弃上清后,使用不含酚红的DMEM/F12培养基重悬,细胞悬液过70um细胞筛以去除大的细胞团块;
3、使用台盼蓝对活细胞进行计数,将细胞悬液的密度调整至150000个细胞/ul,用于后续细胞移植;
4、为在大鼠体内对人多巴胺能神经元样细胞进行追踪,需在体外使用PK67标记试剂盒提前给人多巴胺能神经元样细胞带上荧光标签,操作步骤遵循试剂盒说明书。
图4结果显示,在移植后第16周,依然能够在大鼠纹状体中捕获人多巴胺能神经元样细胞预标记的PKH67绿色荧光信号。
实施例11人多巴胺能神经元样细胞的移植
1、腹腔注射丙泊酚,麻醉大鼠,每只鼠根据体重注射4-5ml左右;
2、使用剃毛器给大鼠的头部去毛,碘伏消毒后,固定于脑立体定位仪上。皮下注射2ml生理盐水以防止术中脱水,且手术全程使用保温垫子保暖;
3、手术刀沿大鼠头部正中划开头皮,暴露出颅骨,找到大鼠脑部前囟位置,它是脑内相对固定的位置,用记号笔在此处做一标记,以它作为原点;
4、按照Paxinos&Watson脑图谱(第6版)确定右脑纹状体内两注射点的位置,其坐标分别为:前囟前1.2mm,中线旁开2.6mm,硬膜下4.4mm;前囟前0.5mm,中线旁开3.0mm,硬膜下4.0mm,用记号笔在这两处分别做标记;
5、使用牙科钻小心钻开这两处的颅骨,5ul微量注射器缓慢进针,到达相对位置后分别注射3ul和2ul细胞悬液或不含酚红的DMEM/F12培养基,注射速度为0.4ul/min,注射完成后在原处停针10min,后缓慢退针;
6、缝合大鼠头部皮肤,使用碘伏在伤口及附近区域消毒。此外,给每只大鼠肌肉注射120000单位青霉素以防止术后感染。将大鼠置于保温垫子上,直至苏醒。术后每天称量大鼠体重,对体重进行检测,并密切关注其状态,如进食情况等。
实施例12行为学实验1:阿朴吗啡诱导的旋转试验
1、在细胞移植前、移植后第4,8,12,16周进行阿朴吗啡诱导的旋转试验;
2、配制阿朴吗啡溶液:在冰上配制,全程避光,使用生理盐水溶解,配制成1mg/ml终浓度即可;
3、称量大鼠体重,按1mg/kg注射量,计算每只大鼠的阿朴吗啡注射量;
4、将大鼠放在测试区域,使其适应周围环境约30min;
5、经颈背部注射阿朴吗啡溶液,诱导大鼠的旋转行为;
6、向未损毁侧(左侧)的旋转被赋值为正值,且仅记录每一次完整的全身旋转。记录每只大鼠在60min内的旋转次数,最终以每小时的净旋转次数表示。每小时旋转大于等于300圈的大鼠被认为造模成功,用于细胞移植/对照。
图3的结果显示,人多巴胺能神经元样细胞的移植能够显著降低帕金森大鼠由于阿朴吗啡诱导的旋转次数。
实施例13行为学实验2:步态试验
1、在细胞移植前、移植后第4,8,12,16周进行步态试验;
2、将大鼠放在测试区域,使其适应周围环境约30min;
3、将大鼠放在平坦的桌面上,轻轻抬起尾巴,使其后腿离开桌面,仅让其前爪接触桌面;
4、以恒定的速度将大鼠沿桌面向后拉1m,记录未损毁侧和损毁侧前爪的调整步数;
5、数据表示为未损毁/损毁侧侧调整步数的百分比。
图3的结果显示,人多巴胺能神经元样细胞的移植能够有效改善帕金森大鼠的步态试验行为学表型。
实施例14取材及大鼠脑片切片
1、腹腔注射丙泊酚,麻醉大鼠;
