Abstract
The invention relates to an acne-removing and repairing composition containing recombinant collagen and application thereof. The invention creatively combines the four components, wherein azelaic acid, nisin and acetylcysteine have potential interaction promotion effects in inhibiting propionibacterium acnes and resisting inflammation, so that the invention has synergistic effects in inhibiting and relieving the formation of acnes and repairing the acnes; the recombinant collagen can repair skin damaged by acnes and inflammations, can provide sufficient nutritional ingredients for the damaged skin to help the skin to repair better, and can further promote the effects of azelaic acid, nisin and acetylcysteine on acne removal, and the four ingredients play roles of acne removal and repair jointly from different angles of different mechanisms. Meanwhile, the composition can well give consideration to excellent acne removal and repair effects and excellent mildness.
Description
Technical Field
The invention belongs to the technical field of acne-removing efficacy products, and relates to an acne-removing repair composition containing recombinant collagen and application thereof.
Background
Acne, also known as whelk, comedo, pimple, is a common and frequent disease state of the skin. The root cause of the formation is a chronic skin disease occurring in the pilosebaceous glands, and the phenomenon of pore blocking is generated due to the vigorous secretion of grease. The cosmetic effect is affected by the abundant sebaceous glands such as head, face, neck and the like.
Propionibacterium acnes is considered to be the most predominant bacterium causing acnes because excessive grease is secreted in the skin, the sebaceous cells are excessively proliferated, the grease is secreted into sebaceous glands and spread in pores, after propionibacterium acnes contacts the grease, triglycerides in the grease are decomposed to form glycerin and fatty acids, the glycerin increases nutrition of bacteria, proliferation of bacteria is caused, and free fatty acids cause keratinization of skin pores, hyperkeratinization, and pore blockage is caused. The topical medicinal products for removing acne include retinoic acid, benzoyl peroxide, and antibiotics. However, retinoic acid increases the photosensitivity of the skin and reds the skin; benzoyl peroxide has certain irritation, and individual patients can have reactions such as redness, desquamation and the like; antibiotics can cause bacterial drug resistance, and the using course of the antibiotics cannot be too long.
The prior art also discloses a few types of cosmetics with acne removing effect, for example, CN107737033A discloses a composition for rapidly removing acnes and acne marks and a preparation method thereof, and the raw materials comprise the following components in parts by weight: 1.2 to 1.8 parts of salicylic acid, 2 to 2.77 parts of arginine, 0.5 to 2 parts of Usnea barbata extract, 1 to 10 parts of glabridin liposome, 0.03 to 0.1 part of O-cymene-5 alcohol, 1 to 12 parts of other acne-removing efficacy components and 75 to 90 parts of other base materials. The product is prepared by combining salicylic acid with arginine with specific content, compounding with scindapsus aureus extract and glabridin liposome with specific content, and adding O-cymene-5 alcohol as a mild and effective antiseptic bactericide and other acne-removing functional components. However, the raw material contains salicylic acid, so that skin and mucous membrane can be stimulated, skin barrier can be damaged, allergic symptoms can be caused, and the raw material is not suitable for people with sensitive skin.
At present, the acne-removing cosmetics on the market are various in variety, the production process and the formula of the cosmetics are basically different in size, and the cosmetics have the effects of protecting and beautifying the human skin to different degrees, but the effects are not ideal, and the cosmetics have no multiple effects of diminishing inflammation, repairing and nursing on facial acnes. And after the products are coated, the products generally lose efficacy in a short time, the functional components are difficult to act on the skin for a long time, the purpose of acne removal is achieved, and the efficacy is difficult to ensure.
Therefore, it is very significant to develop a product which is mild, safe and capable of removing acnes for a long time.
Disclosure of Invention
Aiming at the defects of the prior art, the invention aims to provide an acne-removing and repairing composition containing recombinant collagen and application thereof.
In order to achieve the aim of the invention, the invention adopts the following technical scheme:
In a first aspect, the present invention provides an anti-acne repair composition comprising recombinant collagen, the anti-acne repair composition comprising azelaic acid, nisin, acetylcysteine and recombinant collagen.
