Abstract

本发明属于宠物食品技术领域,特别涉及一种提高宠物猫消化功能的益生菌猫粮及其制备方法。
背景技术
随着宠物猫的普及和宠物主对宠物健康的关注度提高,猫粮的质量和功能性成为重要研究方向。目前市场上的猫粮多注重基础营养的补充,而忽视了猫的特殊消化系统需求和肠道健康。猫作为严格的肉食动物,其肠道较短,对营养物质的吸收能力有限,易受到肠道菌群失衡、消化不良等问题的影响。目前,益生菌因其在提升消化健康方面的潜力而得到了广泛的研究和应用。益生菌在提高宿主肠道健康、维持肠道微生态平衡以及增强营养吸收率方面具有明显的正面作用。然而,益生菌对于温度、湿度和存储条件都具有严格的要求。传统的猫粮生产工艺如高温挤压和烘干可能会破坏益生菌的活性,将益生菌有效地整合进猫粮,同时确保其在生产加工、存储和猫消费过程中的活性和稳定性,是一个技术挑战。
因此,开发一种能够提高猫消化功能、调节肠道菌群平衡的功能性猫粮,对于提升宠物猫的生活质量具有重要意义。

针对现有技术中存在的问题,本发明的目的在于提供一种提高宠物猫消化功能的益生菌猫粮及其制备方法,通过添加特定的益生菌组合物,解决现有猫粮所忽视的宠物猫肠道健康问题,所述益生菌猫粮不仅营养均衡,还能够有效调节宠物猫肠道健康,提升宠物猫肠道消化能力、增强宠物猫的免疫功能。

新鲜鸡肉50-70份,脱骨鸡肉10-25份,鲜鸡肝3-8份,红薯粉3-5份,全蛋粉3-5份,鸡油3-5份,鱼油3-5份,木薯淀粉2-4份,新鲜南瓜0.5-1份,西蓝花0.5-1份,芹菜0.5-1份,胡萝卜0.5-1份,啤酒酵母粉0.1-0.5份,海带粉0.1-0.4份,车前子粉0.1-0.4份,蔓越莓干0.1-0.4份,添加料1-3份;

进一步的,所述添加料包括:牛磺酸、氯化胆碱、氯化钠、氯化钾、磷酸氢钙、果寡糖、甘露寡糖、迷迭香提取物、茶多酚、维生素A乙酸酯、维生素D3、dl-α-生育酚、硝酸硫胺(维生素B1)、核黄素(维生素B2)、烟酸、D-泛酸钙、盐酸吡哆醇(维生素B6)、叶酸、D-生物素、氰钴胺(维生素B12)、甘氨酸铜螯合物、甘氨酸铁螯合物、甘氨酸锌、蛋氨酸锰络合物、碘酸钙、天然类固醇萨洒皂角苷(源自丝兰)。
本发明的第二方面提供了上述提高宠物猫消化功能的益生菌猫粮的制备方法,包括以下步骤:
(1)按照上述的重量份数称取各配料,经斩拌处理将各配料混合均匀;
(2)将混合均匀的配料加入制粒机,成型处理获得猫粮颗粒;
(3)将所述猫粮颗粒进行三段式低温烘焙,得到烘焙粮;
(4)按照上述活菌数,称取动物双歧杆菌乳亚种U9和植物乳杆菌CR12菌粉,溶解得到益生菌混合液;烘焙粮冷却后,使用喷涂设备将益生菌混合液均匀喷涂在烘焙粮表面;完成喷涂后,让猫粮在无菌的流通空气环境下自然风干,即得到所述提高宠物猫消化功能的益生菌猫粮。
进一步的,步骤(3)中三段式低温烘焙的条件为:
90-100℃烘焙30分钟,100-120℃烘焙30分钟,80-100℃烘焙3小时。

