Abstract
本发明公开了一种蛇毒多肽及其对细菌的抑制作用,特别是在抑制或逆转耐药菌耐药性中的应用。本发明的蛇毒多肽稳定、安全、抗菌效果好,能够恢复耐药菌对抗生素敏感性,抑制或逆转耐药菌耐药性,减少抗生素的用量,可以用于制备治疗人或动物由于耐药菌感染所致疾病的药物。

技术领域
本发明涉及生物技术领域,具体涉及蛇毒多肽及其对细菌的抑制作用。
背景技术
蛇毒多肽(Snake Venom Peptide,SVP)是从毒蛇毒腺中分泌出来的一种液体,主要成份是毒性蛋白质,约占干重的90%至95%,内含二十多种酶类和毒素,如蛇毒毒性蛋白Crotamine、神经毒素Hannalgesin。此外还含有一些小分子肽、氨基酸、脂质等,如具有抗肿瘤作用的蛇毒多肽Crotoxin、Bth TX-I等。不同蛇毒的毒性、药理及毒理作用各具特点。
随着毒理学和生物化学的发展,蛇毒的许多活性多肽已得到分离、纯化和测定,其生理效应强,作用广泛,在药物研发中已引起较大关注;特别是在抗肿瘤、抗炎、抗血栓、镇痛和降压等方面,不乏已经开发为药物的先例。
虽然蛇毒多肽在国内外的研究已有很多,但主要是抗炎和抗肿瘤方面的,未见报道过使用蛇毒多肽的抗菌作用。
因此,需要探寻和开发蛇毒多肽作为抗菌多肽的多种应用。
发明内容
为了解决现有技术中存在的上述技术问题之一,本发明提供了一种蛇毒多肽及其对细菌的抑制作用,特别是在抑制或逆转耐药菌耐药性中的应用。
本发明的目的是提供稳定、安全、抗菌效果好的,能够恢复耐药菌对抗生素敏感性的蛇毒多肽,具体来说,本发明特别是提供一种蛇毒多肽及其在逆转耐药菌耐药性中的应用,可以用于制备治疗人或动物由于耐药菌感染所致疾病的药物。
第一方面,本发明提供一种蛇毒多肽,所述蛇毒多肽通过如下步骤制得:
将蛇的唾液进行冻干,得到冻干粉末;
将得到的冻干粉末重悬,煮沸;
将煮沸后的溶液过滤,收集滤液。
上述步骤中,过滤的目的是除去大分子和变性蛋白质,保留小分子多肽物质。
在一些实施方案中,所述蛇毒多肽来源于以下项中的一种或多种蛇:中华眼镜蛇(Chinese Cobra)、短尾蝮蛇(Gloydius brevicaudus)、尖吻蝮蛇(Deinagkistrodon)、竹叶青(Trimeresurus stejnegeri)、银环蛇(Bungarus multicinctus)、金环蛇(Bungarusfasciatus)、原矛头蝮蛇(Protobothrops mucrosquamatus)。
在一些实施方案中,所述蛇为中华眼镜蛇(Chinese Cobra),学名Naja atra,又名舟山眼镜蛇。
在一些实施方案中,所述小分子多肽物质的分子量范围为:3-10KDa。
在一些实施方案中,所述大分子和变性蛋白质的分子量范围为:10001Da以上。
在一些实施方案中,所述冻干粉末用去离子水、超纯水或缓冲液重悬溶解。
在一些实施方案中,所述缓冲液为磷酸盐缓冲液(PBS)。
在一些实施方案中,所述煮沸的时间为3-8min,例如煮沸3min、5min或8min。煮沸时间在3-8min之间,可以保持抗菌有效成分的活性。如果煮沸时间过长,会导致抗菌有效成分失活,MIC升高。
在一些实施方案中,所述过滤为用0.2-0.5μm微孔滤膜过滤,优选地,采用水系微孔滤膜,例如,用0.2μm、0.22μm、0.3μm或0.5μm水系微孔滤膜进行过滤,以除去大分子和变性蛋白。优选地,用0.22μm水系微孔滤膜过滤大分子和变性蛋白。
第二方面,本发明提供一种组合物,所述组合物包括本发明所述的蛇毒多肽。
在一些实施方案中,所述组合物还包括抗生素。
在一些实施方案中,所述抗生素包括四环素类抗生素、头孢菌素类抗生素和/或培南类抗生素。

