技术领域
本发明涉及基因治疗领域。具体而言,本发明涉及一种分离的核酸分子、重组病毒或其应用。更具体而言,本发明的核酸分子、重组病毒可用于治疗或预防与VEGF相关的眼部疾病,特别是湿性年龄相关性黄斑变性。
背景技术
年龄相关黄斑变性(age-related macular degeneration,AMD)是一种中央视网膜的进行性慢性疾病,主要影响视网膜黄斑区域,多发于60岁以上老年人,是老年人不可逆的致盲的主要原因之一(1)。AMD可分为干性和湿性两种。其中,干性为非渗出型,主要表现为玻璃膜疣生成和视网膜色素上皮增生或者脱失。湿性为新生血管型,主要以脉络膜新生血管形成导致出血和渗出(2)。血管可以将营养物质输送到组织和器官并去除分解代谢产物,因此,新生血管供应或血管生成的发展对很多重要的生理过程起着稳态作用。但不受控制的血管生长会促进多种疾病的发生和发展过程,主要包括肿瘤和眼内血管疾病(2)。现有的研究结果显示,眼部的许多疾病都涉及血管的异常发生,其中视网膜和脉络膜新生血管异常增生是眼部疾病视力丧失的主要原因。湿性年龄相关性黄斑变性(wet-AMD)主要为异常的新生血管在黄斑部的视网膜下生长,称为脉络膜新生血管(CNV),导致黄斑区局部水肿或出血引起黄斑区的隆起和局限性色素上皮脱离,最终导致疤痕形成,损伤视网膜感光细胞从而引起视力丧失。
血管内皮生长因子(VEGF)是一种具有重要促血管生成活性的生长因子,可以促进对内皮细胞的有丝分裂和抗凋亡作用,增加血管通透性,促进细胞迁移等,可以调节生理和病理性血管生成过程(3)。VEGF是多种眼病中异常的血管从脉络膜生长到神经视网膜这个过程发生的主要中介因子,在wAMD患者脉络膜新生血管的形成过程中发挥了重要的作用(4)。抑制血管内皮生长因子(VEGF)的活性是治疗新生血管性眼病的标准疗法。
目前比较成熟的治疗方案除了使用光动力疗法(PDT)和激光光凝术等手术疗法以外,还主要包括抗体和重组蛋白类药物治疗。
光动力疗法属于光化学疗法的一种,是用激光照射被化学物质染色的生物组织,产生光化学效应损伤靶组织从而达到治疗目的。此方法治疗wAMD属于利用低强度激光结合光敏剂对视网膜下黄斑部生长的异常新生血管,和由于黄斑区局部水肿或出血引起黄斑区的隆起和局限性色素上皮脱离导致的疤痕进行手术切除治疗(7)。
但是,对于光动力疗法,有些患者一次治疗后病变可复发,可能需要重复治疗。有的患者每年甚至需要治疗3次以上。许多研究证实光敏剂可以在肿瘤的血管组织中大量潴留,沾染光敏剂的血管内皮细胞膜被破坏,使血管内形成血栓和新生血管闭塞(7)。
抗体和重组蛋白类药物治疗通过阻断VEGF与血管内皮细胞表面的VEGFR结合,抑制结合后细胞信号向下游传导,使用VEGF的单克隆抗体或者重组蛋白药物进行针对性的治疗。
VEGF为血管内皮生长因子,是导致wAMD等的多种眼病中异常的血管从脉络膜生长到神经视网膜这个过程发生的主要中介因子。其可以与细胞表面的VEGFR结合,激活血管再生的下游通路。利用竞争性蛋白类药物抑制VEGF的活性,成为wAMD治疗的重要手段。已经用于临床的此类药物包括:阿柏西普、雷珠单抗、贝伐珠单抗、康柏西普等(5)。其中雷珠单抗、贝伐珠单抗是VEGF的单克隆抗体,阿柏西普和康柏西普是VEGFR的结构类似重组蛋白,由VEGF受体1和2胞外结构域和IgG的Fc部分组成(8)。两者都是通过与游离的VEGF特异性结合,阻碍VEGF与细胞表面的VEGFR结合,通过降低结合效率来抑制VEGFR对促血管生成信号向细胞内部的传递(9)。研究证实由于抗体类和重组蛋白类药物的使用,VEGF异常表达导致的眼科疾病的致盲率有所降低。
