Description
技术领域
本发明属于生物技术领域,涉及一种检测抗原抗体结合的方法、产品及应用。
背景技术
单链抗体的结构由一个细长的多肽链组成,包括了完整的抗原结合位点。这种简化的结构更方便工程化设计,可以通过基因工程技术定向设计,以适应不同的疾病特征和治疗需求。
在肿瘤治疗方面,单链抗体可设计成特异性结合肿瘤细胞表面抗原的分子。通过携带单链抗体的治疗性细胞,可实现对癌细胞的精准靶向,实现对癌症细胞的高度选择性打击。此外,单链抗体还可以通过与药物或放射性同位素结合,形成靶向性药物,提高治疗效果同时减少对正常组织的损伤。
在生物医学研究和药物开发中,验证单链抗体与其靶标抗原结合的可靠性是至关重要的一步。然而,由于单链抗体的特殊结构和复杂的抗原结合机制,这一验证过程面临着一系列挑战。由于单链抗体与抗原的结合通常涉及多个相互作用位点,准确地判断结合效力变得更加复杂。单链抗体能否有效识别并结合肿瘤表面抗原的胞外部分至关重要,因为这直接关系到其在治疗或诊断中的效果。
为了应对这些挑战,研究人员采用了一系列先进的实验技术和方法。其中包括:表面等离子共振(SPR)技术、生物传感器技术、晶体学等。但以上技术都面临着成本高昂、操作复杂、技术难度高等方面的挑战。因此,有必要开发一种简单快捷且有效的验证单链抗体于其对应膜抗原细胞表面部分结合能力的方法。
发明内容
为了解决现有技术中存在的技术问题,提供了以下技术方案:
本发明提供了一种双载体系统,所述双载体系统包括:
1)第一载体,所述第一载体包含用于表达抗体的第一表达盒,所述第一表达盒含有编码抗体的第一编码序列;和
2)第二载体,所述第二载体包括用于表达第一载体中抗体靶向的抗原片段的第二表达盒,所述第二表达盒含有编码抗原结合片段的第二编码序列。
术语“载体”可以将编码蛋白质的多核苷酸可操作地插入其中以引起所述蛋白质表达的媒介物。载体可用于转化、转导或转染宿主细胞,以使其在宿主细胞内表达携带的遗传元件,载体的实例包括质粒、阳离子聚合物、树状聚合物、脂质体、多肽等非病毒基因载体,以及病毒载体,所述病毒载体包括慢病毒、腺病毒、腺相关病毒、疱疹病毒、痘病毒、杆状病毒、乳头状瘤病毒、乳多空病毒,噬菌粒,粘粒和人工染色体,如酵母人工染色体(YAC)、细菌人工染色体(BAC)或P1衍生的人工染色体(PAC),噬菌体,如λ噬菌体或M13噬菌体。载体可以含有多种用于控制表达的元件,包含启动子序列、转录起始序列、增强子序列、选择元件和报告基因。另外,载体可以含有复制起点。
进一步,所述第一表达盒还含有编码分泌信号肽的编码序列。
进一步,所述第一表达盒还含有编码第一荧光蛋白的编码序列。
术语“分泌信号肽”是指必须横穿过膜以到达其功能性细胞定位处的蛋白质的高疏水氨基酸序列(例如,优选15至60个氨基酸长)。通过与信号识别颗粒结合,这些序列引导初生蛋白-核糖体复合物到达膜,所述蛋白质在翻译过程中插入至所述膜。信号肽通过多种膜(例如,内质网、线粒体、叶绿体、过氧化物酶体)引导对蛋白质的翻译摄取。非膜蛋白质上的前导信号序列最终通过特定肽酶而被移除。在某些具体的实施方案中,所述分泌信号肽为IFN-γ。
进一步,所述第一荧光蛋白包括GFP、mGFP5、EGFP、D2EGFP、EYFP、BFP、CFP、DsRed、DsRed2、DsRed-express、mRFP1、mCherry、Kaede、RFP。
进一步,所述第二表达盒还含有编码第二荧光蛋白的编码序列。
术语“表达盒”是指遗传物质的构建体,其含有编码序列和足够的调节信息以引导编码序列在体内和/或离体的细胞中恰当转录和/或翻译。可以将所述表达盒插入用于靶向期望的宿主细胞的载体中和/或受试者中。在一些实施方案中,术语“表达盒”是指这样的核酸构建体,其包含与调节元件(例如像,启动子和/或终止信号,并且任选地,影响基因转录或翻译的任何其他核酸序列或其他核酸序列的组合)可操作连接的编码蛋白质的基因或功能RNA。