Azelaic acid has antibacterial, antiinflammatory, hair follicle keratosis regulating, oil secretion inhibiting, pigmentation reducing, and cell proliferation inhibiting effects; nisin can interact with anionic lipids on bacterial cell membranes to cause perturbation of the cell membranes to inhibit bacteria; acetylcysteine can be used as skin conditioner, antioxidant or protease inhibitor in cosmetics and skin care products; the recombinant collagen is collagen prepared by using a DNA recombination technology, can supplement amino acid required by skin, repair damaged skin and provide necessary nutritional ingredients for the skin.

Preferably, the acne-removing and repairing composition comprises, by mass, 4-10 parts of azelaic acid, 0.1-1 part of nisin, 0.5-5 parts of acetylcysteine and 0.01-1 part of recombinant collagen.
Based on the potential interaction among the components, when the four components are combined in a specific mass ratio relationship, the composition has better effects on removing acnes and repairing skin.
The azelaic acid may be 4 parts, 4.5 parts, 5 parts, 5.5 parts, 6 parts, 6.5 parts, 7 parts, 7.5 parts, 8 parts, 8.5 parts, 9 parts, 9.5 parts, 10 parts, etc.; the nisin may be 0.1 part, 0.2 part, 0.3 part, 0.4 part, 0.5 part, 0.6 part, 0.7 part, 0.8 part, 0.9 part, 1 part, etc.; the acetylcysteine may be 0.5 part, 1 part, 1.5 parts, 2 parts, 2.5 parts, 3 parts, 3.5 parts, 4 parts, 4.5 parts, 5 parts, etc.; the recombinant collagen may be 0.01 part, 0.03 part, 0.05 part, 0.08 part, 0.1 part, 0.2 part, 0.3 part, 0.4 part, 0.5 part, 0.7 part, 0.8 part, 1 part, etc. Other specific point values in the numerical ranges are selectable, and will not be described in detail herein.
Preferably, the nucleotide sequence encoding the recombinant collagen is the sequence shown in SEQ ID NO. 1.
Compared with other sources of recombinant collagen, the recombinant collagen from the specific source has better effects on bacteriostasis, anti-inflammation, acne removal and skin repair.

In a second aspect, the invention provides an application of the acne-removing and repairing composition containing the recombinant collagen according to the first aspect in preparing an acne-removing and repairing effect product.
Preferably, the additive amount of the acne removing and repairing composition in the acne removing and repairing effect product is 1-20%, for example 1%, 2%, 3%, 4%, 5%, 6%, 7%, 8%, 9%, 10%, 12%, 14%, 15%, 20%, etc., and other specific point values in the numerical range can be selected, which will not be described in detail herein.
Preferably, the product comprises a cosmetic or pharmaceutical product.
In a third aspect, the invention provides a gel with acne-removing and repairing effects, and the gel comprises the following preparation raw materials in percentage by mass: 1-20% of the acne-removing repair composition in the first aspect, 0.5-5% of a gel, 0.2-2% of a solubilizer, 5-20% of a polyalcohol, 0.1-1% of triethanolamine and 50-90% of water.
The mass percentage of the acne-removing and repairing composition can be selected to be 1%, 2%, 3%, 4%, 5%, 6%, 7%, 8%, 9%, 10%, 12%, 14%, 15%, 20% and the like; the mass percentage of the gel can be selected to be 0.5%, 0.6%, 0.8%, 1%, 1.2%, 1.4%, 1.6%, 1.8%, 2%, 3%, 4%, 5% and the like; the mass percentage of the solubilizer can be selected to be 0.2%, 0.3%, 0.5%, 0.6%, 0.8%, 1%, 1.2%, 1.4%, 1.6%, 1.8%, 2% and the like; the mass percent of the polyol can be selected to be 5%, 6%, 7%, 8%, 9%, 10%, 12%, 14%, 15%, 20%, etc.; the weight percentage of the triethanolamine can be selected to be 0.1%, 0.2%, 0.3%, 0.4%, 0.5%, 0.7%, 0.8%, 0.9%, 1% and the like; the mass percentage of the water can be selected to be 50%, 60%, 65%, 70%, 75%, 80%, 85%, 90% and the like; other specific point values within the above numerical ranges are all selectable, and will not be described in detail herein.