在一些实施方案中,所述蛇毒多肽与抗生素联用。当将本发明的蛇毒多肽与抗生素联用时,可以大大降低最小杀菌浓度,显著减少抗生素的使用量。
在一些实施方案中,所述抗生素包括四环素类抗生素、头孢菌素类抗生素和/或培南类抗生素。

本发明的有益效果至少包括以下之一:
1.筛选蛇毒成分内稳定高效抗菌的小分子多肽
本发明是基于探求防治耐药菌的新型物质功效进行的。本发明使用的蛇毒多肽是将蛇的唾液进行冻干,将得到的冻干粉末重悬,在EP管中煮沸3-8min后,将煮沸后的溶液用0.2-0.5μm水系微孔滤膜进行过滤,除去大分子和变性蛋白质,保留小分子多肽物质,得到所述蛇毒多肽。
2.安全性评价
本发明安全性评价试验研究结果表明,蛇毒多肽在对小鼠的急性毒性实验中的LD50值为200mg/kg。
3.抗菌效果
本发明研究的蛇毒多肽对多种细菌及耐药菌都有杀菌效果,且能恢复多种耐药菌对抗生素的敏感性,例如单用蛇毒多肽对替加环素耐药菌—tet(X4)阳性菌的杀菌浓度为640mg/L,单用替加对替加环素耐药菌—tet(X4)阳性菌的杀菌浓度为>32mg/L,在联用蛇毒多肽和替加环素时,最小杀菌浓度降低8倍以上,可显著减少替加环素的使用量。
本发明提供稳定、安全和高效的蛇毒多肽,这种蛇毒多肽可显著提升抗生素对耐药菌的杀菌效果,可为药物制剂开发提供试验基础。
本发明的蛇毒多肽可用作活性抗菌成分,应用于耐药菌对抗生素敏感性恢复,为蛇毒提供了新的用途,为抗菌药物提供了新的选择,具体来说是蛇毒多肽作为抗菌肽能够减少联合用药的抗生素例如替加环素的使用量以及能够有效提高抗生素例如替加环素的作用效果。

实施例1.小鼠急性毒性实验
本实验参照兽药研究技术指导原则方法进行小鼠急性毒性实验,探究毒性以及LD50,可作为为后续亚急性毒性实验剂量参照。
1.蛇毒多肽的制备
取中华眼镜蛇(Chinese Cobra)的唾液分泌物进行冻干,得到冻干粉末,用超纯水重悬,按用量配制,放置在50mL EP管中煮沸5min,将煮沸后的溶液用0.22μm水系微孔滤膜进行过滤,收集滤液,得到蛇毒多肽。
采用液相色谱质谱联用法(LC-MS/MS)对所得蛇毒多肽进行蛋白分析(苏州普泰生物医药有限公司)。
(1)样品制备
首先通过二硫苏糖醇(DTT)还原蛇毒多肽溶液样品,所有半胱氨酸残基用碘乙酰胺烷基化,并通过脱盐柱或乙醇沉淀进行清洗。然后用测序级修饰胰蛋白酶(Promega)在消化缓冲液(碳酸氢铵100mM,pH8.5)中消化样品。然后通过Nano LC-ESI-MS/MS系统分析溶解的多肽样品。
(2)Nano LC-ESI-MS/MS分析
Nano LC-ESI-MS/MS系统:高效液相色谱(HPLC)系统(安捷伦),填充反相C18的毛细管柱,75μm ID 8cm长度。C18的粒径为3μM,孔径为样品进样时间为20分钟。HPLC溶剂A为97.5%水,2%乙腈,0.5%甲酸。HPLC溶剂B为9.5%水,90%乙腈,0.5%甲酸。从2%溶剂B到90%溶剂B的梯度时间为60分钟,样品上样时间为20分钟,色谱柱洗涤为20分钟。样品进样体积为3μL。HPLC系统与线性离子阱质谱仪(LTQ,Thermo)在线耦合,从HPLC柱中洗脱的样品通过电喷雾电离(ESI)过程直接电离并进入质谱仪。电离电压通常在仪器调谐过程中进行优化,通常在1.5kv-1.8kv范围内。毛细管温度设定为100℃。质谱仪设置为数据依赖模式,以通过低能量碰撞诱导解离(CID)过程获取MS/MS数据。默认碰撞能量为33%,默认充电状态为3。使用1次显微扫描进行一次全扫描,质量范围为350amu至1650amu,然后对9种最强离子进行9次MS/MS扫描,具有全质量范围和3次显微扫描。动态排除功能设置如下:重复计数为1,排除持续时间为1分钟。排除宽度为4Da。
分析结果如表1所示。在表1的肽列表中,列出了通过MS/MS测序鉴定出的多肽(SEQID NO.1-67)。根据鉴定出的多肽序列,分析出这些多肽所对应的蛋白,如表2和图1所示。

