技术领域
本发明涉及抗原结合分子与链霉亲和素突变体的融合蛋白、以及其利用。
背景技术
亲和素与生物素、或者链霉亲和素与生物素之间的亲和性非常高(Kd=10-15-10- 14M),作为生物双分子间的相互作用,为最强的相互作用之一。目前,亲和素/链霉亲和素-生物素相互作用在生物化学、分子生物学或者医学领域被广泛应用。已经设计了将亲和素/链霉亲和素与生物素的高结合能力和抗体分子组合的药物递送方法和预靶向法。关于这些研究,专利文献1中报道了:降低了对天然生物素的亲和性的链霉亲和素突变体、以及对于该对天然生物素呈低亲和性的链霉亲和素突变体具有高亲和性的生物素修饰二聚体。

发明内容
发明所要解决的问题
本发明以提供用于在癌症的治疗或诊断中使用的、识别癌细胞等的分子与链霉亲和素突变体的融合蛋白作为所应解决的课题。进一步地,本发明以提供使用了上述融合蛋白的、癌症的治疗手段或癌症的诊断手段作为所应解决的课题。
用于解决课题的手段
为了解决上述课题,本发明人进行了深入研究,结果选择了分子量小于抗体的分子作为识别癌细胞的分子,调制了上述分子与链霉亲和素突变体的融合蛋白。并且,发现了通过使用上述融合蛋白、和生物素修饰二聚体与酞菁染料的缀合物的光免疫疗法,可抑制癌细胞的增殖(生长),从而完成了本发明。
即,根据本发明,提供以下发明。
<1>融合蛋白,其中,分子量20,000以下的抗原结合分子经由接头序列结合在序列号1(SEQ ID NO:1)中记载的氨基酸序列(但C末端的由Pro-Ser-Ala-Ala-Ser His-His-His-His-His-His构成的氨基酸序列可部分或全部缺失)的N末端侧和/或C末端侧。

<抗原结合分子与链霉亲和素突变体的融合蛋白>
本发明的融合蛋白为下述的融合蛋白,其中,分子量20,000以下的抗原结合分子经由接头序列结合在序列号1中记载的氨基酸序列(但C末端的由Pro-Ser-Ala-Ala-SerHis-His-His-His-His-His构成的氨基酸序列(或C末端的由6个组氨酸构成的氨基酸序列)可部分或全部缺失)的N末端侧和/或C末端侧。在分子量20,000以下的抗原结合分子经由接头序列结合在序列号1中记载的氨基酸序列(但C末端的由Pro-Ser-Ala-Ala-Ser His-His-His-His-His-His构成的氨基酸序列可部分或全部缺失)的N末端侧和C末端侧两者的情况下,该抗原结合分子可为相同的,也可为不同的。
优选地,本发明的融合蛋白为从N末端侧到C末端侧依次具有分子量20,000以下的抗原结合分子、接头序列和序列号1中记载的氨基酸序列(但C末端的由Pro-Ser-Ala-Ala-Ser His-His-His-His-His-His构成的氨基酸序列可部分或全部缺失)的融合蛋白。
序列号1中记载的氨基酸序列为链霉亲和素突变体的氨基酸序列,具体而言,为国际公开WO2015/125820的实施例3(国际公开WO2015/125820的序列号4)(本申请说明书的序列号1)中记载的链霉亲和素突变体LISA314-V2122。
如上所述,本发明的融合蛋白为分子量20,000以下的抗原结合分子与链霉亲和素突变体的融合蛋白。本发明的融合蛋白通过序列号1中记载的氨基酸序列彼此的亲和性而形成四聚体。作为抗原结合分子,例如,在使用具有实施例2中记载的氨基酸序列的蛋白的情况下,本发明融合蛋白的四聚体的分子量为约96kDa。在将如本发明这样的融合蛋白给予生物体以进行癌症治疗的情况下,同时实现向肿瘤的摄取(tumor uptake)良好、清除速度(clearance rate,清除率)良好、以及良好的向肿瘤的渗透(tumor penetration)良好是重要的。设想本发明中上述四聚体的分子量(约96kDa)为可同时实现上述3个参数的分子量。
抗原结合分子的分子量只要为20,000以下即可,但一般而言,抗原结合分子的分子量为4,000以上且20,000以下,优选为4,000以上且10,000以下,更优选为4,000以上且8,000以下。
本发明中的抗原结合分子为与抗体概念不同的分子。相对于IgG抗体的分子量通常为150,000左右,本发明中的抗原结合分子为具有远小于IgG抗体的分子量的分子。作为本发明中的抗原结合分子,也可为模仿抗体而制备的分子。例如,可从将蛋白A的IgG结合结构域(VDNKFNKEQQNAFYEILHLPNLNEEQRNAFIQSLKDDPSQSANLL AEAKKLNDAQAPK)(序列号9)的一部分修饰而制备的分子量约6,000的蛋白中,通过筛选来适当选择与所期望抗原结合的分子。
作为抗原结合分子中的抗原没有特别限定,但优选地,在癌细胞中表达的抗原为优选的。作为在癌症中特异性表达的抗原,可列举出以下的抗原。

作为抗原结合分子的一个实例,可与HER2结合。可列举具有序列号2中记载的氨基酸序列的蛋白。
接头序列只要可实现本发明的效果则没有特别限定,但氨基酸数目优选为5至25个氨基酸,更优选为10至25个氨基酸,进一步优选为15至20个氨基酸。
作为接头序列的具体实例,可列举由甘氨酸残基和丝氨酸残基构成的序列。作为接头序列,可使用例如以[(Gly)m-Ser]n(式中,m表示1~10的整数,n表示1~5的整数)表示的氨基酸序列。作为接头序列的具体实例,可列举Gly-Gly-Gly-Gly-Gly-Ser-Gly-Gly-Gly-Gly-Gly-Ser-Gly-Gly-Gly-Gly-Gly-Ser,但没有特别限定。