wAMD的蛋白类药物,药物结构主要有两个设计方向。其一为VEGF的单克隆抗体,可以特异性与VEGF分子结合。其二为VEGF的受体类似蛋白,主要设计结构为VEGFR1和VEGFR2的与VEGF结合的结构域的组合。
但抗体药和重组蛋白药物由于其游离蛋白的性质,导致药物在体内的半衰期较短,需要患者1-2个月就要进行一次玻璃体内注射(例如老年性黄斑变性患者需要以平均每1-3个月一针的频率进行玻璃体内注射),这无疑对于患者和医护人员都增加了负担,并且还会带来了注射导致的眼内炎症、视网膜脱离和眼出血的风险(6)。并且必须长期坚持用药,确诊2年不用药发展为失明的几率将极大提高。一年内多次的双眼玻璃体内常年持续注射对于病人的依从性和医生都带来比较大的负担。
除了上述两种方法,目前也正在研究开发基因药物治疗。AAV具有独特的特性(例如非致病性)并且具有持久的转基因表达,这使其成为极具吸引力的治疗性基因递送载体。
AAV病毒载体基因治疗的原理是将具有治疗作用的药物结构基因插入到病毒载体的基因组中,药物结构基因随AAV病毒感染被释放到患者体内,并转录表达药物结构蛋白。由于AAV病毒的免疫源性低,不会对宿主致病,可以免受宿主免疫系统攻击,病毒载体中携带的药物结构基因可以在动物体内长时间高效表达药物蛋白,从而达到一针注射就长时间有效的目的。
例如,美国诺华公司开发的治疗脊髓性肌萎缩症(SMA)的药物zolgensma就是利用AAV病毒为递送载体,携带并在体内表达SMN基因,补充体内缺乏的相关蛋白,一次注射目前已经有7.5年有效的相关数据。与渤健的蛋白药诺西那生钠注射液(Spinraza)一年多次注射相比,有更多的优势。
wAMD的基因治疗药物,携带并表达的药物结构基因是表达的药物蛋白的基因基础,也是基因治疗药物的药效的核心内容。有必要对药物结构基因进行进一步改进。
发明内容
本发明的目的在于提供一种改进的药物结构基因,并将其包装到AAV病毒的衣壳中,其表达产物是VEGFR类似物,可以对视网膜处异常新生血管的生长进行抑制。用药时,随着AAV病毒被注射到患者眼内,AAV感染血管细胞后可以表达药物结构蛋白,药物结构蛋白在信号肽的作用下被分泌到细胞外。这种药物结构蛋白,可以与血管内皮细胞表面表达的VEGFR竞争性结合游离的VEGF,从而抑制游离VEGF与细胞表面的VEGFR结合,进而抑制细胞增殖信号的传导,达到抑制细胞增殖的作用。

在一个实施方式中,所述病毒是腺病毒相关病毒(AAV),例如,所述AAV病毒选自以下任一血清型:AAV1、AAV2、AAV3、AAV4、AAV5、AAV6、AAV7、AAV8、AAV9、AAV10或AAV11。
在一个实施方式中,所述病毒是AAV8。
在又一方面,本发明提供上述核酸分子或上述重组病毒在制备用于治疗或预防与VEGF相关的眼部疾病的药物中的应用。
在一个实施方式中,所述与VEGF相关的眼部疾病是眼内新生血管生成的眼部疾病。
在一个实施方式中,所述与VEGF相关的眼部疾病是年龄相关黄斑变性,优选湿性年龄相关性黄斑变性。
在又一方面,本发明提供一种治疗与VEGF相关的眼部疾病的方法,所述方法包括向受试者施用治疗有效量的上述核酸分子或上述重组病毒。所述施用包括玻璃体内注射。
有益效果
本发明能够为与VEGF相关的眼部疾病提供一种长效的药物,注入人体后,目标蛋白表达量更高,亲和性更强,药效能够维持数年乃至终身。

具体实施方式
为了对本发明的技术特征、目的和有益效果有更加清楚的理解,现对本发明的技术方案进行以下详细说明,但不能理解为对本发明的可实施范围的限定。
本发明大体上涉及两个对wAMD进行基因治疗的核酸序列与AAV病毒,这两个核酸序列分别称为DS032和DS035,编码VEGFR类似物。两个AAV病毒分别为AAV-AMDVR032和AAV-AMDVR035。
