Abstract

The invention discloses acne-removing mask mud compounded with tea flowers and green tea and a preparation method thereof, wherein the acne-removing mask mud comprises the following raw materials: tea flower, green tea, kaolin, glycerol, hyaluronic acid, purslane extracting solution, methyl paraben, ethanol and deionized water. When in preparation, the raw materials are mixed according to the weight proportion, fully stirred and uniformly mixed, then subjected to high-temperature sterilization, and bottled and sealed under the sterile condition, thus obtaining the acne-removing mask mud compounded with tea flowers and green tea. The natural tea flower and green tea are adopted as antibacterial components of the acne-removing mask mud, so that the acne-removing mask mud can effectively inhibit the growth and reproduction of propionibacterium acnes, improve acne, has mild and non-irritating effects, and has the effects of resisting oxidation, removing free radicals and the like.

The tea flower and green tea are substantially identical in composition of the ingredients, wherein the tea flower is higher in tea polysaccharide, protein and soluble sugar content than green tea, and green tea is higher in tea polyphenol and caffeine content than tea flower. Unless otherwise indicated, the examples were conducted under conventional experimental conditions or under conditions recommended by the manufacturer's instructions. The reagents used were all conventional products commercially available.
1. Firstly, aiming at tea flowers and green tea which are main antibacterial efficacy components, an antibacterial comparison experiment is carried out, and the specific steps are as follows:
(1) Preparing water leaching solutions of tea flower, tea flower core and green tea respectively, and setting different concentration gradients, namely adding 3g, 6g, 9g and 12g of the raw material powder into each 100mL of distilled water respectively, and filtering after 45min of boiling water bath to obtain the water leaching solution.
(2) The filter paper was perforated by a 6mm puncher to prepare a filter paper sheet. Respectively placing the filter paper sheets into the water leaching solution, using pure water as a reference, soaking for 1h, and sterilizing the filter paper sheets in a high-pressure steam sterilizing pot at 121 ℃ for 20min.
(3) Preparation of thioglycolate solid medium. 2.90g of thioglycolate fluid culture medium powder, 1.50g of agar powder and 100mL of distilled water are sequentially added into a conical flask, the pH value is regulated to 7.0 (+ -0.2) after heating, stirring and dissolving, the mixture is placed into a high-pressure steam sterilization pot for sterilization at 121 ℃ for 20 mm, then a flat plate is poured into a sterile ultra-clean workbench, the inversion is carried out after the culture medium is cooled and solidified, and the mixture is sealed by a sealing film and then placed into a refrigerator at 4 ℃ for standby.
(4) Preparation of thioglycolate liquid medium. 2.90g of thioglycolate fluid culture medium powder and 100mL of distilled water are sequentially added into the conical flask, the pH value is regulated to 7.0 (+ -0.2) after heating, stirring and dissolving, and then the conical flask is placed into a high-pressure steam sterilization pot for sterilization at 121 ℃ for 20 mm, and then placed into a refrigerator at 4 ℃ for storage for standby.
(5) Preparing a bacterial suspension. In a sterile ultra-clean bench environment, a few colonies were gently scraped off on propionibacterium acnes plates with an inoculating loop, placed in a centrifuge tube containing thioglycolate liquid medium, and the turbidity of the bacterial suspension was adjusted to 0.5 mahalanobis turbidity units.
(6) The diameter of the inhibition zone is measured by a filter paper sheet method. In an ultra-clean workbench, 100 mu l of propionibacterium acnes suspension is removed by a liquid-transferring gun and added on a thioglycolate solid culture medium, and is uniformly coated, filter paper sheets soaked in different feed liquid ratios are clamped by sterile forceps and are spread on the culture medium, and three repeated experiments are carried out. Placing the culture dish into an anaerobic bag, placing the anaerobic bag into a constant temperature incubator at 37 ℃ for culturing for 72 hours, and observing the result and measuring the diameter of the inhibition zone.
The antibacterial experiment result shows (see figure 1) that compared with pure water in a control group, the tea flower core and the green tea water extract all have the effect of inhibiting the growth of propionibacterium acnes. Wherein, tea flower, green tea water extract increase along with concentration, and the diameter of the zone of inhibition increases, indicates that antibacterial effect is strengthened. It can be seen that both tea flower and green tea have good inhibitory effect on propionibacterium acnes. In order to fully utilize the advantages of the functional components of the tea flowers and the green tea, the respective biological activities are further exerted, and the tea flowers and the green tea are compounded to realize comprehensive development and utilization of tea resources.
