Abstract
本发明提供了一种具备溶瘤免疫功能的黄芪多糖微针组合物,包含黄芪多糖、蝎毒肽D‑BK、八臂氨基聚合物和微针赋形剂。本发明还提供了上述黄芪多糖微针组合物制备而成的微针。本发明还提供了上述具备溶瘤免疫功能的黄芪多糖微针的制备方法。本发明还提供了上述的具备溶瘤免疫功能的黄芪多糖微针在制备治疗肿瘤疾病的药物中的应用。本发明的微针一方面具有良好的生物安全性,另一方面对肿瘤相关免疫细胞具有原位调节作用,同时包载有东亚钳蝎毒液来源的溶瘤肽和甘露糖修饰的八臂氨基PEG聚合物,从而实现溶瘤免疫疗法和肿瘤抗原主动呈递的协同。
Description
技术领域
本发明属于生物医药领域,涉及一种药物组合物,具体来说是具备溶瘤免疫功能的黄芪多糖微针组合物、微针及其制备方法和应用。
背景技术
黑色素瘤和头颈部肿瘤是两种典型的浅表肿瘤,具有高复发和转移的特点。在过去的30年里,手术一直是治疗病人的有效方法。然而,这些方法只能切除原发肿瘤,不能有效应对肿瘤转移灶。同时,手术切除会造成头面部肿瘤处皮肤的不可逆损伤,严重影响患者的生活质量,因此有必要寻找更好的药物治疗策略。基于免疫检查点的肿瘤免疫疗法已在近十年取得了突破性进展,可使晚期黑色素瘤患者五年生存率从低于5%升至30%,82%头颈部肿瘤患者有效缓解持续时间超过6个月,其作为癌症免疫疗法的主要策略,可显著提高患者缓解率,延长患者生存时间。然而,仍只有30%的患者对免疫检查点疗法产生响应,且78%PD-1抑制剂治疗的患者发展为获得性耐药,因此,免疫检查点疗法只能为一些特定的患者提供持久的治疗,且肿瘤内许多免疫抑制因子可引起肿瘤对免疫检查点抑制剂的抵抗,进而疗效欠佳。因此,发展新型高效且能产生强大而持久的免疫响应的治疗方式对治疗浅表型肿瘤具有重要的意义。
溶瘤免疫治疗作为一种新型肿瘤免疫疗法,可一定程度无视瘤内异质性直接杀伤肿瘤细胞,已被证明可诱导免疫原性细胞死亡,在原位产生大量肿瘤抗原,进而激活抗肿瘤免疫响应,也可与临床现有的免疫检查点抑制剂联用,实现协同的抗肿瘤免疫疗效。然而,溶瘤肽暴露的肿瘤抗原仍面临被快速清除或被酶降解的问题,导致抗原无法被有效呈递从而激活免疫应答。此外,肿瘤免疫微环境中的免疫抑制分子会对细胞毒性T细胞的激活产生负面影响,如细胞毒性T细胞失活或调节性T分化,从而使其丧失识别杀伤肿瘤的能力。若能同时克服肿瘤抗原快速清除以及免疫抑制的微环境,通过溶瘤肽局部给药有望实现更加持久强劲的系统性抗肿瘤免疫响应。另外,临床试验中溶瘤肽的瘤内注射方式存在易出血、易感染、愈合慢等问题,需要开发更安全、便捷的给药形式来替代瘤内注射。
发明内容
针对现有技术中的上述技术问题,本发明提供了具备溶瘤免疫功能的黄芪多糖微针组合物、微针及其制备方法和应用,所述的这种具备溶瘤免疫功能的黄芪多糖微针组合物、微针及其制备方法和应用要解决现有技术中的药物免疫治疗的效果不佳的技术问题。
本发明提供了一种具备溶瘤免疫功能的黄芪多糖微针组合物,包含:

其中的氨基酸结构均为D构型氨基酸;D构型氨基酸更为稳定。
进一步的,所述的八臂氨基聚合物为甘露糖修饰的八臂氨基PEG(8P3),甘露糖修饰的八臂氨基PEG(8P3)的合成过程如下描述:
