Abstract
本发明公开了一种生物墨水、3D打印阿尔兹海默症,类脑模型、方法及应用,类脑模型具有上、中、下三层结构,其中,上、下两层结构中所含有的细胞是活化好的小胶质细胞与星型胶质细胞,中间层所含有的细胞是海马体神经元。在体外利用LPS诱导小胶质细胞与星型胶质细胞产生炎症反应,生物墨水的配方包括海藻酸钠、食用明胶、胶原蛋白、壳聚糖,利用生物3D类器官打印技术制备阿尔兹海默症,类脑模型。该类脑模型能够更好的模拟脑内真实发生的情况。同时,通过3D打印技术将该类器官量产化,从而实现对药物、生物活性物质功能的快速筛选,为下一步针对性的动物实验或临床试验提供思路。

技术领域
本发明涉及一种生物墨水、类器官模型及应用,尤其涉及一种生物墨水、3D打印阿尔兹海默症类脑模型、方法及应用。
背景技术
目前,研究新型药物对某种疾病的治疗作用或生物活性物质对于身体器官、组织的保健功能检测主要是两个方面。
第一种,将药物加入到培养的单一细胞培养皿中去检测药物对于该细胞的作用。对于这种方式,由于人体的器官或疾病的产生是多细胞共同的作用,用单一的细胞培养皿中的一种细胞作为检测无法模拟出真实人体中细胞与细胞之间的系统性交流。同时,由于人体的器官与组织是3D形式,而培养皿中的细胞呈现平面2D的形式生长,无法模拟器官内的细胞生长的最真实状态。
第二种,将药物或生物活性物质注射或喂食给动物。对于这种方式的缺点是,试验周期过长,且在摸索新型药物或生物活性物质的时候需要耗费大量的实验动物和金钱才能将该药物或生物活性物质作用在每一个器官上,进而才能最大程度上找到该药物或生物活性物质的功能。
近年来,3D打印技术逐渐兴起,可根据设计的3D模型来构建生物组织或器官的立体结构,这种技术有潜力用于生物医学研究、生物打印定制的组织和器官、以及药物筛选等应用领域。目前,3D打印技术鲜有应用于阿尔兹海默症的研究,同时,市面上大多数的生物墨水具有内毒素,并不能更好的模拟脑内微环境。基于此,阿尔兹海默症类脑模型3D打印材料的选择是亟需解决的问题,并且,如何使打印后的组织具有无需加药即可具有类似阿尔兹海默症自主加剧的作用,以更好的模拟脑内真实发生情况,将会对阿尔兹海默症的相关研究具有重要意义。
发明内容
为了解决上述技术所存在的不足之处,本发明提供了一种生物墨水、3D打印阿尔兹海默症类脑模型、方法及应用。
为了解决以上技术问题,本发明采用的技术方案是:一种3D打印阿尔兹海默症类脑模型,其特征在于:该阿尔兹海默症类脑模型具有上、中、下三层结构,其中,上、下两层结构中所含有的细胞是活化好的小胶质细胞与星型胶质细胞,胶质细胞与星型胶质细胞按1:1混合到生物墨水中,分别形成上、下两层结构;中间层所含有的细胞是海马体神经元,海马体神经元混合到生物墨水中,形成中间层结构。
进一步地,活化好的小胶质细胞、活化好的星型胶质细胞指的是:经LPS诱导并成功产生炎症反应的小胶质细胞、星型胶质细胞。
一种3D打印阿尔兹海默症类脑模型的制备方法,包括以下制备过程:
S1、活化的小胶质细胞、星型胶质细胞获取:
小胶质细胞、星型胶质细胞先从超低温冰箱中取出来复苏至培养瓶中培养24小时;随后将小胶质细胞、星型胶质细胞通过加入LPS诱导成为炎症的小胶质细胞、星型胶质细胞;
S2、海马体神经元获取:
海马体神经元也从超低温冰箱中取出来复苏至培养瓶中培养24小时,不做任何处理;
S3、生物3D打印准备阶段:
分别对上述海马体神经元细胞、以及活化的小胶质细胞、星型胶质细胞进行离心;