2.实验动物
试验动物为雌雄各半的昆明小鼠,初始体重为18-22g。
药物的配制:用超纯水配置蛇毒多肽(SVP)母液浓度为100mg/mL浓度,即1mL灌服液含有SVP冻干粉末0.1g,煮沸5min后过0.22μm膜除去蛋白沉淀,取清液。
给药方法:经口或注射给药,体积不超过0.1-0.2mL/10g,给药前禁食不禁水8-12h,可一日内多次给予药物,每次间隔2-3h,仍合并为一次剂量计算,若总剂量达到5000mg/kg,仍未引起动物死亡,即停止给药。
3.预实验
设定A、B、C、D、E、F共6组,如表3所示。每组设4只实验动物(雌雄各半),依次进行试验,找出4/4致死剂量和0/4致死剂量。小鼠20g,即0.2mL 100mg/mL蛇毒多肽灌胃体积。得出结果,全数致死量为300mg/kg,全数存活量为100mg/kg。

4.正式实验
60只小鼠禁食不禁水12h后,按体重随机分为6组(n=10),雌雄各半。实验设空白对照组(灌胃生理盐水)和5个给药组(灌胃剂量分别293.5、258.7、227.7、200.6、176.1mg/kg)。灌胃量为10mL/kg,按体重计算各组给药量。灌胃后记录各组小鼠的毒性反应和死亡情况,用Bliss法软件计算半数致死量(LD50),并对死亡小鼠进行大体解剖,检查脏器。计算得出蛇毒多肽的LD50约为200mg/kg。
实施例2.小鼠亚急性毒性实验
本实验根据实施例1急性毒性实验LD50作为参照进行,目的是探究短期SVP作用下,确定SVP的主要毒作用,同时了解有无蓄积作用和作用的靶器官,进入机体各器官引起损害的剂量或浓度。
蛇毒多肽处理方法与实施例1一致,试验动物为雌雄各半的昆明小鼠,初始体重为18-22g。
参照《GB15193.22-2014食品安全国家标准28天经口毒性试验》方法要求,受试物无人体推荐剂量,以LD50的10%作为最高剂量,实施例1得出LD50约为200mg/kg,故设20mg/kg(高剂量)、10mg/kg(中剂量)、5mg/kg(低剂量)3个剂量组,另设空白对照组,采用完全随机方法,将40只雌雄昆明小鼠随机分为4组,每组10只。
小鼠适应性喂养1周后灌胃给药,按10mL/kg体质量1次/天,空白对照组灌胃等量蒸馏水。连续给药28天,试验期间各组小鼠均自由饮水和摄食。
在给药周期内,每日上午观察各组小鼠的外观体征、行为活动、呼吸、粪便、尿液情况等,评估异常的临床体征和行为变化。试验期间测定小鼠体质量(2次/周),做好小鼠体重记录,并测量定食物消耗量(1次/天)。
28天试验周期结束前,各组动物禁食不禁水12h后进行血液样本采集。使用小鼠眼眶静脉丛采血方法收集血样。每组小鼠血液收集两管:空白采血管、EDTA-K2抗凝管各收集1mL以上。小鼠采血后,采用颈椎脱位进行安乐死,并立即将心脏、肝脏、脾脏、肺、肾、脊髓和大脑等器官取出称重,计算相对器官重量(器官重量/体重,%)。称重后将器官置于10%多聚甲醛中固定24h以上后,送检进行病理组织切片检查。
空白全血采集之后室温静置30min,充分凝固后,2500rpm 25℃离心20min,获得血清。所有血清样本采用FAITH-1000全自动生化分析仪和电解质分析仪进行检测。
实验数据采用Graphpad Prism 9.0软件进行统计分析,数据采用平均值±标准差(Standard deviation,SD)或标准误(Standard Error,SE)表示,组间采用学生t-test和单因素方差分析(ANOVA)进行比较,统计学意义P<0.05表示显著差异,P<0.01表示差异极显著。结果如表4-表6所示。