本发明相比现有技术的有益效果为:
1、本发明所述益生菌猫粮中,添加由动物双歧杆菌乳亚种U9和植物乳杆菌CR12组成的益生菌组合物,能够有效调节猫肠道菌群平衡,增强猫肠黏膜屏障功能,从而提升猫的消化能力和整体健康。通过实验验证,本发明的猫粮能显著改善猫的肠道菌群结构,增强肠道免疫功能,提高血清抗氧化能力,及调节血清炎症因子,显示出优良的益生效果;
2、本发明通过特定的配方和加工工艺(三段式低温烘焙+益生菌冷喷涂),不仅能够保护猫粮中维生素、氨基酸和某些生物活性物质不被破坏,改善食品的口感,还可提高可消化性,有效保证了益生菌组合物的活性。

实施例1
本实施例提供了一种提高宠物猫消化功能的益生菌猫粮,包括配料和益生菌组合物,其中,所述益生菌组合物由动物双歧杆菌乳亚种U9和植物乳杆菌CR12组成。
动物双歧杆菌乳亚种U9,保藏编号CGMCC No. 11560;植物乳杆菌CR12,保藏编号CGMCC No. 14000。

本实施例所述添加料购自山东汉欧生物科技有限公司,商品名称为烘焙猫粮添加剂。
所述益生菌猫粮的制备方法为:
(1)按照上述的重量份数称取各配料,加入斩拌机(型号:ZBJ-20,电压:380V,功率:2.25kw)中、斩拌处理10min,将各配料混合均匀。
(2)将混合均匀的配料加入电动制粒机(型号:47511153,电压:220V,功率:650w,制粒直径6mm),成型处理获得猫粮颗粒。
(3)将所述猫粮颗粒进行三段式低温烘焙,得到烘焙粮。
其中,三段式低温烘焙的条件为:90-100℃烘焙30分钟,100-120℃烘焙30分钟,80-100℃烘焙3小时。
(4)按照上述活菌数,称取动物双歧杆菌乳亚种U9和植物乳杆菌CR12菌粉,溶解得到益生菌混合液;烘焙粮冷却后,使用喷涂设备(型号:SYPZ1000,电压:380V,功率:1.5kw)将益生菌混合液均匀喷涂在烘焙粮表面;完成喷涂后,让猫粮在无菌的流通空气环境下自然风干,即得到所述提高宠物猫消化功能的益生菌猫粮。