在本研究中,相比蛋白药阿柏西普,对药物结构序列进行了升级,在DS032序列中增加了VEGFR1的D1区,以增加目标蛋白的表达量,增加了VEGFR2的D4区,以形成更加稳定的二聚体结构。DS035序列中比阿柏西普增加了VEGFR1的D1区,将阿柏西普的VEGRF2的D3区更改为VEGFR1的D3区,使其与VEGF结合的区域与天然蛋白一致,增加效应蛋白的稳定性和生物活性。将药物结构序列设置在AAV病毒表达盒中,提高了药物表达梁,增加了药物稳定性和时效的期限,从而提高药物的依从性和预期效果。

本发明提供一种分离的核酸分子,所述核酸分子包含SEQ ID NO:1-4中任一项所示的核苷酸序列,或者编码包含SEQ ID NO:5-8中任一项所示的氨基酸序列。
该核酸分子可作为VEGFR类似物编码序列,编码对wAMD进行基因治疗的药物结构。药物结构包装到载体病毒,特别是AAV病毒中。用药时,随着AAV病毒被注射到患者眼内,AAV感染血管细胞后可以表达药物结构蛋白,药物结构蛋白在信号肽的作用下被分泌到细胞外。药物结构蛋白可以与血管内皮细胞表面表达的VEGFR竞争性结合游离的VEGF,从而抑制游离VEGF与细胞表面的VEGFR结合,进而抑制细胞增殖信号的传导,达到抑制细胞增殖的作用。
AAV血清型包括AAV1、AAV2、AAV2.5、AAV3、AAV4、AAV5、AAV6、AAV7、AAV8、AAV9、AAV10、AAV11、AAV12、rh10、AAV-DJ及其任何杂合或嵌合AAV。在一些实施方式中,所使用的血清型是基于病毒的趋性,或对感兴趣的靶细胞的感染性。在一个实施方式中,所述病毒是AAV8。
在一些实施方式中,AAV用于通过玻璃体内或视网膜下注射将核酸序列递送至受试者的眼或视网膜细胞中。在一些实施方式中,AAV用于将核酸序列递送至受试者的视网膜细胞中。
与蛋白注射相比,基因疗法的一个优点是基因疗法提供了延长的或持续的治疗剂(例如,本发明的DS032和DS035蛋白)释放,以及在一些实施方式中,不需要多次或重复注射。这种延长的或持续的释放是编码DS032和DS035蛋白的核酸序列的递送所造成,所述核酸序列在体内表达以提供治疗作用。在一些实施方式中,由递送到视网膜细胞中的异源核酸对蛋白的表达可以持续至少1年,超过1年,至少2、3、4、5、10年或更多年,乃至终生。
在一些实施方式中,AAV可以包含衣壳变体蛋白,其增加其对眼中的靶细胞或靶组织(例如,视网膜细胞)的感染性,允许更有效递送编码治疗性转基因的核酸序列至靶细胞或靶组织,在那里治疗性转基因可以表达一段时间(例如,至少1、1.5、2、3、4、5、10年或更多年)。如本申请所公开的基因疗法可以靶向感兴趣的特定组织或细胞类型(例如,感光细胞),这可以有助于最小化脱靶效应,或提供治疗性药物结构的更高靶向性的体内递送。
通过基因疗法在体内延长或持续递送DS032和DS035基因序列,与目前采用的标准疗法(例如,蛋白注射剂或基于非基因疗法的治疗)相比,能够在一段时间内以更少剂量施用药物。
在又一方面,本发明提供上述核酸分子或上述重组病毒在制备用于治疗或预防与VEGF相关的眼部疾病的药物中的应用。在一些实施方式中,所述疾病选自:眼部新生血管疾病,包括年龄相关的黄斑变性(AMD)、湿性AMD、干性AMD、视网膜新生血管形成、脉络膜新生血管形成、糖尿病视网膜病变、增殖性糖尿病视网膜病变、视网膜静脉阻塞、视网膜中央静脉阻塞、视网膜分支静脉阻塞、糖尿病性黄斑水肿、糖尿病性视网膜缺血、缺血性视网膜病变和糖尿病性视网膜水肿及其任何组合。在一个实施方式中,所述与VEGF相关的眼部疾病是年龄相关黄斑变性,优选湿性年龄相关性黄斑变性。