将离心获得的小胶质细胞、星型胶质细胞混合到生物墨水中,记为上样一;
将离心获得的海马体神经元混合到生物墨水中,记为上样二;
S4、设置3D打印参数,上机打印:
以上样一为原料打印出下层结构后,再由上样二为原料打印出中间层结构,再由上样一为原料打印出上层结构,制得阿尔兹海默症类脑模型。
进一步地,细胞复苏培养条件为:用含有89%的DMEM、10%的FBS、1%的双抗在二氧化碳恒温培养箱中分别培养24h,双抗为抗青霉素和链霉素,恒温培养环境为37℃,CO2的体积浓度为5%。

一种3D打印阿尔兹海默症类脑模型的应用,包括但不限于阿尔兹海默症、老年失智症病理进展研究,药物检测,其它生物活性物质检测,药物筛查。
本发明公开了一种生物墨水、3D打印阿尔兹海默症类脑模型、方法及应用,通过3D打印类器官技术,在三维层面上模拟出真实的阿尔兹海默症影响海马体的病理进程打印后的组织具有无需加药即可具有类似阿尔兹海默症自主加剧的作用,能够更好的模拟脑内真实发生的情况。同时,通过3D打印技术将该类器官量产化,从而实现对药物、生物活性物质功能的快速筛选,为下一步针对性的动物实验或临床试验提供思路。此外,创新的提成一种新的生物墨水,改善了传统的生物墨水具有内毒素,且不能更好的模拟脑内微环境的现状。

具体实施方式
下面结合附图和具体实施方式对本发明作进一步详细的说明。
已知,阿尔兹海默症影响记忆功能,会使记忆力发生衰退,而记忆功能受海马体支配,是由于海马体中的神经元死亡造成的,死亡原因是:(1)小胶质细胞、星型胶质细胞被异常活化后,加剧了β淀粉样蛋白的累积导致细胞死亡;(2)小胶质细胞、星型胶质细胞被β淀粉样蛋白等其他介导因素活化后产生的神经毒素因子杀死了海马体神经元。因此,本发明的设计思路主要在小胶质细胞、星型胶质细胞这两种细胞上,同时选择海马体神经元作为检测对象。
激活后的细胞释放的炎症因子是导致细胞死亡的重要原因,小胶质细胞和星型胶质细胞的活化可以通过引发炎症反应来加剧阿尔兹海默症。所以,本发明选用在体外活化好的小胶质细胞、星型胶质细胞作为材料,采用生物3D打印技术建立阿尔兹海默症类脑模型,该类脑模型可以学习小胶质细胞与星型胶质细胞是如何进行交流和协作,自主释放出具有神经毒性的炎症因子进而引起类脑模型中海马体神经元(分管记忆功能区域)的死亡。打印后的类脑模型组织具有无需加药即可具有类似阿尔兹海默症自主加剧的作用,可更好的模拟脑内真实发生情况。
如图1所示,为打印获得的真实的阿尔兹海默症类脑模型,该阿尔兹海默症类脑模型具有上、中、下三层结构,其中,上、下两层结构中所含有的细胞是活化好的小胶质细胞与星型胶质细胞的混合,中间层所含有的细胞是海马体神经元。
阿尔兹海默症类脑模型的建立过程具体有:
S1、活化的小胶质细胞、星型胶质细胞获取:
小胶质细胞、星型胶质细胞先从超低温冰箱中取出来复苏至培养瓶中培养24小时。随后将小胶质细胞、星型胶质细胞通过加入LPS(脂多糖)诱导成为炎症的小胶质细胞、星型胶质细胞,这两种细胞将作为模拟脑内被活化的参与免疫的细胞。
S2、海马体神经元获取:
海马体神经元也从超低温冰箱中取出来复苏至培养瓶中培养24小时,不做任何处理,以模拟脑内海马体中的神经细胞。
S3、生物3D打印准备阶段:
分别对上述活化的小胶质细胞、星型胶质细胞以及海马体神经元细胞进行离心;
将离心获得的小胶质细胞、星型胶质细胞按照1:1混合到生物墨水中,最终细胞浓度为1×106/mL,记为上样一;
将离心获得的海马体神经元混合到同样的生物墨水中,最终细胞浓度为1×106/mL,记为上样二。