The mass percentage of the plant source conditioning agent can be selected to be 1%, 2%, 3%, 4%, 5%, 6%, 8%, 10% and the like; the antioxidant may be selected from 0.05%, 0.08%, 0.1%, 0.15%, 0.2%, 0.25%, 0.3%, 0.4%, 0.5%, etc.; the mass percentage of the aromatic agent can be selected to be 0.1%, 0.15%, 0.2%, 0.25%, 0.3%, 0.4%, 0.5% and the like; other specific point values within the above numerical ranges are all selectable, and will not be described in detail herein.
Preferably, the plant source conditioner comprises an extract of a square flower and/or an extract of kapok.
The invention also creatively discovers that when the extract of the inula flowers and/or the extract of the kapok flowers, especially the combination of the extract of the inula flowers and the extract of the kapok flowers, are further added into the product, the antibacterial, anti-inflammatory and acne-removing effects of the product can be further improved. Wherein the extract of flos Lupuli and the extract of flos kapok have synergistic effect in exerting the above effects.
In a fourth aspect, the present invention provides a method for preparing the gel with acne-removing and repairing effects according to the third aspect, the method comprising the following steps:
Mixing the gel with part of water at 25-45 ℃ to obtain a solution A; mixing the acne-removing repair composition, the solubilizer, the polyol and the residual water at 30-50 ℃ to obtain a solution B; mixing the solution A and the solution B at 20-40 ℃, and adding triethanolamine and the rest preparation raw materials to obtain the gel with acne removing and repairing effects.
The specific point value of 25-45deg.C can be 25deg.C, 28deg.C, 30deg.C, 32deg.C, 35deg.C, 38deg.C, 40deg.C, 42deg.C, 45deg.C, etc.; the specific point value of 30-50deg.C can be 30 deg.C, 33 deg.C, 35 deg.C, 38 deg.C, 40 deg.C, 42 deg.C, 45 deg.C, 48 deg.C, 50 deg.C, etc.; the specific values of 20-40deg.C can be 20deg.C, 22deg.C, 25deg.C, 28deg.C, 30deg.C, 32deg.C, 35deg.C, 38deg.C, 40deg.C, etc.; other specific point values within the above numerical ranges are all selectable, and will not be described in detail herein.

Detailed Description
In order to further describe the technical means adopted by the present invention and the effects thereof, the following describes the technical scheme of the present invention in combination with the preferred embodiments of the present invention, but the present invention is not limited to the scope of the embodiments.
The mass parts of each component in the composition product referred to in the following are calculated according to the actual content of the effective components in the raw materials sold in the market.
The extract of Fanghua involved in the following is derived from the product of Waters biotechnology Co., ltd; kapok extract is derived from the product of the biological sciences company, vortex, lan; nisin is derived from products of guangzhou Tianjia biotechnology limited.
The recombinant collagen referred to in the following was prepared using the product prepared in preparation examples in which pET-28a (+) vector was purchased from Michelin; BL21 (DE 3) chemocompetent cells were commercially available from Merck under the designation CMC0014-4X40 UL.
Preparation example
The preparation example provides recombinant collagen freeze-dried powder, and the preparation method is carried out by referring to the method disclosed in the preparation example 1 in ZL 202310123476.3, and specifically comprises the following steps:
(1) Constructing a recombinant plasmid: according to the nucleotide sequence shown in SEQ ID NO. 1, selecting a pET-28a (+) vector suitable for an expression vector escherichia coli host, and sending the pET-28a (+) vector to a sequencing company for synthesis to prepare a recombinant collagen expression vector pET-28a-1.
(2) Recombinant fermentation strain construction: transferring the pET-28a-1 expression vector into competent cells of escherichia coli (BL 21 (DE 3)) by using a chemical conversion method, screening positive clones in plate colonies after conversion by agarose gel electrophoresis, transferring the screened positive clones to 2mL of LB culture medium, culturing for 12 hours at 37 ℃ and 200rpm, diluting the cultured bacterial liquid by 1 time with glycerol with the volume fraction of 50%, packaging into a freezing tube, and freezing at the temperature of minus 80 ℃ to obtain the recombinant collagen fermentation strain.