根据本发明,进一步提供编码上述的本发明融合蛋白的核酸(例如DNA)。作为本发明核酸的具体实例,可列举编码具有序列号4中记载的氨基酸序列(但C末端的由Pro-Ser-Ala-Ala-Ser His-His-His-His-His-His构成的氨基酸序列可部分或全部缺失)的融合蛋白的核酸。作为本发明核酸的一个实例,可列举具有序列号3中记载的碱基序列的核酸。
编码本发明融合蛋白的核酸(例如DNA)可整合到载体中使用。为了制造本发明的融合蛋白,可将编码本发明融合蛋白的核酸整合到表达载体中,将该表达载体转化到宿主中,由此表达本发明的融合蛋白。即,根据本发明,提供本发明融合蛋白的制造方法,其包括使编码本发明融合蛋白的核酸在宿主中表达的工序。融合蛋白优选可在细菌的包涵体中表达并回收。
在以大肠杆菌为宿主的情况下,作为载体,优选具有复制起点(ori)并且具有用于选择被转化的宿主的基因(例如针对氨苄青霉素、四环素、卡那霉素或氯霉素等药剂的耐药性基因等)。另外,在表达载体的情况下,优选携带使得可在宿主中高效表达本发明的链霉亲和素突变体的启动子,例如lacZ启动子或T7启动子等。作为这样的载体,作为载体的实例,可列举M13系载体、pUC系载体、pBR322、pBluescript、pCR-Script、pGEX-5X-1(Pharmacia)、“QIAexpress系统”(QIAGEN)、pEGFP或pET(这种情况下,优选宿主使用表达T7RNA聚合酶的BL21)等。

在以于CHO细胞、COS细胞、NIH3T3细胞等动物细胞中的表达为目的的情况下,携带在细胞内表达所必需的启动子、例如SV40启动子(Mulligan等人,Nature(1979)277,108)、MMLV-LTR启动子、EF1α启动子(Mizushima等人,Nucleic Acids Res.(1990)18,5322)、CMV启动子等是必不可少的,进一步优选具有用于挑选向细胞内转化的基因(例如可通过药剂(新霉素、G418等)辨别的耐药性基因)。作为具有这样的特性的载体,可列举例如pMAM、pDR2、pBK-RSV、pBK-CMV、pOPRSV、pOP13等。
作为导入载体的宿主细胞没有特别限制,可为原核生物和真核生物中的任一种。例如,可使用大肠杆菌或各种动物细胞等。
在使用真核细胞的情况下,例如可将动物细胞、植物细胞、真菌细胞用于宿主。作为动物细胞,可使用哺乳类细胞,例如CHO细胞、COS细胞、3T3细胞、HeLa细胞、Vero细胞;或者昆虫细胞,例如Sf9、Sf21、Tn5等。在动物细胞中,在以大量表达为目的的情况下,特别优选CHO细胞。载体向宿主细胞中的导入可通过例如磷酸钙法、DEAE葡聚糖法、使用了阳离子脂质体DOTAP(Boehringer Mannheim公司制造)的方法、电穿孔法、脂质转染等方法来进行。
作为植物细胞,例如烟草(Nicotiana tabacum)来源的细胞作为蛋白生产系统而已知,将其进行愈伤组织培养即可。作为真菌细胞,已知酵母,例如酵母(Saccharomyces)属,例如酿酒酵母(Saccharomyces cerevisiae);丝状菌,例如曲霉(Aspergillus)属,例如黑曲霉(Aspergillus niger)。
在使用原核细胞的情况下,可列举大肠杆菌(E.coli),例如JM109、DH5α、HB101等,此外还已知枯草芽孢杆菌。
将这些细胞利用本发明的核酸进行转化,将所转化的细胞在体外进行培养,由此得到本发明的融合蛋白。培养可按照公知的方法来进行。例如,作为动物细胞的培养液,可使用例如DMEM、MEM、RPMI1640、IMDM。此时,可并用胎牛血清(FCS)等血清补液,也可进行无血清培养。培养时的pH优选为约6~8。培养通常在约30~40℃下进行约15~200小时,根据需要增加培养基的交换、通气、搅拌。另外,也可进行用于促进细胞增殖的生长因子的添加。
<癌症治疗剂或癌症诊断剂>
本发明的融合蛋白作为癌症治疗剂或癌症诊断剂是有用的。
根据本发明,提供癌症的治疗或诊断试剂盒,其包含:(1)本发明的融合蛋白;和(2)以下述式(1)表示的化合物或其盐与诊断用物质或治疗用物质的缀合物。
作为抗原结合分子,在使用与存在于癌细胞中的抗原结合的分子的情况下,可通过将本发明的融合蛋白给予患者,可在癌细胞中特异性地聚集链霉亲和素突变体。接着,通过将对链霉亲和素突变体具有亲和性的生物素修饰二聚体与诊断用物质或治疗用物质的缀合物给予患者,可使诊断用物质或治疗用物质准确地向癌细胞中聚集。
或者,也可调制使“本发明的融合蛋白”和“对链霉亲和素突变体具有亲和性的生物素修饰二聚体与诊断用物质或治疗用物质的缀合物”结合而成的复合物,并将该复合物给予患者。

<光免疫疗法>
光免疫疗法是为了在体内破坏特定细胞而使用光敏剂和照射光的治疗方法。光敏剂在暴露于特异性波长的光时产生可诱发附近细胞的凋亡、坏死和/或自噬作用的细胞毒性的活性氧物种(Reactive Oxygen Species)。例如,在日本专利6127045号公报中记载了使细胞死亡的方法,该方法包括:使包含细胞表面蛋白的细胞与治疗有效量的1种或多种抗体-IR700分子接触的步骤,且该抗体与该细胞表面蛋白特异性结合的步骤;以660~740nm的波长、且以至少1Jcm-2的放射剂量(辐射剂量)对该细胞进行照射的步骤;以及在对该细胞照射的约0~8小时后,使该细胞与1种或多种治疗剂接触,由此使该细胞死亡的步骤。在日本特表2017-524659号公报中记载了在患有疾病或病态的受试者中诱发细胞毒性方法,该方法包括:(a)对该受试者给予对治疗有效的药剂,所述药剂包含缀合至与受试者的细胞特异性结合的探针上的IRDye(注册商标)700DX等酞菁染料;以及(b)以有效诱发细胞死亡的量对上述细胞照射适当的激发光。
