Abstract
本发明属于生物医药领域,主要涉及抑制CD47‑SIRPα信号轴基因的siRNA、纳米粒子及其用途。本发明要解决的技术问题是为小核酸药物抗肿瘤药物提供一种新的有效选择。解决问题的技术方案是提供两种能高效地抑制CD47‑SIRPα表达和肿瘤增殖的siRNA。所述的siRNA能高效抑制CD47和/或SIRPα基因的表达,同时所述的siRNA能有效抑制肿瘤细胞增殖。且本发明的DP7‑C/siCD47复合物能有效治疗黑色素瘤肺转移,且DP7‑C/siSIRPα复合物能有效治疗黑色素瘤、结肠癌等多种肿瘤,具有很好的应用前景。

技术领域
本发明属于基因治疗领域,主要涉及抑制CD47-SIRPα信号轴基因表达的siRNA、纳米粒子及其用途。
背景技术
小核酸药物通过反义技术靶向RNA可使特定疾病基因沉默,达到治疗目的。小核酸药物作为一种治疗策略,具有高特异性、高效性、长效性等优势,大大扩展了可用于治疗的靶点的数量和类型。安全高效的递送系统是小核酸药物研发的关键,而筛选出有效的靶点也是小核酸药物研发的重点。
1957年,Burnet和Thomas提出了“免疫监测”假说,认为免疫系统可以监测并消除“外来”病原体,包括表达各种肿瘤特异性抗原和非肿瘤特异性抗原的恶性细胞,以维持一个稳定的环境。肿瘤免疫监视是限制肿瘤生长的重要过程,巨噬细胞在肿瘤细胞的识别和清除过程中起着重要作用。T细胞和自然杀伤细胞(NK)也是免疫系统中重要的效应细胞,在抗肿瘤免疫中也起着关键作用。而肿瘤细胞逃避免疫识别和清除依赖于多种过程,包括诱导免疫抑制肿瘤微环境和降低肿瘤细胞免疫原性。
CD47(分化簇47,也称为整合素相关蛋白,integrin-associated protein)是一种具有糖基化的跨膜蛋白,CD47激活后可以介导细胞增殖、迁移、吞噬以及细胞凋亡,免疫稳态等一系列过程。肿瘤细胞免疫逃逸的一个关键机制是通过过度表达免疫抑制信号分子CD47。CD47被广泛认为是一种“不要吃我”的信号,它有助于在生理条件下维持非恶性细胞的免疫耐受性。但另一方面,这种分子却可以帮助不同癌症类型的癌细胞存活。癌细胞利用CD47的“不要吃我”的功能,在表面表达高于非恶性细胞水平的CD47;大量研究表明,CD47可在不同类型的肿瘤中过度表达,包括骨髓瘤、平滑肌肉瘤、急性淋巴细胞白血病、非霍奇金淋巴瘤、乳腺癌、骨肉瘤、头颈部鳞状细胞癌、脑胶质瘤等等。CD47表达水平与癌症恶化的治疗反应和预后相关。肿瘤细胞表面高表达CD47蛋白后,CD47蛋白与巨噬细胞表面的配体SIRPα结合后会抑制巨噬细胞对肿瘤细胞的吞噬作用,从而使得肿瘤逃避免疫监视,难以抑制肿瘤生长,促进肿瘤转移。抑制CD47在肿瘤细胞中持续高表达可能抑制肿瘤生长和转移,因此靶向抑制肿瘤中CD47的表达是抗肿瘤治疗的可能的有效策略。
肿瘤细胞表面高表达CD47蛋白,而该蛋白与巨噬细胞表面的配体SIRPα结合后会抑制巨噬细胞对肿瘤细胞的吞噬作用从而使得肿瘤逃避免疫监视得以发生转移。研究表明SIRPα是一种属于免疫球蛋白超家族的膜蛋白,主要由巨噬细胞和树突状细胞等髓系细胞表达,通过进一步阐明吞噬抑制的机制,研究人员发现巨噬细胞的SIRPα会与邻近细胞上表达的CD47相互作用,导致SIRPα胞浆免疫受体酪氨酸抑制基序磷酸化。这一过程会募集SHP-1和SHP-2磷酸酶,下游信号级联阻止肌球蛋白-IIA在吞噬细胞突触聚集,导致吞噬抑制。实体肿瘤组织中存在大量的巨噬细胞,而这些巨噬细胞处于免疫抑制状态,促进了肿瘤的发生转移。因此靶向巨噬细胞中SIRPα是抗肿瘤治疗中的潜在靶点。