(3) Fermenting and culturing recombinant fermentation strains: 10 mu L of recombinant collagen fermentation broth is coated on a solid LB plate, cultured for 12 hours in a 37 ℃ incubator, selected and inoculated in 50mL of LB liquid medium containing 50 mu g/mL kanamycin content, placed in a shaking table for culture at 200rpm for 14 hours at 37 ℃, then inoculated in 200mL of LB liquid medium containing 50 mu g/mL kanamycin content at a ratio of 1:100, cultured until the bacterial broth OD=0.6 at 37 ℃, added with IPTG to achieve the final concentration of 1mM, continuously cultured for 4 hours, then centrifuged at 4000rpm for 15 minutes, and the bacterial cells are collected.
(4) Purifying recombinant collagen: the bacterial cells are subjected to ultrasonic disruption and centrifugation, the precipitate is washed by using a TritonX-100 washing solution, the supernatant is removed by centrifugation again, 6M guanidine hydrochloride is added into the precipitate, the pH is regulated to 8, after the precipitate is fully dissolved, the solution is subjected to nickel affinity chromatography resin, 20mM, 50mM, 100mM, 200mM and 500mM concentration gradient imidazole solution is used for eluting in sequence, eluent is concentrated by using a protein concentration column, the concentrated solution is collected and dialyzed overnight (dialysis bag, RC film, 14KD and flat width of 44mM (manufacturer F600112)), then the protein concentration is detected by using a BCA kit, and the solution is uniformly mixed with the mannitol aqueous solution according to the proportion of 0.5mL of mannitol aqueous solution with the volume concentration of 5 percent per 5mg of protein, and then the solution is pre-frozen at-40 ℃ for 10min by using a freeze dryer, and finally freeze-dried at-35 ℃ to obtain the recombinant collagen freeze-dried powder.
Example 1
The embodiment provides an acne-removing and repairing composition which comprises the following components in parts by mass: 5 parts of azelaic acid, 1 part of nisin, 3 parts of acetylcysteine and 0.08 part of recombinant collagen. The components are physically and evenly mixed according to the mass ratio.
Example 2
The embodiment provides an acne-removing and repairing composition which comprises the following components in parts by mass: 4 parts of azelaic acid, 0.5 part of nisin, 5 parts of acetylcysteine and 0.05 part of recombinant collagen. The components are physically and evenly mixed according to the mass ratio.
Example 3
The embodiment provides an acne-removing and repairing composition which comprises the following components in parts by mass: azelaic acid 7 parts, nisin 0.2 parts, acetylcysteine 2 parts and recombinant collagen 0.03 parts. The components are physically and evenly mixed according to the mass ratio.
Comparative example 1
This comparative example provides a composition which differs from example 1 only in that azelaic acid is absent, its reduced mass is apportioned to the mass of nisin and acetylcysteine, the other components and amounts remaining unchanged. The components are physically and evenly mixed according to the mass ratio.
Comparative example 2
This comparative example provides a composition which differs from example 1 only in that nisin is not present, and its reduced mass is apportioned to the mass of azelaic acid and acetylcysteine, the other components and amounts remaining unchanged. This comparative example provides a composition which differs from example 1 only in that it does not contain acetylcysteine, and its reduced mass is apportioned to the mass of azelaic acid and nisin, the other components and amounts remaining unchanged. (1) Mixing the gel with part of water at 30 ℃ to obtain a solution A;
(2) Mixing azelaic acid with 1, 3-propylene glycol and butanediol at 40deg.C, and mixing with nisin, acetylcysteine, recombinant collagen, solubilizer, glycerol and residual water at 40deg.C to obtain solution B;
(3) Mixing the solution A and the solution B at 30 ℃, and adding triethanolamine to obtain the gel with acne removing and repairing effects.
Application examples 2 to 3
The application example provides two gels, and the formula of the gel is different from that of application example 1 only in that the acne-removing repair composition of example 1 is replaced by the acne-removing repair composition prepared in the same amount of examples 2-3, and other components and contents are kept unchanged. The preparation method is reference application example 1.
Comparative application examples 1 to 4
Comparative examples 1 to 4 provided four gels, the formulation of which was different from that of example 1 only in that the acne-removing restoration composition prepared in example 1 was replaced with the same amount of the acne-removing restoration composition prepared in comparative examples 1 to 4, and the other components and contents were kept unchanged. The preparation method is reference application example 1.