S4、设置3D打印参数,上机打印:
以上样一为原料打印出下层结构后,再由上样二为原料打印出中间层结构,再由上样一为原料打印出上层结构,制得阿尔兹海默症类脑模型,打印的真实模型如图1所示。
由此,本发明通过生物3D打印类器官技术,在三维层面上模拟出真实的阿尔兹海默症影响海马体的病理过程,自主加剧阿尔兹海默症作用的特性,可以还原脑内真实情况。应用于阿尔兹海默症、老年失智症的病理进展研究及药物检测或其它生物活性物质检测或药物筛查时,研究结果更加真实且有信服力。同时,通过3D打印技术将该类器官量产化,从而实现对药物、生物活性物质功能的快速筛选,为下一步针对性的动物实验或临床试验提供思路。并且,可减少实验动物的使用,从而减少伦理到的层面上的问题。
作为优选地,在类器官生物3D打印中,细胞和生物墨水结合在一起,一层一层地打印可以创建所需的组织结构。细胞在生物墨水中定位,并根据设计的结构进行排列,最终形成具有生物学功能的组织或类器官。
基于大多数的生物墨水具有内毒素,且不能更好的模拟脑内微环境的现状,本发明还独创的开发一款用于构建中枢神经系统的生物墨水。现在市面上用于接种细胞的生物墨水材料有海藻酸钠、食用明胶,是因为它们具有很好的粘性,然而它们的生物相容性非常差,不利于构建敏感的组织或器官。胶原蛋白具有比较差的粘性但具有良好的生物相容性,因此,本发明将三者结合制备出比较良好的生物墨水材料。将海藻酸钠、食用明胶和胶原蛋白作为基础,加入壳聚糖,用于增加生物相容性的同时模拟脑内组织的孔径大小。
生物墨水的配方如下:
海藻酸钠:2-10wt%;
食用明胶:3-15wt%;
胶原蛋白:1-15mg/mL;
壳聚糖:3-14wt%。
下面结合具体的实验数据,对本发明技术方案的可行性做进一步说明:
1、细胞复苏与培养:从超低温冰箱中分别取出海马体神经元(HT-22细胞MouseHippocampal Neuronal Cell Line,购自sigma-aldrich,货号:SCC129),小胶质细胞(BV-2细胞,购自Accegen,货号:ABC-TC212S))和星型胶质细胞(C8-D1A细胞 [Astrocyte type Iclone],购自ATCC,货号:CRL-2541),并分别复苏在培养瓶中,用含有89%的DMEM、10%的FBS、1%的双抗(抗青霉素和链霉素)在二氧化碳恒温(37℃,CO2的体积浓度为5%)培养箱中分别培养24h。
2、小胶质细胞、星型胶质细胞建立炎症模型
在小胶质细胞与星型胶质细胞培养瓶中加入LPS(eBioscience™Lipopolysaccharide (LPS) Solution (500X)购自Thermal Fisher,货号:00-4976-93)使其最终浓度为100ng/mL,继续分别培养24小时,由LPS诱导产生炎症反应。
3、小胶质细胞的炎症标志物分析
小胶质细胞炎症标志物为细胞内NO的水平,以及细胞内TNF-α,IL-6,IL-1β,INOS基因的表达。细胞内NO的水平利用NO试剂盒检测,上述目的基因用qPCR检测。
检测过程为:将无处理组(未经LPS诱导处理的小胶质细胞)和被LPS处理过的试验组的细胞培养上清液分别收集到两个15mL的离心管中。使用无细胞培养上清液作为背景对照,将待测试NO样本和NO检测试剂盒中的试剂混合。将样本混合物放置在室温下静置一段时间后,使用酶标仪测量反应液的吸光值,波长在540-570nm之间。无处理组与LPS处理组的NO浓度如下:

利用qPCR技术检测mRNA表达水平,构建目标基因扩增的引物,分别有:

TNF-α gene: Forward: GGTGCCTATGTCTCAGCCTCTT;Reverse:GCCATAGAACTGATGAGAGGGAG。
根据RNA提取试剂盒(购买厂家:ThermoFisher、型号:15596026、规格:200mL)的步骤提取细胞RNA后,放入逆转录机中将RNA逆转录为cDNA。cDNA随后按照SYBRGreen Ⅰ荧光反应体系的方法将逆转录产物放入 Real time PCR机中进行扩增。mRNA 的表达结果如下:

4、星型胶质细胞的炎症标志物分析
星型胶质细胞炎症标志物为细胞内谷氨酸的水平细胞内活性氧的水平,以及细胞内GFAP,INOS,C3,TNF-α基因的表达。细胞内谷氨酸的水平利用谷氨酸氧化酶检测试剂盒(Invitrogen的AmpLex Red)检测,上述目的基因用qPCR 检测。
检测过程为:将无处理组和被LPS处理过的试验组的细胞培养上清液分别收集到两个15mL的离心管中。根据谷氨酸氧化酶检测试剂盒的说明步骤进行处理。随后使用酶标仪测量反应液的吸光值。无处理组与LPS处理组的谷氨酸浓度如下:

综上,小胶质细胞的炎症标志物、星型胶质细胞的炎症标志物相较于无处理组,均呈倍数增长,表明小胶质细胞、星型胶质细胞均成功产生炎症反应,这些炎症的小胶质细胞与星型胶质细胞将在未来作为制作阿尔兹海默症的类脑模型的材料。
5、用于生物3D打印的生物墨水的配置
用于生物3D打印的生物墨水是一种特殊的生物材料,通常包括细胞和生物聚合物的混合物。在3D打印过程中,生物墨水通过逐层堆叠或沉积的方式,根据设计的3D模型来构建生物组织或器官的立体结构。
生物墨水的配方如下:
海藻酸钠:2-10wt%;
食用明胶:3-15wt%;
胶原蛋白:1-15mg/mL;
壳聚糖:3-14wt%。
对上述海马体神经元细胞以及LPS诱导后的小胶质细胞、星型胶质细胞离心;将离心获得的小胶质细胞、星型胶质细胞按照1:1混合到生物墨水中,最终细胞浓度为1×106/mL,记为上样一;
将离心获得的海马体神经元混合到同样的生物墨水中,最终细胞浓度为1×106/mL,记为上样二。
6、打印结构的设计
阿尔兹海默症类脑模型组由上、中、下三层组成,其中,上层与下层所含有的细胞是已经建立炎症模型的小胶质细胞与星型胶质细胞的混合,中间层所含有的细胞是海马体神经元。这样设计的意义是,由于小胶质细胞与星型胶质细胞相互作用所导致的炎症进一步加深,会影响到功能性神经元的活性。若中间层的细胞在日后逐渐死亡,证明该阿尔兹海默症类脑模型组织建立成功。
7、设置参数,上机打印
打印机型号为:Regenovo Bio-Architect WS, 购买自杭州Regenovo;
将上样一加入到打印机喷头一中,将上样二加入到打印机喷头二中,打印参数的设计如下:
打印喷头的直径:0.3mm;
打印气动压力:0.4MPa;
打印速度:10mm/s;
打印结构长宽高:6×6×6mm。
打印过程中,喷头一在打印下层后会换喷头二打印中间层结构,中间层结构打印结束后会换喷嘴一打印上层结构。打印获得阿尔兹海默症类脑模型的结构如图1所示(俯视图);
8、海马体神经元细胞存活率检测