在一个实施方式中,药物除了包含上述核酸分子或上述重组病毒之外,还包括药学上可接受的赋形剂。在一些情况下,所述赋形剂包括表面活性剂或稳定剂。在一些情况下,所述表面活性剂选自聚山梨醇酯、十二烷基硫酸钠、月桂基硫酸钠、月桂基二甲基氧化胺、聚乙氧基化醇、聚氧乙烯山梨醇酐、辛苯酚聚醚(octoxynol)、Brij、普朗尼克(pluronic)和聚氧乙烯蓖麻油(polyoxyl caster oil)。在一些情况下,所述药学上可接受的赋形剂包括苯酚、甘露醇、山梨醇或氯化钠。
实施例
本发明提供的技术方案大体上如下所述。
1.设计两个wAMD基因治疗药物的主要药物结构。
2.对药物结构基因序列进行密码子优化,并构建到用于AAV病毒包装的质粒pGOI-AMDVR表达盒中。
3.将质粒pGOI-AMDVR与质粒pHelper和质粒pAAV8-CapRep共转染HEK293细胞,包装病毒AAV-AMDVR。
4.利用ELISA法检测,携带药物结构的AAV病毒感染人脐静脉内皮细胞后,药物结构蛋白的表达量。
5.利用表面等离子共振(Surface plasmon resonance,SPR)技术检测药物结构蛋白与VEGF的亲和力。
6.利用western blot的方法在蛋白水平检测表达药物结构的AAV病毒感染后对人脐静脉内皮细胞增殖相关基因的调控改变。
7.利用CCK8法在细胞水平鉴定表达药物结构蛋白的AAV病毒感染后,对人脐静脉内皮细胞增殖能力的改变。
以下对各步骤进行详细描述。
实施例1.设计wAMD基因治疗药物的主要药物结构。
本研究中,共设计了wAMD基因治疗药物的药物结构序列有两个,药物结构1(DS032)和药物结构2(DS035)。主要设计的理念是利用VEGFR1和VEGFR2蛋白的不同结构域进行多个组合,同时与IgG的Fc段的组合。
实施例2.对药物结构基因序列进行密码子优化,并构建到用于AAV病毒包装的质粒pGOI-AMDVR中。为增加药物结构蛋白基因在未来目标细胞中的表达能力,对基因排列序列进行了密码子优化。
实施例3.将质粒pGOI-AMDVR与质粒pHelper和质粒pAAV8-CapRep共转染HEK293细胞,包装病毒AAV-AMDVR。
1)细胞培养。T75培养瓶中加入3.5×106个细胞,培养基体积为30mL。培养至细胞密度应为90%满),弃掉原细胞培养液,更换为5% FBSDMEM,放置在37℃二氧化碳培养箱中培养1个小时后进行细胞转染。
2)PEI质粒复合物配制。在在3mL opti-DMEM中加入10ug AAV-Helper,10ug AAV-Rep2Cap8,10ug pGOI-AMDVR,充分混匀后加入90uL PEI,震荡混匀后室温静止15min。
3)细胞转染。将PEI质粒复合物全部加入换液后孵育1小时的细胞培养瓶中,轻轻混匀细胞培养基,置于5%CO2,37℃培养箱中继续培养。
4)转染换液。转染24h后进行细胞换液,弃原转染培养基,加入含4uM L-谷氨酰胺和4.5g/L葡萄糖的DMEM 30mL,置于37℃二氧化碳培养箱继续培养。
5)病毒收获液收获。转染换液后的T75培养瓶细胞继续培养72h后进行病毒收获液收获,收获物为细胞培养基上清及细胞沉淀。将病毒收获液置于液氮和37℃水浴锅中反复冻融3次后,使用900rpm 10min离心,离心温度为4℃,吸取上清进行基因组滴度检测。
6)AAV纯化。利用AAV病毒纯化试剂盒(ViraTrap腺相关病毒大量纯化试剂盒(所有血清型)(厂家:Biomiga;货号:BW-V1369)),根据说明书具体要求对AAV病毒进行纯化。