2、暴露心脏,经左心室行主动脉灌注固定:先使用100ml预冷磷酸盐平衡生理盐水冲洗,至右心耳流出的液体清澈,再使用200ml 4%甲醛溶液进行灌注。后取脑,置于4%甲醛溶液中,于4℃继续固定3天;
3、3天后,将脑组织转移至20%和30%蔗糖溶液中梯度沉糖;
4、使用最佳切割温度化合物(optimal cutting temperature compound,OCT)包埋脑组织,后转移至-80℃冰箱冷冻;
5、使用冰冻切片机切片,连续收集覆盖整个纹状体和黑质的冠状切片,切片厚度为30um/张。
实施例15脑片免疫组织化学(Immunohistochemistry,IHC)
1、将脑片置于12孔板,3张/孔;
2、磷酸盐平衡生理盐水清洗脑片3次,每次3min;
3、向每孔加入含3%H2O2的磷酸盐平衡生理盐水,避光敷育15min;
4、磷酸盐平衡生理盐水清洗脑片3次,每次3min;
5、向每孔加入封闭液(含3%牛血清蛋白和1%Triton-X-100的磷酸盐平衡生理盐水),室温敷育2小时;
6、使用抗体稀释缓冲液(含1%牛血清蛋白和0.3%Triton-X-100的磷酸盐平衡生理盐水)稀释一抗,稀释比遵循说明书,向每孔加入适量一抗稀释液,于4℃敷育过夜;
7、弃一抗稀释液,磷酸盐平衡生理盐水清洗脑片3次,每次3min;
8、使用抗体稀释缓冲液稀释过氧化物酶偶联的二抗,稀释比遵循说明书,向每孔加入适量二抗稀释液,常温敷育2小时;
9、弃二抗稀释液,磷酸盐平衡生理盐水清洗脑片3次,每次3min;
10、使用DAB过氧化物酶底物试剂盒进行显色,步骤遵循试剂盒说明书;
11、捞出脑片,贴片于载玻片上,置于阴凉通风处干燥;
12、经75%乙醇2min,95%乙醇2min,无水乙醇2min,二甲苯2min,脱水透明。
13、使用中性树胶封片。
14、使用数字扫描系统VS200采集图片,使用ImageJ软件对图片进行处理。
图3结果显示,相较于对照组大鼠,移植人多巴胺能神经元样细胞16周后,细胞治疗组大鼠的黑质-纹状体通路中多巴胺能神经元的数量显著增加,这一结果与行为学的改善一致。
实施例16脑片免疫荧光(Immunofluorescence,IF)
1、将脑片置于12孔板,3张/孔;
2、磷酸盐平衡生理盐水清洗脑片3次,每次3min;
3、向每孔加入封闭液(含3%牛血清蛋白和1%Triton-X-100的磷酸盐平衡生理盐水),室温敷育2小时;
4、使用抗体稀释缓冲液(含1%牛血清蛋白和0.3%Triton-X-100的磷酸盐平衡生理盐水)稀释一抗,稀释比遵循说明书,向每孔加入适量一抗稀释液,于4℃敷育过夜;
5、弃一抗稀释液,磷酸盐平衡生理盐水清洗脑片3次,每次5min;
6、使用抗体稀释缓冲液稀释带荧光标签的二抗,稀释比遵循说明书,向每孔加入适量二抗稀释液,常温避光敷育2小时;
7、弃二抗稀释液,磷酸盐平衡生理盐水清洗脑片3次,每次5min;
8、捞出脑片,贴片于载玻片上,置于避光通风处干燥;
9、每张载玻片滴上约60ul含DAPI的封片剂,盖上盖玻片封片;
10、使用共聚焦显微镜FV3000采集图片。
图4结果显示,在移植8,12及16周后,移植进大鼠纹状体的人多巴胺能神经元样细胞仍能稳定表达TH,说明其保持了其多巴胺能神经元的属性特征。此外,一些移植的细胞展现出了神经元的形态,主要表现为突触的展开,意味着移植的细胞可能整合进了帕金森大鼠的纹状体并行使成熟多巴胺能神经元的一部分功能。