Application example 4
The application example provides a gel, which comprises the following components:
The preparation method comprises the following steps:
(1) Mixing the gel with part of water at 35 ℃ to obtain a solution A;
(2) Mixing azelaic acid with 1, 3-propylene glycol and butanediol at 30deg.C, and mixing with nisin, acetylcysteine, recombinant collagen, solubilizer, glycerol and residual water at 30deg.C to obtain solution B;
(3) Mixing the solution A and the solution B at 30 ℃, and adding triethanolamine and a conditioner to obtain the gel with acne removing and repairing effects.
Application example 5
The application example provides a gel, and the formula of the gel is different from that of application example 4 only in that the extract of the common bombax flower is lacking, the mass percentage of the extract of the common bombax flower is 8%, and other components and contents are kept unchanged.
Application example 6
The application example provides a gel, and the formula of the gel is different from that of application example 4 only in that kapok extract is absent, the mass percentage of plain flower extract is 8%, and other components and contents are kept unchanged.
Application example 7
The application example provides a gel, the formula of which is different from that of application example 1 only in that the acne-removing repairing composition is lack, the reduced quality is complemented by water, and the other components and the content are kept unchanged.
Test example 1
Safety evaluation:
placing the products prepared in application examples 1-7 and comparative application examples 1-4 in a patch tester, attaching the patch tester with the test object to the forearm flexor side of the tested person (20), and uniformly applying the patch to the skin by lightly pressing the palm for 24 hours; skin irritation and sensitization were observed as shown in Table 1 below 30min, 24h, 48h after removal of the subject plaque tester, respectively, and the observations were recorded.

The results show that the products prepared in application examples 1-7 and comparative application examples 1-4 have a rating of 0, which indicates that the acne-removing repair composition according to the invention has no irritation to skin.
Test example 2
Propionibacterium acnes inhibition assay:
Preparing a sample solution: sample solutions 1 to 11 correspond to application examples 1 to 7 and comparative application examples 1 to 4, respectively, and their formulations differ from application examples 1 to 7 and comparative application examples 1 to 4 in that they do not contain solubilizers, gelling agents and glycerin, and their reduced mass is complemented by water. The preparation method comprises the following steps: mixing azelaic acid with 1, 3-propylene glycol and butanediol at 30deg.C, mixing with nisin, acetylcysteine, recombinant collagen and water at 30deg.C, optionally adding conditioner, and mixing.
Propionibacterium acnes was inoculated into 96-well plates at 1X 10 6/mL, 150. Mu.L of each well was added with 150. Mu.L of the sample solution 1-11, and the culture medium of the same volume was added as a blank, and incubated in a constant temperature incubator at 37℃for 48 hours under anaerobic conditions. The absorbance at 600nm was measured by an ultraviolet spectrophotometer, and the propionibacterium acnes inhibition (%) was calculated according to the following formula, and each group was measured in parallel 5 times, and the average was taken.
Propionibacterium acnes inhibition (%) = (blank absorbance-test absorbance)/blank absorbance x 100%
The results are shown in Table 2.

As shown in table 2, the acne-removing and repairing composition has a remarkable propionibacterium acnes inhibition effect, azelaic acid, nisin and acetylcysteine have a synergistic effect on the propionibacterium acnes inhibition effect, and the extract of inula flowers and the extract of kapok flowers can have the effects and have the synergistic effect.
Test example 3
Propionibacterium acnes inhibition assay:

1X 10 6/mL Raw264.7 cells were inoculated into 24-well plates, 300. Mu.L per well, placed in a 37℃incubator for 24 hours, and the culture supernatant was discarded. 300 mu L of fresh medium is added into the blank group, 300 mu L of fresh medium dissolved with 1 mu g/mL of LPS is added into the LPS group, 300 mu L of sample solution 1-11 dissolved with 1 mu g/mL of LPS is respectively added into the test group, and the test groups are placed in a constant temperature incubator at 37 ℃ for culturing for 24 hours. The supernatant was centrifuged and the concentrations of the inflammatory factors TNF- α and IL-1 were measured according to the test procedure on ELISA kit and each group was assayed 5 times in parallel and the average was taken and the results are shown in Table 3.

As shown in table 3, the acne-removing and repairing composition has a remarkable anti-inflammatory effect, azelaic acid, nisin and acetylcysteine have a synergistic effect on the effect of inhibiting inflammatory factors, and the extract of inula flowers and the extract of kapok flowers can have further effects, and the two have a synergistic effect.