由表4可知,各项脏器的脏器系数差异均不显著(P>0.05);其中高中低剂量组的肾脏的脏器系数均高于空白组的肾脏的脏器系数。

表5的血常规指标结果表明,在Gran(中性粒细胞)中,低剂量组(P<0.05)和高剂量组(P<0.01)与空白对照组表现出差异显著。

如表6所示,高、中、低剂量组各项指标与空白组各项指标差异均不显著。其中高中低剂量组分别都出现ALB(白蛋白)含量升高,白蛋白升高对应血液浓缩而致相对性增高。高剂量组的TG(甘油三酯)的平均值低于TG的正常范围,且超过半数的小鼠的TG值也低于正常范围。
实施例3.溶血性试验
脱纤羊血制备:50mL脱纤羊血(海博生物,产品编号1001339-1)加入氯化钠注射液约10倍量,轻轻摇匀,然后离心,每分钟3000rpm离心10min,除去上清液,沉淀的红细胞再用氯化钠注射液按上述方法洗涤2~3次至上清液不显红色为止。将所得红细胞用氯化钠注射液制成2%的红细胞混悬液,即脱纤羊血。
样品制备:将SVP分别用0.9%氯化钠注射液按倍比稀释的方法制备成2.4g/L、1.2g/L和0.6g/L浓度的供试品溶液,按表7所示进行加样,试管中先加入2%的红细胞混悬液2.5mL,再加入相同体积的纯水、溶剂(0.9%氯化钠注射液或5%葡萄糖注射液)及不同浓度的供试品药液各2.5mL,摇匀后,置37℃恒温培养箱中。前一小时,每隔15min观察1次,1h后,每隔1h观察1次,共观察3h。肉眼观察3h后,将上述样品2500r/min离心5min,用酶标仪在545nm波长处测定各管的吸光度,计算溶血率。按下式计算溶血率:
溶血率(%)=(供试品组吸收度均值-阴性对照组吸收度均值)/(阳性对照组吸收度均值-阴性对照组吸收度均值)×100%。