本发明体外诱导人羊膜上皮干细胞分化为人多巴胺能神经元样细胞的方法,诱导产生的人多巴胺能神经元样细胞具有高度的类神经元形态,且相较于人羊膜上皮干细胞,其神经元及多巴胺能神经元的经典标记物的表达水平显著上升,此外,其多巴胺的分泌水平也显著提高。另一方面,经组合物诱导分化后,细胞仍具有很高的细胞活力,更重要的是,其HLA-A,HLA-B,HLA-DR和HLA-DQ抗原的表达仍然维持在很低的水平,说明分化后的细胞仍保持很低的免疫原性,适合作为移植物进行帕金森病的细胞治疗。动物实验结果表明,向帕金森模型大鼠移植分化得到的人多巴胺能神经元样细胞,能够改善帕金森大鼠的行为学,增加其黑质-纹状体通路的多巴胺能神经元数量。且组织学染色分析表明,移植的人多巴胺能神经元样细胞能在移植区域存活,并整合进大鼠的纹状体中。以上实验结果表明本发明具有良好的临床转化前景。
Claims (9)
1.一种诱导人羊膜上皮干细胞向人多巴胺能神经元样细胞分化的方法,其特征在于,包括以下步骤:
(1)分离人羊膜上皮干细胞,贴壁培养;
(2)将细胞培养液依次更换为诱导组合物-1和诱导组合物-2,诱导人羊膜上皮干细胞分化为人多巴胺能神经元样细胞;
其中,所述的诱导组合物-1为含有抗坏血酸、成纤维细胞生长因子-2和noggin的细胞培养基;
所述的诱导组合物-2为含有成纤维细胞生长因子-8、音猬因子、脑源性神经营养因子、胶质细胞源性神经营养因子、CHIR99021和二丁酰环磷腺苷的细胞培养基。
2.根据权利要求1所述的诱导人羊膜上皮干细胞向人多巴胺能神经元样细胞分化的方法,其特征在于:步骤(1)中,所述人羊膜上皮干细胞为小于P2代次的人羊膜上皮干细胞。
3.根据权利要求1所述的诱导人羊膜上皮干细胞向人多巴胺能神经元样细胞分化的方法,其特征在于:步骤(1)中,细胞贴壁培养24-48小时。
4.根据权利要求1所述的诱导人羊膜上皮干细胞向人多巴胺能神经元样细胞分化的方法,其特征在于:步骤(2)中,先使用诱导组合物-1培养6~8天,后更换为诱导组合物-2继续培养12~16天。
5.根据权利要求1所述的诱导人羊膜上皮干细胞向人多巴胺能神经元样细胞分化的方法,其特征在于:步骤(2)中,所述的诱导组合物-1为含有0.2-5mM抗坏血酸,1-20ng/ml成纤维细胞生长因子-2和10-500ng/ml noggin的细胞培养基;
所述的诱导组合物-2为含有5-100ng/ml成纤维细胞生长因子-8、10-500ng/ml音猬因子,2-50ng/ml脑源性神经营养因子、5-50ng/ml胶质细胞源性神经营养因子、0.3-10uMCHIR99021和10-500uM二丁酰环磷腺苷的细胞培养基。
6.根据权利要求1所述的诱导人羊膜上皮干细胞向人多巴胺能神经元样细胞分化的方法,其特征在于:步骤(1)中,分离人羊膜上皮干细胞,具体过程包括:使用含双抗的磷酸盐平衡溶液多次洗涤羊膜组织后,依次经过酶消化、离心,收获人羊膜上皮干细胞。
7.根据权利要求1~6任一项所述的方法得到的人多巴胺能神经元样细胞。
8.根据权利要求7所述的人多巴胺能神经元样细胞或其细胞制剂在制备治疗和/或改善帕金森病中的用途。
9.一种细胞制剂,其特征在于,包括权利要求7所述的人多巴胺能神经元样细胞以及药学可接受的各类载体。