1)将乙酸酐滴入甘露糖和吡啶的混合物中,在0℃下混合过夜,反应结束后,加入无水乙醇与过量的乙酸酐反应,减压浓缩样品,将化合物倒入冰水中,过滤得到乙酰化甘露糖M2;
2)将叠氮乙醇和M2溶解在二氯甲烷中,于0℃和氮气保护下,逐滴加入三氟化硼二乙醚,温度缓慢升至室温,搅拌过夜。反应结束后,二氯甲烷稀释样品,然后用饱和碳酸氢钠溶液洗涤,然后柱层析纯化,得到固体M3;
3)M3溶于甲醇中,加入甲醇钠与之反应,采用离子交换树脂将pH调节为中性,通过抽滤得到粗产品,柱层析纯化得到M4;
4)将8臂PEG-NH2 8P溶解于水中与(1R,8S,9s)-双环[6.1.0]壬-4-乙炔-9-甲基-丁二酰亚胺碳酸酯的DMSO溶液混合,室温过夜,超滤得到甘露糖修饰8臂PEG-NH2
8P2;
5)将冻干后的8P2溶于水中,在室温下与过量M4的DMSO溶液混合,过夜反应,然后通过超滤和冷冻干燥得到甘露糖修饰的八臂氨基PEG。
进一步的,所述甘露糖修饰的八臂氨基PEG(8P3)能吸附肿瘤抗原以及损伤相关分子模式,并主动靶向引流淋巴结中抗原提呈细胞上的甘露糖受体,促进肿瘤抗原的呈递。
本发明还提供了一种具备溶瘤免疫功能的黄芪多糖微针,由上述的具备溶瘤免疫功能的黄芪多糖微针组合物的原料制备得到。
进一步的,所述微针包含基底和位于基底上的针体;其中,所述针体部分由上述的具备溶瘤免疫功能的黄芪多糖微针组合物的原料制备得到。
进一步的,所述微针为一体式微针;微针基底由透明质酸组成。
本发明还提供了上述的具备溶瘤免疫功能的黄芪多糖微针的制备方法,包括如下步骤:
1)分别配制蝎毒肽D-BK的水溶液、海藻酸钠的水溶液、含黄芪多糖与八臂氨基聚合物和微针赋性剂的水溶液;蝎毒肽D-BK的水溶液的浓度为5mg/mL-20mg/mL、海藻酸钠的水溶液的浓度为5mg/mL-20mg/mL、含黄芪多糖与八臂氨基聚合物和微针赋性剂的水溶液中,每L水溶液中含有黄芪多糖100-300g,八臂氨基聚合物8P3的5-20g,微针赋性剂150-200g;
2)将海藻酸钠的水溶液倒入模具内在4℃下4000转离心5分钟,平转180°后重复;去除微孔外多余物质后,在室温下4000转离心30分钟,平转180°后重复,将其放置在硅胶干燥器中干燥12h;
3)在模具上覆盖蝎毒肽D-BK的水溶液,在4℃下4000转离心2分钟,平转180°后重复,去除微孔外多余的D-BK溶液后,在室温下4000转离心30分钟,平转180°后重复,将其放置在硅胶干燥器中干燥12h;
4)将含黄芪多糖与八臂氨基聚合物的微针赋性剂溶液倒在模具上,在4℃下以4000转离心5分钟,平转180°后重复,然后从模具中去除多余的材料,最后,在模具中加入透明质酸的基底溶液,每100ml基底溶液中含有10-30克的透明质酸,在55℃的干燥箱中对模具进行干燥后脱模得到微针。
本发明还提供了上述的具备溶瘤免疫功能的黄芪多糖微针组合物在制备治疗肿瘤疾病的药物中的应用。
本发明还提供了上述的具备溶瘤免疫功能的黄芪多糖微针组合物在制备治疗肿瘤疾病的药物中的应用。