目前,小核酸药物在实际应用过程中也效果有限,注射小核酸药物后,如何延长药物在体内的存留时间,如何靶向、精准的让治疗性的小核酸药物进入靶细胞发挥治疗功能等方面还面临很大的挑战。而且小核酸药物效果不及预期还可能是由于以下方面的差异:(1)siRNA靶点选择及序列设计的挑战,如特异性、有效浓度、沉默效率、脱靶效应等;(2)药物输送方式(路径、载体、频率、效率);(3)肿瘤细胞内的药物的区域化分布;(4)肿瘤异质性、肿瘤微环境的变化、药物失活、肿瘤细胞药物吸收减少或药物释放增加、肿瘤细胞存活途径的激活和表观遗传变化等等方面都由可能影响小核酸药物的实际使用情况。此外,小核酸药物尤其是RNAi药物,分子到达作用位点需要穿过胞膜作用于胞浆或核内的mRNA,递送难度大,尽管化学修饰能够解决稳定性和免疫原性的问题,但如果不能进入细胞实现胞吞,小核酸药物依然不能发挥药物作用。
DP7多肽(VQWRIRVAVIRK)是用计算机辅助设计新型抗菌肽的筛选方法,通过对模板抗菌肽HH2替换了2个氨基酸得到的具有更高的细菌识别特异性与更强的抗菌活性的抗菌肽。抗菌肽纳米化可以使其获得较好稳定性及减少毒性,这为抗菌肽的应用提供一个新研究方向。
发明内容
本发明要解决的技术问题是为小核酸药物抗肿瘤药物提供一种新的有效选择。
为了解决上述的技术问题,本发明提供了抑制CD47-SIRPα信号轴基因表达siRNA。
本发明提供了一种抑制CD47-SIRPα信号轴基因表达表达的siRNA,所述siRNA序列为抑制SIRPα的siRNA,其序列为:
SIRPαsiRNA正义链(SEQ ID No.1):5’-CCGUGAACCCUAGUGGAAA-3’;

本发明还提供了另一种抑制CD47-SIRPα信号轴基因表达的siRNA,所述siRNA序列为抑制CD47的siRNA,其序列为:

进一步的,上述的抑制CD47基因表达的siRNA和抑制SIRPα基因的siRNA的正义链和反义链的3’端垂悬有dTdT。
在此基础上,本发明还提供了抑制CD47-SIRPα信号轴基因表达的siRNA组,其包括上述的抑制CD47基因表达的siRNA和抑制SIRPα基因的siRNA。
本发明还提供了一种装载了上述的siRNA的纳米粒子。该纳米粒子是由疏水化修饰的多肽装载上述的抑制CD47和SIRPα基因表达的siRNA制备而成;所述多肽的序列为VQWRIRVAVIRK(SEQ ID No.5),所述的疏水化修饰为在多肽的氮末端偶联疏水片段。
其中,上述装载siRNA的纳米粒子中所述的多肽VQWRIRVAVIRK碳端酰胺化修饰为VQWRIRVAVIRK-NH2。
其中,上述疏水片段为甾醇类化合物或饱和直链脂肪酸或PEG或PEG衍生物;其中,所述的甾醇类化合物为胆固醇类化合物或胆酸类化合物;或者所述饱和直链脂肪酸为C6-C20中的至少一种。
其中,上述的甾醇类化合物为丁二酰化胆固醇、胆酸或去氧胆酸中的至少一种;或者所述的PEG衍生物为1,2-二油酰-SN-甘油-3-磷酰乙醇胺-聚乙二醇、二硬脂酰基磷脂酰乙醇胺-聚乙二醇或二棕榈酰磷酯酰乙醇胺-聚乙二醇中的至少一种。

其中,上述装载siRNA的纳米粒子是由疏水化修饰阳离子多肽与所述药物在水中、缓冲液或液态培养基中共孵育5~15min而得;优选的,所述缓冲液为磷酸盐缓冲液、Tris-HCl缓冲液或HEPES缓冲液中的至少一种,液体培养基为RPMI 1640、DMEM双无培养基或Opti-MEM培养基中的至少一种。本发明还提供了上述的抑制CD47-SIRPα信号轴基因表达的siRNA、抑制CD47-SIRPα信号轴基因表达的siRNA组或上述的装载siRNA的纳米粒子在制备治疗肿瘤的药物中的应用。
进一步的,所述的肿瘤为黑色素瘤、结肠癌、乳腺癌、肺癌、脑胶质瘤、胰腺癌、骨肉瘤、头颈部肿瘤等中的至少一种。