通过将本发明的融合蛋白和生物素修饰二聚体与酞菁染料的缀合物给予受试者,以有效诱发细胞增殖的抑制或细胞死亡的量对细胞照射激发光,可诱发细胞增殖的抑制或细胞死亡,来治疗受试者。
优选地,通过将本发明的融合蛋白和生物素修饰二聚体与酞菁染料的缀合物给予受试者,以有效诱发细胞增殖的抑制或细胞死亡的量对细胞照射激发光,可诱发细胞增殖的抑制或细胞死亡,来治疗受试者。
作为受试者,包括人和非人哺乳动物,可列举人或者小鼠等实验动物。作为受试者,优选患有期望诱发细胞增殖的抑制或细胞死亡的疾病的受试者,例如可列举具有癌症或实体瘤的受试者。

实体瘤无论是良性还是恶性通常是指不含包囊的异常细胞块。作为实体瘤,可列举神经胶质瘤、星形细胞瘤、髓母细胞瘤(medulloblastoma)、颅咽管瘤、室管膜瘤、松果体瘤、血管母细胞瘤、听神经瘤、少突神经胶质瘤、脑脊膜瘤、黑色素瘤、神经母细胞瘤(neuroblastoma)和视网膜母细胞瘤。
作为对受试者的给予方法,可列举局部途径、注射(皮下注射、肌肉内注射、皮内注射、腹腔内注射、肿瘤内注射和静脉内注射等)、口服途径、眼途径、舌下途径、直肠途径、经皮途径、鼻腔内途径、阴道途径和吸入途径等,但并不限定于这些。
优选将生物素修饰二聚体与酞菁染料的缀合物、以及本发明的融合蛋白分别以治疗有效量给予。
关于上述的缀合物以及融合蛋白的各自的治疗有效量,为每60千克至少0.5毫克(mg/60kg)、至少5mg/60kg、至少10mg/60kg、至少20mg/60kg、至少30mg/60kg、至少50mg/60kg。例如,在静脉内给予的情况下,为1mg/60kg、2mg/60kg、5mg/60kg、20mg/60kg或50mg/60kg的用量等,例如为0.5~50mg/60kg。在其他实例中,治疗有效量为至少100μg/kg、至少500μg/kg或至少500μg/kg等,至少10μg/kg,例如在肿瘤内给予或腹腔内给予的情况下,为100μg/kg、250μg/kg、约500μg/kg、750μg/kg或1000μg/kg的用量等,例如为10μg/kg~1000μg/kg。在一个实例中,治疗有效量为以局部用溶液给予的10μg/ml、20μg/ml、30μg/ml、40μg/ml、50μg/ml、60μg/ml、70μg/ml、80μg/ml、90μg/ml或100μg/ml等,20μg/ml~100μg/ml之间等,至少500μg/ml等、至少1μg/ml。
可将上述的给予量以1次或多次的分割用量(2、3或4次用量等)或者以单一制剂给予。
生物素修饰二聚体与酞菁染料的缀合物、以及本发明的融合蛋白分别可单独给予,也可在药学上可接受的载体的存在下给予,也可在其他治疗剂(其他抗癌剂等)的存在下给予。
生物素修饰二聚体与酞菁染料的缀合物、以及本发明的融合蛋白可与循环的肿瘤细胞或实体瘤的细胞等靶细胞或靶组织结合。之后,若照射光,则上述缀合物或复合物会吸收光,可损伤或破坏靶细胞或组织。
在光免疫疗法中,照射光的波长优选为660~740nm,例如具有660nm、670nm、680nm、690nm、700nm、710nm、720nm、730nm或740nm的波长。光的照射可通过使用具备近红外(NIR)发光二极管的设备来进行。
光的照射量为至少1J/cm2,例如至少4J/cm2、至少10J/cm2、至少15J/cm2、至少20J/cm2、至少50J/cm2或至少100J/cm2,例如1~500J/cm2。光照射可实施多次(例如2、3、4、5、6、7、8、9或10次)。

<HER2识别蛋白的调制方法>
将Cupid分子(序列号1)(国际公开WO2015/125820)与识别HER2抗原的分子(Lofblom,J.FEBS Lett.584(12):2670-80(2010))(序列号2)融合,将与Psyche分子(上述的Psyche J)和Her2/ErbB2两者具有结合性的蛋白(以下表示为FL)在大肠杆菌中进行合成。FL的基因序列(序列号3)在Eurofins公司进行人工基因合成(序列号4)。使用pET45b作为表达载体,按照规定方法进行表达载体的制备。具体而言,使用引物对(Rv:catggtatatctccttcttaaagttaaac(序列号5),Fw:cgcagcttaattaacctaggctgctgccac(序列号6)),通过PCR反应进行载体的直链化。通过人工基因合成调制的序列使用引物对(Fw:aggagatataccatgGTGGACAACAAATTCAACAAAGAG(序列号7),Rv:gttaattaagctgcgTTAATGATGGTGGTGATGATGCGATG(序列号8)),通过PCR反应进行扩增。PCR反应物均通过琼脂糖凝胶电泳切下条带进行纯化。使用In-Fusion HD克隆试剂盒(TaKaRa Bio),所纯化的载体和插入物的连接反应按照说明书的用法容量实施克隆。