所述市售猫粮的制备方法参照对比例1。
测试例1:肠道菌群结构测试
对上述实施例1及对比例1-4中制备的猫粮,分成5组分别饲喂宠物猫,每组6只猫,每只猫单笼喂养四周,试验前已进行必要的免疫和驱虫处理,猫舍自然通风及照明,室温维持在18℃~26℃,相对湿度保持在40%~70%。每天早、晚各清理粪便1次,每天打扫卫生和消毒猫舍,保持宠物圈舍内的清洁,第四周饲喂结束后,采用代谢笼采集新鲜粪便样品存于-80℃,提取DNA,采用PacBio第三代高通量测序分析肠道菌群结构,主要变化菌群的相对丰度如表1所示:
由表1可知,实施例1制备得到的益生菌猫粮对猫菌群结构影响最明显,食用实施例1的益生菌猫粮后,猫肠道Lactobacillus的相对丰度显著增加,而Peptoclostridium和Collinsella的丰度跟对比例1相比显著降低,与对比例2、对比例3,以及市场上市售的对比例4普通猫粮相比,肠道菌群增加的主要是益生菌,该结果表明,本发明的配方可以改善肠道菌群,起到促进消化的作用。
测试例2:益生菌猫粮对宠物猫肠道屏障功能的影响
本测试例分别测定饲喂对比例1-4猫粮、实施例1益生菌猫粮两组宠物猫,血清中的D-乳酸(D-Lactic Acid, D-LA)和二胺氧化酶(Diamine Oxidase, DAO)的含量。本实施例使用D-乳酸试剂盒、二胺氧化酶试剂盒,均购自北京华英生物技术研究所。
由表2可知,实施例1饲喂益生菌组合物组猫粮显著降低了血清中D-乳酸和二胺氧化酶的含量,D-乳酸是一种由肠道内的某些细菌发酵产生的有机酸。在正常情况下,它以较小的量存在,并可以通过肠道屏障排出体外。然而,如果肠道屏障功能受损,D-乳酸的水平会在血液中升高。这是因为肠道屏障的损伤导致了更多的D-乳酸从肠道进入血液。DAO的主要功能是分解食物和肠道细菌产生的组胺,防止过多的组胺进入血液循环,从而减轻组胺相关的炎症和过敏反应。益生菌组合物的增加可以帮助竞争性抑制有害微生物的生长,从而减少炎症和组胺的产生。益生菌是对宿主有益的活菌,能够改善肠道微生态平衡,增强肠道屏障功能。通过饲喂含有益生菌的猫粮,可以帮助改善肠道黏膜的屏障功能,从而降低D-LA和DAO的血清水平。这表明肠道屏障的完整性得到了改善,减少了有害物质和病原体的穿透,从而维护了宿主的健康状态。
其中,通过比对可以发现,饲喂实施例1益生菌组合物组猫粮的D-LA和DAO的血清水平,要明显低于饲喂对比例2、3单一菌种组猫粮,这说明动物双歧杆菌乳亚种U9和植物乳杆菌CR12两菌株复配,可显著增强宠物猫的肠道屏障功能。
测试例3:益生菌猫粮对猫肠道免疫功能的影响
本测试例分别测定饲喂对比例1-4猫粮、实施例1益生菌猫粮两组宠物猫,血清中的免疫球蛋白A(IgA)的含量。本实施例使用猫免疫球蛋白IgG / A / M测定试剂盒,购自北京华英生物技术研究所。
由表3可知,实施例1益生菌组合物组猫粮显著增加了IgA的含量,IgA是一种重要的抗体,主要在黏膜表面发挥作用,是黏膜免疫系统的关键成分。它在维护肠道健康、预防病原体入侵和调节免疫响应中起着至关重要的作用。且增强了血清中免疫球蛋白G(IgG)和M(IgM)的含量。
测试例4:益生菌猫粮对猫血清抗氧化能力的影响
本测试例分别测定饲喂对比例1-4猫粮、实施例1益生菌猫粮两组宠物猫,血清中的催化酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和总抗氧化能力(T-AOC)含量。本实施例使用过氧化氢酶试剂盒,超氧化物歧化酶试剂盒、谷胱甘肽过氧化物酶试剂盒、总抗氧化能力测试盒,均购自北京华英生物技术研究所。
由表4可知,饲喂实施例1制备的益生菌猫粮导致猫血清中催化酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和总抗氧化能力(T-AOC)含量增加,这些指标主要与体内的抗氧化防御系统有关,对于抵抗氧化应激、预防细胞损伤和维持生理功能的稳定至关重要。CAT能够快速分解氢过氧化物(H2O2),减少氧化损伤;SOD能够将超氧自由基(O2-)转化为氢过氧化物和氧气,是抵御氧化应激的第一道防线;GSH-Px将脂质过氧化物还原为醇,并将H2O2还原为水,保护细胞免受氧化损伤;T-AOC(总抗氧化能力):反映体内抗氧化剂的总水平和整体抗氧化状态。益生菌通过改善肠道微生态平衡,间接提升了机体的抗氧化能力。肠道健康与全身的氧化应激水平密切相关,健康的肠道可以帮助减轻全身的氧化负担。良好的抗氧化状态有助于保护肠道黏膜,提高营养物质的吸收效率,进一步增强机体的健康状况和生理功能。
测试例5:益生菌猫粮对猫血清炎症因子的影响
本测试例分别测定饲喂对比例1-4猫粮、实施例1益生菌猫粮两组宠物猫,血清中的白细胞介素-4 (IL-4)的水平,并显著降低白细胞介素-2 (IL-2)、肿瘤坏死因子-α(TNF-α)和干扰素-γ (IFN-γ)的含量。本实施例使用白细胞介素-4试剂盒,白细胞介素-2试剂盒、肿瘤坏死因子-α试剂盒和干扰素-γ试剂盒,均购自北京华英生物技术研究所。
由表5可知,饲喂实施例1制备的益生菌猫粮显著增加猫血清中白细胞介素-4(IL-4)的水平,并显著降低白细胞介素-2 (IL-2)、肿瘤坏死因子-α (TNF-α)和干扰素-γ(IFN-γ)的含量。IL-4是一种主要由Th2细胞产生的细胞因子,对于调节免疫反应和促进抗体产生具有关键作用。IL-4的增加表明免疫系统正向Th2型反应倾斜,这种反应通常与抗寄生虫免疫和过敏反应相关。在非过敏背景下,IL-4的增加有助于促进B细胞分化和IgE以及其他类型抗体的产生,增强机体对特定病原体的免疫防御。IL-2:主要由Th1细胞产生,是免疫系统中T细胞增殖和活化的关键因子。IL-2水平的降低可能反映了对Th1型免疫反应的抑制,这种反应通常与细胞介导的免疫防御相关。TNF-α:是一种主要由巨噬细胞产生的炎症性细胞因子,对于启动炎症反应和抵抗感染至关重要。然而,TNF-α的过度表达与多种炎症和自身免疫疾病的发展有关。TNF-α水平的降低可能有助于减少慢性炎症和降低某些自身免疫条件的风险。IFN-γ:同样由Th1细胞产生,是强烈的促炎细胞因子,对抗病毒和某些细菌感染至关重要。IFN-γ水平的降低可能表明对Th1型免疫反应的调节,有助于减轻与过度免疫反应相关的炎症状况。