本发明还提供了一种抗肿瘤药物,是由上述的抑制CD47-SIRPα信号轴基因表达的siRNA、抑制CD47-SIRPα信号轴基因表达的siRNA组或上述的装载siRNA的纳米粒子添加药学上可接受的成分制备而成。进一步的上述药物还可以含有药学上可以接受的辅助性成分。
本发明的有益效果在于:本发明提供了效果很好的,针对CD47-SIRPα信号轴基因表达的siRNA分子和siRNA分子组。本发明还进一步地使用了抗菌肽衍生物DP7-C作为本发明针对CD47和/或SIRPα基因的siRNA分子的递送系统,制得了新的纳米药物。本发明CD47siRNA和SIRPαsiRNA能分别高效抑制CD47和SIRPα基因的表达,mRNA水平均可显著下调50%以上。本发明提供的装载了针对SIRPα基因的siRNA和针对CD47基因的siRNA分子的DP7-C纳米胶束具有高效的siRNA传输能力,可有效地局部递送其中的靶向巨噬细胞的siRNA以用于实体瘤以及肺部肿瘤转移灶等肿瘤的治疗肿瘤的治疗。此外,在体内外均表现出较高的安全性,而且制备简便,成本较低,在肿瘤基因治疗上有很好的应用前进。

具体实施方式
CD47-SIRPα通路是2000年以来被鉴定的介导肿瘤细胞免疫逃逸的通路之一。SIRPα属于Ig超家族成员,是一种主要表达在髓系造血细胞如巨噬细胞、单核细胞、树突状细胞和粒细胞表面的跨膜蛋白;CD47是SIRPα最主要的配体。CD47又称为整合素相关蛋白,是一种五重跨膜糖蛋白,在正常细胞表面广泛表达,通过与SIRPα结合,释放一种"别吃我"的信号,巨噬细胞由此避免误伤正常细胞。然而狡猾的肿瘤细胞也会表达高水平的CD47蛋白,进而通过与SIRPα结合,逃避吞噬细胞的清除。目前,CD47-SIRPα信号通路的抑制剂主要分为三类:(1)通过阻断靶细胞上的CD47分子来抑制通路活性的分子;(2)通过阻断免疫效应细胞上的SIRPα分子来抑制途径活性的分子;(3)CD47成熟所需的QPCTL酶的抑制剂。肿瘤组织中存在大量的巨噬细胞,而这些巨噬细胞处于免疫抑制状态,促进了肿瘤的发生转移。因此靶向巨噬细胞中CD47或SIRPα是抗肿瘤治疗中潜在的理想靶点。近日,辉瑞开发的1类新药PF-07901801注射液获批临床,拟开发用于治疗晚期血液系统恶性肿瘤患者。PF-07901801为一款在研的CD47-SIRPα重组融合蛋白,由抗体的Fc区和SIRPα蛋白的工程变体组成。该药旨在阻断CD47-SIRPα相互作用,通过这种阻断作用,促进巨噬细胞(一种免疫细胞)对癌细胞的吞噬作用。但安全性和有效性有待临床试验进一步评估。
本发明针对CD47-SIRPα通路,在分析CD47和SIRPα基因及相互作用的基础上,经过设计和大量的前期优选,获得了效果优秀的CD47 siRNA和SIRPαsiRNA分子:

由于小核酸药物后药物如何在体内存留足够长的时间,如何让治疗性的小核酸精准进入靶向细胞发挥治疗功能,并最大程度的避免误伤正常细胞等方面还面临很大的挑战。因此,需要为此siRNA搭配适当的递送系统,以保证其发挥自身的作用。
为此,本发明进一步地使用了抗菌肽衍生物DP7-C作为本发明针对CD47-SIRPα通路基因的siRNA分子的递送系统,制得了新的纳米药物。
本发明一个实施例中使用的DP7-C为丁二酰化胆固醇修饰DP7多肽,其肽结构为:

DP7-C的合成及DP7-C纳米粒子的制备可参考中国专利文献CN107446019 A记载的方法进行。
本发明DP7-C中的抗菌肽DP7分子量小,可以方便的使用Fmoc固相多肽合成法合成,与胆固醇经化学合成偶联方法简单易行。DP7-C在水溶液中能自组装成纳米胶束,在水溶液中呈正电,有较好的单分散性和Zeta电位,冻干再溶解也很稳定。