如表7所示,实验结果发现高剂量组(1组)有轻微溶血现象,其溶血率为11%,中剂量组与低剂量组均无溶血现象。
实施例4.热稳定性和酸碱稳定性实验
本实验将SVP用不同温度、酸碱度进行处理,探究SVP的稳定性。
蛇毒多肽处理方式同实施例1。
热稳定性:将SVP同实施例1处理,并配置成2.56g/L的溶液,(即每份加1mL原液和3mL灭菌水)平分成九份,分别置于室温(25℃)、60℃、65℃、70℃、75℃、80℃、90℃和100℃的水浴中加热1小时,及121℃高温灭菌处理15min后冷却至室温。利用大肠杆菌(Escherichia Coli)ATCC25922作为指示菌评价药物的有效性,利用最小抑菌浓度法(MIC)检测其抗菌活性,以室温下的样品作为对照,对比不同温度处理后SVP残余的抗菌活性,并使用GraphPad Prism绘图软件进行作图。
酸碱稳定性:将2.56g/L的SVP溶液分成十一份后(即每份加1mL原液和3mL灭菌水),利用HCL和NaOH将其pH调节至2-12,1个小时后将各管pH调回初始管pH值,并定容至8mL(即配置成1.28g/L),以ATCC25922、tet(X4)阳性大肠杆菌菌株a232、a62、J27ab-2(含有tet(X4)的耐药基因)作为指示菌,利用最小抑菌浓度法检测其抗菌活性,以初始管的样品作为对照,对比不同酸碱性处理后的SVP残余的抗菌活性,并使用GraphPad Prism绘图软件进行作图。
SVP的热稳定性和酸碱稳定性实验结果
如图2所示,SVP在不同温度下处理1h后的结果显示,90℃以下,SVP溶液无明显眼观变化,a232、a62、J27ab-2等tet(X4)阳性大肠杆菌菌株对SVP的MIC值与常温下MIC值均为320mg/L;SVP在90℃、100℃、121℃下明显有蛋白质变性沉淀生成,其MIC值升高至>1280mg/L,抗菌活性明显下降。
如图3所示,使用不同pH处理SVP 1h后,J27ab-2、a232、a62等tet(X4)阳性大肠杆菌菌株对SVP的MIC都为320mg/L,且SVP溶液眼观无明显变化,无明显蛋白沉淀,抗菌活性无明显变化。
如图4所示,SVP在不同温度下处理1h后的结果显示,90℃、100℃、121℃处理后明显有蛋白质变性沉淀生成在,90℃以下处理SVP,ATCC25922对SVP的MIC值为仍然是640mg/L,抗菌活性无明显变化,但在90℃及以上处理之后的MIC值大于2560mg/L,抗菌活性明显下降。
如图5所示,对SVP不同pH处理1h后,其对ATCC25922的MIC都为640mg/L,且SVP溶液眼观无明显变化。
发现SVP对于酸碱的耐受能力较强,2-12pH处理1h后的MIC值和杀菌效果均无明显变化,耐酸碱能力强,而耐高温能力较弱,在90℃及以上处理1h后其有蛋白沉淀析出,MIC值明显升高,抗菌活性明显降低。
实施例5.蛇毒多肽对各类耐药菌的抗菌效果评价
本实验使用SVP与多种抗生素联合用药治疗病原菌和多重耐药病原菌,并通过FICI指数评价其联合抗菌效果。
菌液制备:将各类菌株提前划线接种至20mL MHB(Mueller-Hinton Broth)肉汤,200rpm,37℃摇床培养6-8h,并通过比浊仪调比浊度至0.5(0.4-0.6左右),通过倍比稀释1000倍即使用浓度。
SVP药物配制:同实施例1。
实验步骤:
(1)将新鲜MHB肉汤倒入90mm平皿,使用排枪吸入50μL新鲜肉汤至96孔板第一列和F排第六孔(6×6)。
(2)使用移液枪吸入50μL SVP至A1-A6,并以SVP浓度2倍降序从A横排加至E横排以此类推,F组不加SVP。
(3)使用移液枪吸入50μL抗生素至第6竖排,并以抗生素浓度2倍降序从第6竖排加至第2竖排,第一竖排不加抗生素,H1孔即为空白对照。
(4)吸取100μL配置好的菌液至每孔(6×6)。
FICI判读标准:FICI≤0.5:协同效应;0.5<FICI<1:相加效应;1≤FICI<4:无关效应;FICI≥4:拮抗效应。
1.J27ab-2,耐替加环素(Tigecycline)tet(X4)阳性大肠杆菌菌株,对替加环素和SVP的MIC分别为8mg/L和320mg/L。结果如表8所示。

如表9所示,单用SVP的最小杀菌浓度(MBC)是640mg/L,单用替加环素的最小杀菌浓度在32mg/L以上,在80mg/L的SVP联合用药替加环素用量下降低至4mg/L(降低8倍以上),在160mg/L的SVP联合用药替加环素用量下降至1mg/L。
2.34AB替加环素耐药菌—tet(X5)阳性鲍曼氏不动杆菌(Acinetobacterbaumannii)菌株,对替加环素和SVP的MIC分别为4mg/L和160mg/L。结果如表10所示。

如表11所示,单用SVP的最小杀菌浓度是160mg/L,单用替加环素的最小杀菌浓度在32mg/L以上,在80mg/L的SVP联合用药下替加环素用量降低至2mg/L(降低16倍以上)。

如表13所示,单用SVP的最小杀菌浓度为1280mg/L,单用替加环素的最小杀菌浓度在1mg/L,在160~640mg/L的SVP联合用药替加环素MBC下降低至0.5mg/L,用量降低了2倍。
4.21f31er,耐头孢类ESBL阳性大肠杆菌菌株,对头孢噻肟和SVP的MIC分别为>1024mg/L和640mg/L。结果如表14所示。

如表15所示,21f31er对头孢噻肟的MBC为>1024mg/L,在160mg/L的SVP联合用药头孢噻肟的MBC下降低至64mg/L(降低8倍以上)。
5.AD21R,NDM-5阳性大肠杆菌菌株,对美罗培南和SVP的MIC分别为32mg/L和320mg/L。结果如表16所示。

