基于数据挖掘刘瑞芬教授治疗原发性痛经的用药规律研究,刘学红,公开了治疗原发性痛经的药用规律,主要的中药共16味,分别为当归、川芎、黄芪、续断、木香、白芥子、香附、延胡索、白芍、肉桂、吴茱萸、红花、五灵脂、小茴香、蒲黄、桃仁。按照从高到低顺序排列,出现频次最多的为川芎-当归组合(94 次),其次为当归-白芍组合(92 次),列出频次≥76 次的药对(具体见19页表12上方)。
发明内容
本发明要解决的技术问题是提供一种改善痛经的复合茶包、制备方法和应用,有效的改善痛经。
本发明实施例提供一种改善痛经的复合茶包,包括如下重量百分含量组分,雪莲花多糖8~25%,月季花黄酮5~25%,玫瑰黄酮8~25%,玳玳花总黄酮10~25%,三七花皂苷10~25%。
优选的,包括如下重量百分含量组分,雪莲花多糖15~20%,月季花黄酮20~25%,玫瑰黄酮15~20%,玳玳花总黄酮15~20%,三七花皂苷15~20%。

优选的,所述雪莲花多糖的制备方法为,雪莲花粉末进行乙醇回流以进行脱脂处理,抽滤,干燥,然后用小分子有机溶剂进行抽滤处理,所述小分子有机溶剂为乙醇、或/和丙酮(优选为体积浓度95%乙醇、100%乙醇和丙酮);干燥,加入水进行浸提;然后离心,减压浓缩,然后进行脱蛋白处理;将脱蛋白处理后的样品离心,在上清液中加入无水乙醇,静置,离心,收集沉淀物,加水复溶,冷冻干燥,得到雪莲花多糖。
优选的,所述月季花黄酮的制备方法为,干燥的月季花粉末进行乙醇回流提取,过滤,减压浓缩,加水悬浮后,加石油醚进行洗涤,水相加乙酸乙酯萃取,将乙酸乙酯部分蒸干后加乙醇溶解,得到月季花黄酮。
优选的,所述玫瑰黄酮的制备方法为,将玫瑰花用乙醇浸泡,浓缩,得到玫瑰浸膏,然后进行超临界萃取,干燥后,得到玫瑰黄酮。
优选的,所述玳玳花总黄酮的制备方法为,将玳玳花粉末加水进行超声波萃取,过滤,降压浓缩,加水溶解,冷冻干燥,得到粗提物;然后将粗提物进行大孔树脂柱层析,得到洗脱液,减压浓缩,冷冻干燥,得到玳玳花总黄酮。
优选的,所述三七花皂苷的制备方法为,将三七花和乙醇混合,浸提,滤液减压浓缩,喷雾干燥,得到三七花皂苷。
本发明提供一种所述的改善痛经的复合茶包的制备方法,将雪莲花多糖、月季花黄酮、玫瑰黄酮、玳玳花总黄酮和三七花皂苷混合,得到改善痛经的复合茶包。
本发明提供一种所述的改善痛经的复合茶包的应用,所述改善痛经的复合茶包用于制备改善痛经的物质。
本发明的有益效果是,该复合茶包在一定配比范围内有着协同作用,使得改善痛经的功效更加突出,同时还具有养阴生津、活血化瘀、美容养颜等功效,是一种功效突出、方便饮用的日常饮品。此外,本发明采取的是纯天然药物,不添加任何的有毒添加剂及成瘾性药物,无毒副作用,不成瘾,增加机体免疫力,改善胃肠道微生态环境。本发明作为改善痛经的复合茶包,采用绿色植物作为原料,可方便随身携带、饮用便捷、整体配方合理有效,长期服用可改善痛经问题、提升身体免疫力,其制备方法操作简单,原料优质,适于推广以改善女性痛经问题。
雪莲花性温,味微苦,入肝、脾、肾三经。具有散寒除湿、活血通络、温火助阳、抗炎镇痛、缩宫等功效,民间常用于治疗妇女小腹冷痛、闭经和胎衣不下等症。本发明经过提取,得到雪莲花多糖。