实施例4.利用ELISA法检测,药物结构蛋白的表达量。
(1)原理:AAV-AMDVR病毒感染易感细胞会在细胞内过表达药物结构蛋白,并在信号肽的作用下,分泌到细胞外。利用VEGF蛋白进行包板,将不同浓度的感染后细胞蛋白裂解液与之共孵育,通过ELISA实验对细胞蛋白裂解液中游离的药物结构蛋白进行定量检测,分析目的药物结构蛋白与配体的亲和力和表达的总量,进而作为体外药效指标。
(2)步骤:以500ng/mL VEGF蛋白包被ELISA专用酶标板,每个孔中加入100μL,4℃过夜培养;用PBST(含0.1%吐温的PBS)洗板4次,用含5%BSA的PBST在37℃封闭2小时;用PBST洗板4次后,梯度加入250ng/mL—1.953ng/mL标准品,绘制标准品曲线。加入梯度稀释的细胞蛋白裂解液(原倍/2倍/4倍/8倍/16倍稀释),每个稀释梯度各两个复孔;加入梯度稀释的阿柏西普(0.05pM-400pM)作为阳性对照,每个稀释梯度各两个复孔。37℃孵育2小时后,用PBST洗板4次。加入兔抗人酶标二抗,1:30000稀释,37℃孵育1.5小时;PBST洗4次,加入显色液A,显色液B各50ul,加入50uL终止液,孵育15分钟,在450nm和630nm读取吸光度。
(3)结果:携带两种种药物结构蛋白基因的AAV病毒,感染效应细胞后均可表达药物结构蛋白。表达量情况及表达量结果统计见表1。

实施例5.利用表面等离子共振(Surface plasmon resonance,SPR)技术检测药物结构蛋白与VEGF的亲和力。
(1)原理:Biacore T200是基于表面等离子共振(Surface plasmon resonance,SPR)技术的高通量分子水平的相互作用分析系统。将表达纯化的VEGF分子通过氨基偶联的方法固定在生物传感芯片上,将待测的药物结构蛋白以一定浓度流过芯片表面,若二者之间有相互作用,则芯片表面的折射率发生变化从而导致共振角的改变。通过实时检测共振角的变化,来对药物结构蛋白与VEGF的分子间相互作用的动力学和亲和力信息进行收集,确定两者的亲和能力。
(2)步骤:利用氨基偶联试剂盒中的EDC/NHS活化芯片,用醋酸钠稀释VEGF并将其固定在芯片上,用乙醇胺进行封闭。准备好药物结构蛋白,并分别配制成浓度为85nM,8.5nM,0.85nM的分析物溶液。药物结构蛋白溶液流经芯片,通过通道的响应值判断分析物和芯片表面是否有非特异性结合。阿柏西普溶液作为阳性对照。
(3)结果:SPR法检测发现,两种药物结构蛋白DS032和DS035均可以与靶效应蛋白结合,结果见图1。
实施例6.利用western blot的方法在蛋白水平检测表达药物结构的AAV病毒感染后对人脐静脉内皮细胞增殖相关基因的调控改变。
(1)原理:HUVEC细胞为人脐静脉内皮细胞,是血管内皮的模型细胞,可以组成性表达VEGF因子,并且在细胞表面表达VEGFR。当携带药物结构基因的AAV-AMDVR病毒,感染HUVEC细胞后,在细胞内表达药物结构蛋白AMDVR,并在信号肽的作用下分泌到细胞培养上清培养基中。药物结构蛋白AMDVR,可以与HUVEC细胞表达在细胞外的游离VEGF因子竞争性结合,抑制VEGF-VEGFR的结合以及信号的传导,导致细胞增殖必须的标志蛋白表达下调。利用western blot法检测标志蛋白的表达水平,来判断次标志蛋白表达是否下调,进而判断AAV递送的药物结构基因表达的药物结构蛋白的对血管内皮细胞增殖的抑制效果。