进行克隆、序列分析,将确认整合了目标基因的质粒载体导入感受态细胞BL21(DE3)(ECOS感受态大肠杆菌BL21(DE3),NIPPON GENE公司)进行转化。将在100mL的2×YT培养基中培养过夜的培养液接种(食菌)到1升培养液中,在37℃下进行培养,在600nm的OD值变为0.5~0.8的时间点添加IPTG以使最终浓度为0.5mM,在37℃下进行4小时培养后,通过离心(7500×g,在4℃下20分钟)回收菌体。
关于从菌体中回收IB的方法如下所示。向B-PER(Thermo SCIENTIFIC公司)中添加Benzonase(Merck)并悬浮后,在室温下培养10分钟,通过离心来分离不溶性级分(以下将包涵体表示为IB)和可溶性级分,回收IB。然后,将所回收的IB用稀释10倍的B-PER缓冲液(未添加Benzonase)再悬浮并离心后,弃去上清,将IB的洗涤重复3次,在3次洗涤后,将通过离心回收的IB用超纯水(MilliQ)再悬浮,向1.5mL管中各分注1mL,在-80℃下冷冻保存。
关于IB的变性和解旋如下所示。向IB中添加变性缓冲液(denature buffer)(6M盐酸胍(guanidium HCl)、200mM NaCl、50mM Tris-HCl、1mM EDTA;在4℃下pH 8.0),通过移液模式(pipetting)进行溶解,边用旋转器搅拌边在4℃下培养过夜,离心(15,000×g,在4℃下20分钟)并回收上清。用变性缓冲液调制以使所回收的上清的蛋白浓度(OD280 nm)为30至50mg/mL,边搅拌稀释缓冲液(PBS或200mM NaCl,50mM Tris-HCl,1mM EDTA;在4℃下pH8.0)边将经浓度调制的变性蛋白溶液以成为500倍稀释的分量滴加。之后在4℃下培养24~48小时,通过Ni-NTA柱(cOmplete,Merck)进行纯化。接着,使用离心式超滤过滤器,进行浓缩直至变为5~10mg/mL,使用凝胶过滤纯化柱(HiLoad 16/60Superdex 75pg,GEHealthcare),用PBS进行凝胶过滤纯化。纯化级分的结果显示于图1。
<FL的性能评价>
关于所纯化的FL,使用SPR(BiacoreT200,Cytiva)对于与HER2(ErbB2)的结合活性进行分析。在传感器芯片CM5(Cytiva)上用胺偶联法(胺偶联试剂盒,Cytiva)进行Her2抗原(重组人ErbB2/Her2 Fc嵌合蛋白,R&D SYSTEMS)的固定化。接着,使用FL的稀释系列(1E-08M至6.25E-10M的2倍稀释系列)实施结合活性的测定。实施结果显示于图2。结果确认到显示更稳定的结合。
关于纯化的FL,使用SPR(BiacoreT200,Cytiva)对于与Psyche的结合活性进行分析。在传感器芯片CM5(Cytiva)上用胺偶联法将FL固定化,与将Psyche稀释系列(1E-08M至6.25E-10M的2倍稀释系列,5个系列)作为分析物的结合活性的分析用单循环动力学法实施。实施结果显示于图3。结果确认到显示更稳定的结合。
<使用FL和光敏剂Psyche的细胞毒活性>
在96孔板上接种Her2阳性细胞(SK-BR-3,KPL-4)以使细胞数为1×104个细胞/孔、培养液为50μL/孔,进行过夜培养。将FL与光敏剂Psyche(上述制造例6中制造的化合物8)混合以使摩尔比为1:2(以下表示为复合物),制备从10μg/mL起2倍稀释系列的12个系列。稀释系列调整后,将复合物添加至细胞中,24小时后,从培养平板底面以100J/cm2用发出690nm的光的LED进行光照射。之后,培养24小时,使用细胞计数试剂盒-8(同仁化学公司)进行活细胞数的比较。用法用量按照使用说明书,在添加试剂的1.5小时于37℃、CO2的培养箱中进行培养,之后测定450nm的吸光度,计算平均值并背景校正后,以对照作为100%,计算了各条件下的细胞增殖与对照的比例。实施结果显示于图4。确认到FL与光敏剂Psyche化合物的复合物具有浓度依赖性的细胞毒性。
<体内实验>
将在含有10%FBS的DMEM中进行培养的KPL-4细胞以5×106至10×106个细胞/身体,向裸鼠(5~10周龄)的皮下或者第二乳腺进行移植。移植后,向肿瘤块的体积达50至200mm3的小鼠,将FL与光敏剂Psyche的复合物以75至150μg/身体,从尾静脉进行给予。作为对照药,将赫赛汀(中外制药)以150μg/身体从尾静脉进行给予。在给予5至6小时后,对于给予FL与光敏剂Psyche复合物的小鼠,对肿瘤部分以230J/cm2使用发出690nm的光的LED(USHIO公司),对肿瘤块部分进行光照射。光照射后,进行体重的测定、肿瘤块体积的测定。具体的肿瘤块体积测定,用游标卡尺测定肿瘤块的长径和短径,将通过计算式:(短径)2×长径×1/2计算的值作为肿瘤块的体积。实施结果显示于图5。确认到FL与光敏剂Psyche化合物的复合物具有肿瘤增殖抑制效果。
<使用FL光敏剂Psyche的体内实验2皮下移植肿瘤的治疗>
将在含有10%FBS的DMEM中进行培养的KPL-4细胞以5×106至10×106个细胞/身体,向裸鼠(5~10周龄)的皮下进行移植。移植后,向肿瘤块的体积达到500至800mm3的小鼠,将FL与光敏剂Psyche的复合物以150μg/身体,从尾静脉进行给予。如图6A中所示,在药剂给予起20小时后和68小时后,使用发出690nm的光的LED(USHIO公司),对小鼠的肿瘤部分以230J/cm2在麻醉下进行光照射。光照射后,进行体重的测定、肿瘤块体积的测定。具体的肿瘤块的体积测定,用游标卡尺测定肿瘤块的长径和短径,将通过计算式:(短径)2×长径×1/2计算的值作为肿瘤块的体积。如图6B、图6C中所示,从光照射后立刻起可见肿瘤回缩,在14周后,肿瘤消失,皮肤损伤也消失,确认到治疗效果。