本发明还提供了上述的具备溶瘤免疫功能的黄芪多糖微针组合物与PD-L1抗体联合在制备治疗肿瘤疾病的药物中的应用。
进一步的,所述的甘露糖修饰的八臂氨基PEG(8P3),能吸附肿瘤抗原以及损伤相关分子模式,并主动靶向引流淋巴结中抗原提呈细胞上的甘露糖受体,促进肿瘤抗原的呈递。
具体的,所述的肿瘤为黑色素瘤或者头颈部肿瘤。
本发明的黄芪多糖微针是通过顶端所负载的带负电荷的海藻酸钠将阳离子多肽吸附到针尖附近,首先,刺入皮肤后可在肿瘤部位率先释放多肽,快速裂解肿瘤细胞,释放大量肿瘤抗原,激活免疫。随后,微针快速溶解,其中包载的聚合物快速吸附肿瘤抗原,防止其被快速清除,靶向将抗原呈递给抗原提呈细胞。其次,黄芪多糖的原位释放能够改善肿瘤免疫抑制微环境,激活免疫细胞,促进细胞毒性T细胞对肿瘤细胞的识别与杀伤能力。最后,体内实验表明该类多功能黄芪多糖微针能够有效抑制黑色素瘤荷瘤小鼠的肿瘤生长,具有高效、稳定等优点,因此可用于制备抗黑色素瘤药物。
本发明的黄芪多糖微针中的微针赋形剂聚乙烯醇与聚维酮的组合的作用为:支撑微针成型,使其具备硬度穿刺皮肤。
本发明构建了黄芪多糖微针,装载溶瘤肽D-BK,以及甘露糖修饰的八臂氨基PEG聚合物,通过经皮给药的方式激活抗肿瘤免疫。
本发明和已有技术相比,其技术效果是积极和明显的。
1、本发明构建了可溶性黄芪多糖微针,使微针具有良好的生物安全性,进入肿瘤部位后能快速溶解释放药物,同时对肿瘤相关免疫细胞具有原位调节作用。同时包载有东亚钳蝎毒液来源的溶瘤肽和甘露糖修饰的八臂氨基PEG聚合物,从而实现溶瘤免疫疗法和肿瘤抗原主动呈递的协同。
2、本发明是一种以黄芪多糖为基质材料的微针,作为一种透皮的新型给药模式。黄芪多糖作为中药黄芪中的有效成分,能够作为一种免疫调节剂重塑免疫抑制微环境,并且溶解度较好,能够作为微针的赋形剂之一。
3、本发明的黄芪多糖微针具备好的生物相容性、快速的药物释放效率以及提高患者对给药的顺应性等优点,能避免传统溶瘤免疫治疗瘤内注射所带来的出血、感染等风险,在替代瘤内药物注射方面具有广阔的前景。
4、本发明的黄芪多糖微针能将溶瘤肽装载于微针尖端,率先快速释放诱导免疫原性细胞死亡,针体于3分钟内完全溶解,将装载药物完全释放,且具备刺穿皮肤的能力。
5、本发明的黄芪多糖微针中包载的甘露糖修饰的八臂氨基PEG能吸附肿瘤抗原以及损伤相关分子模式,并主动靶向引流淋巴结中抗原提呈细胞上的甘露糖受体。
6、本发明的黄芪多糖微针显著抑制C57小鼠皮下B16F10黑色素瘤的生长,显著增强了溶瘤免疫效果。
7、本发明将通过重塑淋巴结-肿瘤免疫抑制微环境的方式增强溶瘤免疫治疗,从局部诱发全身免疫响应,具有较好的应用前景。
8、本发明通过基于黄芪多糖的微针的给药方式将同时递载溶瘤肽与PEG聚合物,以局部给药的方式实现溶瘤免疫疗法和肿瘤抗原主动呈递的协同。

具体实施方式
以下结合具体实施例,对本发明作进一步说明。应理解,以下实施例仅用于说明本发明而非用于限定本发明的范围。
实施例1本发明提供的蝎毒肽D-BK的结构如下:
其中的氨基酸结构均为D构型氨基酸;上述的结构的化合物能够通过Fmoc固相合成法合成。上述的Fmoc固相合成法为现有技术,在此不再赘述,图1为本发明黄芪多糖微针组合物中的多肽D-BK的质谱。
实施例2本发明具备溶瘤免疫功能的黄芪多糖微针组合物中的甘露糖修饰的八臂氨基PEG的合成
制备步骤:
将乙酸酐滴入甘露糖(5.4g,1当量)和吡啶(50mL)的混合物中,与之在0℃下混合过夜。采用薄层色谱(TLC)(PE:EA=3:1)和硫酸乙醇比色法对反应进行监测。反应结束后,加入无水乙醇与过量的乙酸酐反应,减压浓缩样品,将化合物倒入冰水中,过滤得到乙酰化甘露糖M2;
将叠氮乙醇(98%)(0.304g,2当量)和M2(1.0g,1当量)溶解在二氯甲烷中,于0℃和氮气保护下,逐滴加入三氟化硼二乙醚(48%)(2.6mL,4当量)。温度缓慢升至室温,搅拌过夜。采用薄层色谱法(石油醚:乙酸乙酯=3:1)和硫酸乙醇比色法对反应进行监测。反应结束后,二氯甲烷稀释样品,用饱和碳酸氢钠溶液洗涤。然后柱层析纯化(石油醚:乙酸乙酯=10:1→5:1),得到固体M3;
M3(614mg,1当量)溶于甲醇中充分搅拌,加入甲醇钠(397mg,5当量)与之反应2h,采用薄层色谱法(二氯甲烷:甲醇=10:1)和硫酸乙醇比色法监测反应。采用离子交换树脂将pH调节为中性,通过抽滤得到粗产品。最后,柱层析纯化得到M4(二氯甲烷:甲醇=15:1);
合成甘露糖修饰8臂PEG-NH2:将8臂PEG-NH2(Mw:10000,1当量)溶解于水中与(1R,8S,9s)-双环[6.1.0]壬-4-乙炔-9-甲基-丁二酰亚胺碳酸酯(2当量)的DMSO溶液混合,室温过夜。超滤得到甘露糖修饰的八臂氨基PEG;
将冻干后的8P2(1当量)溶于水中,在室温下与过量的M4(3当量)在DMSO中混合过夜。最后,通过超滤和冷冻干燥得到8P3。

实施例3本发明具备溶瘤免疫功能的黄芪多糖微针的制备
制备步骤:
首先用海藻酸钠溶液(10mg/mL)在PDMS模具凹槽上,然后在4℃下以4000转离心5分钟2次。去除微孔外多余物质后,在室温下4000转离心2次30分钟,在硅胶干燥器中干燥12小时。
然后在模具上覆盖多肽D-BK溶液,在4℃下4000转离心2分钟。除去多余的多肽D-BK溶液,在室温下以4000转离心两次30分钟,在干燥器中干燥12小时。
配制溶液,每L水溶液中,含有75g PVA(聚乙烯醇)、75g PVP(聚维酮)、200g APS(黄芪多糖)和10g 8P3(甘露糖修饰的八臂氨基PEG)。
将以上溶液倒在模具上,在4℃下以4000转离心5分钟两次,然后从模具中去除多余的材料。最后,在模具中加入透明质酸(HA)的基础溶液,在模具中加入透明质酸的基底溶液,每100ml基底溶液中含有10克的透明质酸,在55℃的干燥箱中对模具进行干燥后脱模得到微针。
具体的,每次离心使模具水平旋转180°,以确保材料完全填充所有微孔。
实施例4本发明具备溶瘤免疫功能的黄芪多糖微针的形态学与机械强度表征