Claims (4)
1.一种提高宠物猫消化功能的益生菌猫粮,包括配料和益生菌组合物,其特征在于,所述益生菌组合物包括动物双歧杆菌乳亚种U9和植物乳杆菌CR12;
动物双歧杆菌乳亚种U9,保藏编号CGMCC No. 11560;
植物乳杆菌CR12,保藏编号CGMCC No. 14000;
各配料按重量份数计,包括:
新鲜鸡肉50-70份,脱骨鸡肉10-25份,鲜鸡肝3-8份,红薯粉3-5份,全蛋粉3-5份,鸡油3-5份,鱼油3-5份,木薯淀粉2-4份,新鲜南瓜0.5-1份,西蓝花0.5-1份,芹菜0.5-1份,胡萝卜0.5-1份,啤酒酵母粉0.1-0.5份,海带粉0.1-0.4份,车前子粉0.1-0.4份,蔓越莓干0.1-0.4份,添加料1-3份;
所述益生菌猫粮中动物双歧杆菌乳亚种U9的活菌数为1×1010 CFU/Kg~2×1010 CFU/Kg,植物乳杆菌CR12的活菌数为1×1010 CFU/Kg~2×1010 CFU/Kg。
2.根据权利要求1所述的益生菌猫粮,其特征在于,所述添加料包括:牛磺酸、氯化胆碱、氯化钠、氯化钾、磷酸氢钙、果寡糖、甘露寡糖、迷迭香提取物、茶多酚、维生素A乙酸酯、维生素D3、dl-α-生育酚、硝酸硫胺、核黄素、烟酸、D-泛酸钙、盐酸吡哆醇、叶酸、D-生物素、氰钴胺、甘氨酸铜螯合物、甘氨酸铁螯合物、甘氨酸锌、蛋氨酸锰络合物、碘酸钙、天然类固醇萨洒皂角苷。
3.一种如权利要求1或2所述提高宠物猫消化功能的益生菌猫粮的制备方法,其特征在于,所述制备方法包括以下步骤:
(1)按照如权利要求1所述的重量份数称取各配料,经斩拌处理将各配料混合均匀;
(2)将混合均匀的配料加入制粒机,成型处理获得猫粮颗粒;
(3)将所述猫粮颗粒进行三段式低温烘焙,得到烘焙粮;
(4)按照如权利要求1所述活菌数,称取动物双歧杆菌乳亚种U9和植物乳杆菌CR12菌粉,溶解得到益生菌混合液;烘焙粮冷却后,使用喷涂设备将益生菌混合液均匀喷涂在烘焙粮表面;完成喷涂后,让猫粮在无菌的流通空气环境下自然风干,即得到所述提高宠物猫消化功能的益生菌猫粮。
4.根据权利要求3所述的制备方法,其特征在于,步骤(3)中三段式低温烘焙的条件为:
90-100℃烘焙30分钟,100-120℃烘焙30分钟,80-100℃烘焙3小时。