基于该DP7-C递送系统,局部递送靶向巨噬细胞的CD47 siRNA和SIRPαsiRNA可用于实体瘤的治疗。本发明涉及到的DP7-C纳米胶束除了高效的siRNA传输能力,其在体内外也均表现出较高的安全性。在本发明的实例中,装载有本发明抑制CD47-SIRPα信号轴基因表达的siRNA的DP7-C纳米胶束,成功实现对肿瘤细胞的高效siRNA导入,并有效抑制多种肿瘤模型的生长,得到一种新型的CD47 siRNA和SIRPαsiRNA药物。
以下将结合附图和实施例详细说明本发明,应当说明的是,实施例是对本发明的技术方案的进一步说明,并不表明本发明仅限于这些实施例。
本发明实施例使用的主要试剂:
(1)小干扰RNA(siRNA,上海吉玛公司合成)。
(2)CFDA SE细胞增殖与示踪检测试剂盒(C0051,碧云天)。
(3)CD47抗体(ab175388,abcam)。
(4)TUNEL检测试剂盒(A113-01,诺维赞)。
本发明所涉及到的细胞系:
(1)小鼠单核巨噬细胞系:RAW264.7(购买自ATCC,美国典型培养物保藏中心)。
(2)小鼠黑色素瘤细胞系:B16F10(购买自ATCC,美国典型培养物保藏中心)。
(3)小鼠结肠癌细胞系:MC38、CT26(购买自ATCC,美国典型培养物保藏中心)。
实施例1CD47表达量与多种免疫细胞浸润程度呈高度相关性
通过生物信息学(http://timer.cistrome.org/)分析,在冷肿瘤(肾上腺皮质癌、膀胱癌、脑胶质瘤、肺腺癌、黑色素瘤、甲状腺癌)和热肿瘤(结直肠癌、乳腺癌)等多种肿瘤中,CD47的表达量与多种免疫细胞浸润程度呈高度相关性(图1),表明CD47是一种重要的免疫治疗靶点,其具备广泛的应用潜力,可用于多种“冷”和“热”肿瘤。
实施例2SIRPα表达量与多种免疫细胞浸润程度呈高度相关性
首先LPS将BMDM诱导为诱导为M1型BMDM。用CFDASE绿色荧光染料对M1型BMDM进行标记。其次,使用pHrodoTM Red-SE红色荧光染料将肿瘤细胞进行标记。当肿瘤细被巨噬吞噬时,可通过绿色和红荧光共定位胞进行标记。
实验结果(见图2A)显示:与SIRPα蛋白孵育后的MC38肿瘤细胞被巨噬细胞吞噬的数量与共培养时间成正比,且在共培养后2h达到饱和,未处理组被巨噬细胞吞噬的MC38肿瘤细胞没有随着共培养时间的延长而增加。通过生物信息学(http://timer.cistrome.org/)分析,在在冷肿瘤(肾上腺皮质癌、膀胱癌、脑胶质瘤、肺腺癌、黑色素瘤、甲状腺癌)和热肿瘤(结直肠癌、乳腺癌)等多种肿瘤病人中,SIRPα的表达量与多种免疫细胞浸润程度呈高度相关性,在调控免疫细胞的反应中起着关键的调控作用,提示SIRPα在调节免疫细胞浸润和肿瘤抗性中的重要作用(图2B)。
实施例3抑制CD47的siRNA的筛选
趋化因子受体CD47存在于免疫细胞表面,能引导免疫细胞到达炎症和肿瘤部位。表达CD47的癌细胞会产生免疫抑制的微环境,治疗性阻断CD47信号可以抑制炎症细胞聚集、浸润,从而逆转肿瘤微环境的免疫抑制状态并且激活抗肿瘤CD8+T细胞应答。
本发明中靶向CD47基因,通过设计和筛选得到的抑制CD47基因的siRNA(siCD47),以期得到具有很好抑制和抗肿瘤效果的siCD47。
在实验前期,我们通过软件设计并前期优选后,选出了三条较佳的针对CD47基因的siRNA。首先,将合成的三条靶向CD47基因的siRNA分别与DP7-C孵育制备成DP7-C/siCD47复合物。使用的DP7-C参考中国专利文献CN107446019 A记载的方法进行的合成得到。