术语“抗原”是指能够作为激活人体免疫反应的抗原物质,一般为膜蛋白,但也可包含其他可引起人体免疫反应的物质。
术语“蛋白质”、“蛋白”和“肽”指氨基酸残基聚合物(氨基酸序列),不意味着分子的特定长度。该术语也指或包括任何多肽的修饰(例如翻译后),例如糖基化、乙酰化、磷酸化等。
如本说明书和权利要求中所用,除非上下文另有明确说明,否则单数形式“一个”、“一种”、“该”和“所述”包括复数指代。例如,术语“抗体”包括多种抗体,包括它们的混合物。如本文所用,术语“包含”或“包括”意在指组合物和方法包括列举的要素,但不排除其他要素。
术语“表达”是指基因产物的产生。如本文所用,“表达”是指多核苷酸转录成mRNA的过程和/或所转录的mRNA随后翻译成肽、多肽或蛋白质的过程。如果多核苷酸来源于基因组DNA,则表达可包括在真核细胞中剪接mRNA。
本发明提供了一种宿主细胞,所述宿主细胞含有前面所述的第一载体或第二载体。
进一步,所述宿主细胞包括大肠杆菌、枯草芽孢杆菌、酵母细胞、昆虫细胞、哺乳动物细胞、或其组合。

术语“NK细胞”是指在非常规免疫中涉及的淋巴细胞亚群。可以借助某些特征和生物学特性,例如表达特异性表面抗原(包括针对人类NK细胞的CD56和/或CD16)、在细胞表面上不存在α/β或γ/δTCR复合物、通过激活特定细胞溶解机构而结合并且杀死不能表达“自我”MHC/HLA抗原的细胞的能力、杀死肿瘤细胞或表达针对NK活化受体的配体的患病细胞的能力、以及释放称为刺激或抑制免疫应答的细胞因子的蛋白分子的能力来鉴定NK细胞。使用本领域熟知的方法,任何这些特征和活性可以用于鉴定NK细胞。术语NK细胞还将涵盖NK细胞的任何亚群。
本发明提供了一种基于前面所述的双载体系统的检测抗原抗体结合的方法,所述方法包括如下步骤:
1)将第一载体转染到宿主细胞中,编码序列表达后收集培养上清液;
2)将第二载体转染到宿主细胞中;
3)将步骤1)中的上清液与步骤2)中的宿主细胞孵育,通过检测第一荧光蛋白与第二荧光蛋白的共定位情况判断抗体是否与其对应抗原片段相结合。
在某些具体的实施方案中,所述对应抗原片段是指对应抗体其制备目的的抗原片段,即抗体与其目的抗原片段在理论上应当结合。
本发明提供了一种组合物,所述组合物包括前面所述的方法中所使用的细胞或试剂。
本发明提供了一种根据前面所述的双载体系统、前面所述的宿主细胞在抗原抗体结合检测中的应用。

获取抗体的编码序列、抗体的抗原结合片段的编码序列、分泌信号肽的编码序列、第一荧光蛋白的编码序列、第二荧光蛋白的编码序列;
制备第一序列,第一序列的具体组成包括:
抗体的编码序列C端连接第一荧光蛋白的编码序列、N端连接分泌信号肽的编码序列,或抗体的编码序列N端连接第一荧光蛋白的编码序列、第一荧光蛋白的编码序列N端连接分泌信号肽的编码序列;
制备第二序列,第二序列的具体组成包括:抗体的抗原结合片段的编码序列的N端或C端连接第二荧光蛋白的编码序列;
将第一序列、第二序列分别组装在载体上,得到所述的双载体系统。

一种装置,所述装置包括:单个或多个处理器,和存储器,所述存储器用于存储单个或多个计算机程序,当所述单个或多个计算机程序被所述单个或多个处理器执行时实施:
操作1,获取抗体的编码序列、抗体的抗原结合片段的编码序列、分泌信号肽的编码序列、第一荧光蛋白的编码序列、第二荧光蛋白的编码序列;
操作2,制备第一序列,第一序列的具体组成包括:
抗体的编码序列C端连接第一荧光蛋白的编码序列、N端连接分泌信号肽的编码序列,或抗体的编码序列N端连接第一荧光蛋白的编码序列、第一荧光蛋白的编码序列N端连接分泌信号肽的编码序列;
制备第二序列,第二序列的具体组成包括:抗体的抗原结合片段的编码序列的N端或C端连接第二荧光蛋白的编码序列;
操作3,将第一序列组装在第一载体上,将第二序列组装在第二载体上;
操作4,将第一载体转染到宿主细胞中,第一序列表达后收集培养上清液;
操作5,将第二载体转染到宿主细胞中;