4.1实验方法:将微针置于离子溅射镀膜机中进行表面镀金后,用扫描电镜观察了微针的形貌。将FITC标记的多肽和Cy5标记的聚合物加入到微针内,通过激光共聚焦和荧光显微镜对比了不含海藻酸钠的微针的药物分布,结果显示具备海藻酸钠的微针中多肽能够富集于微针尖端。采用质构分析仪分析其机械强度。当探针(直径为10毫米的不锈钢圆筒)接触试样时(触发力达到0.05N时),以30毫米/分钟的速度向下按压使试样变形40%,然后探针返回起始位置。由此得到力-位移图。将微针插入SD大鼠皮肤,并在不同时间点取出后,用光学显微镜观察微针的形态。

4.2实验结果
采用扫描电镜、荧光显微镜和激光共聚焦观察微针形态。所有的微针针尖都呈四棱锥型,这表明药物的掺入不会影响微针的形态,也不会导致针尖凹陷(图6B)。海藻酸钠是一种可生物降解的材料,常用于微针的制备,其被封装在微针的针尖中,利用其负电性质可以静电吸附并富集针尖中的阳离子多肽D-BK。如果没有海藻酸钠的存在,则无法观察到针尖的吸附效果。用FITC荧光探针对D-BK进行标记,并将其包裹在纳米颗粒中,通过荧光显微镜可以直观地观察到尖端吸附效果。此外,用Cy5探针标记甘露糖修饰的八臂氨基PEG。另外,通过共聚焦Z轴三维重建观察微针中的药物分布,结果显示,8P3在MNs中分布均匀,而D-BK与海藻酸钠一起定位于针尖。据报道,微针可以有效地刺穿皮肤,以每针超过0.1牛的力输送药物。此外,载药微针的机械强度检测结果显示,贴片具有0.188N/针的机械强度,能够满足最小穿皮力(0.1N/针)。将微针按压于大鼠皮肤上,在不同时间点剥离,光学显微镜能观察到微针逐渐溶解在皮肤中,且所有针尖在3分钟内消失,基底材料保留。此外,黄芪多糖微针具有粘性,溶解后有丝状物出现。实施例5本发明具备溶瘤免疫功能的黄芪多糖微针组合物中的甘露糖修饰的八臂氨基PEG的抗原吸附功能验证
5.1实验方法:采用超声裂解法(20w,开10s/关10s,三次,共1min)提取黑色素瘤B16F10细胞和头颈部肿瘤SCC7细胞的肿瘤细胞裂解液。8P3分别与肿瘤细胞裂解液孵育过夜。未吸附的游离蛋白用超滤管(截留分子量100kDa)在4000rpm下超滤3次,每次30min。采用Zetasizer Nano ZS仪器检测了吸附蛋白前后的8P3的zeta电位。利用透射电子显微镜(TEM)鉴定吸附蛋白前后的8P3的形态。将8P3与肿瘤细胞裂解液的混合物超滤后,收集下清通过离心浓缩仪浓缩,将浓缩物在NP40缓冲液中重悬后离心,收集上清液,取200μg蛋白,设置对照组为未经超滤处理的肿瘤细胞裂解液,随后通过液质联用仪LC-MS对处理后的上清液中蛋白进行鉴定。

5.2实验结果
采用透射电镜、Zeta电位检测和LC-MS验证黄芪多糖微针中的甘露糖修饰的八臂氨基PEG的抗原吸附功能。首先,透射电镜显示了8P3的实际尺寸,与肿瘤裂解液共孵育前后的8P3,它们的粒径都在进入肿瘤引流淋巴结的合适范围内(10-100nm)。Zeta电位的变化表明,8P3可以成功地以静电吸附的形式抓取肿瘤抗原。为了进一步鉴定8P3所吸附的肿瘤突变蛋白,新抗原以及损伤相关分子模式DAMPs,我们收集培养液进行蛋白质组学分析,鉴定得到8P3能直接捕获肿瘤细胞死亡时所释放的一系列,从而激活免疫。实施例6本发明具备溶瘤免疫功能的黄芪多糖微针组合物中的甘露糖修饰的八臂氨基PEG进入引流淋巴结靶向树突状细胞的功能验证