复合物的制备过程具体为将siCD47用DEPC水溶解至终浓度为1μg/μl,吸取2μgsiCD47,在200μl培养基中混匀siCD47,再加入10μg DP7-C,与siCD47室温孵育10分钟后形成复合物,再将复合物轻轻加入B16F10细胞中。通过细胞转染及qPCR实验比较,选出了和DP7-C配合使用后,敲减效率最高的siCD47-2(之后实例中标记为siCD47),与NS组、DP7-C组、DP7-C/siScramble组相比,均具有统计学差异(***p<0.001)。因此选择siCD47-2进行后续体内体外抗肿瘤实验(图3)。siCD47-2的核苷酸序列为:

我们进一步验证了优选的siCD47对CD47 mRNA和蛋白的敲减效率,结果如图4所示,DP7-C/siCD47处理组显著下调了B16F10细胞中CD47基因的mRNA表达水平(图4A,***p<0.001)和蛋白表达水平(图4B)。
实施例4SIRPαsiRNA序列的筛选
经过前期的设计和优选,得到三条候选靶SIRPα基因的siRNA,然后在小鼠单核细胞RAW264.7中筛选出干扰效果最好的SIRPαsiRNA。
(1)细胞铺板:将生长状态良好的RAW264.7细胞按照2×106的细胞量接种到6孔板内,过夜培养。
(2)细胞转染:细胞转染前,先将孔中培养基换为双无培养基。然后DP7-C(10μg)分别设计的三条SIRPαsiRNA(2μg)共孵育15min后加入相应的孔中,每组设置三个复孔。同时将未经任何处理的孔设置为空白对照组,以单独的DP7-C,DP7-C/siScramble为材料对照组。连续转染6小时后,吸出孔中培养基,加入1ml含10%胎牛血清的新鲜培养基继续培养至24h。
(3)总RNA提取、逆转录及qPCR检测:采用Trizol法提取RNA,提取过程中为了防止RNA降解,采用RNAase free的枪尖和EP管;采用cDNA逆转录试剂盒对RNA进行逆转录,加样过程中为了防止RNA降解,采用RNAase free枪尖和200μl的RNAase free PCR管,总体系为20μl。上机检测。
结果如图5所示,DP7-C/siSIRPα-2处理组显著下调了RAW264.7细胞中SIRPα基因的mRNA表达水平,其干扰效率达到50%以上,与NS组(**p<0.01)、DP7-C组(**p<0.01)、DP7-C/siScramble组(*p<0.05)相比,均具有统计学差异,且比其他的SIRPαsiRNA敲减效率更高。
经过前期优选,我们获得了一个和DP7-C复合形成的纳米粒,沉默效率高的SIRPαsiRNA序列,即SIRPαsiRNA-2:

后续实验均选用该条siRNA作为有效siRNA展开实验。同时,本实验证明DP7-C良好的巨噬细胞递送效率及SIRPαsiRNA靶点的有效性。
实施例5体外DP7-C/siCD47复合物抑制肿瘤细胞增殖
肿瘤细胞表面高表达CD47蛋白,而该蛋白与巨噬细胞表面的配体SIRPα结合后会抑制巨噬细胞对肿瘤细胞的吞噬作用,从而使得肿瘤逃避免疫监视得以发生转移。因此,本项目中我们靶向CD47基因,通过不同基因发挥不同抗肿瘤机制来进一步验证DP7-C/siCD47复合纳米粒的有效性。
首先,将siCD47用DEPC水溶解为1μg/μl,吸取2μg siCD47与200μl培养基中混匀,再加入10μg DP7-C,室温孵育10分钟后得到复合物,将复合物轻轻加入培养的B16F10细胞中。持续培养细胞一周,弃去培养基,用1×PBS轻轻清洗。用4%多聚甲醛固定后,1×PBS轻轻清洗细胞三次,用浓度为1%的结晶紫对细胞进行染色,10分钟后弃掉结晶紫染料,计数细胞克隆数。克隆形成实验结果表明,体外在B16F10细胞中转染DP7-C/siCD47复合纳米粒可以显著抑制B16F10细胞增殖(见图6A)。对各组克隆形成数进行统计分析如图6B,相比于NS组158±7,DP7-C组157±11,DP7-C/siScramble组156±6形成的克隆数,DP7-C/siCD47组(39±7)显著抑制了细胞生长,且均有统计学意义(***p<0.001)。