(2)步骤:用HUVEC细胞在6孔板中培养到90%融合,用PBS洗涤细胞3次后,加入新鲜培养基。利用无血清培养基将AAV-AMDVR1-6病毒进行稀释,分别以5×104vg/cell、5×105vg/cell、5×106vg/cell的梯度感染HUVEC细胞,每个稀释度设置4个复孔,放入细胞培养箱中孵育4h,然后更换为新鲜培养基,感染72h后,通过细胞刮刀收集细胞。向收集到的细胞中加入60uL细胞裂解液(1mM PMSF),冰上裂解30分钟,然后在4℃、13000rpm离心10分钟,将上清转移到新的1.5mL的EP管中。向其中加入15uL 5×SDS-PAGE上样缓冲液,沸水煮10分钟。将样品进行SDS-PAGE电泳,电泳完成后,取出胶块,用双蒸水漂洗下层胶,除去残留的电泳液。将胶块上的蛋白条带转移到PVDF膜上,转膜结束后将PVDF膜用双蒸水漂洗,除去残留的电转液,将PVDF膜转移至5%脱脂乳溶液中,室温摇床封闭2小时;分别孵育Cyclin E1、BCL2、PCNA和GAPDH的单克隆抗体,4℃摇床过夜孵育。利用5%的PBST洗涤PVDF膜5次,5分钟/次,然后室温孵育相应的辣根过氧化物酶标记的二抗,孵育时间为1小时。孵育结束后用5%的PBST洗涤PVDF膜5次,5分钟/次。配制增强化学发光法(ECL)显色液,加至PVDF膜,通过扫膜仪成像,计算蛋白条带灰度值,并结合内参确定蛋白表达变化情况。
(3)结果:Western Blot实验结果显示,携带药物结构的AAV病毒感染血管内皮细胞,对细胞增殖效应蛋白有抑制作用;携带药物结构的AAV病毒感染对效应蛋白的抑制作用有量效关系;不同的药物结构对效应蛋白的抑制作用能力有所不同。结果见图2。
实施例7.利用CCK8法鉴定表达药物结构蛋白的AAV病毒感染后,对人脐静脉内皮细胞增殖能力的改变。
(1)原理:Cell Counting Kit-8(简称CCK-8)是一种基于WST-8的广泛应用于细胞增殖和细胞毒性的检测试剂。WST-8,化学名:2-(2-甲氧基-4-硝基苯基)-3-(4-硝基苯基)-5-(2,4-二磺酸苯)-2H-四唑单钠盐,是一种类似于MTT的化合物,它在电子载体1-甲氧基-5-甲基吩嗪鎓硫酸二甲酯(1-Methoxy PMS)存在的情况下,被线粒体中的脱氢酶还原为具有高度水溶性的橙黄色甲瓒产物(formazan)。细胞增殖越多越快,颜色越深;细胞毒性越大,则颜色越浅,对于同样的细胞,颜色的深浅与活细胞的数量成正比,因此可利用这一特性直接进行细胞增殖和毒性分析。通过CCK-8试剂盒结合酶标仪检测AAV-AMDVR感染HUVEC细胞中甲瓒的量,衡量rAAV-FLT1等基因药物对HUVEC细胞增殖的影响。
(2)步骤:在96孔板中加入100uL的HUVEC细胞悬液(约2000个细胞),在37℃、5%CO2细胞培养箱中培养过夜。向细胞孔中分别加入5×104vg/cell、5×105vg/cell、5×106vg/cell的AAV-AMDVR1-6病毒。分别在病毒感染24、48、72小时时,每孔加入10uLCCK-8溶液,在37℃、5%CO2细胞培养箱中继续培养2-4小时,使用酶标仪,检测样品450nm处吸光值。
(3)结果:CCK8实验结果显示,携带不同药物结构基因的AAV病毒,感染效应细胞HUVEC(静脉血管内皮细胞)后,对细胞的增殖具有抑制作用,且存在一定的量效关系。具体结果见图3。
实施例8.小鼠造模与体内表达量与药效研究
(1)原理:采用激光光凝法诱导小鼠脉络膜新生血管(CNV)模型,初步考察供试品单次玻璃体腔注射给予后对小鼠CNV的可能作用。