<表达载体的构建>
对于上述<HER2识别蛋白的调制方法>中调制的表达载体,使用PCR,通过使目标基因序列缺失的位点特异性变异导入法,制备了用于制备具有去除了序列号4中记载的氨基酸序列的C末端的11个氨基酸“PSAASHHHHHH”的序列号10中记载的氨基酸序列的FL2-G5Sx3-del5的表达载体。具体而言,使用引物对(Fw:AAGTCAAATAACGCAGCTTAATTAACCTAG(序列号11),Rw:TGCGTTATTTGACTTTGGTAAAGGTGTCATG(序列号12)),将表达载体作为模板进行PCR反应。之后,反应液的DpnI处理在37℃下进行1小时,用该反应液转化大肠杆菌并进行克隆。克隆后,进行序列分析,确认整合了序列号13中记载的基因序列,将载体名设为pET45-FL2-G5Sx3-del5。
接着,进行了FL2-G5Sx3-del5中连接识别HER2抗原的分子与Cupid-del5的氨基酸接头的长度不同的蛋白表达载体的构建。具体而言,将组合4个甘氨酸和1个丝氨酸的肽作为基本单元,制备了具有1个单元、2个单元、3个单元的长度的接头的表达载体。制备方法如下。
用引物对(Fw:GCGGAAGCCGGTATTACCGGGACCTGGTC(序列号14),Rw:TTTCGGAGCTTGAGCATCATTCAGTTTC(序列号15))进行了pET45-FL2-G5Sx3-del5的直链化。如下合成互补链的寡聚物,以使得表达各长度的氨基酸接头,95℃反应10分钟,通过自然冷却进行退火。

接着,使用In-Fusion HD克隆试剂盒(TaKaRa Bio),将所纯化的载体与经退火的寡聚物的连接反应按照说明书的用法容量实施克隆。确认整合了表达具有序列号22、序列号23、序列号24中记载的各接头长度的蛋白的基因序列(序列号25、序列号26、序列号27)的表达载体,分别命名为pET45-FL2-G4Sx1-del5、pET45-FL2-G4Sx2-del5、pET45-FL2-G4Sx3-del5。
<蛋白表达和纯化法的研究>
首先,推进FL2-G5Sx3-del5的表达、纯化的研究。将序列排列被确认为正确的质粒载体导入感受态细胞BL21(DE3)(ECOS感受态大肠杆菌BL21(DE3),NIPPON GENE公司)中进行转化。将在100mL的2×YT培养基中培养过夜的培养液接种(食菌)到1升的培养液中,在37℃下进行培养,在600nm的OD值变为0.5-0.8的时间点添加IPTG以使最终浓度为0.5mM,在37℃下进行4小时培养后,通过离心(7500×g,在4℃下20分钟)回收菌体。

关于IB的变性和解旋如下所示。向IB中添加变性缓冲液(0.1MTris-HCl,pH8.5,6M盐酸胍,10mM EDTA),通过移液模式进行溶解,边用旋转器搅拌边在4℃下培养过夜,离心(15,000×g,在4℃下20分钟)并回收上清。用变性缓冲液调制以使所回收的上清的蛋白浓度(OD280 nm)为30-50mg/mL,边搅拌稀释缓冲液(PBS或200mM NaCl,50mM Tris-HCl,1mMEDTA;在4℃下pH 8.0)边将经浓度调制的变性蛋白溶液以成为500倍稀释的分量滴加。之后,在4℃下培养24~48小时,通过Ni-NTA柱(cOmplete,Merck)进行纯化。接着,使用离心式超滤过滤器,进行浓缩直至变为5~10mg/mL,使用凝胶过滤纯化柱(HiLoad 16/60Superdex75pg,GE Healthcare),用PBS进行凝胶过滤纯化。
<FL2-G5Sx3-del5的解旋研究>
对于FL2-G5Sx3-del5,用与<HER2识别蛋白的调制方法>中IB的变性和解旋中记载的相同的再折叠缓冲液尝试解旋。明确了在稀释时大量凝集,难以形成4聚体。为了解决该问题,推进通过pH的4聚体形成的优化(图7)。在于再折叠缓冲液(50mM磷酸钠,0.4M精氨酸盐酸盐,pH5.5)中边搅拌边将经浓度调制的变性蛋白溶液以成为40-80倍稀释的分量滴加的情况下,在早期确认到4聚体形成(图7A)。通过进一步在4℃下培养48小时,确认到大部分的单体消失。另外,确认到在pH 5.5下形成4聚体的FL-del5之后即使被置换为pH 6.8-pH7.5的缓冲液也维持4聚体(图7B)。
接着,进行离心(12,000×g,在4℃下20分钟),除去不溶化的蛋白,回收上清,使用离心式超滤过滤器,进行浓缩直至变为5-10mg/mL,使用凝胶过滤纯化柱(HiLoad 16/60Superdex 75pg,GE Healthcare),用凝胶过滤缓冲液(0.1M磷酸钠、0.2M精氨酸盐酸盐,pH6.8)进行凝胶过滤纯化。
<FL2-G4Sx1-del5、FL2-G4Sx2-del5和FL2-G4Sx3-del5的表达和解旋>
对于FL2-G4Sx1-del5、FL2-G4Sx2-del5和FL2-G4Sx3-del5,也用与FL2-G5Sx3-del5相同的方法调制IB,使用pH 5.5的再折叠缓冲液,通过进行解旋来调制4聚体(图8)。