月季花味甘、性温,入肝肾经具有良好的活血调经、疏肝解郁、活血祛瘀、消肿解毒之功效。本发明经过提取,得到月季花黄酮。
玫瑰花有理气解郁,活血散瘀,调经止痛,镇静、安抚、抗抑郁,行气活血,调和脏腑的功效。本发明经过提取,得到玫瑰黄酮。
玳玳花具有强心、镇静、疏肝理气、消食、化痰等功效,玳玳花作为茶饮,微苦,香气浓郁。本发明经过提取,得到玳玳花总黄酮。
三七花性甘凉、具有清热平肝、生津止渴、活血化瘀等功效,具有抗炎、镇痛、镇静、活血化瘀、抗心律失常、清咽润喉和保护肝脏的药理学活性。三七花安全性实验结果表明其安全无毒、无明显脏器损伤和遗传毒性。本发明经过提取,得到三七花总皂苷。
具体实施方式
下面结合实施例对本发明的实施方式作进一步详细描述。以下实施例用于说明本发明,但不能用来限制本发明的范围。
实施例1
本实施例1中的一种改善痛经的复合茶包的必要构成成分包括雪莲花多糖、月季花黄酮、玫瑰黄酮、玳玳花总黄酮、三七花皂苷。
其中雪莲花多糖的制备过程为:称取6.82g藏雪莲花,加入体积浓度95%的乙醇,于85℃水浴锅中冷凝回流3h进行脱脂处理。抽滤后将样本粉末置于60℃的干燥箱中烘干,然后再先后使用体积浓度95%乙醇、100%乙醇和丙酮对样本进行抽滤处理,风干后将样本粉末和蒸馏水按1:40的料液比在90℃条件下搅拌浸提3h,总共提取3次。将提取后的样本在3000rpm条件下离心10 min,收集上清液进行减压浓缩处理(65℃)。随后,利用Sevag法对水提物进行脱蛋白处理,重复5次。将脱蛋白处理后的样本在3000 rmp下离心10min,随后合并上清液,加入3倍体积的无水乙醇,4℃条件下静置48 h。3500rpm离心15min后收集沉淀物,以蒸馏水复溶后对样本进行真空冷冻干燥,得到藏雪莲花多糖。
其中月季花黄酮的制备过程为:将采集的月季花洗净,用滤纸吸干,取33.33g月季花于60 ℃烘干,粉碎,过筛,将烘干样品放入广口瓶中,备用。置于圆底烧瓶中,按一定的液料比例,加入不同浓度的乙醇溶液摇匀后,在一定的温度下进行加热回流提取一定时间,过滤至圆底烧瓶中,于55 ℃减压浓缩至干,加水600 mL悬浮,加石油醚洗涤3次,弃去石油醚部分,水相再加乙酸乙酯萃取3 次,合并乙酸乙酯部分,水浴蒸干,加乙醇溶解后定量转移至容量瓶中,乙醇稀释至刻度即得供试品溶液。
其中玫瑰黄酮的制备过程为:将刚采摘的玫瑰花洗净,用滤纸吸干,取玫瑰花53.33g与95%乙醇按照1:2倍重量比于常温浸泡24 h,经真空浓缩制成玫瑰浸膏。取玫瑰浸膏装入超临界萃取仪的萃取缸中,按照二氧化碳→萃取缸→精柱→分离I→分离Ⅱ→萃取缸的流程分别调整好萃取的温度和压力,各参数稳定时开始计时,3h后停机,首先分离出玫瑰粗油和部分(大量)玫瑰黄酮溶液;将玫瑰粗油收集后按体积比3:2添加夹带剂,再按以上流程进行2次萃取,最终产物是玫瑰精油和玫瑰黄酮溶液(少量);把2次萃取获得的玫瑰黄酮溶液合并收集,经喷雾干燥制成粉末状的玫瑰黄酮固体产物。