如表17所示,AD21R对美罗培南的MBC为>64mg/L,在160mg/L的SVP联合用药美罗培南MBC下降低至8mg/L(降低8倍以上)。
本发明的实验结果表明,蛇毒多肽对多种耐药菌有明显的抑菌和杀菌效果,与多种抗生素联用后能够极大地降低抗生素的最小抑菌浓度和最小杀菌浓度,即降低抗生素的使用量,为耐药菌逆转耐药机制研究提供了理论基础。
本发明的技术方案不限于上述具体实施例的限制,凡是根据本发明的技术方案做出的技术变形,均落入本发明的保护范围之内。
Claims (10)
1.一种蛇毒多肽,其特征在于,所述蛇毒多肽通过如下步骤制备:
将蛇的唾液进行冻干,得到冻干粉末;
将得到的冻干粉末重悬,煮沸;
将煮沸后的溶液过滤,收集滤液。
2.根据权利要求1所述的蛇毒多肽,其特征在于,所述蛇毒多肽来源于以下项中的一种或多种蛇:中华眼镜蛇(Chinese Cobra)、短尾蝮蛇(Gloydius brevicaudus)、尖吻蝮蛇(Deinagkistrodon)、竹叶青(Trimeresurus stejnegeri)、银环蛇(Bungarusmulticinctus)、金环蛇(Bungarus fasciatus)、原矛头蝮蛇(Protobothropsmucrosquamatus)。
3.根据权利要求1所述的蛇毒多肽,其特征在于,所述小分子多肽物质的分子量范围为:3-10KDa。
4.根据权利要求1所述的蛇毒多肽,其特征在于,所述冻干粉末用去离子水、超纯水或缓冲液重悬溶解;优选地,用超纯水或磷酸盐缓冲液重悬溶解。
5.根据权利要求1所述的蛇毒多肽,其特征在于,所述过滤为用0.2-0.5μm微孔滤膜过滤。
6.一种组合物,其特征在于,所述组合物包括权利要求1至5中任一项所述的蛇毒多肽。
7.根据权利要求6所述的组合物,其特征在于,所述组合物还包括抗生素,
优选地,所述抗生素包括四环素类抗生素、头孢菌素类抗生素和/或培南类抗生素;
更优选地,所述四环素类抗生素为甘氨酰环素类抗生素;进一步优选地,所述甘氨酰环素类抗生素包括替加环素和环孢素;
更优选地,所述头孢菌素类抗生素包括头孢噻肟、头孢唑肟、头孢曲松、头孢地秦、头孢他啶、头孢哌酮、头孢匹胺、头孢甲肟、头孢磺啶、头孢克肟、头孢特仑酯、头孢他美酯、头孢布烯、头孢地尼、头孢泊肟酯、头孢托仑匹酯;
更优选地,所述培南类抗生素包括美罗培南、亚胺培南、帕尼培南、厄他培南、比阿培南、多尼培南。
8.权利要求1-5中任一项所述的蛇毒多肽或权利要求6或7所述的组合物在制备预防或治疗细菌感染的药物中的用途。
9.根据权利要求8所述的用途,其特征在于,所述细菌为耐药菌;
优选地,所述耐药菌包括对四环素类抗生素、头孢菌素类抗生素和/或培南类抗生素耐药的细菌;
更优选地,所述四环素类抗生素为甘氨酰环素类抗生素;优选地,所述甘氨酰环素类抗生素包括替加环素和环孢素;
更优选地,所述头孢菌素类抗生素包括头孢噻肟、头孢唑肟、头孢曲松、头孢地秦、头孢他啶、头孢哌酮、头孢匹胺、头孢甲肟、头孢磺啶、头孢克肟、头孢特仑酯、头孢他美酯、头孢布烯、头孢地尼、头孢泊肟酯、头孢托仑匹酯;
更优选地,所述培南类抗生素包括美罗培南、亚胺培南、帕尼培南、厄他培南、比阿培南、多尼培南;
更优选地,所述耐药菌包括J27ab-2tet(X4)阳性大肠杆菌菌株、34AB tet(X5)阳性鲍曼不动杆菌菌株、ATCC25922大肠杆菌菌株、21f31er ESBL阳性大肠杆菌菌株、AD21R NDM-5阳性大肠杆菌菌株。
10.根据权利要求8所述的用途,其特征在于,所述蛇毒多肽与抗生素联用;
优选地,所述抗生素包括四环素类抗生素、头孢菌素类抗生素和/或培南类抗生素;
更优选地,所述四环素类抗生素为甘氨酰环素类抗生素;进一步优选地,所述甘氨酰环素类抗生素包括替加环素和环孢素;