6.1实验方法:将骨髓来源树突状细胞BMDC接种于玻璃底培养皿中培养过夜。将FITC标记的8P2和8P3分别作用BMDC细胞1h和2h,采用Hoechst33342(5×10-6M)染色5min,通过GE DeltaVision OMX SR拍照。将细胞接种于12孔板培养过夜,8P2-FITC和8P3-FITC处理后,收集细胞,移入管中400x g离心4min,PBS洗涤后,通过流式细胞术分析细胞。B16F10小鼠瘤内注射Cy7标记的8P3后。分别于2h、4h、8h、72h分离腹股沟淋巴结,进行冷冻切片,DAPI染色后,通过共聚焦激光扫描显微镜(CLSM)检测Cy7的荧光。

6.2实验结果
我们通过高分辨以及流式细胞术分析了8P3对树突状细胞BMDC的靶向作用。通过高分辨拍摄了BMDC对FITC标记的8P2以及8P3的摄取。与8P2相比,8P3所修饰的甘露糖靶头能以受体介导的内吞作用的方式增强其被摄取能力,并具有时间依赖性,流式细胞术展现一致的结果。对瘤内注射Cy7标记的8P3的C57黑色素瘤小鼠在不同时间点腹股沟淋巴结冷冻切片进行荧光成像,显示淋巴结内产生了Cy7荧光,且2h荧光最强,随时间逐渐变弱。表明8P3-Cy7能够进入小鼠腹股沟淋巴结内靶向免疫细胞。
实施例7具备溶瘤免疫功能的黄芪多糖微针的体内抗肿瘤实验
7.1模型建立
雌性C57小鼠,右后背部皮下接种B16F10细胞(5×105),约6天形成肿瘤。
7.2评价黄芪多糖微针的体内抗肿瘤作用
实验分组:①空白组;②空白微针组;③蝎毒肽D-BK微针组;④黄芪多糖APS微针组;⑤甘露糖修饰的八臂氨基PEG聚合物8P3组;⑥APS+8P3联合给药组;⑦D-BK+APS联合给药组;⑧D-BK+8P3联合给药组;⑨D-BK+8P3+APS联合给药组。给药方式:微针瘤内给药的形式,按压于肿瘤部位三分钟使其完全溶解,两天给药一次,给药四次。评价指标为:①以瘤体积2000立方毫米为终点,观察肿瘤生长曲线;②跟踪记录实验过程中小鼠一般健康状况,饮食、饮水和体重变化,比较副作用。
图7为具备溶瘤免疫功能的黄芪多糖微针显著抑制C57小鼠皮下B16F10黑色素瘤的生长且无毒副作用。(A)B16F10黑色素瘤生长曲线,与空白组与空白微针组相比,单独含有APS、8P3和D-BK的微针对肿瘤生长有轻微的抑制作用。虽然APS和8P3对肿瘤细胞没有直接杀伤作用,但APS的抑制作用提示肿瘤微环境的改善与肿瘤生长密切相关。8P3产生的抑瘤作用可能是由于微针产生的机械损伤诱导肿瘤应激从而提供了一部分抗原。8P3使产生的抗原没有被迅速清除,而是被有效内化和提呈。与D-BK联用组可产生明显的肿瘤生长抑制,是由于D-BK能够诱导肿瘤细胞坏死,产生更强的肿瘤抑制率。值得注意的是,三药联合组对肿瘤的抑制作用最强。(B)给药后小鼠体重无明显变化,微针并未对小鼠产生副作用。(C)B16F10黑色素瘤C57小鼠生存率,三药联合组治疗后的小鼠的生存期最长。(n=5)**p<0.01,***p<0.001。
