Abstract
本发明公开了一种抑菌促愈合伤口敷料,包括面层以及均匀吸附在面层上的粉剂,所述面层由粘胶纤维和化纤编织而成,具有细密的微孔结构;所述粉剂由海藻多肽、褐藻寡糖、复合维生素和聚乙二醇组成。该敷料贴敷在伤口处,能够使微孔内的粉剂缓慢释放至伤口内,其中的聚乙二醇能够快速吸收伤口渗出物并保持伤口环境潮湿,海藻多肽、褐藻寡糖结合维生素能够有效促进伤口内部组织修复,加快伤口愈合。
Description
技术领域
本发明涉及创伤医疗用品技术领域,具体涉及一种抑菌促愈合伤口敷料。
背景技术
敷料是常用的创伤医疗用品,其功能是吸收创伤部位的渗出液并且将伤口部分与外界隔离以保护伤口免受外界灰尘、细菌等的污染。传统敷料例如棉布,虽然可以快速吸收渗出液,但是吸收能力有限、保水性差、易残留碎屑,也容易粘连伤口造成二次创伤,进而导致伤口愈合时间延长。
随着材料技术的发展,敷料种类越来越多,功能也越来越多,例如,辅料中增加了抗菌成分,促愈合成分等,通过将抗菌和促愈合成分等功能性成分与载体成分混合或者表面涂布的方式制成敷料,在使用时,功能性成分与伤口部分接触,发挥抗菌促愈合作用。然而这类敷料仍然存在以下问题:载体成分与功能性成分的适配性较差,载药量有限或者药物释放能力有限;载体成分一般吸水性差,对伤口渗出液的吸收能力有限。这些问题导致需要频繁更换敷料,敷料使用成本增加,患者顺从性也变差。
发明内容
本发明的目的是真对现有技术中的上述不足,提供一种抑菌促愈合伤口敷料,能够快速吸收伤口渗出液,并保持伤口环境湿润,缓慢释放药物促进伤口愈合,减少更换频率。
本发明技术方案详述如下:
一种抑菌促愈合伤口敷料,包括面层以及均匀吸附在面层上的粉剂,所述面层由粘胶纤维和聚烯烃纤维编织而成,聚烯烃纤维表面具有微孔结构;所述粉剂由抗菌制剂、海藻多肽、褐藻寡糖、维生素、成纤维细胞生长因子和聚乙二醇组成。
粘胶纤维是以例如木材、芦苇、甘蔗渣、棉短绒等中的天然纤维为原料,经过化学反应将天然纤维素分离出来,制成可溶性纤维素黄原酸酯,再溶于稀碱液制成粘胶,经湿法纺丝而制成。由于粘胶纤维在水中膨润度较高,利于吸收伤口渗出液,而聚烯烃纤维吸湿性差因此又能够防止面层与伤口粘连。聚烯烃纤维表面具有微孔结构,粘胶纤维表面较为粗糙,横截面呈粗糙的锯齿状结构,加上编织的网眼,使得整个面层具有不同大小的孔洞(网眼、锯齿之间缝隙、微孔)以供吸附粉剂,载药量增加。在贴敷到伤口处后,面层吸附的粉剂更容易缓慢释放,延长药物作用时间。
所述粉剂通过少量添加海藻多肽、褐藻寡糖等成分,可有效减少抗菌制剂的用量,达到满意的抗菌效果。
海藻多肽是以天然海藻为原料,经过破碎、生物酶解、提纯、浓缩、离心喷雾干燥等工艺得到的小分子海藻肽粉。海藻多肽具有抗菌、促进伤口愈合等功效。褐藻寡糖是褐藻胶(海藻酸钠)经降解得到的低分子寡糖,聚合度20以下,主要由β-1,4-D甘露糖醛酸和其C-5差向异构体α-1,4-L古洛糖醛酸通过β1,4糖苷键连接而成。褐藻寡糖分子量低,水溶性强,稳定性高,具有抑菌、抗炎、抗氧化等功能。聚乙二醇能够快速吸收伤口渗出物并保持伤口环境潮湿。粉剂覆盖伤口后可随着渗出液的浸渍缓慢释放至伤口组织,通过海藻多肽、褐藻寡糖、成纤维细胞生长因子配合维生素发挥其抗菌促愈合功能。
