由于体内复杂的微环境,蛋白类药物在注射进入体内后容易被体内的多种蛋白酶所降解,因而半衰期较短。人尿激肽原酶是从新鲜人尿中提取的一种蛋白酶,在高血压、微循环障碍及轻-中度急性血栓性脑梗死等疾病的治疗中具有较高的应用价值。在静脉注射后血浆中人尿激肽原酶的浓度迅速降低,在家兔、犬和健康成人体内的消除半衰期分别是120分钟、40分钟和156-197分钟。在轻-中度急性血栓性脑梗死的患者中用药需每次将0.15PNA单位,溶于50ml或100ml氯化钠注射液中,静脉滴注30分钟,每日1次,持续3周为一个疗程。从降低给药次数提高患者的用药依从性的角度考虑,人尿激肽原酶需要长效化改造以延长其在体内的循环时间。
聚乙二醇(PEG)是FDA批准的药用材料,通过化学键将聚乙二醇修饰在多肽/蛋白药物分子上,可用于制备长效多肽/蛋白药物。尽管有些药物分子的活性在PEG化之后降低很多,但PEG化带来的延长的体内半衰期可有效弥补这一部分。比如在实验小鼠中,IFNα2a的半衰期只有0.7小时,PEG化之后半衰期延长到51小时,同时体外活性测试显示PEG化的IFNα2a的活性只有IFNα2a的7%。但是临床试验显示,PEG化IFNα2a的疗效可以持续一个星期。相比于IFNα2a每周三次的给药方案,PEG化IFNα2a的每周一次给药频率有效提高了病人的依从性和延长了药物的作用时间。蛋白药物的聚乙二醇化修饰的主要优势包括:1)PEG修饰增加了药物分子的分子量,降低了药物分子被肾小球滤过的频率,延长了药物分子在体内的循环时间;2)柔性的PEG链可保护药物分子的酶活位点,降低其被蛋白酶降解的几率;3)PEG分子可屏蔽药物分子上的免疫原性位点,降低药物分子在体内的免疫原性。
专利CN201910046203.7公开了重组人激肽释放酶的聚乙二醇化修饰物,但与未修饰的激肽释放酶和市售的尤瑞克林相比,激肽释放酶的聚乙二醇化修饰物半衰期均未得到明显改善,而且生物活性也有明显的降低,只保留了原蛋白活性的80-90%。
发明内容
发明目的:本发明旨在提供一种半衰期长、比酶活高、降低脑梗死效果好的人尿激肽原酶的化学修饰物。
技术方案:本发明所述的人尿激肽原酶的化学修饰物,所述化学修饰为聚乙二醇修饰,所述聚乙二醇的活化基团为羧酸NHS活性酯。
优选地,所述聚乙二醇修饰采用直链型或分支型的聚乙二醇。
优选地,所述直链型的聚乙二醇的结构为:

本发明提供的PEG化制备方法,在无水纯有机溶剂DMSO中进行,不影响修饰后人尿激肽原酶的体外酶活。
有益效果:与现有技术相比,本发明具有如下显著优点:所述人尿激肽原酶的化学修饰物相对于未修饰的人尿激肽原酶来说,多修饰聚乙二醇化的人尿激肽原酶,比酶活不但没有降低反而有所提高,体内半衰期明显延长,在体内可长时间保持较高的血药浓度,在降低神经功能评分和脑梗死范围方面,表现更优异。