操作6,将操作4中的上清液与操作5中的宿主细胞孵育,通过检测第一荧光蛋白与第二荧光蛋白的共定位情况输出抗体与其对应抗原片段是否结合的结果。

本发明实施例中所使用的主要仪器耗材和实验试剂的具体信息见表1和表2。

本发明实施例的设计思路如图1所示,由图1可知本发明的设计思路为:分别在scFv的N端融合IFN-γ分泌信号肽,C端融合GFP蛋白。转染HEK293-T细胞,IFN-γ-scFv-GFP表达后将其分泌到培养上清中;将抗原胞内端融合RFP蛋白,命名为antigen-RFP。转染HEK293-T细胞,antigen-RFP表达后定位于细胞膜上;将上清液与antigen-RFP的转染细胞进行孵育,通过检测GFP与RFP的共定位情况判断scFv是否可以与其对应抗原相结合。
如本文所用,术语“融合蛋白”和语法等同物是指包含衍生自至少两种单独的蛋白质的氨基酸序列的蛋白质。至少两个单独的蛋白质的氨基酸序列可以通过肽键直接连接;或者可以通过氨基酸接头可操作地连接。因此,术语融合蛋白涵盖其中例如蛋白A的氨基酸序列通过肽键直接连接至蛋白B的氨基酸序列(蛋白A-蛋白B)的实施方案,以及其中例如蛋白A的氨基酸序列通过氨基酸接头可操作地连接至蛋白B的氨基酸序列(蛋白A-接头-蛋白B)的实施方案。
实施例1分泌性scFv融合GFP的表达
1、实验材料
①293T细胞
②PEI转染试剂
③DMEM培养基
④胎牛血清
⑤GFP ELISA试剂盒
2、实验方法(以Her-2靶点为例)
1)分泌型scFv融合GFP的结构设计
将IFN-γ分泌信号肽连接于单链抗体(scFv)N端,同时将GFP连接于scFv C端(如图2)。通过基因合成手段将目的片段(具体片段组合形式为IFN-γ-scFv-GFP)合成再构建至plenti慢病毒表达载体上。
2)转染293T细胞
1.提前一天将约7.5×106个293T细胞铺至10cm培养皿中,置于5% CO2的37℃温箱中孵育约16h~24h;
2.转染当天,提前1h更换无血清高糖DMEM培养基:弃上清,加入5mL 37℃预热好的DMEM完全培养基;
3.制备PEI-DNA混合物(即PEI转染试剂与IFN-γ-scFv-GFP片段的plenti慢病毒表达载体的混合物):准备2只1.5ml EP管,分别加入250μL无血清DMEM。其中1只EP管中加入10μg IFN-γ-scFv-GFP片段的plenti慢病毒表达载体并混匀。另一只EP管中加入40μl PEI溶液,混匀后加入前一只EP管中,室温孵育20分钟;
4.孵育结束后,均匀滴入10cm培养皿中,“8”字混匀;置于5% CO2的37℃温箱中孵育6h;
5.孵育结束后更换DMEM完全培养基,继续培养24小时。
3)融合GFP的分泌型scFv(IFN-γ-scFv-GFP)表达鉴定
1.荧光显微镜观察:荧光显微镜下观察GFP是否表达;
2.ELISA检测:
(1)在对应孔中加入50μL样品以及标准品;
(2)向每个孔中加入50μL的抗体混合物;
(3)密封板子,在室温下的板子摇床上孵育1小时,转速设置为400rpm;
(4)用350μL的1×洗涤缓冲液洗涤每个孔。倒出液体,然后向每个孔中加入350μL的1×洗涤缓冲液PT进行洗涤。重复此过程三次。在最后一次洗涤后,倒转板子并将其抹在干净的纸巾上,以去除多余的液体;
(5)向每个孔中加入100μL的TMB显色液,并在400rpm的板子摇床上黑暗中孵育10分钟;
(6)向每个孔中加入100μL的停止液。在摇床上摇动1分钟混合;
(7)利用吸光光度计,读取450nm下各孔的OD值。
3、实验结果
1)转染表达融合GFP的分泌型scFv(即IFN-γ-scFv-GFP)之后,通过荧光显微镜观察GFP是否成功表达。结果显示转染24小时后GFP便开始表达(图3)。