可选或优选的,上述敷料中,所述聚烯烃纤维为聚乙烯纤维或聚丙烯纤维。聚乙烯纤维和聚丙烯纤维具有憎水性强、比重小、强度和弹性高、成本低等优点,且在制备微孔结构时更容易形成微孔。
可选或优选的,上述敷料中,所述聚烯烃纤维的制备方法是将氯化钠与聚烯烃进行共混,再经熔融纺丝得到共混纤维,将共混纤维放置在水中待共混纤维表面的氯化钠溶解使表面形成微孔,最终获得聚烯烃纤维。
可选或优选的,上述敷料中,所述抗菌制剂为壳聚糖季铵盐。壳聚糖季铵盐是由壳聚糖分子经过季铵化反应得到的产物,生物相容性好生物可降解性较好。
可选或优选的,上述敷料中,所述壳聚糖季铵盐分子量10000~20000Da,季铵化度50%~60%。该分子量和季铵化度下的壳聚糖季铵盐具有较适当的释放速率和时间,用于本发明粉剂中与其他成分配合利于提高药效和减少副作用。
可选或优选的,上述敷料中,所述复合维生素由维生素C、维生素A、维生素D、维生素E和维生素K组成。复合维生素有利于促进伤口愈合。
可选或优选的,上述敷料中,所述聚乙二醇由分子量为1000~1500的聚乙二醇和分子量6000~8000的聚乙二醇按质量比1:5~8混合而成。分子量为1000-1500的聚乙二醇,呈固态蜡状,分子量6000~8000的聚乙二醇为粉末状,与其他粉体成分混合后能够使整个粉剂既保持一定的塑形性不至于过度松散而造成面层吸附量少,又不会使粉剂过湿导致生产时吸附困难。
可选或优选的,上述敷料中,还包括胶粘层,所述面层包括上表面和下表面,所述胶粘层设于面层的上表面并向两端各延伸出一段,胶粘层均布透气孔,面层的下表面吸附粉剂。
与现有技术相比,本发明具有如下有益效果:
本发明面层是由抗菌聚烯烃纤维和粘胶纤维编织而成,同时满足了吸收渗液和不粘连伤口的要求,另外配合粘胶纤维的粗糙表面和聚烯烃纤维的微孔,使得面层对粉剂的吸附更强,载药量增加,粉剂药物释放缓慢持久,可有效减少敷料更换次数,患者顺从性更好。
粉剂中富含吸湿、抗菌和促愈合成分,与伤口创面接触后能够快速吸收渗出液并保持湿润微环境,同时进一步降低感染,粉剂药物缓慢释放至伤口组织促进伤口愈合,能使敷料一次性使用时间更长,减少更换频率。相较于常规促敷料,本发明的带粉剂敷料使用后伤口恢复时间能够有效缩短。
具体实施方式
下面结合较佳的具体实施例对本发明技术方案进行详细解释和说明,以使本领域技术人员能够更好地理解本发明并予以实施。
除特殊说明外,本发明实施例中所用到的仪器、试剂等均为现有市售产品,所用方法均为本领域常规实验方法。
实验面层:氯化钠2.5%(质量百分含量)与聚乙烯余量。称取各原料,将纺丝机预热至所需温度,原料添加到纺丝机中,进行熔融纺丝,得到共混纤维。共混纤维放置到密封容器,加入蒸馏水,密封放置3天,取出,干燥待用。共混纤维与粘胶纤维编织成面层布。
对照面层:将纺丝机预热至所需温度后,添加聚乙烯到纺丝机中,进行熔融纺丝,得到聚乙烯纤维。聚乙烯纤维与粘胶纤维编织成对照面层布。
对照面层和实验面层分别进行以下载药操作:面层布周边用机械抓手固定拉平,经过粉剂填装箱体,箱体内有喷头将粉剂均匀喷洒在面层布表面,喷洒过程中机械抓手带动面层布进行振动以将粉末尽量填充到孔隙中。喷洒结束后,将面层布折叠成多层以达到所需厚度(一般2-5层),再用平板向下压面层布进行定型,最后在表面粘压带透气孔的胶粘层,再裁成所需大小,获得成品敷料。