实施例6DP7-C/siSIRPα复合物用于B16F10小鼠黑色素实体瘤的治疗
癌症的发生发展与炎症有着密切的关系,而肿瘤相关巨噬细胞(TAM)是肿瘤相关验证过程中的一种主要的细胞群,大约占到实体肿瘤的50%。巨噬细胞表达SIRPα,通过敲减SIRPα来阻断CD47/SIRPα相互作用、直接杀伤肿瘤效应、抗体依赖的细胞吞噬作用、抗原提呈和T细胞免疫反应、抗体依赖的细胞毒作用和补体依赖的细胞毒作用来诱导抗肿瘤作用。
为了进一步表征DP7-C胶束在肿瘤治疗中的应用潜力,同时验证DP7-C/siSIRPα复合物的抗肿瘤效果,我们建立了B16F10小鼠黑色素皮下移植瘤模型,并使用DP7-C递送SIRPαsiRNA治疗。
(1)第0天时,6-8周龄雌性C57小鼠皮下接种0.2mL细胞悬液(约含2.5×106个B16F10细胞)。
(2)接种第8天时,待肿瘤长至40mm3,小鼠被随机分为4组,每组8只并标记。
(3)小鼠分别瘤内注射生理盐水(空白对照组)、空白DP7-C胶束(50μg)、DP7-C/siScramble复合物(50μg/10μg)、或DP7-C/siSIRPα复合物(50μg/10μg),按照隔一天给药一次,共给药5次的实验方案。每两天测量一次小鼠肿瘤,并做好记录。
(4)在第5次给药结束后5天,断颈法处死所有的小鼠,剥离各组小鼠肿瘤,并称重。
DP7-C/siSIRPα复合物经过瘤内注射治疗B16F10皮下移植肿瘤模型的效果如图7所示。其中,图7A显示了各组小鼠皮下肿瘤体积的统计结果。DP7-C/siSIRPα复合物治疗组的平均肿瘤体积为296.8±86.89mm3,生理盐水对照组体积为881.7±330.9mm3(***p<0.001),空载DP7-C组体积为778.1±299.9mm3(***p<0.001),DP7-C/siScramble复合物为639.9±202.3mm3(*p<0.05)。结果表明,DP7-C/siSIRPα复合物能有效抑制小鼠的肿瘤生长,与其他各组相比均具有统计学差异。
各组小鼠的肿瘤平均重量见图7B,DP7-C/siSIRPα复合物治疗组的平均肿瘤重量为0.61±0.18g,生理盐水对照组重量为2.48±0.72g(***p<0.001),空载DP7-C组重量为0.41±0.1g(***p<0.001),DP7-C/siScramble复合物为2.37±0.23g(*p<0.05)。DP7-C/siSIRPα复合物治疗组的小鼠肿瘤平均重量明显低于其他治疗组,具有统计学差异。该研究结果表明,靶向肿瘤微环境中巨噬细胞的SIRPα靶点是一种有效的抗肿瘤治疗策略。
实施例7系统性递送DP7-C/siCD47复合物治疗小鼠黑色素瘤肺转移模型
将6-8周龄左右的C57雌鼠经过检疫一周后饲养于SPF实验动物房中,在肿瘤细胞接种前一天观察小鼠状态。收集常规培养的B16F10肿瘤细胞并计数,无血清的DMEM基础培养基重悬细胞使得细胞浓度为5×106个/ml。于SPF动物房中超净台内以胰岛素注射针进行小鼠尾静脉注射,每只100μl肿瘤细胞悬液(5×105个细胞/只),小鼠接种完毕后随机分为6组,并做好分组标记。接种完成第七天,按照隔一天给药一次,连续给药7次的实验方案,分别静脉给药生理盐水(NS组)、空白DP7-C组(60μg)、DP7-C/siScramble组(60μg/12μg)、或DP7-C/siCD47组(60μg/12μg)。每两天测量一次小鼠肿瘤,并做好记录。自接种后第21天,断颈法处死所有的小鼠,剥离各组小鼠肿瘤,并称重。
