Description

Culture medium for grifola frondosa cultivar and preparation method thereof
Technical Field
The invention belongs to the technical field of edible fungus culture, and particularly relates to a culture medium for grifola frondosa cultivars and a preparation method thereof.
Background
Grifola frondosa is an edible fungus integrating nutrition, delicacy and health, and the fresh fruiting body contains more than 20% of protein, more than 50% of carbohydrate and rich flavor amino acids. The polysaccharide component in the grifola frondosa is developed into an anti-tumor drug, which is one of edible fungi with drug word size processing products in China. In recent years, the research shows that the grifola frondosa polysaccharide has the effect of reducing blood sugar, and is the strongest in various edible and medicinal fungi such as ganoderma lucidum, corious versicolor, phellinus linteus and the like.
In edible fungus strain production, mycelium pure cultures obtained by fruiting body tissue separation and spore separation or mycelium separation in auricularia auricula and mushroom wood bases are generally called mother seeds, and transferred strains are also called primary seeds. The pure culture of mycelium obtained by mother seed transplanting and expanding culture is called stock seed, also called secondary seed. The stock is mainly used for producing cultivars, and can also be directly used as cultivars. Pure mycelium cultures obtained by transplanting and expanding culture of stock seeds are called cultivars, or production (used) seeds, also called tertiary seeds, and the cultivars are directly used for cultivation. The cultivation seed production aim is: 1) Further expands the amount of the strain and saves the strain cost; 2) The strain is quickly adapted to a culture formula, growth conditions and nutrients are provided for the quick growth of mycelium of the strain, and the mycelium growth time and cultivation period are shortened.
The prior art discloses a high-yield grifola frondosa culture medium and a preparation method thereof (refer to Chinese patent CN 108925366A), wherein the culture medium comprises 15-25 parts of bran, 10-15 parts of leaves, 10-15 parts of wood dust, 5-10 parts of sandy loam, 5-10 parts of rice bran, 10-15 parts of cotton seed hulls, 15-25 parts of soybean meal, 1-2 parts of glucose, 3-5 parts of sunflower seed meal, 2-4 parts of peanut meal, 0.1-0.3 part of folic acid and 60-130 parts of water. Although the patent increases the yield, the hypha growth rate is not increased and the growth cycle is shortened.

As can be seen from tables 1 and 2, the average growth rate of hyphae in the culture mediums prepared in examples 1 to 7 is much faster than that in comparative examples 1 to 3, the growth rate of hyphae is fast, the feeding speed is fast, the bag filling time of hyphae is early, and the growth period of cultivars is shortened; the yield of Grifola frondosa was also higher than that of comparative examples 1 to 3. Compared with examples 1-7, the comparative example 1 has no caragana microphylla added, the average growth speed of hyphae in the prepared culture medium is slower, and the yield of the grifola frondosa is lower, which indicates that the caragana microphylla can promote the growth of the hyphae and improve the yield of the grifola frondosa; compared with examples 1-7, the comparative example 2 has no soybean meal, the average growth speed of hyphae in the prepared culture medium is very slow, and the yield of the grifola frondosa is low, which shows that the soybean meal is used as a source of vitamin B1, thereby accelerating the growth of the hyphae and improving the yield of the grifola frondosa; compared with examples 1 to 7, comparative example 3 was free of bagasse, and the average growth rate of mycelia in the prepared medium was slightly slow and the yield was low, because sugar contained in bagasse was used as a carbon source, the carbon-nitrogen ratio was adjusted, the growth of mycelia was promoted, and the yield of Grifola frondosa was increased. Bagasse contains a large amount of beneficial bacteria and active substances, such as water-soluble humic acid and crude protein, and enhances the fertility of the matrix.