不同成分粉剂配方如下表所示:
备注:例7中聚乙二醇用PVP K30替代;聚乙二醇分子量1000:分子量6000按1:8配比。
以例1粉剂进行载药量实验,面层面积为1平方米(总计3层),粉剂喷洒时间和操作过程相同,结果表明实验面层相较于对照面层的载药量增加15%。
抑菌效果实验
经过37℃培养24h的金黄色葡萄球菌、大肠杆菌、绿脓杆菌和肺炎克雷伯菌,用移液枪转入锥形瓶内,液体培养基条件下37℃摇床振荡培养至对数生长期。
向培养至对数生长期的各菌种培养瓶中添加等量粉剂(按50mL培养基体积添加0.2g的标准添加),设置空白对照组。继续摇床振荡培养。取24h后的菌液,计算抑菌率。
抑菌率(%)=(对照组OD值-实验组OD值)/实验组OD值×100%。
结果如下表所示:
抑菌实验结果表明,该辅料中的粉剂对金黄色葡萄球菌、大肠杆菌、绿脓杆菌和肺炎克雷伯菌都具有较好的抑制作用。
敷料对烧伤伤口的治疗实验
实验动物:Ⅱ度烧伤SD大鼠模型。
分组:分为5组,每组8只。创面消毒后去除坏死表皮,渗出物先用无菌纱布吸干,再经一次消毒后贴敷敷料,敷料应盖过创面1-2cm。
对照组:普通抗菌敷料(市售)。实验组:例2、例4、例5、例6粉剂制作的敷贴。各组饲养条件相同,敷料更换以避免和减轻皮肤糜烂为原则。观察每组创面愈合程度、记录创面面积。创面愈合率=愈合面积/原始创面面积×100%。
创面愈合率(%)统计结果见下表:
实验结果表明,在创面愈合率方面,本发明粉剂配方能够更加快速地促进伤口愈合。

Claims (9)
1.一种抑菌促愈合伤口敷料,其特征在于,包括面层以及均匀吸附在面层上的粉剂,所述面层由粘胶纤维和聚烯烃纤维编织而成,聚烯烃纤维表面具有微孔结构;所述粉剂由抗菌制剂、海藻多肽、褐藻寡糖、复合维生素和聚乙二醇组成。
2.根据权利要求1所述的敷料,所述聚烯烃纤维为聚乙烯纤维或聚丙烯纤维。
3.根据权利要求2所述的敷料,其特征在于,所述聚烯烃纤维的制备方法是将氯化钠与聚烯烃进行共混,再经熔融纺丝得到共混纤维,将共混纤维放置在水中待共混纤维表面的氯化钠溶解使表面形成微孔,最终获得聚烯烃纤维。
4.根据权利要求1所述的敷料,其特征在于,所述抗菌制剂为壳聚糖季铵盐或纳米银。
5.根据权利要求4所述的敷料,其特征在于,所述壳聚糖季铵盐分子量10000~30000Da,季铵化度50%~60%。
6.根据权利要求4或5所述的敷料,其特征在于,所述粉剂中,各成分质量百分比为:
壳聚糖季铵盐0.2%~0.6%;
海藻多肽15-20%;
褐藻寡糖0.1-0.5%;
复合维生素0.1-0.2%;
聚乙二醇余量。
7.根据权利要求6所述的敷料,其特征在于,所述复合维生素由维生素C、维生素A、维生素D、维生素E和维生素K组成。
8.根据权利要求6所述的敷料,其特征在于,所述聚乙二醇由分子量为1000~1500的聚乙二醇和分子量6000~8000的聚乙二醇按质量比1:5~8混合而成。
9.根据权利要求1所述的敷料,其特征在于,还包括胶粘层,所述面层包括上表面和下表面,所述胶粘层设于面层的上表面并向两端各延伸出一段,胶粘层均布透气孔,面层的下表面吸附粉剂。