小鼠黑色素瘤肺转移模型的肺整体照片如图8A所示,与其他三组相比,小鼠经DP7-C/siCD47处理后明显抑制了黑色素瘤的肺转移(n=7/组)。图8B显示各组小鼠的平均肺肿瘤结节数量,DP7-C/siCD47组的肺肿瘤结节数量为24.7±4,与NS组170±11相比,肺结节数减少85.5%,差异具有统计学意义(****p<0.0001);同时相比于DP7-C组159±11,DP7-C/siScramble组154±15,差异均具有统计学意义(****p<0.0001)。同时,跟NS组0.4±0.03g(***p<0.001),DP7-C组0.35±0.01g(***p<0.001),DP7-C/siScramble组0.33±0.03g(*p<0.05)相比,DP7-C/siCD47治疗组的肺组织重量显著降低至0.25±0.001g(图8C)。此外,各组肺组织HE染色结果显示,DP7-C/siCD47处理组较其他三组表现出较少的肿瘤负荷,并且基本保持一个良好的肺组织形态(图8E)。治疗结束后,我们检测了各处理组小鼠淋巴结中不同亚型巨噬细胞的比例,结果如图8D所示:与NS组(*p<0.05)、DP7-C组(*p<0.05)和DP7-C/siScramble组(***p<0.001)相比,DP7-C/siCD47治疗后淋巴结中M1型巨噬细胞比M2型巨噬细胞的比例明显提高,这表明沉默CD47表达能增强抗肿瘤免疫反应。
为了进一步阐明探究CD47与引起的抗肿瘤机制,即CD47与巨噬细胞之间的关系,在体外对肺组织切片进行不同亚型巨噬细胞的免疫荧光检测。结果如图9A所示,DP7-C/siCD47肺组织切片中检测到明显升高的CD206+M1巨噬细胞和降低的F4/80+M2巨噬细胞荧光强度。此外,TUNEL法检测肺组织切片上结果显示,与其他三组相比,DP7-C/siCD47处理组的肿瘤组织中显示出强烈的凋亡信号(图9B)。
实施例8系统性递送DP7-C/siCD47及DP7-C/siSIRPα复合物的安全性评价实验
在给药治疗期间,我们每天对小鼠的精神状态及进食状况进行观察,小鼠的精神状态及进食状况未出现明显异常。给药治疗结束后,断颈处死小鼠,取心肝脾肾进行病理HE染色,显微镜观察主要脏器是否有病理性变化。与对照组相较,小鼠的心肝脾肺肾无明显病理性变化(图10,11)。结果初步表明,尾静脉注射DP7-C/siCD47及DP7-C/siSIRPα治疗小鼠皮下B16F10黑色素瘤肺转移模型治疗完成后,小鼠体重无异常下降,主要脏器无明显病理性变化,安全性较好。
我们知道吞噬细胞是免疫监测的第一道防线,在“吃我”和“不吃我”信号之间维持平衡,而CD47-SIRPα轴是这维持一平衡状态的关键角色。CD47是属于免疫球蛋白家族,充当“不要吃我”信号。当CD47与吞噬细胞上表达的SIRPα结合时,启动免疫受体酪氨酸抑制基序介导的抑制信号传导,以阻断吞噬细胞的清除。在许多癌症中,肿瘤细胞通过该信号轴来逃避吞噬细胞的免疫监视。相应地,CD47的高表达也与一系列癌症的不良预后相关。因此,CD47-SIRPα信号轴成为肿瘤免疫治疗的重要靶点。本发明提供的两种siRNA可以通过阻断CD47和/或SIRPα以抑制CD47-SIRPα信号轴作用的策略,均能有效发挥抗肿瘤效应,具有很好的应用前景。
Claims (16)
1.抑制CD47-SIRPα信号轴基因表达表达的siRNA,其特征在于,所述siRNA序列为抑制SIRPα的siRNA:
SIRPαsiRNA正义链:5’-CCGUGAACCCUAGUGGAAA-3’;