Test example 4
Sensory evaluation test:
160 oilpox muscle females and male volunteers with the age range of 18-40 years old are selected, randomly divided into 8 groups, 20 people in each group and men and women in each half, the products of application examples 1,4-6 and comparative application examples 1-4 are used in each group, the products are used once after facial cleaning every morning and evening, the use is continued for 60 days, other acne removing component products are not used in the period, and the use amount and the operation are ensured to be basically consistent. The mildness and vaccinia restoration effect of the product were scored after 60 days, wherein the highest score was 5 and the lowest score was 1, the scoring interval was 1-5, and the evaluation results are shown in table 4 (average score):

From table 4, it can be seen that the composition and the cosmetic prepared by the composition have excellent acne-removing and repairing properties, wherein the recombinant collagen plays an important role in improving the acne-removing and repairing effects and mildness of the product, the plant extract added into the cosmetic can further improve the acne-removing and repairing effects of the composition, and the extract of the inula flower and the extract of the kapok have a certain synergistic effect in improving the acne-removing and repairing effects of the product.
The applicant states that the technical solution of the present invention is illustrated by the above embodiments, but the present invention is not limited to the above embodiments, i.e. it does not mean that the present invention must be implemented by the above embodiments. It should be apparent to those skilled in the art that any modification of the present invention, equivalent substitution of raw materials for the product of the present invention, addition of auxiliary components, selection of specific modes, etc., falls within the scope of the present invention and the scope of disclosure.
The preferred embodiments of the present invention have been described in detail above, but the present invention is not limited to the specific details of the above embodiments, and various simple modifications can be made to the technical solution of the present invention within the scope of the technical concept of the present invention, and all the simple modifications belong to the protection scope of the present invention.
In addition, the specific features described in the above embodiments may be combined in any suitable manner, and in order to avoid unnecessary repetition, various possible combinations are not described further.
Claims (10)
1. The acne-removing repair composition containing the recombinant collagen is characterized by comprising, by mass, 4-10 parts of azelaic acid, 0.1-1 part of nisin, 0.5-5 parts of acetylcysteine and 0.01-1 part of recombinant collagen; the nucleotide sequence of the coded recombinant collagen is shown as SEQ ID NO. 1.
2. Use of the acne-removing repair composition containing recombinant collagen according to claim 1 for preparing an acne-removing repair efficacy product.
3. The use according to claim 2, wherein the acne-removing and repairing composition is added to the acne-removing and repairing efficacy product in an amount of 1-20%.
4. Use according to claim 2, wherein the product comprises a cosmetic or pharmaceutical product.
5. The gel with the acne-removing and repairing effects is characterized by comprising the following preparation raw materials in percentage by mass: the anti-acne repair composition of claim 1, 1-20%, carbomer 0.5-5%, solubilizer 0.2-2%, polyalcohol 5-20%, triethanolamine 0.1-1% and water 50-90%.
6. The gel with acne-removing and repairing effects according to claim 5, wherein the solubilizer comprises PPG-26-butanol polyether-26 and/or PEG-40 hydrogenated castor oil.
7. The gel with acne-removing and repairing effects according to claim 5, wherein the polyhydric alcohol comprises any one or a combination of at least two of propylene glycol, butylene glycol, or glycerin.
8. The gel with acne-removing and repairing effects according to claim 5, wherein the preparation raw materials of the gel further comprise, in mass percent: 1-10% of plant source conditioner, 0.05-0.5% of antioxidant or 0.1-0.5% of aromatic agent or the combination of at least two.
9. The gel with acne-removing and repairing effects according to claim 8, wherein the plant source conditioner comprises a inula flower extract and/or a kapok flower extract.
10. The method for preparing the gel with acne-removing and repairing effects according to any one of claims 5 to 9, wherein the preparation method comprises the following steps:
mixing carbomer with part of water at 25-45deg.C to obtain solution A; mixing the acne-removing repair composition, the solubilizer, the polyol and the residual water at 30-50 ℃ to obtain a solution B; mixing the solution A and the solution B at 20-40 ℃, and adding triethanolamine and the rest preparation raw materials to obtain the gel with acne removing and repairing effects.