本发明使用以下缩写:
KN:人尿激肽原酶
PEG:聚乙二醇
M-SCM-20K:分子量为20000Da的甲氧基聚乙二醇羧甲基琥珀酰亚胺酯,结构为直链型,具体结构式如式I所示,其中n为455;
MPEG2-NHS-40K:分子量为40000Da的二(甲氧基聚乙二醇)-琥珀酰亚胺酯,结构为支链型,具体结构式如式Ⅱ所示,其中n为455;
实施例1
1.1、ZSPKN的制备
1)称取M-SCM-20K 100mg,KN 100mg,然后加入无水DMSO 20mL,在40℃水浴锅中搅拌反应6h;
2)加入纯水40mL终止反应,开始用截留分子量7kDa的透析袋开始透析,透析48h;
3)透析完成后,开始用截留分子量10kDa的超滤管开始超滤浓缩,重复4次,制备得到单修饰产物ZSPKN,4℃冰箱中保存。
1.2、ZSPKN的纯化
色谱法条件:色谱柱:HiTRAP Q FF 5mL;缓冲液A:20mM Tris-HCl,pH=8.0;缓冲液B:缓冲液A+1M NaCl;流速1mL/min;检测波长为280nm。
上样:上述单修饰产物ZSPKN用0.5M的NaOH溶液调节至pH 8.0,结合至Q离子交换柱。
平衡:缓冲液A冲洗5个柱体积,洗脱出未反应的PEG。
收集:用85%缓冲液A+15%缓冲液B洗脱3个柱体积,收集洗脱峰样品为ZSPKN纯化样品;用70%缓冲液A+30%缓冲液B洗脱2个柱体积,收集洗脱样品为未反应的人尿激肽原酶。
再生:用100%缓冲液B冲洗1个柱体积。
ZSPKN纯化样品的HPLC-SEC纯度分析图如图1所示。
实施例2
2.1、ZMPKN的制备
1)称取M-SCM-20K 200mg,KN 50mg,加入无水DMSO 30mL,在40℃水浴锅中搅拌反应6h。
2)加入纯水60mL终止反应,开始用截留分子量为7kDa的透析袋开始透析,透析48h。
3)透析完成后,开始用截留分子量50KDa的超滤管开始超滤浓缩,重复4次,制备得到多修饰产物ZMPKN,于4℃冰箱中保存。
2.2、ZMPKN的纯化
色谱法条件:色谱柱:博格隆Chromdex 200PG;缓冲液C 0.02M PB+0.15M NaCl,pH7.0;检测波长为280nm。
上样:上述修饰产物ZMPKN直接上样;
收集:用缓冲液C直接进行洗脱,收集洗脱峰样品,得到ZMPKN纯化样品。