2)采用GFP ELISA检测试剂盒对上清中融合GFP的分泌型Her-2scFv进行检测。标准曲线见图4。通过计算,表达融合GFP的分泌型Her-2scFv的293T上清中,GFP的含量为554.73pg/mL。证明融合GFP的分泌型Her-2scFv可以正常表达,并可以成功分泌到上清中。
实施例2膜表面抗原融合RFP的表达(以Her-2为例)
1、实验材料
①293T细胞
②PEI转染试剂
③DMEM培养基
2、实验方法
1)胞内融合RFP的膜蛋白抗原的结构设计
将RFP蛋白融合于膜蛋白抗原的胞内端(如图5)。通过基因合成手段将目的片段(antigen-RFP)合成再构建至Plenti慢病毒表达载体上。
2)转染293T细胞
1.提前一天将约1×106个293T细胞铺至6孔板中,置于5% CO2的37℃温箱中孵育约16h~24h;
2.转染当天,提前1h更换无血清高糖DMEM培养基:弃上清,加入1mL 37℃预热好的DMEM完全培养基;
3.制备PEI-DNA混合物(即PEI转染试剂与antigen-RFP片段的plenti慢病毒表达载体的混合物):准备2只1.5ml EP管,分别加入50μL无血清DMEM。其中1只EP管中加入2μg质粒(即antigen-RFP片段的plenti慢病毒表达载体)并混匀。另一只EP管中加入8μl PEI溶液,混匀后加入装有质粒的EP管中,室温孵育20分钟;
4.孵育结束后,均匀滴入6孔板中,8字混匀;置于5% CO2的37℃温箱中孵育6h;
5.孵育结束后更换DMEM完全培养基,继续培养24小时。
3)融合RFP的膜蛋白抗原(antigen-RFP)的表达鉴定
共聚焦荧光显微镜观察RFP是否可以表达在细胞膜上。
3、实验结果
共聚焦结果显示,RFP可以表达于细胞膜上。证明RFP和Her-2融合后可以正常表达,而且Her-2可以将RFP带到细胞膜上(图6)。
实施例3scFv与相应抗原的结合验证(以Her-2为例)
1、实验材料
①PBS
②多聚甲醛
③BSA
2、实验方法
①六孔板中转染RFP融合的Her-2的293T为实验组,野生型293T为对照组。弃去上清,并用4℃遇冷的PBS清洗一遍;
②各孔加入1mL 4%的多聚甲醛,室温固定10分钟;
③弃去多聚甲醛,用PBS清洗一遍;
④每孔加入1mL 5% BSA,室温封闭1h;
⑤弃去BSA封闭液,每孔加入1ml表达分泌型融合GFP的Her-2scFv,过夜孵育;
⑥结束孵育后,弃掉上清,PBS清洗三遍后置于共聚焦显微镜下观察。
3、实验结果
实验结果显示,在实验组中GFP可以与RFP重合且定位于细胞上,而在对照组细胞膜上没有GFP的附着(图7)。说明融合GFP的Her-2scFv可以特异性的结合Her-2蛋白,本发明的检测方法能够快速有效地检测到单链抗体与抗原的结合情况。
本方法中分泌性抗体表达24小时,同时抗原表达24小时,抗原抗体结合验证24小时。总共用时48小时。与现有常用方法相比,省去了抗体纯化的时间和费用。
实施例4其他scFv与相应抗原的结合验证
1、实验材料
与实施例3相同
2、实验方法
将实施例3中的实验方法中转染RFP融合的Her-2的293T实验组变为:转染RFP融合的IL13Ra2、转染RFP融合的Trop2、转染RFP融合的MSLN;将实施例3中的实验方法中分泌型融合GFP的Her-2scFv变为:分泌型融合GFP的IL13Ra2 scFv、分泌型融合GFP的Trop2 scFv、分泌型融合GFP的MSLN scFv。其余步骤不变。
3、实验结果
实验结果显示,在实验组中GFP可以与RFP重合且定位于细胞上,而在对照组细胞膜上没有GFP的附着,结果如图8、图9、图10所示。说明融合GFP的IL13 Ra2 scFv可以特异性结合IL13 Ra2蛋白、融合GFP的Trop2 scFv可以特异性结合Trop2蛋白、融合GFP的MSLNscFv可以特异性结合MSLN蛋白,充分说明了本发明的检测方法能够快速有效地检测到单链抗体与抗原的结合情况。
本发明所使用的蛋白其编码序列如表3所示。