其中玳玳花总黄酮的制备过程为:玳玳花3.64g粉碎后,加纯水[液料比 5 ∶1(mL/g)],超声波功率为600W,超声萃取4h,过滤,45 ℃水浴减压旋转浓缩,浓缩至褐色浸膏。经少量纯水重新溶解后,滤纸过滤,冷冻干燥处理得到粗提物样品,4 ℃冰箱备用。取大孔树脂 D101,加入无水乙醇浸泡24h激活。之后将激活的大孔树脂装入长25 cm,直径1.5 cm的层析柱中,用无水乙醇冲洗至洗脱液滴入纯水中无浑浊现象,再用纯水洗脱至洗脱液澄清,管柱上方留1 cm~2 cm 高的液面。称取得到的粗提物样品用少量纯水溶解后,湿法上样。先用纯水200 mL洗脱后,再用25 %的乙醇-水溶液洗脱,此洗脱液经减压浓缩,冷冻干燥后即为玳玳花黄酮。
其中三七花皂苷的制备过程为:以52.9%体积分数乙醇、料液比(m:v)分别为1:21.8、1:50.8重复浸提2次,称取4.44g三七花进行浸提,滤液采用多级闪蒸器在40 ℃、-0.08 MPa条件下减压浓缩至原体积的1/3,以温度120 ℃、风速75%、进料速度8 mL/min喷雾干燥,得到黄绿色的三七花提取物三七花皂苷。
将上述制备得到的雪莲花多糖、月季花黄酮、玫瑰黄酮、玳玳花总黄酮、三七花皂苷按比例(雪莲花多糖15%,月季花黄酮25%,玫瑰黄酮20%,玳玳花总黄酮20%,三七花皂苷20%)进行配伍总含量为2克的茶包。
对比例1
称取当归25g、白芍25g、香附25g、川芎15g、红花12.5g,将上述原料混合,加入10倍水,在30℃下进行浸提60min,过滤,将滤渣和8倍水混合后,在70℃下浸提15min,过滤,合并两次滤液,减压浓缩,干燥,得到提取物。
对比例2
称取雪莲花6.82g,月季花33.33g,玫瑰花53.33g,玳玳花3.64g,三七花4.44g,将上述原料混合,加入10倍水,在30℃下进行浸提60min,过滤,将滤渣和8倍水混合后,在70℃下浸提15min,过滤,合并两次滤液,减压浓缩,干燥,得到提取物。
对比例3
改善痛经的复合茶包的制备方法,包括如下步骤:
(1)首先采摘新鲜的雪莲花6.82g、月季花33.33g、玫瑰花53.33g、玳玳花3.64g和三七花4.44g。
(2)将采摘的雪莲花、月季花、玫瑰花、玳玳花和三七花放入浓度75%盐水中浸泡1小时,将上述的花从盐水中取出。
(3)将浸泡过后的雪莲花、月季花、玫瑰花、玳玳花和三七花进行蒸发处理,蒸发的温度控制在115℃,持续30min,再进行干燥处理;用烘干机分两次高温烘焙,一次烘焙温度90℃,摊花厚度1.5cm,时间25min;二次烘焙75℃,摊花厚度3.0cm,时间25min。
(4)将处理后的雪莲花、月季花、玫瑰花、玳玳花和三七花一起放入混合搅拌设备中进行充分搅拌混合。
(5)最后将搅拌混合的材料放在日光下暴晒,时间控制在48h,然后将晒干处理后的混合物摊放冷却后,用符合卫生标准的复合包装材料按需包装。
将所述雪莲花、所述月季花、所述玫瑰花、所述玳玳花和所述三七花分类分批送入烘炒机内,烘炒时间控制在15min;然后将烘炒好的原料分类分批送入粉碎机进行粉碎,并过100目筛网。
实验例1
随机抽取湖南中医药大学第一附属医院年龄在15-40岁之间的痛经人群,共200人,分为4组,每组50人,饮用实施例1茶包1份,对比例1、对比例2、对比例3各一份。坚持每天日间服用一次,加水200ml,共计3个月,疗效判定标准如表1所示。

①显效:痛经状况明显改善,饮用复合茶包后痛经程度评分减少3分以上;②好转:痛经状况有所改善,饮用复合茶包后痛经程度评分减少3分以下;③无效:痛经状况无明显改善,饮用复合茶包后痛经程度评分无改变。调查结果如下表2所示。