图8为具备溶瘤免疫功能的黄芪多糖微针对免疫细胞产生的影响。(A)微针给药后肿瘤内巨噬细胞M1/M2比例增加。(B)微针给药后肿瘤CD4+以及CD8+T细胞浸润比例增加。(C)微针给药后肿瘤以及淋巴结内树突状细胞成熟比例增加。(D)微针给药后肿瘤内细胞毒性T细胞以及辅助T细胞比例增加,Treg细胞比例减少。(n=3)**p<0.01,***p<0.001。
实施例8具备溶瘤免疫功能的黄芪多糖微针与PD-L1抗体的联合体内抗肿瘤实验
8.1模型建立
雌性C57小鼠,右后背部皮下接种B16F10细胞(5×105),约6天形成肿瘤。
8.2评价黄芪多糖微针与PD-L1抗体的联合治疗体内抗肿瘤作用
实验分组:①空白组;②PD-L1抗体给药组;③D-BK+8P3+APS微针组;④D-BK+8P3+APS微针与PD-L1抗体联用组;评价指标为:①以瘤体积2000立方毫米为终点,观察肿瘤生长曲线;②跟踪记录实验过程中小鼠一般健康状况,饮食、饮水和体重变化,比较副作用;③实验中途分析淋巴结与肿瘤内树突状细胞成熟比例、肿瘤内M1/M2巨噬细胞比例、CD4+以及CD8+T细胞浸润;④实验终点分析CTLs、Th1以及Treg细胞比例。
图9为具备溶瘤免疫功能的黄芪多糖微针与PD-L1抗体联合治疗显著抑制C57小鼠皮下B16F10黑色素瘤的生长且无毒副作用。(A)给药后小鼠体重无明显变化,微针并未对小鼠产生副作用。(B)B16F10黑色素瘤生长曲线,与空白组相比单独抗PD-L1抗体对肿瘤仅产生轻微抑制作用。微针组以及在微针组中,加入抗PD-L1均能在18天内对肿瘤产生强大的生长抑制作用。(C)B16F10黑色素瘤小鼠生存曲线,在微针联合组中,加入抗PD-L1也能有效抑制停药后肿瘤体积的反弹,显著延长小鼠的生存时间(n=7)**p<0.01,***p<0.001。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员,在不脱离本发明方法的前提下,还可以做出若干改进和补充,这些改进和补充也应视为本发明的保护范围。
Claims (11)
1.一种具备溶瘤免疫功能的黄芪多糖微针组合物,其特征在于包含如下重量份的原料:
2.根据权利要求1所述的具备溶瘤免疫功能的黄芪多糖微针组合物,其特征在于,所述黄芪多糖与黄芪多糖微针组合物的质量比为1:10-3:10;所述的微针赋形剂为聚乙烯醇和聚维酮的组合物;所述的八臂氨基聚合物为无修饰的八臂氨基PEG聚合物或甘露糖修饰的八臂氨基PEG聚合物。
3.根据权利要求1所述的具备溶瘤免疫功能的黄芪多糖微针组合物,其特征在于,所述的蝎毒肽D-BK的结构如下:
其中的氨基酸结构均为D构型氨基酸。
4.根据权利要求1所述的具备溶瘤免疫功能的黄芪多糖微针组合物,其特征在于,所述的八臂氨基聚合物为甘露糖修饰的八臂氨基PEG,甘露糖修饰的八臂氨基PEG的制备过程如下所述:
1)将乙酸酐滴入甘露糖和吡啶的混合物中,在0℃下混合过夜,反应结束后,加入无水乙醇与过量的乙酸酐反应,减压浓缩样品,将化合物倒入冰水中,过滤得到乙酰化甘露糖M2;