(2)步骤:32只(16只/性别)眼前节、眼底无明显异常的C57BL/6J小鼠,按体重随机分为4组,每组8只(4只/性别),分别双眼玻璃体腔注射给予溶媒对照品(PBS缓冲液)、市售对照品(40mg/mL)、DS032和DS035给药体积为2μL/眼,以给药当天为D1(/>给药当天为D32)。D29,所有动物双眼眼底激光以诱导CNV。试验期间,所有动物每天进行临床观察;接收时、分组前、D1(给药前)、D8、D15、D22、D29(造模前)、D36及D43称重;分组前、D1(给药前)、D29(造模前)、D36及D43进行一般眼科检查;分组前、D29(造模后即刻,仅眼底照相)、D36及D43进行眼底照相和血管荧光造影检查。
(3)结果:DS032和DS035在小鼠CNV模型实验中,携带DS032的AAV8病毒给药后5周(D36)和给药后6周(D43),2E7 GC/眼及2E9 GC/眼的剂量下,均能降低三级光斑率,改善荧光渗漏,给药后6周2E9 GC/眼的剂量下光斑渗漏平均分显著低于溶媒对照组(p≤0.05),药效更明显,接近(40mg/mL,8μg/眼)。携带DS0352的AAV8病毒给药后5周(D36)和给药后6周(D43),2E7 GC/眼及2E9 GC/眼的剂量下,均能降低三级光斑率,改善荧光渗漏,给药后6周2E7 GC/眼的剂量下光斑渗漏平均分显著低于溶媒对照组(p≤0.05),药效更明显,接近/>(40mg/mL,8μg/眼)。此实验中,在给药36、43天后,两个药物序列在AAV病毒载体包装递送后,均表现出具有与对照药物/>给药3、10天后接近的药效结果,说明具有更加持久的药效。数据见表2和图4。

2.根据权利要求1所述的核酸分子,其特征在于,所述核酸分子还包含SEQ ID NO:9所示的信号肽编号核苷酸序列。
3.根据权利要求1所述的核酸分子,其特征在于,所述核酸分子包含SEQ ID NO:1所示的核苷酸序列。
4.根据权利要求1-3中任一项所述的核酸分子,其特征在于,所述核酸分子编码包含SEQ ID NO:5或7的氨基酸序列。
5.一种分离的核酸分子,其特征在于,所述核酸分子包含SEQ ID NO:4所示的核苷酸序列。
6.根据权利要求5所述的核酸分子,其特征在于,所述核酸分子还包含SEQ ID NO:10所示的信号肽编号核苷酸序列。
7.根据权利要求5所述的核酸分子,其特征在于,所述核酸分子包含SEQ ID NO:2所示的核苷酸序列。
8.根据权利要求5-7中任一项所述的核酸分子,其特征在于,所述核酸分子编码包含SEQ ID NO:6或8的氨基酸序列。
9.一种重组病毒,其特征在于,所述重组病毒包含权利要求1-8中任一项所述的核酸分子。
10.根据权利要求9所述的重组病毒,其特征在于,所述重组病毒还包括以下多核苷酸表达盒内的调控元件的组合:
(1)AAV2型病毒的反向末端重复序列ITR1
(2)CMV启动子
(3)CMV增强子
(4)Kozak序列
(5)VEGFR1信号肽
(6)BGH PloyA序列。
11.根据权利要求10所述的重组病毒,其特征在于,所述病毒是腺病毒相关病毒(AAV),例如,所述AAV病毒选自以下任一血清型:AAV1、AAV2、AAV3、AAV4、AAV5、AAV6、AAV7、AAV8、AAV9、AAV10或AAV11。
12.根据权利要求11所述的重组病毒,其特征在于,所述病毒是AAV8。
13.权利要求1-8中任一项所述的核酸分子或权利要求9-12中任一项所述的重组病毒在制备用于治疗或预防与VEGF相关的眼部疾病的药物中的应用。
14.根据权利要求13所述的应用,其特征在于,所述与VEGF相关的眼部疾病是眼内新生血管生成的眼部疾病。
15.根据权利要求13或14所述的应用,其特征在于,所述与VEGF相关的眼部疾病是年龄相关黄斑变性,优选湿性年龄相关性黄斑变性。