食品的研发是一个复杂的过程,同时由于影响因素众多,所以试验具有偶然性,食品的组分的种类和含量均对食品的功能产生不可预料的影响。发明人在大量的试验下偶然发现由包括实施例在内的技术方案制备的改善痛经的复合茶具有有效改善痛经症状的作用,有效率≥89%,而对比例1改善痛经症状的效果明显不如实施例1组,因此可知本申请的配方,经过特殊的提取工艺后,可以有效的提高效果。本发明的五种原料在一定含量配比范围内具有协同作用,能够有效的改善痛经,而且还具有养阴生津、活血化瘀、美容养颜等功效。
综上,本发明是一种非常理想的保健饮品,口味佳,整体加工工艺方法简单,原料简单且成本低,适于推广。

1.一种改善痛经的复合茶包,其特征是,包括如下重量百分含量组分,雪莲花多糖8~25%,月季花黄酮5~25%,玫瑰黄酮8~25%,玳玳花总黄酮10~25%,三七花皂苷10~25%。
2.如权利要求1所述的改善痛经的复合茶包,其特征是,包括如下重量百分含量组分,雪莲花多糖15~20%,月季花黄酮20~25%,玫瑰黄酮15~20%,玳玳花总黄酮15~20%,三七花皂苷15~20%。
3.如权利要求1所述的改善痛经的复合茶包,其特征是,包括如下重量百分含量组分,雪莲花多糖15%,月季花黄酮25%,玫瑰黄酮20%,玳玳花总黄酮20%,三七花皂苷20%。
4.如权利要求1-3任一项所述的改善痛经的复合茶包,其特征是,所述雪莲花多糖的制备方法为,雪莲花粉末进行乙醇回流以进行脱脂处理,抽滤,干燥,然后用小分子有机溶剂进行抽滤处理,所述小分子有机溶剂为乙醇、或/和丙酮;干燥,加入水进行浸提;然后离心,减压浓缩,然后进行脱蛋白处理;将脱蛋白处理后的样品离心,在上清液中加入无水乙醇,静置,离心,收集沉淀物,加水复溶,冷冻干燥,得到雪莲花多糖。
5.如权利要求1-3任一项所述的改善痛经的复合茶包,其特征是,所述月季花黄酮的制备方法为,干燥的月季花粉末进行乙醇回流提取,过滤,减压浓缩,加水悬浮后,加石油醚进行洗涤,水相加乙酸乙酯萃取,将乙酸乙酯部分蒸干后加乙醇溶解,得到月季花黄酮。
6.如权利要求1-3任一项所述的改善痛经的复合茶包,其特征是,所述玫瑰黄酮的制备方法为,将玫瑰花用乙醇浸泡,浓缩,得到玫瑰浸膏,然后进行超临界萃取,干燥后,得到玫瑰黄酮。
7.如权利要求1-3任一项所述的改善痛经的复合茶包,其特征是,所述玳玳花总黄酮的制备方法为,将玳玳花粉末加水进行超声波萃取,过滤,降压浓缩,加水溶解,冷冻干燥,得到粗提物;然后将粗提物进行大孔树脂柱层析,得到洗脱液,减压浓缩,冷冻干燥,得到玳玳花总黄酮。
8.如权利要求1-3任一项所述的改善痛经的复合茶包,其特征是,所述三七花皂苷的制备方法为,将三七花和乙醇混合,浸提,滤液减压浓缩,喷雾干燥,得到三七花皂苷。
9.一种如权利要求1-8任一项所述的改善痛经的复合茶包的制备方法,其特征是,将雪莲花多糖、月季花黄酮、玫瑰黄酮、玳玳花总黄酮和三七花皂苷混合,得到改善痛经的复合茶包。
10.一种如权利要求1-8任一项所述的改善痛经的复合茶包的应用,其特征是,所述改善痛经的复合茶包用于制备改善痛经的物质。