2)将叠氮乙醇和M2溶解在二氯甲烷中,于0℃和氮气保护下,加入三氟化硼二乙醚,温度升至室温,搅拌过夜,反应结束后,二氯甲烷稀释样品,然后用饱和碳酸氢钠溶液洗涤,然后柱层析纯化,得到固体M3;
3)M3溶于甲醇中,加入甲醇钠与之反应,采用离子交换树脂将pH调节为中性,通过抽滤得到粗产品,柱层析纯化得到M4;
4)将8臂PEG-NH2 8P溶解于水中与(1R,8S,9s)-双环[6.1.0]壬-4-乙炔-9-甲基-丁二酰亚胺碳酸酯的二甲基亚砜DMSO溶液混合,室温过夜,超滤得到甘露糖修饰8臂PEG-NH2 8P2;
5)将冻干后的8P2溶于水中,在室温下与过量M4的DMSO溶液混合,过夜反应,然后通过超滤和冷冻干燥得到甘露糖修饰的八臂氨基PEG。
5.一种具备溶瘤免疫功能的黄芪多糖微针,其特征在于,由权利要求1-4中任一项所述的权利要求1所述的具备溶瘤免疫功能的黄芪多糖微针组合物的原料制备得到。
6.根据权利要求5所述的具备溶瘤免疫功能的黄芪多糖微针,其特征在于,所述微针包含基底和位于基底上的针体;其中,所述针体部分由权利要求1-4中任一项具备溶瘤免疫功能的黄芪多糖微针组合物的原料制备得到。
7.根据权利要求6所述的具备溶瘤免疫功能的黄芪多糖微针,其特征在于,所述微针为一体式微针;微针基底由透明质酸组成。
8.如权利要求5-7任一项所述的具备溶瘤免疫功能的黄芪多糖微针的制备方法,其特征在于,包括如下步骤:
1)分别配制蝎毒肽D-BK的水溶液、海藻酸钠的水溶液、含黄芪多糖与八臂氨基聚合物和微针赋性剂的水溶液;蝎毒肽D-BK的水溶液的浓度为5mg/mL-20mg/mL、海藻酸钠的水溶液的浓度为5mg/mL-20mg/mL、含黄芪多糖与八臂氨基聚合物和微针赋性剂的水溶液中,每L水溶液中含有黄芪多糖100-300g,八臂氨基聚合物8P3的5-20g,微针赋性剂150-200g;
2)将海藻酸钠的水溶液倒入模具内在4℃下4000转离心5分钟,平转180°后重复;去除微孔外多余物质后,在室温下4000转离心30分钟,平转180°后重复,将其放置在硅胶干燥器中干燥12h;
3)在模具上覆盖蝎毒肽D-BK的水溶液,在4℃下4000转离心2分钟,平转180°后重复,去除微孔外多余的D-BK溶液后,在室温下4000转离心30分钟,平转180°后重复,将其放置在硅胶干燥器中干燥12h;
4)将含黄芪多糖与八臂氨基聚合物的微针赋性剂溶液倒在模具上,在4℃下以4000转离心5分钟,平转180°后重复,然后从模具中去除多余的材料,最后,在模具中加入透明质酸的基底溶液,每100ml基底溶液中含有10-30克的透明质酸,在55℃的干燥箱中对模具进行干燥后脱模得到微针。权利要求1-4任一所述的具备溶瘤免疫功能的黄芪多糖微针组合物在制备治疗肿瘤疾病的药物中的应用。
9.权利要求5-7任一所述的具备溶瘤免疫功能的黄芪多糖微针组合物在制备治疗肿瘤疾病的药物中的应用。
10.权利要求1-4任一所述的具备溶瘤免疫功能的黄芪多糖微针组合物与PD-L1抗体联合在制备治疗肿瘤疾病的药物中的应用。
11.根据权利要求9-10任一所述的应用,其特征在于,所述的肿瘤为黑色素瘤或者头颈部肿瘤。