SIRPαsiRNA反义链:5’-UUUCCACUAGGGUUCACGG-3’。
2.抑制CD47-SIRPα信号轴基因表达的siRNA,其特征在于,所述siRNA序列为抑制CD47的siRNA:
CD47 siRNA正义链:5’-GGAAUGACCUCUUUCACCA-3’;
CD47 siRNA反义链:5’-UGGUGAAAGAGGUCAUUCC-3’。
3.根据权利要求1或2所述的抑制CD47-SIRPα信号轴基因表达的siRNA,其特征在于:所述的抑制CD47基因表达的siRNA和抑制SIRPα基因的siRNA的正义链和反义链的3’端垂悬有dTdT。
4.抑制CD47-SIRPα信号轴基因表达的siRNA组,其特征在于:包括权利要求1和2或者权利要求3所述抑制CD47基因表达的siRNA和抑制SIRPα基因的siRNA。
5.装载siRNA的胶束,其特征在于是由疏水化修饰的多肽装载权利要求1~3任一项所述的抑制CD47-SIRPα信号轴基因表达的siRNA,或者权利要求4所述的抑制CD47-SIRPα信号轴基因表达的siRNA组制备而成;所述多肽的序列为VQWRIRVAVIRK,所述的疏水化修饰为在多肽的氮末端偶联疏水片段。
6.根据权利要求3所述的装载siRNA的纳米粒子,其特征在于所述的多肽VQWRIRVAVIRK碳端酰胺化修饰为VQWRIRVAVIRK-NH2。
7.根据权利要求3所述的装载siRNA的纳米粒子,其特征在于所述疏水片段为甾醇类化合物或饱和直链脂肪酸或PEG或PEG衍生物;其中,所述的甾醇类化合物为胆固醇类化合物或胆酸类化合物;或者所述饱和直链脂肪酸为C6-C20中的至少一种。
8.根据权利要求3所述的装载siRNA的纳米粒子,其特征在于所述的甾醇类化合物为丁二酰化胆固醇、胆酸或去氧胆酸中的至少一种;或者所述的PEG衍生物为1,2-二油酰-SN-甘油-3-磷酰乙醇胺-聚乙二醇、二硬脂酰基磷脂酰乙醇胺-聚乙二醇或二棕榈酰磷酯酰乙醇胺-聚乙二醇中的至少一种。
9.根据权利要求3所述的装载siRNA的纳米粒子,其特征在于所述的多肽的氮端与疏水片段偶联的方式为通过疏水片段上的-CO-OH与多肽上的-NH2酰胺化反应生成;优选的,所述疏水化修饰的多肽结构为:
10.根据权利要求3所述的装载siRNA的纳米粒子,其特征在于所述的多肽结构中的R为:
中的至少一种。
11.根据权利要求3所述的装载siRNA的纳米粒子,其特征在于所述的疏水化修饰多肽和小核酸按质量比3~5︰1作为原料制备而成。
12.根据权利要求3所述的装载siRNA的纳米粒子,其特征在于是由疏水化修饰多肽与siRNA共孵育制得。
13.根据权利要求3所述的装载siRNA的纳米粒子,其特征在于是由所述的疏水化修饰阳离子多肽与所述药物在水中、缓冲液或液态培养基中共孵育5~15min而得;优选的,所述缓冲液为磷酸盐缓冲液、Tris-HCl缓冲液中的至少一种,液体培养基为RPMI 1640、DMEM双无培养基或Opti-MEM培养基中的至少一种。
14.权利要求1或2任一项所述的抑制CD47或SIRPα基因表达的siRNA或权利要求3~11任一项所述的装载siRNA的纳米粒子在制备治疗肿瘤的药物中的应用。
15.根据权利要求12所述的应用,其特征在于所述的肿瘤为黑色素瘤、结肠癌、乳腺癌、肺癌、脑胶质瘤、胰腺癌、骨肉瘤、头颈部肿瘤等中的至少一种。
16.抗肿瘤药物,其特征在于由权利要求1~3任一项所述的抑抑制CD47-SIRPα信号轴基因表达的siRNA表、权利要求4所述的抑制CD47-SIRPα信号轴基因表达的siRNA组,或权利要求5~15任一项所述的装载siRNA的纳米粒子添加药学上可接受的成分制备而成。