<使用FL2-G5Sx3-del5、FL2-G4Sx1-del5、FL2-G4Sx2-del5和FL2-G4Sx3-del5与光敏剂Psyche的细胞毒活性的确认>

<试剂等>

<重组ZHER2:342-Cupid-His的调制>
重组ZHER2:342-Cupid-His蛋白与上述<HER2识别蛋白的调制方法>中记载的相同,是通过使用变性剂的变性和直接稀释再折叠法从大肠杆菌包涵体制造的。关于重组ZHER2:342-Cupid-His蛋白的表达和纯化,迄今已详述(Sugiyama A,Kawamura T,Tanaka T等人,Cupidand Psyche system for the diagnosis and treatment of advanced cancer.Proc JpnAcad Ser B Phys Biol Sci.2019;95:602-611.doi:10.2183/pjab.95.041)。简而言之,将大肠杆菌株BL21(DE3)用表达载体pET45b(+)-ZHER2:342-Cupid-His转化,作为包涵体(IB)表达重组ZHER2:342-Cupid-His蛋白。使纯化IB可溶化于变性缓冲液。将可溶化的IB溶液使用离心分离(12,000×g,在4℃下15分钟)澄清化,用再折叠缓冲液直接使用40倍稀释进行再折叠。在4℃下培养72小时后,使用凝胶过滤柱(HiLoad 16/600Superdex 75pg,#28989333,Cytiva,Marlborough,MA,USA),纯化在凝胶过滤缓冲液中再折叠的四聚体重组ZHER2:342-Cupid-His蛋白。四聚体重组ZHER2:342-Cupid-His蛋白的纯度通过非还原(无沸腾和无还原剂)SDS-PAGE(TGX Stain-Free FastCast 12%,#1610185,Bio-Rad,Hercules,CA,USA)分析。四聚体重组ZHER2:342-Cupid-His蛋白与HER2和Psyche-Ax-SiPC的结合活性使用基于Biacore T200(Cytiva,Marlborough,MA,USA)的表面等离子体共振进行分析。
<体内乳腺癌肿瘤模型实验>
<动物模型的调制>
将KPL-4细胞株维持在DMEM(低葡萄糖)(FUJIFILM Wako Pure ChemicalCorporation,Osaka,Japan)中补充有10%FBS、100U/mL青霉素和100μg/mL链霉素(#15140122,Thermo Fisher Scientific,美国,MA,Waltham)的培养基中(Kurebayashi J,Otsuki T,Tang CK等人,Isolation and characterization of a new human breastcancer cell line,KPL-4,expressing the Erb B family receptors and interleukin-6.Br JCancer.1999;79:707-717.doi:10.1038/sj.bjc.6690114)。将KPL-4细胞(750万)皮下移植到BALB/cSlc-nu/nu裸鼠(Sankyo Labo Service Corporation,INC,东京,日本)大腿部。皮下肿瘤增殖通过使用游标卡尺测定肿瘤体积(0.5×长度×宽度2)来监测,作为治疗相关毒性的指标监测了动物体重。20只小鼠的肿瘤大小在移植后44天增加,达到约400mm3。将这20只小鼠分别随机分类成Kadcyla(注册商标)组和ZHER2:342-Cupid-His-Ax-SiPC组的2组的10只小鼠。
<体内实验计划>
在第0天将Kadcyla(注册商标)和ZHER2:342-Cupid-His-Ax-SiPC(图10A)注入至异种移植模型小鼠。本试验中,为了评价肿瘤的复发,将在最初的治疗起约19日后设定为观察期间。在肿瘤复发的情况下,在初次治疗前的同一时期、即癌细胞增殖的复发确认期(初次治疗后第63天)的44天后,实施第2次治疗。最后,在治疗后第97天,第2次治疗后经过19天以上后,将异种移植模型小鼠宰杀。图10B显示实验计划的示意图。
<治疗药的调制>
按照制造商的指示制成Kadcyla(注册商标)。ZHER2:342-Cupid-His和Psyche-Ax-SiPC的调制为已报道的(Yamatsugu K,Katoh H,Yamashita T等人,Antibody mimeticdrug conjugate manufactured by high-yield Escherichia coli expression andnon-covalent binding system.Protein Expr Purif.2022;192:106043.doi:10.1016/j.pep.2021.106043)。简而言之,将以5mM的浓度可溶化于二甲基亚砜中的Psyche-Ax-SiPC作为储备溶液在-80℃下保存。为了调制络合物,将ZHER2:342-Cupid-His和Psyche-Ax-SiPC在冰上的暗处以1:2的摩尔比混合10分钟。使用磷酸缓冲盐水来稀释ZHER2:342-Cupid-His-Ax-SiPC的浓缩物。
<模型小鼠的治疗>