Claims (10)
1.一种双载体系统,其特征在于,所述双载体系统包括:
1)第一载体,所述第一载体包含用于表达抗体的第一表达盒,所述第一表达盒含有编码抗体的第一编码序列;和
2)第二载体,所述第二载体包括用于表达第一载体中抗体靶向的抗原片段的第二表达盒,所述第二表达盒含有编码抗原片段的第二编码序列。
2.根据权利要求1所述的双载体系统,其特征在于,所述第一表达盒还含有编码分泌信号肽的编码序列;
优选地,所述第一表达盒还含有编码第一荧光蛋白的编码序列;
优选地,所述第一荧光蛋白包括GFP、mGFP5、EGFP、D2EGFP、EYFP、BFP、CFP、DsRed、DsRed2、DsRed-express、mRFP1、mCherry、Kaede、RFP;
优选地,所述第二表达盒还含有编码第二荧光蛋白的编码序列;
优选地,所述第二荧光蛋白包括GFP、mGFP5、EGFP、D2EGFP、EYFP、BFP、CFP、DsRed、DsRed2、DsRed-express、mRFP1、mCherry、Kaede、RFP;
优选地,所述第一荧光蛋白与第二荧光蛋白荧光颜色不相同;
优选地,所述抗原片段为膜蛋白抗原片段;
优选地,所述抗体的N端连接分泌信号肽、所述抗体的C端连接第一荧光蛋白,或所述抗体的N端连接第一荧光蛋白、第一荧光蛋白的N端连接分泌信号肽;
优选地,所述抗原片段的N端或C端连接第二荧光蛋白;
优选地,所述抗体包括单克隆抗体、多克隆抗体、线性抗体、单链抗体、多特异性抗体、纳米抗体、结构域抗体、抗体片段。
3.一种宿主细胞,其特征在于,所述宿主细胞含有权利要求1或2所述的第一载体或第二载体;
优选地,所述宿主细胞包括大肠杆菌、枯草芽孢杆菌、酵母细胞、昆虫细胞、哺乳动物细胞、或其组合;
优选地,所述哺乳动物细胞包括T细胞、B细胞、NK细胞;
优选地,所述哺乳动物细胞为T细胞。
4.一种基于权利要求1或2所述的双载体系统的检测抗原抗体结合的方法,其特征在于,所述方法包括如下步骤:
1)将第一载体转染到宿主细胞中,编码序列表达后收集培养上清液;
2)将第二载体转染到宿主细胞中;
3)将步骤1)中的上清液与步骤2)中的宿主细胞孵育,通过检测第一荧光蛋白与第二荧光蛋白的共定位情况判断抗体是否与其对应抗原片段相结合。
5.一种组合物,其特征在于,所述组合物包括权利要求4所述的方法中所使用的细胞或试剂。
6.一种根据权利要求1或2所述的双载体系统、权利要求3所述的宿主细胞在抗原抗体结合检测中的应用。
7.一种权利要求1或2所述的双载体系统的构建方法,其特征在于,所述构建方法包括:
获取抗体的编码序列、抗体的抗原结合片段的编码序列、分泌信号肽的编码序列、第一荧光蛋白的编码序列、第二荧光蛋白的编码序列;
制备第一序列,第一序列的具体组成包括:
抗体的编码序列C端连接第一荧光蛋白的编码序列、N端连接分泌信号肽的编码序列,或抗体的编码序列N端连接第一荧光蛋白的编码序列、第一荧光蛋白的编码序列N端连接分泌信号肽的编码序列;
制备第二序列,第二序列的具体组成包括:抗体的抗原结合片段的编码序列的N端或C端连接第二荧光蛋白的编码序列;
将第一序列、第二序列分别组装在载体上,得到所述的双载体系统。
8.权利要求1或2所述的双载体系统、权利要求7所述的构建方法在制备细胞产品中的应用。
9.权利要求1或2所述的双载体系统、权利要求4所述的抗原抗体结合检测的方法、权利要求7所述的构建方法在计算机模型或装置中的应用。
10.一种装置,其特征在于,所述装置包括:单个或多个处理器,和存储器,所述存储器用于存储单个或多个计算机程序,当所述单个或多个计算机程序被所述单个或多个处理器执行时实施:

操作4,将第一载体转染到宿主细胞中,第一序列表达后收集培养上清液;
操作5,将第二载体转染到宿主细胞中;
操作6,将操作4中的上清液与操作5中的宿主细胞孵育,通过检测第一荧光蛋白与第二荧光蛋白的共定位情况输出抗体与其对应抗原片段是否结合的结果。