实施例3
3.1.YSPKN的制备
1)称取MPEG2-NHS-40K 200mg,KN 100mg,加入无水DMSO 20mL,在40℃水浴锅中搅拌反应6h。
2)加入纯水40mL终止反应,开始用截留分子量为7kDa的透析袋开始透析,透析48h。
3)透析完成后,开始用截留分子量30kDa的超滤管开始超滤浓缩,重复4次,制备得到单修饰产物YSPKN,于4℃冰箱中保存。
3.2YSPKN的纯化
色谱法条件:色谱柱:HiTRAP Q FF 5mL;缓冲液A:20mM Tris-HCl,pH=8.0;缓冲液B:缓冲液A+1M NaCl;流速1-2mL/min;检测波长为280nm。
上样:上述单修饰反应产物YSPKN用0.5M的NaOH溶液调节至pH 8.0,结合至Q离子交换柱。
平衡:缓冲液A冲洗5个柱体积,洗脱出未反应的PEG;
收集:用85%缓冲液A+15%缓冲液B洗脱3个柱体积,收集洗脱样品为YSPKN纯化样品;用70%缓冲液A+30%缓冲液B洗脱2个柱体积,收集洗脱样品为未反应的人尿激肽原酶。
YSPKN纯化样品的HPLC-SEC纯度分析图如图3所示。
实施例4
4.1YMPKN的制备
1)称取MPEG2-NHS-40K 500mg,KN 50mg,然后加入无水DMSO 20mL,然后在40℃水浴锅中搅拌反应24h。
2)加入纯水40mL终止反应,开始用截留分子量为7KDa的透析袋透析,透析48h;
3)透析完成后,用截留分子量50kDa的超滤管超滤浓缩,重复4次,制备得到多修饰产物YMPKN,于4℃冰箱中保存。
4.2.YMPKN的纯化
色谱法条件:汇研生物Focurose 75PG色谱柱,缓冲液C:0.02M PB+0.15M NaCl,pH7.0,检测波长为280nm。
上样:上述多修饰产物YMPKN直接上样;
收集:用缓冲液C直接进行洗脱,收集洗脱峰样品,得到ZMPKN纯化样品。
YMPKN纯化样品的HPLC-SEC纯度分析图如图4所示。
实施例5
1.PEG化修饰后KN的鉴定
2.ZSPKN、ZMPKN、YSPKN和YMPKN的SEC-HPLC纯度检测。
采用SEC-HPLC对实施例1-4的产物ZSPKN、ZMPKN、YSPKN和YMPKN纯化样品进行评价。采用Sepax SRT-SEC-300(7.8*300mm,5μm)色谱柱,以0.15mol/L磷酸盐缓冲液与乙腈体积比=9:1为流动相,流速1ml/min,进行SEC-HPLC分析,检测波长为280nm,进样量20μL,分析时间20min,ZSPKN、ZMPKN、YSPKN和YMPKN纯化样品分析结果如图1-4所示,纯度分别为90.34%,92.53%,93.03%,98.29%。
3.SDS-PAGE
以4-12%梯度胶(购自于金斯瑞,SurePAGETM预制胶,Bis-Tris)在200V,25mA下对ZSPKN,ZMPKN,YSPKN,YMPKN纯化样品进行SDS-PAGE电泳,采用考马斯亮蓝显色,结果如图5和图6所示。
人尿激肽原酶分子量为35kDa,制备ZSPKN采用分子量为20kDa的PEG分子,由于PEG分子的水合作用,实际表观分子量会增大约1.5-2.5倍,因此PEG化后的人尿激肽原酶单修饰产物分子量约为65-85kDa,SDS-PAGE如图5所示,结果显示在分子量为66.2kDa左右有单一条带,表明一个人尿激肽原酶偶联一个PEG。
制备YSPKN采用分子量为40kDa的PEG分子,由于PEG分子的水合作用,实际表观分子量会增大约1.5-2.5倍,因此PEG化后的人尿激肽原酶单修饰产物分子量约为95-135kDa,SDS-PAGE如图6所示,结果显示在分子量为150kDa左右有单一条带,表明一个人尿激肽原酶偶联一个PEG。
ZMPKN实际表观分子量均显示大于116kDa以及YMPKN实际表观分子量显示大于250kDa,表明两者均为多修饰产物。
4.PEG平均修饰度测定
采用LC-MS/MS方法对PEG化修饰前后KN的酶切肽段进行检测,通过比较发生改变的修饰前后的肽段,得出PEG的修饰比例。
首先,分别对未PEG化和PEG化蛋白供试品经变性还原、烷基化后、酶切、淬灭,得到肽段。肽段具体制备过程为:
参比品(KN)前处理:
称取约0.78mg KN置1.5mL的EP管中,加入780μL的超纯水,混匀,即得,浓度为1mg/mL。
PEG化修饰后KN前处理:
取实施例1的产物ZSPKN纯化样品1mg,加入1mL超纯水,混匀,即得,浓度为1mg/mL。
取实施例2的产物ZMPKN纯化样品1mg,加入1mL超纯水,混匀,即得,浓度为1mg/mL。
取实施例3的产物YSPKN纯化样品1mg,加入1mL超纯水,混匀,即得,浓度为1mg/mL。
取实施例4的产物YMPKN纯化样品1mg,加入1mL超纯水,混匀,即得,浓度为1mg/mL。
换液:加入200μL纯水到超滤管中,13000g,离心2min,去除超滤管中少量的甘油。加入100μL 8M盐酸胍,再加入100μL前处理的参比品或PEG化修饰后KN 13000g离心5min。加入200μL 8M盐酸胍,13000g离心15min。
变性还原:加入100μL的8M盐酸胍,2μL 500mM的DTT(终浓度约10mM),混匀。将混合液在56℃的温度下孵育30min。
烷基化:加入4μL 500mM IAM溶液(终浓度约20mM),室温,避光,反应30min。
酶切:把反应液13000g离心10min。加入200μL100mM Tris-HCl,13000g离心10min,重复2次。用100μL100mM Tris-HCl复溶。加入4μL 0.5μg/μL Trypsin和2μL0.5μg/μL LysC,37℃孵育,过夜。把反应液从超滤膜上吸到1.5mL离心管中,再加入100μL100mM Tris-HCl,13,000g离心10min,将离心液吸到1.5mL离心管中,合并。
淬灭:加入20μL10%甲酸溶液,备用。
肽段经UHPLC分离后,用质谱进行数据采集,Biopharma Finder分析软件将实验采集的一级质谱图和关联的二级质谱图与理论肽段分子量和二级碎片离子进行比对,根据发生改变的PEG修饰前后肽段的强度,从而得到肽段PEG的修饰比例。
PEG修饰比例的计算方式,面积归一化法校正后的特征肽段峰面积除以参比样品对应肽段峰面积,再用1减去计算数值最终乘以百分之一百得到。公式如下:

Thermo UHPLC-Q Exactive Plus分别对发生PEG化修饰前后KN的酶切肽段进行检测,Thermo Biopharma Finder软件进行数据分析,确认KN中7个含有赖氨酸的肽段和N端伯氨基,为PEG化的潜在修饰位点。
PEG平均修饰度和EG偶联数结果如表1所示:

实施例6
实施例1-4的PEG化修饰后KN的比酶活测定
称取适量实施例1-4的PEG化修饰后KN(ZSPKN、ZMSPKN、YSPKN和YMPKN纯化样品),溶解在0.2M Tris缓冲液中(pH8.0),制成待测样品溶液。取0.2mL待测样品溶液分别加入4支试管中,同时加入4mL0.2 M Tris缓冲液,混匀,置于37度孵育5分钟,于第一管中加入0.4mL 50%醋酸溶液,其余3管中加入0.4mL底物溶液(1.5mM S-226),立即摇匀,置37度水浴中反应15分钟。然后在第一管中加入底物溶液0.4mL,第二、三、四管中各加入50%醋酸溶液0.4mL,使用紫外-可见分光光度仪,于405nm波长处,以第一管为空白,测定吸光度,吸光度应控制在0.1~0.2之间。
根据测得的吸光度值,计算得到待测样品的效价。待测样品溶液中的蛋白浓度采用BCA方法测定。同一样品的效价与其蛋白浓度的比值即为比酶活。
根据上述方法,测得实施例1-4的PEG化人尿激肽原酶的比酶活如表2所示:

由上表可以得出,相对于未修饰人尿激肽原酶来说,ZSPKN、YSPKN和YMPKN的比酶活得到提高,而ZMSPKN比酶活有所降低,其中YMPKN比酶活最高,相比于未修饰人尿激肽原酶的比酶活提高了10%,由此说明,直链型单修饰聚乙二醇化的人尿激肽原酶以及分支型单/多修饰聚乙二醇化的人尿激肽原酶相对于未修饰的人尿激肽原酶来说比酶活不但没有降低,反而还得到提高。
实施例7
ZSPKN、ZMPKN、YSPKN和YMPKN药代动力学研究
采用125I同位素标记示踪法对PEG修饰前后的人尿激肽原酶的体内血药浓度进行了研究。
30只ICR小白鼠雄鼠,分为5组,分别为KN组、ZSPKN组、ZMPKN组、YSPKN组、YMPKN组,每组6只。
KN组:单次静脉注射给予0.35PNA单位/mL125I-注射用尤瑞克林(冻干制剂,商品名为凯力康,广东天普生化医药股份有限公司生产)0.2mL;分别在给药前0h,给药后0.083h、0.25h、0.5h、0.75h、1h、2h、4h、6h、8h、12h、24h、48h采集血样至EDTA-K2抗凝管中,共采集12个时间点,分析获得放射性药代动力学参数(以沉淀放射性计)。
ZSPKN组:单次静脉注射给予0.35PNA单位/mL125I-ZSPKN注射液(取实施例1产物ZSPKN纯化样品,加入0.9%氯化钠注射液至所需浓度,配成ZSPKN注射液)0.2mL,每组6只,分别在给药前0h,给药后0.083h、0.25h、0.5h、0.75h、1h、2h、4h、6h、8h、12h、24h、48h、72h、96h、120h、144h采集血样至EDTA-K2抗凝管中,共采集16个时间点,分析获得放射性药代动力学参数(以沉淀放射性计)。