将Kadcyla(注册商标)和ZHER2:342-Cupid-His-Ax-SiPC以300μg和150μg给予。注射20小时后,对ZHER2:342-Cupid-His-Ax-SiPC组的小鼠以230J cm-2照射690nm的发光二极管灯(Yamato Scientific公司,东京,日本)。在最初光照射48小时后,再次同样地照射肿瘤。
<病理学分析>
从小鼠获得来自异种移植肿瘤部位的皮肤试样以及来自包括肺、心脏、肾脏、肝脏和消化道在内的其他重要器官的试样,将这些试样在4%多聚甲醛磷酸缓冲液(#163-20145,FUJIFILM Wako Pure Chemical Corporation,Osaka,Japan)中在4℃下固定24小时,然后按照标准组织病理学程序包埋于石蜡中。H&E染色通过将组织病理学试验片在室温下浸泡于二甲苯(#241-00091、FUJIFILM和光纯化学株式会社,大阪,日本)中10分钟而脱石蜡化,然后通过在乙醇(#057-00451,FUJIFILM和光纯化学株式会社,大阪,日本)中浸泡而再水合。按照制造商的方案将苏木精(#6187-4P,Sakura Finetek Japan,Tokyo,Japan)和伊红(#8660,Sakura Finetek Japan,Tokyo,Japan)溶液用于H&E染色。将染色后的载玻片浸泡于乙醇中,接着浸泡于二甲苯中,由此再次脱水。使用包含Marinol(#4197193;MutoPure Chemicals,Tokyo,Japan)的玻璃盖玻片,覆盖染色后的载玻片。使用OLYMPUScellSens标准系统(OLYMPUS,Tokyo,Japan)对H&E染色载玻片进行组织学试验。
<统计分析>
将p值小于0.05(p<0.05)视为具有统计学意义。全部图表、计算以及统计分析均使用Microsoft Excel(Microsoft Corp.,Redmond,WA,USA)用的统计软件包来进行。
(2)结果
(2-1)局部AMDC治疗(通过ZHER2:342-Cupid-His-Ax-SiPC的治疗)使异种移植小鼠模型的肿瘤体积快速减少,但在一部分病例中会导致复发。
为了建立HER2阳性乳腺癌小鼠异种移植模型,对BALB/cSlc-nu/nu裸鼠,皮下移植KPL-4细胞。20只小鼠的肿瘤体积在KPL-4细胞注射后增加,在细胞移植44天后达到400±110mm3(125至551mm3)的平均体积。将KPL-4异种移植物模型小鼠随机分为2个处理组(每组n=10)。在组1的第0天注射Kadcyla(注册商标)(平均±SD:417±88),在组2的第0天注射ZHER2:342-Cupid-His-Ax-SiPC(平均±SD:384±126)。两组均在第1天(注射后间隔24小时)和第3天(注射后间隔72小时)进行照射。在Kadcyla(注册商标)组和ZHER2:342-Cupid-His-Ax-SiPC组中,各个小鼠的肿瘤体积未见统计上的显著差异。
为了评价局部AMDC疗法的效果,测定了Kadcyla(注册商标)和ZHER2:342-Cupid-His-Ax-SiPC治疗后的KPL-4异种移植物模型小鼠中的肿瘤体积随时间的变化。图11显示Kadcyla(注册商标)和ZHER2:342-Cupid-His-Ax-SiPC注射后的KPL-4异种移植物模型小鼠中的肿瘤体积的变化。两组均在初次治疗后发现肿瘤大小的缩小。在治疗后第4天(p<0.001)、第7天(p<0.001)、第11天(p<0.001)、第14天(p<0.01)、第17天(p<0.05),Kadcyla(注册商标)组和ZHER2:342-Cupid-His-Ax-SiPC组之间,肿瘤体积的减少存在显著差异(图11A)。
KPL-4异种移植模型小鼠中的肿瘤体积在Kadcyla(注册商标)处理后逐渐减少,在注射后21天仅1例减少至0mm3(图11B和D)。另一方面,在相同的异种移植模型中,肿瘤的大小在ZHER2:342-Cupid-His-Ax-SiPC处理后快速减少。进一步地,约一半小鼠的肿瘤在ZHER2:342-Cupid-His-Ax-SiPC注射11天后消失(图11C和D)。平均肿瘤体积在治疗后20日左右以及复发检查期间最小。但是,在观察期间以后,异种移植模型小鼠的肿瘤体积持续,在5/10例中再次逐渐增大。根据情况,在ZHER2:342-Cupid-His-Ax-SiPC组中,在注射2个月后,肿瘤大小更大(第63天的平均体积:381±296mm3)。另一方面,在Kadcyla(注册商标)组中,观察到肿瘤大小的稳步下降。
这些数据表明,短期的局部AMDC治疗对HER2阳性乳腺癌具有强力的抗肿瘤效果,但在一部分病例中,可能导致初次治疗后的肿瘤复发风险。
(2-2)通过第2次局部AMDC治疗使复发肿瘤迅速缩小并完全消失。
为了研究重复局部AMDC治疗的有效性,在最初治疗的63天后(肿瘤观察期间的44天后),将ZHER2:342-Cupid-His-Ax-SiPC预缀合物注射到异种移植物模型小鼠中,测定复发肿瘤的大小。图12显示第2次ZHER2:342-Cupid-His-Ax-SiPC治疗后的5个肿瘤复发病例的大小变化。与第1次ZHER2:342-Cupid-His-Ax-SiPC处理的情况相比较,异种移植物模型小鼠中的肿瘤体积,在第2次ZHER2:342-Cupid-His-Ax-SiPC处理后更快速地减少。进一步地,肿瘤在第2次ZHER2:342-Cupid-His-Ax-SiPC注射的7天后在所有小鼠中均消失(图12A和B),在第2次ZHER2:342-Cupid-His-Ax-SiPC治疗后,即使肿瘤复发起超过1个月,也未观察到再增殖。在第1次ZHER2:342-Cupid-His-Ax-SiPC治疗后第0天和第97天之间,肿瘤大小有显著差异。进一步地,ZHER2:342-Cupid-His-Ax-SiPC重复治疗组的块的大小,比最初治疗97天后的Kadcyla(注册商标)治疗组的块的大小更小(图12C)。这些结果暗示了重复局部AMDC治疗使复发肿瘤迅速且完全地消散。