打印后用10%的CaCl2溶液对阿尔兹海默症类脑模型组织交联处理10分钟,然后将打印组织加入细胞培养液并放入恒温培养箱继续培养,在第3、4、5、6、7、8天对中间层海马体神经元细胞进行MTT细胞存活率检测。无处理组指的是未经LPS诱导处理的小胶质细胞与星型胶质细胞为原料3D打印出的模型,除小胶质细胞与星型胶质细胞的处理方式不同外,其余与阿尔兹海默症类脑模型的均相同。检测结果如下:

根据存活率检测结果可知,在第3、4天阿尔兹海默症类脑模型组织中海马体神经元细胞出现增多现象,这可能是因为在炎症浓度较低的水平条件下,活化的小胶质细胞与星型胶质细胞会对海马体神经元产生促进增长、分裂的作用;随着炎症浓度水平的不断提高,在第5天海马体神经元的存活率开始降低,且在第5天后表现出持续降低的特征,即中间层的细胞在日后逐渐死亡,证明该阿尔兹海默症类脑模型组织建立是成功的。
上述实施方式并非是对本发明的限制,本发明也并不仅限于上述举例,本技术领域的技术人员在本发明的技术方案范围内所做出的变化、改型、添加或替换,也均属于本发明的保护范围。
Claims (6)
1.一种3D打印阿尔兹海默症类脑模型,其特征在于:该阿尔兹海默症类脑模型具有上、中、下三层结构,其中,上、下两层结构中所含有的细胞是活化好的小胶质细胞与星型胶质细胞,胶质细胞与星型胶质细胞按1:1混合到生物墨水中,分别形成上、下两层结构;中间层所含有的细胞是海马体神经元,海马体神经元混合到生物墨水中,形成中间层结构;
其中,活化好的小胶质细胞、活化好的星型胶质细胞指的是:经LPS诱导并成功产生炎症反应的小胶质细胞、星型胶质细胞;
生物墨水的配方如下:
海藻酸钠:2-10wt%;
食用明胶:3-15wt%;
胶原蛋白:1-15mg/mL;
壳聚糖:3-14wt%。
2.一种如权利要求1所述的3D打印阿尔兹海默症类脑模型的制备方法,其特征在于:包括以下制备过程:
S1、活化的小胶质细胞、星型胶质细胞获取:
小胶质细胞、星型胶质细胞先从超低温冰箱中取出来复苏至培养瓶中培养24小时;随后将小胶质细胞、星型胶质细胞通过加入LPS诱导成为炎症的小胶质细胞、星型胶质细胞;
S2、海马体神经元获取:
海马体神经元也从超低温冰箱中取出来复苏至培养瓶中培养24小时,不做任何处理;
S3、生物3D打印准备阶段:
分别对上述海马体神经元细胞、以及活化的小胶质细胞、星型胶质细胞进行离心;
将离心获得的小胶质细胞、星型胶质细胞混合到生物墨水中,记为上样一;
将离心获得的海马体神经元混合到生物墨水中,记为上样二;
其中,小胶质细胞、星型胶质细胞按照1:1混合到生物墨水中,最终细胞浓度为1×106/mL;
海马体神经元混合到生物墨水中的最终细胞浓度为1×106/mL;
S4、设置3D打印参数,上机打印:
以上样一为原料打印出下层结构后,再由上样二为原料打印出中间层结构,再由上样一为原料打印出上层结构,制得阿尔兹海默症类脑模型。
3.根据权利要求2所述的3D打印阿尔兹海默症类脑模型的制备方法,其特征在于:细胞复苏培养条件为:用含有89%的DMEM、10%的FBS、1%的双抗在二氧化碳恒温培养箱中分别培养24h,双抗为抗青霉素和链霉素,恒温培养环境为37℃,CO2的体积浓度为5%。
4.根据权利要求3所述的3D打印阿尔兹海默症类脑模型的制备方法,其特征在于:在小胶质细胞与星型胶质细胞中加入LPS使其最终浓度为100ng/mL,继续分别培养24小时,由LPS诱导产生炎症反应。
5.根据权利要求4所述的3D打印阿尔兹海默症类脑模型的制备方法,其特征在于:3D打印参数具体有:
打印喷头的直径:0.3mm;
打印气动压力:0.4MPa;
打印速度:10mm/s;
打印结构长宽高:6×6×6mm。