YSPKN组:单次静脉注射给予0.35PNA单位/mL125I-YSPKN注射液(取实施例3产物YSPM纯化样品,加入0.9%氯化钠注射液至所需浓度,配成YSPKN注射液)0.2mL,每组6只,分别在给药前0h,给药后0.083h、0.25h、0.5h、0.75h、1h、2h、4h、6h、8h、12h、24h、48h采集血样至EDTA-K2抗凝管中,共采集12个时间点,分析获得放射性药代动力学参数(以沉淀放射性计)。
YMPKN组:单次静脉注射给予0.35PNA单位/mL125I-YMPKN注射液(取实施例4产物YMPKN纯化样品,加入0.9%氯化钠注射液至所需浓度,配成YMPKN注射液)0.2mL,每组6只,分别在给药前0h,给药后0.083h、0.25h、0.5h、0.75h、1h、2h、4h、6h、8h、12h、24h、48h采集血样至EDTA-K2抗凝管中,共采集12个时间点,分析获得放射性药代动力学参数(以沉淀放射性计)。
得到的具体药代动力学参数如表3所示,给药血动曲线如图7所示:

结果表明:与未修饰KN组相比,YMPKN组、ZMPKN组半衰期明显延长,分别达到22.087h和26.187h,且在小鼠体内一直保持较高的血药浓度,可持续120h以上,而YSPKN组与ZSPKN组并没有得到改善,半衰期反而下降到3.891h和3.22h。由此可知,多修饰聚乙二醇化的人尿激肽原酶相对于未修饰的人尿激肽原酶来说半衰期明显提高,最高可达26.187h,且在小鼠体内一直保持较高的血药浓度,可持续达120h以上。
实施例8
药效学性质研究
采用三氯化铁法制备大鼠脑缺血再灌注模型,缺血3h后评分,选取造模成功的9~11分动物分为6组,每组8只,分别为模型对照组、KN组、ZSPKN组、ZMPKN组、YSPKN组以及YMPKN组。
KN组:静脉注射市售注射用尤瑞克林组尾静脉给予市售的注射用尤瑞克林(商品名凯力康,广东天普生化医药股份有限公司生产)15.5×10-3PNAu/kg,给药1次。ZSPKN组:静脉注射ZSPKN注射液(取实施例1产物ZSPKN纯化样品,加入0.9%氯化钠注射液至所需浓度,配成ZSPKN注射液)15.5×10-3PNAu/kg,给药1次。ZMPKN组:静脉注射ZMPKN注射液(取实施例2产物ZMPKN纯化样品,加入0.9%氯化钠注射液至所需浓度,配成ZMPKN注射液)15.5×10-3PNAu/kg,给药1次。YSPKN组:静脉注射YSPKN注射液(取实施例3产物YSPM纯化样品,加入0.9%氯化钠注射液至所需浓度,配成YSPKN注射液)15.5×10-3PNAu/kg,给药1次。YMPKN组:静脉注射YMPKN注射液(取实施例4产物YMPKN纯化样品,加入0.9%氯化钠注射液至所需浓度,配成YMPKN注射液)15.5×10-3PNAu/kg,给药1次。模型对照组给予等体积生理盐水,试验终点为药后7d。检测指标如下:
(1)神经功能缺陷评分:
分别于治疗前、治疗后1、3、5d观察动物行为障碍程度并进行评分(16分)。评分标准见表4。