(2-3)局部AMDC治疗对HER2阳性异种移植肿瘤模型显示出强健的效果,而不会引起皮肤变性。
为了评价针对局部AMDC治疗的病理学反应,在Kadcyla(注册商标)和ZHER2:342-Cupid-His-Ax-SiPC治疗后第97天,进行了KPL-4异种移植物模型小鼠的组织学检査。在组织学上,在第97天于异种移植KPL-4肿瘤部位周围的皮下区域,在Kadcyla(注册商标)处理(n=10)或第2次AMDC处理(n=10)小鼠的任一者中,均未观察到残存肿瘤细胞(图13和图14)。在任一小鼠中,均未在淋巴结或远端脏器处观察到转移性肿瘤(图13和图14)。由此,得出在两组中均实现了病理学上完全缓解的结论。根据进一步的组织学观察,在Kadcyla(注册商标)处理小鼠的皮下区域中频繁观察到与包含血铁黄素的巨噬细胞巢的肉芽肿性反应和/或局部钙化(6/10[小鼠#1、2、3、7、8和10],60%)(图13)。在Kadcyla(注册商标)处理小鼠中的1只(小鼠#2)中,还观察到被肉芽肿组织包围的皮下包囊。相对地,在第2次AMDC处理组(1/10[小鼠#6],10%)中很少观察到肿瘤肉芽肿性反应(图14(p<0.001,卡方检验)。Kadcyla(注册商标)治疗组的肉芽肿性反应的频率暗示了Kadcyla(注册商标)治疗比AMDC治疗更可能诱发伴随出血性和坏死性的变化的高水平的组织障碍。第2次局部AMDC治疗后,明显实现了异种移植KPL-4肿瘤的病理学上的完全缓解。进一步地,除了在2只小鼠(小鼠#2和6)中观察到的显微镜下的局限性肝坏死外,在重要脏器中未观察到组织学上明显的副作用(图14)。
(2-4)测定了Kadcyla(注册商标)和ZHER2:342-Cupid-His-Ax-SiPC处理后的异种移植物体重随时间的变化。Kadcyla(注册商标)治疗后的异种移植物模型小鼠的体重在所有的各个小鼠之间显示出中等程度增加的倾向(图15A)。1只异种移植物ZHER2:342-Cupid-His-Ax-SiPC处理小鼠在肿瘤复发病例中,从最初处理的3周后起体重减少,但在第2次处理后,体重快速增加(图15B)。复发实例中的其他的异种移植物ZHER2:342-Cupid-His-Ax-SiPC处理小鼠即使在第2次处理后体重也减少,但这可能是由组织病理学检査中偶发性发现的无关的感染症引起的。这些结果显示出,局部AMDC对异种移植模型小鼠具有强力的治疗效果,副作用也比Kadcyla(注册商标)治疗少。
Claims (14)
1.融合蛋白,其中,分子量20,000以下的抗原结合分子经由接头序列结合在序列号1中记载的氨基酸序列的N末端侧和/或C末端侧,但C末端的由Pro-Ser-Ala-Ala-Ser His-His-His-His-His-His构成的氨基酸序列可部分或全部缺失。
2.权利要求1所述的融合蛋白,其从N末端侧到C末端侧依次具有分子量20,000以下的抗原结合分子、接头序列和序列号1中记载的氨基酸序列,但C末端的由Pro-Ser-Ala-Ala-Ser His-His-His-His-His-His构成的氨基酸序列可部分或全部缺失。
3.权利要求1或2所述的融合蛋白,其中,抗原结合分子为与癌细胞中表达的抗原结合的分子。
4.权利要求1至3中任一项所述的融合蛋白,其中,抗原结合分子为与Her2结合的分子。
5.权利要求1至4中任一项所述的融合蛋白,其中,抗原结合分子具有序列号2中记载的氨基酸序列。
6.权利要求1至5中任一项所述的融合蛋白,其中,接头序列由甘氨酸残基和丝氨酸残基构成,且氨基酸残基的数目为5至25个。
7.权利要求1至6中任一项所述的融合蛋白,其中,接头序列为以[(Gly)m-Ser]n表示的氨基酸序列,式中,m表示1~10的整数,n表示1~5的整数。
8.权利要求1至7中任一项所述的融合蛋白,其具有序列号4中记载的氨基酸序列,但C末端的由Pro-Ser-Ala-Ala-Ser His-His-His-His-His-His构成的氨基酸序列可部分或全部缺失。
9.核酸,其编码具有序列号4中记载的氨基酸序列的融合蛋白,但C末端的由Pro-Ser-Ala-Ala-Ser His-His-His-His-His-His构成的氨基酸序列可部分或全部缺失。
10.癌症治疗剂或癌症诊断剂,其包含权利要求1至8中任一项所述的融合蛋白。
11.癌症的治疗或诊断试剂盒,其包含:(1)权利要求1至8中任一项所述的融合蛋白;和(2)以下述式(1)表示的化合物或其盐与诊断用物质或治疗用物质的缀合物:

13.权利要求1至8中任一项所述的融合蛋白的制造方法,其包括使编码权利要求1至8中任一项所述的融合蛋白的核酸在宿主中表达的工序。
14.权利要求13所述的方法,其中,使上述融合蛋白在细菌的包涵体中表达并回收。