(2)脑梗死测定:
将脑组织切片进行脑水肿检测拍照后,置2%红四氮唑(TTC)溶液中,37℃孵育5min,梗死组织呈白色,非梗死组织为红色。采用Image J软件进行脑梗死面积测定,并计算梗死面积占全脑面积的百分比。
实验结果
1.1对大鼠行为学评分的影响
在神经功能缺陷评分中,模型对照组大鼠神经功能出现显著障碍。与模型对照组比较,YSPKN及YMPKN组可显著降低药后3d、7d评分(P<0.05);KN组可显著降低药后3d、7d评分(P<0.05~0.01);ZSPKN及ZMPKN组相对模型组有降低神经功能评分趋势,但无显著性差异,结果见表5。

1.2对大鼠脑梗死的影响
与模型对照组相比,YSPKN组、YMPKN组、KN组能显著降低脑梗死范围,改善率分别为31.0%(P<0.05)、32.4%(P<0.05)、31.6%(P<0.05)。ZSPKN及ZMPKN组相对模型组有改善大鼠脑梗死趋势,改善率分别为18.0%、24.1%,但无显著性差异。结果见表6。

Claims (9)
1.一种人尿激肽原酶的化学修饰物,其特征在于,所述化学修饰为聚乙二醇修饰,所述聚乙二醇的活化基团为羧酸NHS活性酯。
2.根据权利要求1所述的人尿激肽原酶的化学修饰物,其特征在于,所述聚乙二醇采用直链型或分支型的聚乙二醇。
3.根据权利要求2所述的人尿激肽原酶的化学修饰物,其特征在于,所述直链型的聚乙二醇的结构为:
平均分子量为5kDa-40kDa。
4.根据权利要求2所述的人尿激肽原酶的化学修饰物,其特征在于,所述分支型的聚乙二醇的结构为:
平均分子量为10kDa-80kDa。
5.根据权利要求1-4任一权利要求所述的人尿激肽原酶的化学修饰物,其特征在于,所述聚乙二醇修饰为单修饰或多修饰,多修饰聚乙二醇化的人尿激肽原酶中,每个人尿激肽原酶单体平均偶联3-15个聚乙二醇分子。
6.根据权利要求5所述的人尿激肽原酶的化学修饰物,其特征在于,所述多修饰聚乙二醇化的人尿激肽原酶中,每个人尿激肽原酶单体平均偶联3-8个聚乙二醇分子。
7.根据权利要求5所述的人尿激肽原酶的化学修饰物,其特征在于,所述多修饰聚乙二醇化的人尿激肽原酶中聚乙二醇通过酰胺键与人尿激肽原酶中N端伯氨基和赖氨酸残基的氨基连接。
8.根据权利要求5所述的人尿激肽原酶的化学修饰物,其特征在于,所述多修饰聚乙二醇化的人尿激肽原酶的制备方法为:
将人尿激肽原酶冻干粉溶解在二甲亚砜中,加入聚乙二醇粉末,加热反应,产物经色谱柱纯化制得,其中,所述聚乙二醇的摩尔投料量为人尿激肽原酶的8-10倍。
9.根据权利要求5所述的人尿激肽原酶的化学修饰物,其特征在于,单修饰聚乙二醇化的人尿激肽原酶的制备方法为:
将人尿激肽原酶冻干粉溶解在二甲亚砜中,加入聚乙二醇粉末,加热反应,产物经色谱柱纯化制得,其中,所述聚乙二醇的摩尔投料量为人尿激肽原酶的2-4倍。
